        Adenovirus에 의해서 발현된 BMP-2가 치주인대세포의 분화에 미치는 영향

        The regeneration of lost periodontal tissue is a major goal of therapy. Periodontal ligament cell(PDL) is a specialized connective tissue that connects cementum and alveolar bone to maintain and support teeth in situ and preserve tissue homoeostasis. Bone morphogenetic proteins(BMPs) have shown much potential in the reconstruction of the periodontum by stimulate new bone and new cementum formation. Limitiations of BMP administration to periodontal lesions is high dose delivery, BMP transient biological activity, and low bioavailability of factors at the wound site. Gene delivery method can be alternative treatment strategy to deliver BMPs to periodontal tissue. The purpose of this study is to investigate efficiency of BMP-2 gene delivery with cell-based therapy using PDL cells. PDL cell were transduced with adenoviruses encoding either BMP-2 or Lac-Z gene. To evaluate osteogenic activity of expressed BMP-2 on PDL cells, we investigated secreted BMP-2, cellular activity, ALPase, produced mineralized nodules. To evaluate collagen scaffold as carrier for transduced cell delivery, we examined morphology and secreted BMP-2 of transducd PDL cells on it. BMP-2 transducd PDL cells produced higher levels of BMP-2, ALPase, mineralized nodules than non transduced cells. Cellular activity of transduced cells was showed similar activity to non transduced cells. Transduce cells attached on collagen scaffold secreted BMP-2 at 7day and was showed similar morphology to non transduced cells. These results demonstrated that transduced PDL cells produced biologically active BMP-2 and collagen scaffold could be carrier of transducd cells.

        단위가축영양 : 수퇘지의 정액 성상과 혈중 항산화물 농도에 대한 통마늘분말 급여의 영향

        본 연구는 통마늘분말의 급여가 수퇘지의 정액 성상과 혈중 항산 화물 농도에 미치는 영향을 구명하기 위해 12개월령의 듀록종 수퇘지 9두를 공시하여 시험사료만을 급여한 대조구(4두)와 시험사료에 0.3% 통마늘분말을 첨가하여 급여한 실험구(5두)를 이용해 13주간 수행되었다. 통마늘분말의 급여가 수퇘지의 정액생산량과 정자농도에 미치는 영향을 8주간에 걸쳐 평가한 결과, 정액생산량에서는 통마늘분말 급여 여부에 따른 차이를 보이지 않았으나, 수퇘지 정액의 평균 총정자수는 0~4주의 평균 총정자수는 통마늘분말 급여여부에 상관없이 차이가 없었다. 그러나, 5~8주에 채취된 정액의 평균 총정자수는 통마늘분말을 급여한 실험구가 대조구에 비해 유의적으로 높게 나타났다(P<0.05). 통마늘분말 급여가 수퇘지의 성욕에 미치는 영향을 알아보기 연속 사정횟수와 전체 사출정액에 대한 총정자수를 조사한 결과, 통마늘분말을 급여한 실험구가 대조구에 비해 사정횟수와 총정자수가 유의적으로 높게 나타났다(P<0.05). 수퇘지에 대한 통마늘분말의 급여가 정장 및 혈장 내 polyphenol 농도에 미치는 영향에서는 정장 내에서의 polyphenol 농도는 통마늘분말 급여여부에 상관없이 차이를 보이지 않았다. 그러나, 혈장 내에서의 polyphenol 농도는 9주, 12주 및 13주차에 통마늘분말을 급여한 실험구가 대조구에 비해 유의적으로 높은 수치를 나타내었다(P<0.05). 본 실험의 결과를 종합하면, 정액채취용 수퇘지에 대한 통마늘분말의 급여는 성욕의 향상에 영향을 미쳐 정액채취빈도와 정액내 총 정자수를 증가시키고, 혈액 내 항산화물 농도를 높이는 것으로 나타나, 통마늘분말이 정액채취용 수퇘지의 성적을 향상시키는데 사료적 가치가 있는 것으로 사료된다. The objective of this study was to determine the effects of dietary supplementation of whole garlic powder (WGP) on semen characteristics and blood antioxidant level in boars. For this study, nine Duroc boars of 12 months age were used. Semen and blood samples were collected for 13 weeks, once in each week. The boars were fed the basal diet (BD; control) or BD supplemented with 3% WGP. There were no significant differences in the semen volume and sperm concentration between control and WGP group on all collection weeks. However, total sperm number per ejaculate was higher in the WGP group than that in the control group on collection weeks 6, 7 and 8 (P<0.05). Also, on collection weeks 5, 6, 7 and 8, mean of total ejaculated sperm numbers per boar were significantly higher in the WGP group compared to control group (P<0.05). On the other hand, ejaculation frequency per boar (boar`s libido) and total ejaculated sperm number per boar were significantly increased in the WGP group compared to the control group, respectively (P<O.05). Although there was no difference in polyphenol level in seminal plasma between two treatment groups, polyphenol level in blood serum was significantly higher in the WGP group on collection weeks 9, 12 and 13 (P<0.05). These results indicate that dietary supplementation of 3% WGP improves boar libido and semen productivity such as ejaculation frequency per boar, total sperm number per ejaculate, mean of total ejaculated sperm number per head. and elevate the blood level of antioxidant (polvphenon in boar serum.

        매실과 홍삼 부산물이 스트레스에 대한 육계 생체 반응에 미치는 영향

        This study was conducted to test the efficacy of plum (Prunus mume) and red ginseng (Panax ginseng C.A Meyer) marc as stress inhibitors under heat stress and lipopolysaccharide (LPS) challenge in broilers by investigating their effects on blood biochemical parameters, immunoglobulin concentration and splenic cytokine mRNA expressions. A total of one hundred ninety-two 1-d-old male broiler chicks (Ross 308) were divided into 2 stress conditions (heat and LPS) experiments. Each experiment was divided into 4 treatment groups with 8 replicates of 3 birds in each group. NC (negative control, no immune substances), PC (positive control, 25 ppm β-glucan), PM (1% plum marc) and RGM (3% red ginseng marc) treatments were administered with respective substance through water supplementation. During heat stress, The Ca/Mg ratio in PM and RGM was significantly decreased in comparison with that of NC (P<0.05). The immunoglobulin M was significantly lower in PM than in NC (P<0.05). Expression patterns of splenic cytokine mRNAs (IL-1, IL-2 and IL-6) were similar over the treatment. Expression rates of IL-1 and IL-2 in PM were significantly decreased in comparison with NC. Also, expression rates of IL-1, IL-2 and IL-6 were significantly lower in RGM than in NC (P<0.05). In conclusion, the dietary supplementation of plum and red ginseng marc improved coping ability to heat stress by preventing Ca/Mg ratio increment and by inhibiting inflammatory response in broiler chicks. However, it is necessary to determine optimal dietary level of red ginseng marc for improving growth performances in broiler chickens. These results suggest the possibility that plum and red ginseng marc could be used as the stress inhibitor under heat stress and inflammatory response in broiler chicks.

        급성 심근경색증 환자에서 심근 Troponin T 의 임상적 유용성

        N/A Objectives : Cardiac troponin T is an isoform of structurally bound proteins in myocardium. We evaluated cardiac troponin T and other cardiac enzymes as the diagnostic markers of the acute myocardial infarction. Methods: Twenty eight confirmed acute myocardial infarction patients and 12 non-anginal chest pain patients were enrolled. Concentrations of cardiac troponin T, CK, CK-MB and enzymatic activities of CK, LDH and LDH isoenzyme pattern were measured every 6 hours during the first 24 hours and then every 24 hours for 9 days after the onset of chest pain. Results: There were two peaks in the curve of serially measured serum cardiac troponin T(16.4±5.3 hour, 89.1±19.8 hour). Serum cardiac troponin T was elevated in 8 of 9 cases at 6 hours after the onset of chest pain. The sensitivity of cardiac troponin T for detecting acute myocardial infarction was 100% for 12 hours to 144 hours after onset of chest pain. Cardiac troponin T was not detected in all of 12 non-anginal chest pain patients confirmed by coronary angiography and left ventriculogrophy. Both early and late peaks of cardiac troponin T were related to the area under time-curve of cardiac troponin T curve. Conclusion : Compared to CK and CK-MB measurements, cardiac troponin T measurement has a few advantages in the diagnosis of acute myocardial infarction, such as longer time window and estimating infarct size. Therefore, cardiac troponin T appeares to be one of the most useful serum markers for detecting acute myocardial infarction.

        홍삼 부산물이 스트레스에 대한 산란계 생체반응에 미치는 영향

        This study was conducted to determine the possible use of Red Ginseng marc as stress inhibiter in thermal stress (temperature humidity index 86) and lipopolysaccharide (LPS)-exposed laying hens by investigating their effects on laying performance, blood biochemical parameters, immunoglobulin concentration and serum superoxide dismutase (SOD) like ability. A total of forty-five 52-wk-old laying hens (ISA Brown) were divided into 3 treatment groups with 5 replicates of 3 birds in each group. NC (negative control, no immune substances), PC (positive control, β-glucan 25 ppm) and RGM (Red Ginseng Marc 3%) were added in feed with respective substance. Egg production in RGM was significantly increased in comparison with NC groups for 8 weeks (P<0.05). On blood biochemical parameters, effects of ambient temperature is definite by showing significant difference in aspartate aminotransferase and others (P<0.05), but RGM both before and after thermal stimulation have no significant difference in comparison with other groups. And for 3 weeks after thermal stimulation, laying performance was also not significantly different among treatments. Immunoglobulin M content and SOD like activities after challenge with LPS were higher in the RGM and PC than NC (P<0.05). In conclusion, although ineffective as inhibiter in thermal stress, dietary supplementation of Red Ginseng marc improved SOD like activity and immune system by regulating immunoglobulin content in laying hens. These findings have laid the foundation for future studies of immunomodulation in laying hens fed Red Ginseng Marc and of evaluation of heat stress inhibitor.

        This study was conducted to test the efficacy of Ulva pertusa kjellman as immunomodulators under lipopolysaccharide(LPS) challenge in broilers by investigating their effects on serum superoxide dismutase(SOD) like ability, immunoglobulin concentration, and splenic cytokine mRNA expression. A total of ninety six1-d-old male broiler chicks(Ross 308) were divided into 4 treatment groups with 8 replicates of 3 birds in each group. NC(negative control, no immune substances), PC(positive control, β-glucan 25 ppm), Ulva P(Ulva pertusa kjellman Powder 3%), and Ulva E(Extract form Ulva pertusa kjellman 0.3%) were added in feed with respective substance. Heparinized venous blood and spleens were collected from five birds per dietary treatment at 5 wk of age. The SOD like activities in Ulva P and Ulva E were significantly increased in comparison with other groups(P<0.05). The immunoglobulin M content was lower in the Ulva E than other groups(P<0.05). Expression patterns of splenic cytokine mRNA in the IL-1β, IL-2 and IL-6 were similar. Expression rate of IL-1β, IL-2 and IL-6 in Ulva pertusa kjellman(Ulva P, Ulva E) were decreased(P<0.05) in comparison to other groups. Expression rate of IL-1β, IL-2 and IL-6 were significantly lower in groups treated with Ulva E than Ulva P. In conclusion, dietary supplementation of Ulva pertusa kjellman improved SOD like activity and affect immune system by inhibiting inflammatory response in broiler chicks. In addition, it is more effective to use Ulva pertusa kjellman extract than powder for immunomodulatory function. These results suggest the possibility that Ulva pertusa kjellman could be used as the immunomodulator for inflammatory response in broiler chicks.

        Emphysematous gastritis is a rare infection of the stomach wall with high mortality rate. It is caused by gas forming organisms and may arise by local spread through the mucosa or hematogenous dissemination from distant focus. Clinical manifestation includes acute abdomen with systemic toxicity, and diagnosis is based on radiologic demonstration of gas within the gastric wall. Treatment should be aimed to cover gram-negative organisms and anaerobes using wide-spectrum intravenous antibiotics, and sometimes surgical management may be needed in order to enhance survival. Herein, we report a case of emphysematous gastritis in a patient with end stage renal disease on hemodialysis. (Korean J Gastroenterol 2011;58:38-41)
