        투수성 매질 내에서의 지하수위 상승에 따른 TCE 거동특성 및 오염물 이동량 변화 연구

        In this study, three dimensional and two dimensional laboratory experiments were conducted to investigate the effect of water table rising on DNAPL migration, contaminants mass discharge ($M_d$), and residual NAPL distribution. The accumulation of TCE in unsaturated zone was observed in both two and three dimensional experiments. This implies DNAPL sources could exist in unsaturated zone at contaminated sites. It has been investigated that the TCE concentration is proportional to the areal ratio of residual TCE. This means the residual TCE obviously could affect the TCE concentration in aquifer system. The results of the two-dimensional experiment indicated that the contaminant sources in unsaturated zone could lead the $M_d$ increasing with water table rising and the source zone heterogeneity could also highly affect the $M_d$.

        순천만 자연생태공원 관리를 위한 중요도.만족도 분석

        해안생태계 지역은 휴양이나 관광을 위한 기회 제공과 더불어 자연자원의 보전이라는 두 가지 가치가 상충하는 공간이다. 하지만 대부분 이러한 지역에서는 자연자원의 중요도를 높이 평가하여 관리전략을 세우다 보니 방문객의 만족도 면이 간과되고 있는 실정임에도 불구하고, 방문객들의 지속적인 증가로 1970년대 이후부터 이를 보완하기 위한 수단으로 중요도 만족도 분석이 실시되고 있다. 본 논문은 현재 기조성되어 있는 순천만 자연생태공원의 방문객을 대상으로 관리전략이 상충되는 주요 관리요소에 대한 중요도와 실제 방문객들이 느끼는 만족도 사이의 차이를 주중과 주말로 나누어 분석하였다. 분석 결과, 자연생태공원이나 시 차원에서 많은 관심과 투자를 기울이고 있었지만 홍보효과 측면에서 그 효용성이 떨어졌다. 따라서 안내시설이나 홍보교육을 위한 시설과 인력구축이 이루어져야 한다. 또한 주말과 주중의 방문객의 동반형태가 달라지기 때문에 차별화된프로그램이 필요하며, 연령대에 따른 정보 교육 통제를 위한 가이드투어 프로그램을 운영하여 시공간별 이용객 통제가 이루어져야 한다. 그리고 조류출현에 대한 만족도 부분에서는 조류 출현에 대한 사람들의 기대심리를 충족시켜줄 수 있도록 관찰 교육 전시를 위한 사육종 공간이 조성되어야 할 것이다. 방문객의 긍정적인 만족도는 제한된 지역에서의 관리전략 수립시 매우 중요한 결과물로 제공될 수 있다. 따라서 중요도 만족도가 상충하는 해안생태계지역의 관리우선순위를 통한 관리 전략의 필요성과 연관된 지침을 세우는데 기초 자료로 제공될 수 있을 것으로 사료된다. The coastal area is the place where two conflicting values provide opportunities for recreation, tourism and conservation of the natural environment. Most of these coastal areas have been disregarded in terms of tourism because of the high value placed on natural resources. Management strategy has usually been first on the priority list of natural. resources. However, due to the significant number of visitors, the Important-Satisfaction Analysis(ISA) was applied as a mean of complement since the 1970s. This study analyzed the difference between visitor satisfaction and the importance of the main management strategy targeting visitors to Sun-cheon Bay Ecological Park on weekdays and weekends. Results show that Sun-cheon Bay Ecological Park and the city government have paid a lot of attention and invested a lot of money, but efficiency of publicity didn't come up to their afford. Therefore, we should prepare information facilities, public education facilities and human power. Also needed are visitors' temporal-spacial control to set specific programs and a guide for information education control. It means visitors' company forms change depends on weekday and weekend. In addition, a breeding space for birds should be built for observation, education and exhibition to help meet visitor expectations. Visitors' positive satisfaction might be provided in establishing strategy as a very important measure in limited area. In conclusion, this study might be provided as preliminary data when the management strategy and related guidelines are established through the management priority of coastal regions where importance and satisfaction conflict.

        수도권 내 개발계획관련 요소와 산림파편화 관계분석을 통한 관리방안

        Recently, reckless exploitation of land resources without much consideration for the environmental value of the land has been witnessed to accommodate the ever-increasing demands for regional development. Fragmentation due to land development is a major reason for the declining biodiversity in forest ecosystems. The purposes of this study were (1) to investigate the relationship between the factors of land development and forest fragmentation in 13 watersheds of a metropolitan area and (2) to suggest a forest management plan through the relationship. We carried out a factor analysis to determine explanatory axes of forest fragmentation, and then conducted a correlation analysis between the factor scores and the factor of land development, such as the rate of built-up areas, road density, number of built-up patches, and area of housing developments. The first explanatory axis represented stability of landscape highly related with the rate of the built-up area and road density. The second axis represented the level of fragment highly related with a number of built-up patches. Forest fragmentation patterns of the 13 watersheds were classified for the similarity in forest fragmentation. This study presents the forest management plans including distribution and level of land development and forest conservation.

      • Construction of Expression Vectors Containing Portable PL-Promoter of $\lambda$ Phage

        본 실험에서는 prokaryotic promoter중에서 강력하다고 알려진 phage $\lambda$의 PL-promoter를 끼어 넣은 유전자 운반체를 개발하여 온도로써 이의 발현을 조절할 수 있는 대장균 속에 이주시켜 유전자 발현이 강력하면서도 온도로써 그 발현을 조절할 수 있는 host-vector system을 확립하였다. $\lambda$ phage의 PL-promoter와 N gene이 함유된 BamHI--BglII fragment가 pBR322의 BamHI site에 삽입된 pPL81에서 N gene의 일부를 포함하는 부위인 899bp를 ClaI을 처리하여 제거하고 다시 연결시켜 pPL82를 개발하였으며, pPL82는 PL-promoter와 N gene의 일부를 가지므로 fused protein을 얻을 수 있다. 또한 pPL81을 HpaII로 처리하면 PL-promoter와 operator만을 갖는 273 bp가 나오며 이를 cohesive end가 같은 pBR322의 ClaI site에 삽입하였다. PL-promoter가 시계반대방향으로 들어간 것을 pPL83, 시계방향인 것을 pPL84이라 명명 하였으며 이들 plasmid는 EcoRI과 HindIII를 처리하면 PL-promoter만 있는 약 300 bp의 DNA조각을 얻을 수 있다. PL-promoter와 구조 유전자와의 거리에 따른 PL-promoter의 효율을 $\beta$-lactamase 효소의 활성도로 조사한 결과 pPL81보다는 pPL82가 pPL82보다는 pPL83이 높았다. In this study, series of expression vectors that contain PL-promoter of phage lambda were constructed. The PL-promoters in those plasmids were able to be controlled by temperature in the host Escherichia coli which contained temperature sensitive CI857 gene on the chromosome. Plasmid pPL81, which was constructed with the fragment containing the N gene and its PL-promoter of phage lambda into the BamHI site of pBR322, was digested with ClaI for the deletion of 899 bp and self-ligated to construct pPL82. The plasmid pPL82 is useful for constructing gene fusions whose product contained amino acids encoded by N gene. Besides the construction of pPL82, HpaII fragment containing only oLpL region of pPL81 was inserted into the ClaI site of pBR322. The plasmid with the PL promoter in the counterclockwise orientation was named as pPL83, and that in the clockwise orientation was named as pPL84. These plasmids carry portable promoter that can be separated by codigestion with EcoRI and HindIII restriction endonucleases. Since the, $\beta$-lactamase genes of pPL81, pPL82 and pPL83 are located at the downstream of the PL-promoter, the $\beta$-lactamase can be induced at $42^{\circ}C$. The comparison of the production of $\beta$-lactamase indicated that PL-promoter of pPL82 was more efficient than pPL81, and that of pPL83 was the most efficient. These results indicated that when the structural gene is closely located to the PL-promoter, the influence of PL-promoter is more effective.

        삵의 서식지 적합성 평가를 위한 분석단위 설정 및 보전지역 선정 - 충청도 지역을 중심으로 -

        본 연구는 환경부 보호종으로 지정된 삵을 대상으로 다양한 단위면적의 격자 및 유역을 평가단위로 설정하여 충청도 지역의 서식지 적합성 평가를 실시하기 위해 수행되었다. 이미 선진국에서는 GAP(Gap Analysis Program), 서식지 평가절차 및 서식지 적합성지수의 활용 등을 통해 종의 보전을 위한 서식지 평가 및 관리정책을 수립하고 있는 상황이며, 국내에서도 서식지 보전을 위한 다양한 평가방법들이 제안되고 있다. 그러나 국내 연구들은 아직 평가단위에 대한 고찰이 부족하고 분석 결과를 광범위하게 적용하는데도 문제가 있는 실정이다. 본 연구에서는 표고, 경사, 산림, 토지피복, 도로, 하천 등의 환경변수를 로지스틱 회귀모형을 활용하여 유역단위와 격자단위별로 서식지 적합성 평가를 실시하였다. 또한, 이 결과를 비교하여 광역적인 지역단위에 적합한 서식지 적합성 평가단위를 확인하였다. 평가 결과, 종의 발견지점 자체에 대한 환경인자 평가보다 주변의 일정 면적을 함께 고려하여 서식지를 평가하는 것이 적합하며, 그 단위면적은 격자단위 평가의 경우 반경 100m 유역단위의 경우 $1km^2$가 삵의 서식지 적합성을 평가하는데 가장 효과적임을 확인할 수 있었다. 본 연구는 기존의 서식지 보전을 위한 다양한 보전지역 선정 방법을 보완할 수 있는 방법론을 제안하고 있으며, 이 결과는 전국적으로 보전가치가 높은 종의 보전을 위한 관리정책에 활용될 수 있을 것으로 판단된다. The purpose of this study is to compare a habitat suitability grid unit included within a radius of 100m and $1km^2,\;2km^2,\;4km^2$ watershed units in order to predict suitable habitats for Chungcheong province's endangered leopard species(Prionailurus bengalensis). Other developed countries have carried out habitat assessment and established management policies for species conservation using such methods as HEP(Habitat Evaluation Procedures), HSI(Habitat Suitability Index) and GAP(Gap Analysis Program), etc. In accordance with these studies, many evaluation methods for habitat conservation have been proposed in Korea, but these studies are lacking in consideration of analytic units and general application of analysis results. This study predicted leopard habitat using a logistic regression analysis according to analytic units by data from 56 location and 8 sources of environmental data, including elevation, slope, forest area, land cover, roads, water, broadleaf trees, and human habitation. Moreover, the habitat suitability assessment unit was confirmed by a model comparison process encompassing model explanation. verification, and application on a regional scale. Results showed that assessment methods that took into consideration areas in and around the location points were beneficial in predicting habitat and that the assessment unit was appropriate for a 30m grid unit including areas within a radius of 100m and a $1km^2$ watershed unit in Chungcheong Province. This study suggests a method for regional habitat conservation to complement existing conservation area selection methods, and the results are expected to be used in conservation area selection and ecosystem management policies for endangered species.

        보전가치평가를 위한 경관생태학적 지표의 활용 및 적용

        The purpose of this paper is to assess conservation value of forests and cultivated areas from the landscape ecological point of view. The main focus of landscape ecology estimates the reciprocal relationships between spatial patterns and ecological processes. This paper sets the criteria for conservation value and classify the conservation value based on the criteria. Forests and cultivated areas in Seo-Gu, Incheon Metropolitan City was selected as the case study area. In this research, factors such as patch size and connectivity have been selected in order to consider the landscape ecological aspect, gradient for the environmental physics aspect and to consider the environment-ecological aspect, grade of environmental-oriented land suitability analysis Map developed by the ROK Ministry of Environment has been utilized. GIS methodologies have been adopted to calculate the relationships among the above variables. Through literature review, the following evaluation criteria have been adopted: (1) based on island biogeography and metapopulation dynamics theory, patch size criteria are set 2ha and 10ha; (2) connectivity was set the degree of connection with surrounding grids; and (3) gradient of 20 degrees. Conclusively, this paper suggests that local conditions, landscape ecology and physical environment aspects should be considered to develop an estimation framework of the conservation value.

        산림패치의 특성이 조류 종 다양성에 미치는 영향분석 -충청지역을 중심으로-

        Urban development typically results in many species being confined to small, isolated and degraded habitat fragments. Fragment size and isolation underpin many studies of modified landscape to prevent biodiversity loss. However, habitat characteristics such as vegetation structure and edge effects are less frequently incorporated in planning. The relative influence of biogeographic (e.g. size, isolation) and vegetation parameters on assemblages is poorly understood, but critical for conservation management. In this study, the relative importance of biogeographic and vegetation parameters in explaining the diversity of forest-interior dwelling birds in forest fragments in Chungcheong Province Area. Fragment size and vegetation characteristics were consistently important predictors of bird diversity. Forestinterior bird richness was influenced by fragment size (0.437), wood age (0.332), wood diameter (0.068), and patch shape (-0.079). To preserve bird diversity of Chungcheong Province Area, it is important to consider differing responses of bird diversity to landscape change, move beyond a focus primarily on spatial attributes (size, isolation) to recognize that landscape change also has profound effects on habitat composition and quality. The result is very useful for long-term aspect of biodiversity conservation plan in regional scale.
