      • Immunoaffinity Purification and Receptor Assay for Polymerized Human Serum Albumin of Pre-S2 Peptide on Hepatitis B Virus

        대장균내에서 과량 발현된 재조합 프리-S2 펩티드를 면역친화력 크로마토그래피에 의해 분리 정제하였으며, 이 펩티드에 대한 중합 인혈청 알부민(pHSA)의 결합을 연구하였다. 프리-S2 부위의 pHSA수용체 활성을 검출하는 방법을 프리-S2-$\beta$ 갈락토시다제 융합단백질을 이용하여 개발하였으며, 민감도는 10-100 p mol의 프리-S2에 해당되었다. 또한 이 방법은 프리-S2 농도 결정 뿐 아니라 pHSA수용체 활성을 중화시킬 수 있는 인자들을 측정하는데 이용될 수 있다. Recombinant pre-S2 peptide was purified to apparent homogeneity from E. coli by immunoaffinity chromatography using monoclonal antibody and studied for its binding characteristics with monoclonal antibody and pHSA (polymerized human serum albumin). A simple and sensitive assay method for detecting the activity of pHSA receptor on pre-S2 peptide of hepatitis B virus surface antigen (HBsAg) has been developed using pre-S2-$\beta$-galactosidase fusion protein. The assay can be used for determining pre-S2 or factors neutralizing pHSA receptor activity. The sensitivity of the assay is about 10 to 100 pmol of pre-S2.

        B 형 간염바이러스 표면항원 중 Pre - S2 부위의 항원구조 및 중합 알부민 수용체의 특성

        Pre-S2 region (amino acids 120-174) of hepatitis B virus surface antigen contains immunodominant and disulfide bond-independent epitopes. Using pre-S2 peptides (recombinant peptides 121-174/adw2, 134-170/adw2 and synthetic peptide 120-145/ayw), epitope and polyalbumin receptor region on pre-S2 were studied. Polyalbumin receptor assay and enzyme immunoassay provided evidences that the region of at least 12 residues (134-145) of pre-S2 was shared by epitope for an anti-pre-S2 monoclonal antibody and polyalbumin receptor. This region was shown to be hydrophilic and a possible β-sheet.

        유전자 재조합 방법에 의해서 대장균으로 부터 재조합 인체 인터루킨-2 (rH IL-2, $Ser^{125}$-rH IL-2)를 과발현 시켰으며, 이 재조합 인체 인터루킨-2를 순수분리하여 그 생화학적 특성을 조사하였다. 분리 및 정제는 세포봉입체의 분리, 우레아 추출, 용해 및 젤여과 크로마토그라피 등을 사용하였으며, 정제된 인터루킨-2의 원형재현은 투석법을 사용하였다. 정제된 재조합 인터루킨-2의 순도는 전기영동 및 HPLC로 확인하였으며, 또한 결정을 얻음으로써 재확인하였다. 아미노말단 아미노산분석 및 아미노산배열순서를 일부 결정함으로서 정제된 재조합 인체 인터루킨-2가 천연 인터루킨-2와 동일한 일차구조를 갖고 있음을 확인하였다. The purification was carried out by methods involving isolation of inclusion body, urea extraction, solubilization and gel filtration chromatography. The renaturation of the product was achieved by extensive dialysis against the storage buffer. The purity was confirmed by SDS-PAGE and HPLC. Amino terminal amino acid analysis and partial amino acid sequence analysis showed that the primary structure of the recombinant protein (rH IL-2) was found to be identical with natural IL-2. The purity and the conformation of the rH IL-2 were al so confirmed by obtaining crystals of the recombinant protein.

        칫솔질을 이용한 구강간호가 중환자실 환자의 구강상태 및 인공호흡기 관련 폐렴에 미치는 효과

        Purpose: The purpose of this study was to examine the effect of tooth-brushing oral care on oral health and ventilator-associated pneumonia of patients in Intensive Care Unit (ICU). Methods: A total of 74 participants were recruited from a medical, surgical, or neurosurgical ICU at S Hospital in Seoul, Korea, from September of 2010 to January of 2011. An experimental group (n= 36) received oral care with tooth-brushing while a control group (n=38) received oral care with cotton ball and gauze. In both group, the oral care was done three times a day at least one minute for 7 days. Oral health was examined by the Modified Oral Assessment Guide before the intervention and each night. Results: There is no difference between the groups in aspect of passage of time (p = .603), interaction between time and intervention (p =.300), the performance intervention (p = .766), and the incidence of VAP (p = .486). Conclusion: The effect of oral care with tooth-brushing on oral health and VAP was not different from that of usual oral care in ICU. However, further study is necessary due to high attrition in this study.

      • Antigenic Structure and Polyalbumin Receptor Characteristics of Pre-S2 Region of Hepatitis B Virus Surface Antigen

        B형 간염바이러스(HBV) 표면항원 단백질(HBsAg) 중 55개의 아미노산으로 구성된 pre-S2 부위를 재조합 대장균으로부터 순수 분리하였다. 이 부위의 펩티드 절편들(121-174/adw2, 134-170/adw2, 120-145/ayw)은 anti-pre-S2(adw2) 단일클론항체 및 중합 인혈청 알부민에 결합하는 결과를 보였으며 이는 세 펩티드의 공통서열인 134-145 부위에 항원 결정부위 및 중합 인혈청 알부민 수용체가 존재하는 것을 암시한다. 134-145 부위는 HBV의 여러 serotype들에서 보존된 서열을 포함하며, 2차구조로 예측 결과 ${\beta}$-sheet 형성의 높은 가능성을 보인다. 또한 anti-pre-S2 단일클론항체가 pre-S2의 중합 인혈청 알부민 수용체 활성을 저해시키는 결과는 HBV 중화항체에 대한 항원 결정부위와 중합 인혈청 알부민 수용체 부위가 같거나 공통서열을 가짐을 뒷받침한다.         장애통합어린이집 보육교사와 특수교사의 아동 인권에 대한 중요도 인식과 실행도 비교

        본 연구는 보육교사와 특수교사의 아동 인권에 대한 중요도 인식과 실행도의 실태를 파악하고 이를 높일 수 있는 방법을 제시하는 것을 목적으로 시행되었다. 경기, 서울시 소재 47개 국공립, 법인, 민간 장애통합어린이집에 근무하는 보육교사와 특수교사 각 128명으로 총 256명을 대상으로 설문조사를 시행하였다. 수집된 자료는 대응 표본 t 검증과 연속적인 비교 방법으로 분석되었다. 본 연구의 결과, 첫째, 보육교사와 특수교사 두 집단의 아동 인권에 대한 중요도 인식과 실행도의 전체 평균에서 두 집단 모두 아동 인권에 대한 중요도 인식이 실행도에 비해 높은 점수로 나타났다. 둘째, 보육교사와 특수교사의 집단 간 아동 인권에 대한 중요도 인식과 실행도는 전체 평균에서, 두 집단이 중요도 인식에서 유의한 차이를 보이지 않았으나, 실행도에서는 보육교사 집단이 특수교사 집단에 비해 통계적으로 유의하게 높은 정도를 보였다. 셋째, 보육교사와 특수교사가 인식하는 아동에 대한 인권 향상을 위해 가장 필요한 요소에 대한 개방형 질문 분석 결과, 교사들은 아동의 인격체 존중과 개인별 아동발달에 대한 이해·교사의 근무환경개선·교사교육·교사의 인식 개선 및 실천 순으로 필요성을 제안하였다. This study aimed to investigate how about the recognition of importance and implementation of child right was between child care and education teachers and special education teachers. And how different in the recognition of importance and implementation of child right was between two groups in inclusive settings. This study was intended to suggest the strategies to improve the recognition and implementation of child right. The 256 public and private child care and education teachers(128) and special education teachers(128) in charge of inclusive classes were participated at surveys in Seoul and Gyeonggi Province. The data were analyzed by paired t-test and constant-comparative methodology. The results of this study were as follows: First, as to the comparison of the recognition of importance and implementation of child right, the recognition of importance showed higher than the implementation in both groups. Second, as to the differences of the recognition and implementation of child right, there were no differences of the recognition of importance between two groups. In the implementation, child care and education teacher group showed higher than special education teacher group. Third, as to open question analysis on the urgent needs to improve the recognition of importance and implementation of child right, two group teachers mentioned orderly the respecting the child as human being itself, the understanding the individual child development, the making the work conditions better, carrying out the various teacher education, and improving the teacher’s recognition and practice.

        퍼즐 게임 플레이에 나타난 엔트로피 감소의 시뮬레이션

        본 논문은 엔트로피 개념을 적용해 퍼즐 게임 플레이를 분석함으로써 퍼즐 게임 플레이의 원동력을 고찰하였다. 엔트로피는 하나의 닫힌 시스템에서 시간이 흐름에 따라 증가하는 에너지의 불가용성과 무질서의 정도를 의미한다. 이때 닫힌 시스템의 엔트로피의 증가 정도는 이 시스템을 하나의 전체 즉, 場으로 볼 때만 파악가능하다. 퍼즐 게임 역시 플레이어가 하나의 기호를 조작하면 場 전체가 변화하는 닫힌 계 이다. <비주얼드>에서 플레이어는 자리바꿈이라는 기호 조작을 통해 플레이 場을 지속적으로 자리바꿈이 가능한 場으로 변화시킨다. 이것은 엔트로피의 관점에서 에너지의 가용성을 높이는 플레이 행위로 불 수 있다. 한편 <테트리스>에서 플레이어는 기호 조작을 통해 빈 공간 없이 기호들을 플레이 場 하단에 쌓는다. 이것은 엔트로피의 관점에서 질서를 추구하는 플레이 행위로 볼 수 있다. 이와 같은 퍼즐 게임의 플레이는 엔트로피 증가로 나아가는 물리적 세계에서 질서를 추구하는 인간의 삶의 과정을 시뮬레이션하는 행위로 볼 수 있으며 이는 플레이의 원동력이 된다. This Study analyzes dynamic of a puzzle game play by applying entropy law. Entropy is a concept that a quantitative measure of the amount of thermal energy not available to do work in a closed system. And amount of entropy can be measured only if we see the closed system as whole, the field. Puzzle game is also closed system. When player moves an object in game, it change a relationship among objects in play field. In <Bejeweled>, through an act of position change, player sustains a play field active. In respect of an entropy, this kind of play is considered as pursue of usability of the energy. In <Tetris>, player piles up objects without empty space. In respect of an entropy, this kind of play is considered as pursue of the order. Likewise, puzzle game play can be considered as simulation of a human's pursue of the order in an entropy increasing physical world. And this pursue is a driving force of puzzle game play.

        도시 건축에서의 지역성과 역사성 -텐덴자, 이탈리아 건축운동(1965-1985)을 중심으로-

        In today’s urban architecture, historicity and locality are impending issues both in preservation of modern buildings and construction of postmodern buildings. The duality implied by historicity and locality in urban architecture dates back to Tendenza, the Italian architectural movement that unfolded from the 1960s to the 1980s. This paper, a study on Tendenza, which suggested the importance of historicity and locality in urban architecture as a new architectural ‘trend’, is summarized as follows. First, the authors have searched out that Tendenza has succeeded in preservation of the history and the tradition asserted by Rationalism which occurred during the fascist period, the Neorealistic architecture and the Neoliberty movement after World War II, adapting these criteria to the contemporary time based on rationality. From this point of view, seeking to innovate modern architecture Tendenza can be evaluated as a Neorationalism movement. Second, the authors summarized that Tendenza emphasized thepreservation of the traces of history and the memory of the community in spaces and places. To realize them in architecture, it proposed a study on the tangible form and types of cities and buildings as a methodology. The so-called ‘Urban Typomorphology’ which is the methodology of Tendenza, emphasizes the history and traditions of communities, cities, and citizens rather than the nation-state, nation, and people. In addition, Tendenza provided the basis for the architectural movement that emphasized the duality, hybridity, and emotions that go beyond uniformity, unity, and reason which are the characteristics of modern architecture. In this aspect, Tendenza has a tendency to be regarded as a postmodern movement. Third, the authors have revealed that Tendenza has the characteristics of innovating modern architecture, while at the same time, it transcends the modern architecture towards ‘postmodern’ architecture. It is a bridge movement with the duality that connects the modern and postmodern architecture. The urban architecture of today has the impending issues, like locality and historicity similar to Tendenza’s.
