        꿩의 정자형성기와 비형성기의 정소내 Sertoli cell의 형태적변화

        The morphological changes of Sertoli cells of the Korean native pheasant were studied in the active and inactive spermatogenic phases. Twenty-four male of the pheasants were studied in the active (April~June) and inactive(August~March) phase. These data are useful in studying the male genital organs of the Korean native pheasant. Light microscopic morphological changes of the Sertoli cells were studied on paraffin-embedded sections stained with hematoxylin-eosin stain. Ultrastructural changes of Sertoli cells were investigated of ultrathin section using electron microscope. Results are summarized as follows: During the active phase, the average diameter of seminiferous tubule was $245.33{\pm}29.93{\mu}m$ and was largely decreased by $94.50{\pm}14.10{\mu}m$, and the thickness of interstitial tissue was comparatively increased during the inactive phase. During the active phase, in the cytoplasmic process of Sertoli cell and lipid droplets appeared disperse. Well-developed smooth Endoplasmic Reticulum and microtuble were observed in the cytoplasmic process. The nuclei of Sertoli cells were adjacent to the basement membrane. The size of nuclei was reduced and nuclei of Sertoli cells were densely packed within the tubule. Few collagen fibers, fibroblast and various sizes of lipid droplets were observed in the interstitial cell of the seminiferous tubule.

        Monosodium Glutamate가 초생추 대퇴골 근위골단에 미치는 영향에 관한 투과 및 주사전자현미경적 연구

        This study was carried out to investigate the ultrastructural changes of the proximal epiphyseal plate of the femur in young chickens that had been treated with monosodium glutamate(MSG). Eighty 1-day old broiler chickens(Hubbard strain) were divided into control and experimental groups. The experimental group received daily administration of MSG(3mg/g of body weight in 0.75% saline) per orally for 1, 3, 6, 9, 12, 15, 18 and 21 days, and were sacrificed with exanguination. The control group received an equal volume of 0.75% saline. For the transmission electron microscopy, the prehypertrophic cartilage zone of epiphyseal plate was cleaved, fixed with 2% glutaraldehyde(containing 0.2% ruthenium red), postfixed with 1 % osmium tetroxide, embedded in Epon 812, and stained with uranyl acetate and lead citrate. For the scanning electron microscopy, the calcified zone of epiphyseal plate was cleaved and coated with gold palladium. The results obtained were as follows; 1. On transmission electron microscopic examination, the sacculation decreased from 12 day to 21 day MSG administrated groups, and the vesiculation decreased in 18 and 21 day MSG administrated groups in rough endoplasmic reticulum of chondrocytes in prehypertrophic cartilage zone. The ruthenium red binding particles in pericellular rim, territorial matrix and interterritorial matrix increased from 9 day to 21 day MSG administrated groups, but the crystalloid materials decreased. 2. On scanning electron microscopic examination, the trabecular formation and calcospherites of calcification zone decreased in 18 and 21 day MSG administrated groups. The resorption cavities widened from 15 day to 21 day MSG administrated groups.

        한우 대퇴골의 연골내골화에 관한 조직화학적 연구

        The present study was undertaken to provide basic data on fetal endochondral ossification for the native Korean cattle femur. This study was determined to the both earliest stages of chondrification and ossification by histochemical methods. The forty-five pairs of femur, from a series of embryos and fetuses ranging from 11 to 100mm in crown-rump(C-R) length, were used. These samples were divided into 9 groups. The groupings were based on C-R length, as the first grouping being 11~20mm, the second grouping being 21~30mm and so on. The results were as follows: 1. Alcianophility and PAS reaction were markedly increased in the perichondrium and interterritorial matrix in the 3rd group(C-R length 31~40mm). These reactions were decreased in the territorial matrix and in the adjacent area to the cartilage canal in the 5th group(C-R length 51~60mm). 2. Calcium deposits and collagen substances were observed initially in the 3rd group(C-R length 31~40mm). The calcium deposits and calcified cores were observed in the trabeculae of the proximal epiphyseal plate of the femur in the 8th group(C-R length 81~90mm).

        한우 자궁경부의 임신경과에 따른 형태학적 및 조직화학적 연구

        This study was performed to investigate the effect of gestation on the histological and histochemical changes of the uterine cervix of the native Korean cattle. A total of 110 cows obtained from the Chonhuk abattoir were divided into six groups. The 1st group was of 9 non-pregnant cows and 101 singleton pregnant cows were grouped into 5 groups from pregnant I to pregnant V according to gestation periods by means of crown rump length measures. For light microscopy the tissues were fixed in 10% neutral formalin and processed routinely for paraffin sections The $6{\mu}m$ sections were taken and stained with H-E, Alcian blue pH 1.0, Alcian blue pH 2.5, Alcian blue pH 2.5/PAS, PAS reaction, toluidin blue, and trichrome. The results obtained were as follows: 1. The cervical lengths and widths were increased in relation to advancing gestation. 2. The cervical folds of the pregnant groups were increased and complicated with many branches, and the cervical muscosal epithelia were increased in according to advancing gestation. 3. As advancing gestation, the tunics muscularis of cervix was increased following moderate distribution of fibroblast and vascularity, meanwhile decreased mast cells were found. 4. The cervical mucosubstance was composed of mixed mucopolysaccharides; the acid mucus was increased from the early pregnant state but the neutral mucus was found after pregnant IV, and the mucus was stained deeply in each staining in the fold cavity in according to gestation state.

        한우 태자의 견갑골 발생에 관한 조직학적 연구

        This study was undertaken to establish the sequence of development of ages and its time of the fetal endochondral ossification in the scapula of the Korean native cattle. This study was also designed to confirm through histological observation the earliest stages of both chondrification and ossification. Thirty eight scapulae, a series of embryos and fetuses from the pregnant Korean native cattle ranging from 11 to 110mm in crown-rump (C-R) length, were used. The following results were obtained. The ossification center was observed in the supra- and infra- spinous fossa in the 5th group (CRL 51-60mm), that was markedly ossified in the 6th group (CRL 61~70mm) by Alizarin red S stain. The chondrogenic center of scapula was observed in the 1st group (CRL 11~20mm). The primary ossification center was presented in the 4th group (CRL 41~50mm). In the 5th group(CRL 51~60mm), the endochondral ossification progressed actively. Alcianophility was markedly increased in the interterritorial matrix in the 3rd group (CRL 31~40mm. However this reaction was markedly decreased in the interterritorial matrix the adjacent portion to the marrow cavity and trabecula in the 5th group (CRL 51~60mm).

        한우의 흉골 발생에 관한 형태학적 연구

        This study was undertaken to obtain basic data of the sternal development in Korean native cattle from the earliest sternal formation to the ossification using histological and histochemical methods. Thrity three sterna were collected from a series of embryos and fetuses ranging from 11 to 225mm (estimated age 37~120days) in crown rump length. The bilateral sternal bars were observed in the 2nd group(CRL 21~3mm) of Korean cattle embryos. Those bars initiated to be fused in the 3rd group (CRL 31~4mm) and completed in the 7th group(CRL 71~80mm). The ossification centers were detected in the 8th group(CRL 81~90mm) also bilateral ossification centers were found in the same group. The typical epiphyseal plates, endochondral bone and calcium deposit were found in the 9th group(CRL 91~100mm). Osteocytes, osteoblasts, osteoclasts and myeloid cells appeared in ossification centers in the 10th group(more than CRL 101mm). The alcianophility responded markedly in the 9th group that was decreased and showed slightly positive reaction in territorial matrix of the 10th group. Marked positive reaction to PAS was observed in bony trabeculae in the 10th group. The positive reaction to calcium deposit by trichrome stain was observed initially in the hypertrophied zone of epiphyseal plate in the 9th group and was conspicuous in the calcified zone of epiphyseal plate in the 10th group. The 1st positive reaction to the von Kossa stain was observed in the 9th group.

        한국산 꿩의 고환수출소관의 구조

        The morphology of the ductuli efferentes of the Korean native pheasants were observed in order to obtain a basic data for further studying reproductive physiology and other male genital organs. The mature (14-16 months after hatching) male pheasants were used in this study. The specimens from pheasants were collected on a monthly basis. The general morphological changes of the ductuli efferentes were observed with hematoxylineosin stain, and semithin section by light microscope. The ultrastructural changes of the ductuli efferentes were investigated with ultrathin section by transmission electron microscope. The results obtained are summarized as follows : 1. During the breeding season, the average height of ductuli efferentes epithelium was $23.45{\pm}2.34{\mu}m$ and was largely decreased by $17.85{\pm}2.01{\mu}m$ during the non-breeding season. The thickeness of interstitial tissue was comparatively increased during the non-breeding season. 2. During the breeding season, the epithelial cells of ductuli efferentes were well developed. During the non-breeding season, epithelial layer and lumen of ductuli efferentes, were markedley reduced compared with those of breeding season. 3. Morphological changes of the ductuli efferentes underwent periodic changes paralleling to the spermatogenic cycle. 4. At least two different cell types were identified in the epithelium of ductuli efferentes, namely non-ciliated and ciliated cells. 5. The ciliated cells possess many vesicles, slightly smaller than those of the non-ciliated cells. 6. The ciliated cells contained numerous mitochondria, smooth and rough endoplasmic reticulum, Golgi complex, lysosome, and oval nuclei. The non-ciliated cells had a irregular nuclei and a cytoplasm containing few organelles. 7. During the breeding season, a number of vesicles, rough and smooth endoplasmic reticulum, Golgi complex, and mitochondria were distinctively showed in the epithelial cells but in the non-breeding season only a few observed.

        허혈양상화와 K<sub>ATP</sub> 통로가 허혈후 재관류된 흰쥐의 골격근육에서 SOD 활성 및 apoptosis에 미치는 영향

        Ischemic preconditioing (IPC), i.e., a preliminary brief episode of ischemia and reperfusion, has been shown to reduce the cell damage induced by long ischemia and reperfusion. Superoxide radical which is produced during reperfusion after ischemia was recognized as a factor of the ischemic injury and it is dismutated into $H_2O_2$ and $O_2$ by two types of intracellular superoxide dismutase (SOD), Cu,Zn-SOD in cytoplasm and Mn-SOD in mitochondria. Recently oxygen free radicals are suggested to induce the apoptosis, however mechanism of the reduced apoptosis by ischemic preconditioing was unknown, while many studies performed in mammalian heart indicated that ATP-sensitive $K^+$ ($K_{APT}$) channel activation related with the protective effects. The aim of present study is to investigate 1) whether IP upregulate the Cu,Zn-SOD and Mn-SOD activities, and 2) whether ischemic preconditioning decreases apoptosis via $K_{APT}$ channel activation in timely reperfused skeletal muscle after long ishemia. The experimental animals, Sprague-Dawley rats weighing 250~300g, were divided into 8 groups; 1) control group, 2) ischemic preconditioning only groups, 3) pinacidil, a $K_{APT}$ channel opener, treatment only groups, 4) glibenclamide, a $K_{APT}$ channel blocker, treatment only groups, 5) ischemia groups, 6) ischemia after IPC groups, 7) ischemia and pinacidil treatment groups, and 8) IP and ischemia after glibenclamide pretreatment groups. Animals of the control group were administered with the vehicle (DMSO) alone. Pinacidil (1mg/kg) was administered intravenously 5 minutes after initiation of ischemia, and glibenclamide (0.5mg/kg) was injected intravenously 20 minutes before IPC. In rats that were ischemic preconditioned, the left common iliac artery was occluded for 5 minutes followed by 5 minutes of reperfusion by three times using vascular clamp. Ischemia was done by occlusion of the same artery for 4 hours. The specimens of left rectus femoris muscle were obtained immediately (0 hour), 12 hours, 24 hours after drug administrations, IP or ischemia and reperfusion. The immunoreactivities of SOD and its alterations were observed by use of sheep antihuman Cu,Zn-SOD and Mn-SOD antibodies on the $10{\mu}m$ cryosections. The incidencies of apoptosis were observed by TUNEL methods with in situ apoptosis detection kit on $6{\mu}m$ paraffine section. The results obtained were as follows : 1. After IPC, immunoreactivities of Cu,Zn-SOD mainly in the small-sized fibers were increased by 24 hours, that of Mn-SOD at 0 hour and 24 hours. 2. No significant changes in immunoreactivities of SOD was observed in the pinacidil and in the glibenclamide treatment only groups, and in the ischemia only groups. 3. The immunoreactivities of the Cu,Zn-SOD were increased in the ischemia after IPC groups and the ischemia and pinacidil treatment groups. 4. The immunoreactivities of the Cu,Zn-SOD in the IPC and ischemia after glibenclamide pretreatment groups were not increased except for the 12 hours reperfusion group. But, Mn-SOD immunoreactivities were increased in the 0 hours, 12 hours and 24 hours after reperfusion. 5. In the control group, the IPC only groups, and the pinacidil treatment only groups, negative or trace apoptotic reactions were observed, but the positive apoptotic reaction occured in the glibenclamide treatment groups. 6. Moderate or many number of apoptosis were revealed in the ischemia groups, and also the IPC and ischemia after glibenclamide pretreatment group except for 12 hours and 24 hours after reperfusion. However, the incidence of apoptosis was decreased in the ischemia after IPC groups and in the ischemia and pinacidil treatment groups. 7. There is a coincidence between the increase of Cu,Zn-SOD immunoreactivities and the decrease of apoptosis in the presence of ischemia and reperfusion. These results suggest that the protective effec

        자돈 축추골 몸통의 골화에 관한 조직학적 연구

        The histological and histochemical study of the axis body of piglet was performed in order to clarify the ossification process. These experimental animals collected from after birth to 52 days were used for this study. The results were summarized as follows; 1. The epiphysis, the intercentrum 2 and the centrum of proatlas were appeared in the axis body and dens of piglet after birth. 2. In 39 days of age piglet, the ossification of the intercentrum 2 was first observed but the epiphysis was already initiated. The centrum of proatlas was observed cranioventral part of the dens of the 52 days piglet axis. 3. The notochordal remnants revealed distinctively in the part between the centrum 1 and the centrum2, in the apical cartilaginous tissue of dens and in the caudal cartilaginous part of body of axis. 4. Intramembranous ossification occurred in the adjacent area of perichondrium of centrum 2. These ossified trabeculae show the woven bone configuration.

        한우의 무릎관절 발생에 관한 조직학적 연구

        The morphogenesis of the stifle joint in Korean cattle embryos and fetuses was observed by radiography, alizarin red S stain and light microscopy. Fourty-eight(48) embryos and fetuses, ranging from 11mm to 160mm in crown-rump length (C-R L), were used for this study. The experimental samples were divided into twelve separate groups according to their C-R L. The first to the nineth group ranged from 11-100mm in C-R L, spaced at 10mm intervals. The tenth to the twelfth group ranged in C-R L from 101mm to 160mm, and were spaced at 20mm intervals. The results were as follows; 1. The first appearance of the interzone between the femur and tibia was formed in the second experimental group. The patellar ligament and cruciate ligament appeared in the third group and the patella, menisci and synovial cavity appeared in the fourth group. 2. The lateral and medial menisci were first obseved in the fourth group, and these structures showed crescent shape in the fifth group of the fetuses. 3. The first appearance of the femoro-tibial joint cavity was in the third group. The fifth group contained the first appearance of the patello-femoral cavity. Also, the femoro-tibial cavity was divided into two cavities by the anterior and posterior cruciate ligament.
