        신제품 설계 및 품질 개선 측면에서 본 무상보증기간 중 항만시설장비 고장 분석

        우리나라에서 항만은 수출입 화물의 99.7% 이상을 처리하는 국가 핵심 물류거점이다. 특히 항만시설장비는 컨테이너터미널의 운영에 필수적인 요소이다. 본 연구는 고장 전파에 의한 고장이 상대적으로 적은 장비 운용 초기인 무상 보증기간동안 항만시설장비에서 발생한 실제 고장 데이터를 최초로 분석하였다. 또한, 항만시설장비 중 트랜스퍼크레인이나 컨테이너크레인이 아닌 스프린터캐리어의 고장 데이터를 국내에서 처음으로 다루었다. 본 논문에서는 장비 고장 실제 데이터를 통계적으로 분석하여 그 결과를 정비 업무 경력자들의 기존 인식뿐 아니라 선행 연구 결과와 비교하고 그 차이의 원인을 설명하였다. 본 연구는 무상보증기간에 순수하게 장비 자체의 이유로 발생한 고장만을 분석함으로써, 그 결과를 장비 품질개선과 신제품 설계에 활용할 수 있으며, 컨테이너터미널의 운영에 적용하여 생산성과 효율성을 높이는데 기여할 수 있다. Ports are national key logistics bases that process more than 99.7% of import and export freights in South Korea. Port facility equipment is particularly an essential element for operating container terminals. This research newly analyzes the actual failure data collected from port facility equipment during the warranty period, an initial operating period with relatively few failures due to failure propagation, In addition, this study deals with the failure data of sprinter carrier, rather than transfer crane or container crane, for the first time in South Korea. This research statistically analyzes actual failure data, compares its results with those of a literature survey and a preliminary interview with experienced mechanics, and explains the reasons for their differences. By analyzing only the failures caused by the equipment itself during the warranty period, this paper provides useful results that can be used for quality improvements and new product design and applied to operations of container terminals to improve their productivity and efficiency.

        장애인 교통수단 효율성에 대한 핵심 결정요인 분석

        노약자나 장애인 등 교통약자를 위한 수요응답형 교통서비스에 대한 수요는 지속적으로 증가해왔다. 수요응답형 교통서비스 운영자들은 높은 운영비에도 불구하고 요금이 법률로 규제되어있는 탓에 운영에 있어서 높은 효율성을 확보할 필요가있다. 수요응답형 교통서비스에 대한 포괄적인 효율성 평가가 필수적임에도 불구하고, 이 분야에 대한 연구는 많지 않다. 본 연구의 목적은 수요응답형 교통서비스의 운영 효율성에 영향을 주는 변수를 파악하고, 수요응답형 교통서비스의 경쟁력을 향상시킬 방안을 제시하는 것이다. 본 연구는 수요응답형 교통서비스 운영자의 효율성을 분석하고 운영 효율성에 영향을 미치는 요인을 발굴하기 위한 새로운 체계를 제시한다. 본 연구 결과는 최소예약시간, 최대예약시간, 운영경험이 운영효율성에 영향을 미침을 보여준다. 그러나, 선행연구 결과와는 달리 오너쉽의 형태는 운영 효율성에 영향을 주지 않았다. 본 연구는 정책입안자와 서비스 운영자가 효율적인 경영전략을 수립하기 위한 가이드라인을 제시한다. The demand for demand-responsive transit (DRT) services has continuously increased for vulnerable transport users such as the elderly and disabled. DRT operators must maximize efficiency because their fares are regulated by law despite their high operating costs. Although a comprehensive evaluation of the efficiency of DRT services is imperative, little work has been done in this area. The main objective of this research is to identify the variables that significantly affect the operational efficiency of DRT service providers and propose ways to improve the competitiveness of DRT services. This study presents a new approach to analyzing the efficiency of DRT operators and investigating the determinants of their efficiency. The results indicate the significant impacts of minimum reservation time, maximum reservation time, and operational experience on efficiency. Unlike previous studies, however, ownership type does not significantly affect efficiency. This study presents guidance for policy makers and managers who want to develop transparent management strategies.

        Centella asiatica 추출물을 함유한 주름 개선 화장품의 효능 평가

        Although it has been reported that extracts of Centella asiatica (C. asiatica), a pharmacological plant containing various pentacyclic triterpenes, exert wound healing and collagen synthesis in skin cells, there are only a few clinical studies of the effect of C. asiatica extracts on human skin. In this study we have examined the anti-wrinkle and lifting effects of cosmetics containing extracts of C. asiatica on eleven healthy subjects. Compared to before and after treatment, the visual grade of wrinkle was improved 3.05% after 4 weeks. Ra, Rz and Rp parameters were significantly reduced after 4 weeks (p<0.05). The dermal density was significantly improved after 2 and 4 weeks (p<0.05). Skin sagging was significantly improved immediately after using the product, after 2 and 4 weeks. Adverse reactions of subjects were not observed during the course of the study. These observations indicate that C. asiatica extracts have the antiwrinkle and lifting effects on human skin and can be usefully applied to the anti-aging cosmetic industry.

        인간 섬유아세포주에서 resveratrol에 의한 GADD45 alpha 유전자 발현 증가와 이를 통한 DNA 수복

        In skin, dermal fibroblasts are a primary center of organization of dermis. Decrease of dermal fibroblasts through DNA damage by UV, oxidative stress and chemicals induces disorder of skin structure and homeostasis. Therefore, DNA damage protection and evasion are important in cosmetics. Resveratrol (trans-3, 4`, 5-trihydroxystilbene) is a polyphenol occurred naturally in grape and various medicinal plants. Although it has been shown that resveratrol decreases reactive oxygen species (ROS) by direct and indirect mechanism and increases the DNA repair activity, its detailed molecular mechanism has not been understood, thoroughly. Here we show that resveratrol plays a critical role in activation of the DNA repair activity by inducing GADD45α in human dermal fibroblasts (HDFs). Using DNA microarray analysis, qRT-PCR and promoter assay, we demonstrated that resveratrol increases expression of GADD45α in HDFs. We also found that resveratrol increases G2/M arrest and the DNA repair activity. The results here suggest that up-regulated expression of GADD45α by resveratrol attenuates cell death thought G2/M arrest and increase of the DNA repair activity in HDFs.

        백모 모구의 유전자 발현 프로파일링을 통한 백모화 기전 분석

        Hair bulb is composed of follicular melanocytes, keratinocytes, and dermal papilla fibroblasts and their relationship play an important role in the morphogenesis, growth and graying of the hair. To investigate the cause of graying hair in molecular levels, in this study, we analyzed changes of gene expression in hair bulb obtained from Korean male aged 20`s-30s`. We identified mRNA expression profiles in between graying and black hair bulb using DNA microarray analysis. We found 67 genes with more than 2-fold changes in their expression levels in graying hair bulb as compared with black hair bulb. Among them, 30 genes were up-regulated and 37 genes were down-regulated. The function of 67 genes turned out to be associated with melanogenesis, cell-cell interaction, regulation of calcium concentration, cytokine, keratin and serine type protease inhibition. These results indicate that graying is modulated by changes of the gene expression pattern in hair bulb, regulating various cellular functions.

        Centella asiatica (C. asiatica) plays a key role in traditional Ayurvedic medicine, treating a myriad of conditions. C. asiatica`s ancient reputation earned its place in modern alternative medicine and it has even been examined in a number of clinical studies. C. asiatica has many other practical uses because of its ability to improve circulation and strengthens vessel walls, varicose and spider veins. Also it is often used as an active ingredient in the cosmetic industry. The purpose of this study is to analyse the effect of obesity treatment on abdominal obese with C. asiatica extracts. This research divided 20-30`s women who have more than 30% higher percent body fat. Here we present that the group treated with C. asiatica extracts showed significant differences in changes of body compositions and concentrations of triglyceride in blood in comparison with the control group. The C. asiatica treatment has positive influence upon physical slimming, body circumference reduction and so on. These imply that if this ingredient is largely used for abdominal obesity management, it could be more helpful to preventing obesity related disease.

        In this study, we examined the fast soothing effect of Centella asiatica (C. asiatica) extracts on the red spot, the most typical symptom of skin irritation after short inflammation, and its general effects on the skin. C. asiatica has been reported to use for various purposes such as skin and scar treatment, memory reinforcement, and others. For the study, 18 men and women who are in 20`s and have the inflammatory acne on the skin with easily noticeable red spots were selected. Gel masks, emulsion lotion, and toner which were made with or without extracts of C. asiatica were applied to their faces. The results of this study show that extracts of C. asiatica have the whitening effect by alleviating the red spots on the face and reducing the melanin index, improves the pores, and enhances the overall acne grade. Therefore this study may provide a useful information for not only improving the inflammatory acne skin by using the C. asiatica extracts but also developing cosmetics related to the skin reproduction after soothing, given that the cosmeceuticals has come to the spotlight in the cosmetics industry recently.

        산화적 스트레스 모델로써 Hydrogen peroxide에 대한 인간 모유두 세포의 유전자 발현 변화 분석

        Human dermal papillas (HDPs) located at the base of the hair follicle play an important role in hair follicle. It has been reported that cellular apoptosis of hair follicle is induced by various stimuli, including oxidative stress. However, there are only few studies on cellular mechanism of hydrogen peroxide (H2O2)-stimulated HDPs. In this study, we investigated the mechanism of it through analysis of the gene expression profiling using DNA microarray in H2O2-stimulated HDPs. H2O2 decreased cell viability in a dose-dependent manner and caused G2/M arrest in HDPs. Using DNA microarray analysis, we found 207 genes with expression changes in H2O2-stimulated HDPs. Among them, 42 genes were up-regulated and 165 genes were down-regulated. These genes were associated with the regulation of cell proliferation, cell division, cell cycle and the DNA damage repair. Taken together, this study demonstrates that the oxidative stress, such as H2O2, could regulate the expression of genes associated various cellular mechanisms in HDPs.

        레몬 및 유칼립투스 에센셜오일의 피부 상재균에 대한 항산화 및 항균 효과

        레몬 오일과 유칼립투스 오일은 노화방지, 살균작용과 같은 다양한 효능을 가진 것으로 알려져 있다. 그러나 특정 피부 상재균에 대한 항균 효과와 항산화 효과는 아직 연구가 미비한 상황이다. 따라서 본 연구에서는 DPPH 라디칼 소거능 실험과 paper disc 방법을 이용하여 단일 오일들과 혼합오일의 항산화와 항균 활성을 검증하고자 하였다. 레몬 오일은 높은 DPPH 소거능을 보였으나 유칼립투스 오일은 아주 약한 소거능을 보였다. 두 오일을 혼합한 경우에는 효과가 증대되지 않았다. Paper disc 기법을 이용한 항균 효과 시험에서는 두 오일 모두 Candida albicans (C. albicans)와 Propionibacterium acnes (P. acne)에서 항균력이 우수하며 농도의존적인 효능이 있음을 알 수 있었다. 또한, 두 오일을 혼합 하였을 때 P. acne에 대하여 항균 활성이 상승됨을 알 수 있었다. 본 연구의 결과를 종합적으로 볼 때, 레몬오일과 유칼립투스 오일의 혼합 오일은 천연성분으로서 균으로 야기된 문제성 피부나 여드름 피부를 개선시키는데 효과적일 것으로 사료된다. Lemon and eucalyptus oils were known to have various biological effects such as anti-aging and sterilizing action. However these essential oils were not well studied about their antioxidant activity and antimicrobial activity against specific skin flora. In this study, we investigated antioxidant activity and antimicrobial activity of lemon and eucalyptus oils by using DPPH radical scavening activity and paper disc method. Lemon oil showed the high DPPH scavenging activity, while eucalyptus oil did not. Blending oils of lemon and eucalyptus had little enhancememt on antioxidant activity. Paper disc method demonstrated that each oil had a high antimicrobial effect against C. albicans and P. acnes in a concentration dependent manner. The blending oils had enhancememt on antimicrobial effect against P. acnes. In conclusion, the blending oil of lemon and eucalyptus can be used as a more effective natural agents for cure of skin trouble and acne.

        발아 검은쌀 올리고펩타이드의 각질형성세포에서 Hyaluronan Synthase 발현과 피부 탄력 개선 효과

        본 연구에서는 발아 검은 쌀로부터 분리한 올리고펩타이드($Oligosproutin^®$)의 피부 탄력 개선 효과에 대하여 연구하였다. 올리고펩타이드가 각질형성세포의 유전자 발현에 미치는 영향을 알아보고자 DNA microarray를 수행하였다. 각질형성세포에 올리고펩타이드를 처리하여 유전자의 발현이 2배 이상 증가하는 유전자는 745개이고, 2배 이상 감소하는 유전자는 1011개로 나타났다. 이 중 hyaluronan synthase 2 (HAS2)의 유전자 발현이 2배 이상 증가함을 확인하였다. 올리고펩타이드를 처리한 각질형성세포에서 세포활성 증가, HAS2 유전자 발현 증가, 세포내 항산화 효과를 확인하였다. 올리고펩타이드를 함유한 O/W 에멀젼을 이용한 임상실험에서 피부의 탄력 개선 효과를 확인하였다. 본 연구를 통하여 발아 검은 쌀로부터 분리한 올리고펩타이드는 피부 탄력 개선을 주는 항노화 화장품 개발에 응용될 수 있을 것이라 기대된다. In this study, the effect of oligopeptide ($Oligosproutin^®$) from sprouted black rice was evaluated for possible improvement in skin elasticity. We examined the changes in gene expression on oligopeptide-treated HaCaT cells using DNA microarray analysis. As a result, oligopeptide treatment showed a differential expression ratio of more than 2-fold : 745 genes were activated and 1011 genes were repressed. One of the most interesting findings is a 2-fold increase in hyaluronan synthase 2 (HAS 2) gene expression by oligopeptide. We also found that oligopeptide increased cell proliferation, HAS2 mRNA expression and intracellular ROS scavenging activity in HaCaT cells. A human clinical study which oil-in-water emulsion with oligopeptide was topically applied showed significant increase in skin elasticity. These results suggest that the sprouted black rice oligopeptide ($Oligosproutin^®$) can be effective anti-aging ingredient for cosmetics.
