당고특대황(唐古特大黃)의 주증(酒蒸) 여부가 혈관이완 기전에 미치는 영향

Objectives: The aim of this study was to evaluate the differential mechnism of vasodilation of alcohol steamed Rhei Tangutici Radix et Rhizoma. (ART) and Rhei Tangutici Radix et Rhizoma. (RT) in rat thoracic aorta. Methods: Rat aortic ring preparations were mounted in organ baths with oxygenated (95% O2-5% CO2) Krebs-Ringer bicarbonate solutions at 37±0.5 ℃and subjected to contractions or relaxations. Results: ART exerted vasorelaxation on phenylephrine(PE)-induced contraction in a dose dependent manner. Vasorelaxation effects of ART and RT were endothelium-independent. In the Ca2+-free high KCl (60 mM) baths, the contraction of aortic rings induced by accumulative addition of Ca2+ (0.3-10.0mM) was significantly reduced by pre-treatment with both ART and RT for 10 min. The magnitude of vasodilatation was biggerin ART. Moreover, verapamil (0.001uM) and diltiazem (10uM), voltage operative Ca2+channel blockers, attenuated the relaxation effect of ART but not that of RT. In the absence of extracellular Ca2+, pre-incubation of the aortic rings with RT (1.0㎎/㎖) significantly reduced the contraction caused by PE but not that of ART. K+ channel inhibitors such as glibenclamide (Gli, 10-5M), tetraethylammonium (TEA, 1mM) and 4-aminopyridine (4-AP, 0.2mM) significantly reduced the ART`s vasorelaxation efficacy, but not that of RT. However, the relaxation effects of ART and RT were not inhibited by pre-treatment with indomethacin (10-5M), and atropine (10-6M). Conclusions: These results suggest that the endothelium-independent relaxation is due to inhibition of Ca2+ influx via the suppression of Ca2+ release from intracelluar store in RT but via both voltage operative Ca2+channel blockage and K+ channel activation in ART.
HMC05의 배양 Chinese Hamster Lung 세포를 이용한 염색체이상 시험

Objectives: We investigated genetic toxicity of HMCO5 in relation to chromosome aberration test on Cultured Chinese Hamster Lung (CHL) in the presence and absence of S-9 mix. Methods: Experimental groups were divided into two groups: with S-9mix (+S) or without S-9 mix (-S). -S group was also divided 2 series by treatment hours (6 hr: 6-S; or 24 hr; 24-S). Each group treated with vehicle only (complete culture medium), HMCO5 (1,250, 2,500, 5,000 ㎍/㎖), and cyclophosphamide monohydrate (CPA) and ethylmethanesulfonate (EMS), respectively. Results: HMC05 did not show any aberrant metaphase. However, there were significant (p<0.01) aberrant metaphases with CPA in S+ and with EMS in S-. Conclusions: These results indicate that HMC05 fomula does not show any toxicity in chromosome aberration test.

Objectives: We investigated genetic toxicity of HMCO5 using the Micronucleus Test in bone marrow cells of mice and Bacterial Reverse Mutation Assay in plate incorporation method according to OECD Guidelines and KFDA Guidelines. Methods: 1. Micronucleus test: The male rats were divided into 5 groups, respectively; G(1), treated with distilled water: G(2), treated with 1250mg/kg HMC05: G(3), treated with 2500mg/kg HMC05, G(4), treated with 5,000mg/kg HMC05; G(5), treated with Cyclophosphamide·H2O. Sterilized distilled water and HMC05 were administered for two consecutive days. Cyclophosphamide·H2O was administered once on the day of 2nd administration. 2. Bacterial Reverse Mutation Aassay: Experimental groups were divided into two groups: with S-9mix(+S) or without S-9mix(-S). Each group treated with sterilized distilled water only, HMCO5(62, 185, 556, 1,667, 5,000㎍/plate) and, positive vehicles(Sodium azide, 2-Aminoanthracene, 4-Nitroquinoline N-oxide, ICR 191), respectively. Results: HMC05 did not show any changes in the number of micronucleated polychromatic erythrocytes(MNPCE) among 200 polychromatic erythrocytes compare to negative control. However, there were significant (p<0.01) increase with CPA in MNPCE. In Bacterial Reverse Mutation Aassay, no significant increases in the number of revertant colonies compared to (삭제) negative control were detected in all concentrations of HMC05. Conclusions: These results indicate that HMC05 did not show any genotoxicity against in Micronucleus test and Bacterial Reverse Mutation Aassay.
갈근(葛根)의 항산화작용에 의한 위점막 보호효과와 내인성 Sulfhydryl 화합물의 영향

Ethanol induces compoundhemorrhagic gastric lesions and causes a dose-dependent decrease in the concentration of endogenous nonprotein sulfhydryls in rat gastric mucosa. Sulfhydryl-containing drugs protect rats from ethanol - induced gastric lesions. Based on this findings, we investigated the involvement of sulfhydryl compounds in the antioxidant effects of Puerariae radix, a traditional herbal medicine, against ethanol - induced gastric lesions in the absence and presence of iodoacetamide(IDA. sulfhydryl blocking agent) in rats. respectively. Because of the known role of sulfhydryls in gastric cytoprotection, its role in gastric antioxidation was of intrest. In vitro, Puerariae radix extract(PRE) reduced linoleic acid autooxidation and exert DPPH radical scavenging effect. In vivo. PRE increased antioxidants(SOD, catalase. GSH) and reduced lipid peroxide level in ethanol-induced gastric mucosal lesions. But treatment with PRE plus IDA significantly inhibit the antioxidant effects such as SOD and GSH but did not affect catalase levels. These results suggest that Puerariae radix may play roles in the gastric cytoprotection through antioxidant effects and increase of SOD activity and GSH level are dependent of endogenous sulfhydryls.

Objectives and methods : This research aims to study about conversion system of channels. In the present study, we investigated the movement and conversion of channels on the base of three step theory(三才論), Yeak(易), Hado.Laksea(河圖洛書) and five elements motion and six kinds of factors(Six-Qi). Results and Conclusions : The organization of meridian is composed of the following three parts: hand and foot, Yin and Yang, and the viscera and bowals. It is play an important role in energy flow and its conversion. The law governing energy conversion is divided into three groups i.e. taiyin-yangming channel, shaoyin-taiyang channel and jueyin-shaoyang channel group. Those are composed of Deadea(對待) of Six-Qi, making the body homeostasis. Taken together, we suggest that the conversion system of meridian is founded on the unity between the human body and nature which provides the medical workers with a necessary method of thinking in treating diseases.
