식용버섯 자실체를 이용하여 새로운 식후 혈당상승억제 물질을 개발하기 위하여 노랑느타리버섯 자실체에 함유되어있는 α-glucosidase 저해물질을 증류수로 추출하여 식후 혈당 상승억제효과를 조사하였다. α-Glucosidase 저해물질을 함유한 노랑느타리버섯 물추출물은 streptozotocin으로 당뇨를 유발시킨 당뇨 쥐에서 장기 30 mg/kg, 150 mg/kg 경구투여(2일)와 단기 500 mg/kg, 1,000 mg/kg 경구투여(120분)시 모두, 농도 의존적으로 식후 혈당상승억제 효과를 보였다. Following preparation of water extracts of Pleurotus cornucopiae fruiting body-containing α-glucosidase inhibitor, their antihyperglycemic effects were examined using streptozotocin-induced diabetic Sprague-Dawley (SD)-rats. The water extracts from Pleurotus cornucopiae showed dosage-dependant antihyperglycemic effects on the streptozotocin-induced diabetic SD-rats after oral administration to 120 min on the short time test and 4 days on the long time test, respectively. The water extracts from Pleurotus cornucopiae fruiting body showed dosage-dependent hypoglycemic action after administration to 120 min and 4 days in the SD-rat and streptozotocin-induced diabetic SD-rat.

In order to develop new valuable pear marc nuruk and further, reduce environmental pollution by pear marc from pear juice processing, we prepared pear marc nuruk by incubation of Aspergillus oryzae into pear marc containing 50% of moisture at 30oC for 7 days. α-Amylase and glucoamylase activities of the pear marc nuruk were 320.2 IU and 442.8 IU, respectively and its acidic protease activity was showed 142.6 IU. After brewed makgeolli by using the pear marc nuruk, cooked rice and Saccharmyces cerevisiae, its physicochemical characteristics was investigated. Ethanol content of pear marc nurukmakgeolli was 6.8% after fermentation at 25oC for 10 days and also pear marc makgeolli showed 45.6% of antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity. In conclusion, pear marc nuruk had high amylase activity and pear marc-makgeolli had also good fermentation characteristics and antihypertensive ACE inhibitory activity. Therefore, it has the potential to become a new nuruk for brewing makgeolli.
영가철 충진 복극전해조를 이용한 TCE 정화기법에 관한 연구

영가철을 이용한 투수성 반응벽체는 지하수를 처리하는 원위치정화기술로 국내외적으로 연구가 활발히 진행되고 있다. 본 연구는 기존 영가철을 이용한 투수성 반응벽에 직류전원을 공급하여 효율이 향상된 TCE 처리기술 개발을 목적으로 하였다. 주문진규사로만 충진한 컨트롤컬럼, 영가철과 주문진규사로 충진한 영가철컬럼(영가철:주문진규사 = 1:2(v/v)) 그리고 영가철 컬럼에 직류전원을 공급한 복극전기분해컬럼, 이 세 컬럼의 운전을 통해 TCE 분해효율을 비교하였다. 실험 결과, 영가철를 충진하고 직류전원을 공급한 복극전기분해컬럼이 영가철만을 사용한 컬럼에 비해 높은 TCE의 환원효율을 나타내었다. 이는 영가철 입자가 미세전극으로 작용하여 전자의 이동을 촉진시킨 것에 기인한 것으로 보인다. 따라서 영가철이 반응매질로서의 역할만 할 때보다 반응매질의 역할과 동시에 전극으로서 작용했을 때, TCE 분해효율이 더 높게 나타났음을 알 수 있었다. Permeable Reactive Barriers (PRBs) using zero valent iron (ZVI, Fe0) is a promising technology for in-situ remediation of trichloroethylene (TCE) forming dense non aqueous phase liquid (DNAPL). The objective of this study is to develop an enhanced treatment method of trichloroethylene-contaminated groundwater using ZVI packed bed with direct current (D.C.). A column experiment was performed to investigate degradation efficiency of TCE that was performed in three different combination of control (only sand), ZVI column (ZVI:sand, packing ratio 1:2(v/v)) and bipolar column (ZVI:sand=1:2(v/v) with electric current) in the test columns. As the results of this study, the degradation efficiency of TCE was improved with simultaneous application of both bipolar column compared to that used ZVI column. Because ZVI particles are isolated and individual particles act like small electrodes. In this experiment, it was indicated a basic material for application of bipolar packed bed as electro-PRBs that was effective degradation of TCE.
효모 Pichia burtonii Y257-7에 의한 α-Glucosidase 저해제의 생산 및 식후 혈당 상승 억제 효과

In order to develop a new anti-diabetic α-glucosidase inhibitor, we compared the α-glucosidase inhibitory activity of the cell-free extracts of 48 strains of yeasts isolated from Korean fermented foods, and found that Pichia burtonii Y257-7 exhibited the highest α-glucosidase inhibitory activity of 55.6%. The α-glucosidase inhibitor was maximally produced when Pichia burtonii Y257- 7 was cultured in LB broth (initial pH of 6.0) at 28oC for 24 h. The α-glucosidase inhibitor, partially purified by Sephadex G-50 gel permeation chromatography and systematic solvents extraction, revealed potent hypoglycemic effects in normal rats and streptozotocin-induced diabetic rats after the oral administration of starch.
