rf PECVD법으로 증착된 a-C:H 박막의 결합구조에 미치는 보조가스의 영향

본 연구에서는 rf PECVD(13.56MHz)법을 이용하여 $CH_4$가스에 소량의 보조가스($O_2$와 $N_2$)를 혼합하여 a-C:H 박막을 얻었다. 이렇게 얻어진 박막의 증착속도는 rf power 증가에 따라서 증가하다가 200W에서는 다시 감소하였으며, 산소와 질소가스의 유량이 증가함에 따라 감소하였다. FT-IR분석으로 계산된 박막내의 수소함량은 rf power 증가와 산소 및 질소첨가량의 증가에 따라 감소하였다. 산소가스 첨가 시에는 C=O 결합이 생성되며, 질소가스 첨가 시에는 C=N 결합이 생성됨을 FT-IR 분석을 통하여 알 수 있었다. 이와 같이 산소와 질소를 보조가스로 첨가할 경우에 스퍼터링 효과로 박막내의 수소함량 감소와 더불어 a-C:H 박막의 구조 변화를 일으킬 수 있을 것으로 생각된다. Raman 분석결과 산소와 질소를 첨가함에 따라서 I(sub)D/I(sub)G비가 증가하였고, D line과 G line의 위치가 높은 파수 쪽으로 이동하였으며, D line의 폭은 넓어지는 반면에 G line의 폭은 감소됨을 보였다. 이것은 산소와 질소의 첨가로 박막내의 수소함량 감소, 결합각의 disorder 감소 및 micro-crystallite 흑연의 형성에 의한 것이라고 판단된다.

Objective : The purpose of this research is to examine the effects of Pinelliae Rhizoma (PR) extract on immune cells and cytokines in ovalbumin (OVA)-induced asthmatic mice. Methods : In vivo, C57BL/6 mice were sensitized and handicapped by OVA for 12 weeks. During this experiment, the one group was then treated with PR extract for the 8 weeks (3 times per week) and analyzed by ELISA and flow cytometer. Results : In vivo, there were significant decreases in eosinophils, IL-4, IL-5, IL-13, IgE in BALF (bronchoalveolar lavage fluid) compared with that of control group. However, $IFN-{\gamma}$ in BALF of GS group increased significantly, compared with that of control group. Additionally, the population of $CD3e^-/CCR3^+,\;CD69^+/CD3e^+,\;IgE^+/B220^+,\;CD11b^+/Gr-l^+$ cells in the PR group decreased, compared with that of control group. Conclusion : The results of this study support a role for PR as an effective treatment for asthma in its experimental success in significantly decreasing inflammation and asthma reactions, and in increasing $IFN-{\gamma}$, which helps prevent such reactions.

The effect of IAA on lysine decarboxylase(LDC)activity, diamine oxidase(DAO) activity and the polymine level in seedlings of soybean were investigated. The seedlings of soybean were treated with IAA (10^-8M, 10^-6M, 10^-4M)and LDC activity, DAO activity and polyamine levels of cotyledon, elongating hypocotyl, elongated hypocotyl and the root were measured after 4, 6, 8, 10 days. Increased concentration of IAA stimulated the LDC activity and increased the level of polyamine in the seedlings, while reducing the DAO activity and polyamine level was increased in the seedlings. The light also stimulated the IAA effect on polyamine level and DAO activity, and polyamine level was higher in the seedlings grown in the light than those in the dark. Cadaverine was the most abundant polyamine in the seedlings of soybean. The levels of diamines, cadaverine, putrescine were higher in the clongated hypocotyl and root than in the elongating hypocotyl, but the levels of spermidine and spermine were relatively higher in the cotyledon than in the other parts. Accumulation of diamines by high concentration of IAA is postulated to be a phenomenon of inhibition in seedling growth.

Addition of plant growth hormones [0.01 ㎎/L NAA and 0.2 ㎎/L benzyladenine (BA)] to a woody plant medium stimulated the adventitious bud formation of poplar explants during culture. Endogenous IAA content increased rapidly at the initial culture stage and then decreased, being followed by rapid increment again at the late culture. But the content of trans-zeatin riboside (t-ZR) increased continuously during the culture. Cytoplasmic soluble proteins were analyzed by one- and two- dimensional SDS-PAGE. Increased amount of 40 kD band was detected by one-dimensional electrophoresis using Coomassie Blue staining during the culture and tow distinctive proteins whose mol wt is 40,000 were detected by two-dimensional electrophoresis using autoradiography and these proteins were synthesized continuously prior to the adventitious bud formation. When the midvein segments were transferred to the actinomycin D-containing medium, the spots of adventitious bud-related proteins (ABRPs) did not disappeared but weakened in intensity. So, it is concluded that genes coding for the ABRPs are regulated to some degree at the transcriptional level. Also, they were not observed in BA-free medium, suggesting that these proteins be regulated by cytokinin, which made them possible to form the adventitious bud.
옥수수 중배축으로부터 분리한 원형질체에서 IAA 와 Zeatin 에 의한 세포질 Ca2+ 농도의 변화

Ca^2+ is implicated as a second messenger in coupling various stimuli such as hormone, gravity and light. To determine whether or not plant hormones mobilize calcium with different action, we investigated the cytosolic Ca^2+ changes by IAA and zeatin in the protoplasts isolated from elongating mesocotyl of maize. IAA increased the influx of Ca^2+ due to the calcium channel opening, which was confirmed by using verapamil, calcium channel blocker. On the other hand, zeatin increased the cytosolic Ca^2+ by promoting the efflux of Ca^2+ derived from cellular organelles. These results suggest that different calcium flux induced by IAA and zeatin plays a role in appropriate response resulting in increase of cell elongation or repression of cell elongation.
