        Bt 벼의 토양미생물상 영향 비교평가

        경제적 및 농업적 장점은 유전자 변형 작물 재배면적의 증가를 가져왔다. 그러나 유전자 변형 작물의 상업적 재배전에 유전자 변형 작물의 인간건강 및 환경에 미칠 잠재적 위해성에 대한 면밀한 검토가 필수적이다. 본 연구에서는 Bt벼의 토양미생물 군집에 미치는 영향을 조사하였다. 토양화학성분을 분석한 결과, Bt벼와 낙동벼 근권토양 간 화학 성분의 유의성 있는 차이는 없는 것으로 조사되었다. 재배전, 재배초기, 최고 분얼기의 토양미생물 군집밀도를 조사했을 때 Bt벼 근권토양의 세균, 방선균, 진균 군집밀도는 낙동 벼와 유사한 수준으로 나타났다. 시기별 DGGE 분석결과 Bt 벼 근권토양 전체미생물상은 낙동벼와 차이가 없는 것으로 조사되었다. Pyrosequencing을 통한 Bt벼와 낙동벼의 미생 물 군집조성을 조사한 결과 주요 미생물상 분포에 있어서도 매우 유사한 양상을 나타내었다. 위의 결과들을 종합해볼 때 Bt 재배에 따른 토양미생물상에 미치는 영향은 미미한 것으로 사료된다. 수확 후 벼 잔존물이 토양환경에 미치는 영향에 대해서는 좀 더 연구가 진행되어야 할 것이다. The cultivation of genetically modified (GM) crops has increased due to their economic and agronomic advantages. Before commercialization of GM crops, however, we must assess the potential risks of GM crops on human health and environment. The aim of this study was to investigate the possible impact of Bt rice on the soil microbial community. Microbial communities were isolated from the rhizosphere soil cultivated with Bt rice and Nakdong, parental cultivar and were subjected to be analyzed using both culture-dependent and molecular methods. The total counts of bacteria, fungi, and actinomycetes in the rhizosphere of transgenic and conventional rice were not significantly different. Denaturing gradient gel electrophoresis (DGGE) analysis of PCR-amplified 16S rRNA genes revealed that the bacterial community structures during cultural periods were very similar each other. Analysis of dominant isolates in the rhizosphere cultivated with Bt and Nakdong rice showed that the dominant isolates from the soil of Bt rice and Nakdong belonged to the Proteobacteria, Cloroflexi, Actinobacteria, Firmicutes, and Acidobacteria. These results indicate that the Bt rice has no significant impact on the soil microbial communities during cultivation period. Further study remains to be investigated whether the residue of Bt rice effect on the soil environment.

        BACKGROUND: This paper describes the successful discrimination of GM crops from the respective wild type (WT) controls using spectroscopy and chemometric analysis. Despite the many benefits that GM crops, their development has raised concerns, particularly about their potential negative effects on food productionand the environment. From this point of view, the introduction of GM crops into the market requires the development of rapid and accurate identification technologies to ensure consumer safety. METHODS AND RESULTS: The development of a GM crop discrimination model using spectroscopy involved the pre-processing ofthe collected spectral information, the selection of a discriminant model, and the verification of errors. Examples of GM versus WT discrimination using spectroscopy are available for soybeans, tomatoes, corn, sugarcane, soybean oil, canola oil, rice, and wheat. Here, we found that not only discrimination but also cultivar grouping was possible. CONCLUSION: Since for the determination of GM crop there is no pre-defined pre-processing method or calibration model, it is extremely important to select the appropriate ones to increase the accuracy in a case-by-case basis.

        The studies about the effect of planted GM crops on soil microorganisms have been continued. Although many studies have been carried out over the past decades, they provided contradictory information about the effect on soil microorganisms even for the same GM crop due to the diversity of soil environment, which means the effect of GM crops on soil microorganisms should be considered in many aspects. In this study, we investigated the effect of the drought tolerant rice, MSRB2-Bar-8 which expresses CaMSRB2 gene on soil microorganisms based on the culture-dependent and culture-independent methods. For this, soil chemical analysis, population density of soil microorganisms, denaturing gradient gel electrophoresis (DGGE) and pyrosequencing were analyzed with rhizosphere soils of GM and the non-GM counterpart of Ilmi rice. There was no significant difference in soil chemicals between GM and non-GM rice. The microbial community densities of the GM soils were found to be within the range of those of the non-GM rice. The DGGE patterns of the total soil microorganisms for GM and non-GM rice were also similar. In the pyrosequencing analyses, Proteobacteria and Cloroflexi were dominant at seedling stage while Cloroflexi showed dominance over Proteobacteria at maturity stage in both soils. In UPGMA dendrogram, two soils were grouped by time period showing difference in soil microorganisms by time period; however, such difference was not the difference in soil microorganisms between GM and non-GM. In conclusion, the results from this study imply that the effect of MSRB2-Bar-8 cultivation on soil microorganisms is not significant.

        경피 간침 생검후 혈종발생에 대한 압박 및 비압박 안정의 영향

        N/A Percutaneous needle biopsy of the liver is known to be a commonly utilized and safety procedure that can be performed at the patients bedside. But the compression of the biopsy site makes the patient uncomfortable, on the other hand intrahepatic or subcapsular hematoma formation could be serious and a life threatening complication. Recently, real-time ultrasound technigues have been used for early detection of such complication. We compared blood pressure, pulse rate and hematoma incidence between the compression group (45 patients) and the noncompression group (49 patients) after percutaneous liver biopsy. The results were as follows: 1) Intrahepatic hematoma developed in one case in each group and their diameter were less than 10 mm. 2) There was no significance in hematoma incidence between the 2 groups. 3) There was no significant difference in blood pressure and pulse rate between the two groups. In conclusion, we think that noncompression after percutaneous liver biopsy is a safe method.

        본 연구의 목적은 Bt 배추의 토양미생물 군집에 미치는 영향을 조사하기 위한 것이다. 토양미생물 조사에 앞서 토양화학성분을 분석한 결과, Bt 배추와 일반품종 배추 근권토양 간 화학성분의 유의성 있는 차이는 없는 것으로 조사되었다. 생육 최대 성숙기의 토양미생물 군집밀도를 조사했을 때 Bt 배추 근권토양의 미생물 군집밀도 범위가 일반 배추 근권토양의 미생물 군집밀도 범위내에 속하는 것으로 나타났다. 월별 DGGE 분석결과 일반품종 배추 근권토양 미생물 군집에 비해 Bt 배추 근권토양 미생물 군집의 변이는 없었다. 형질전환 벡터 유래 유전자부위를 이용하여 유전자의 수평이동성을 조사했을 때 Bt 근권토양 유래 DNA에서는 벡터 유래 유전자가 검출되지 않았다. The aim of this study was to investigate the possible impact of Bt Chinese cabbage on the soil microbial community. Microbial communities were isolated from the rhizosphere of one Bt Chinese cabbage variety and four varieties of conventional ones and were subjected to be analyzed using both culture-dependent and molecular methods. The total counts of bacteria, fungi, and actinomycetes in the rhizosphere of transgenic and conventional Chinese cabbages were observed to have an insignificant difference. Denaturing gradient gel electrophoresis (DGGE) analysis of PCR-amplified 16S rRNA genes revealed that the bacterial community structures were very similar to each other and this genetic stability of microbial communities was maintained throughout the culture periods. Analysis of dominant isolates in the rhizosphere of transgenic and conventional Chinese cabbages showed that the dominant isolates from the soil of transgenic Chinese cabbage belonged to the Bacilli and Alphaproteobacteria, while the dominant isolates from the soil of conventional cabbage belonged to the Holophagae and Planctomycetacia, respectively. These results indicate that the Bt transgenic cabbage has no significant impact on the soil microbial communities.

        논둑에 분포하고 있는 식생구조에 대해 식물사회학적 연구를 수행하였다. 총 59개 조사구에 대한 식생자료의 분석은 Zurich-Montpellier 학파의 방법을 이용하였다. 논둑 식생 조사 결과 3군단, 3군집, 3군락으로 구분되었다: 쑥-수크령군단(바랭이-돌피군집, 쑥-개망초군락, 띠군락, 한삼덩굴-돌콩군락), 참억새-새군단(억새군집), 미국가막사리-개기장군단(고마리군집). DCA분석에서 식생단위와 환경과의 상관관계를 보면, 쑥-개망초군락과 띠군락은 토양 경도와 양의 상관관계, 습한 논둑에 서식하는 고마리군집과 서식폭이 넓은 억새군집은 전질소와 높은 상관관계를 나타내었다. The phytosociological study was carried out to investigate the structural characteristics of paddy levee vegetation in South Korea. The vegetation data of total 59 releves were analyzed by the Zurich-Montpellier school`s method. 6 syntaxa (3 associations and 3 communities) of paddy levee were recognized: Echinochlo-Digitaretum ciliaris ass. nov. hoc., Artemisia princeps-Erigeron annus community, Imperata cylindrica v. koenigii community, Glycine soja-Humulus scandens community, Miscantheum sinensis f. purpurascens ass. nov. hoc,, Polygonetum thunbergii Lohm. et Miyawaki 1962. Detrended correspondence analysis(DCA) showed that Artemisia princeps-Erigeron annus community and Imperata cylindrica v. koenigii community were positively correlated with soil hardness. Polygonetum thunbergii Lohm. et association and Miscantheum sinensis f. purpurascens ass. nov. hoc. was intimately correlated with high soil total nitrogen.

        BACKGROUND: Soybean [Glycine max (L.) Merrill] is legume and an important oil crop worldwide. This study was conducted to evaluate the possible impact of transgenic soybean cultivation on the soil microbial community. METHODS AND RESULTS: Microorganisms were isolated from the rhizosphere soils. Microbial community was identified based on the culture-dependent and molecular biology methods. The total numbers of bacteria, fungi, and actinomycete in the rhizosphere soils cultivated with transgenic and non-transgenic soybeans were similar to each other, and there was no significant difference between transgenic and non-transgenic soybeans. Dominant bacterial phyla in the rhizosphere soils cultivated with transgenic or non-transgenic soybeans were Actinobacteria, Firmicutes, and Proteobacteria. The microbial communities in transgenic and non-transgenic soybean soils were characterized using the denaturing gradient gel electrophoresis (DGGE). The DGGE profiles showed the different patterns, but didn`t show significant difference to each other at 0.05 significance level. DNAs were isolated from soils cultivating transgenic or non-transgenic soybeans and analyzed for persistence of transgenes in the soil by using PCR. PCR analysis revealed that there were no amplified γ-tmt and bar gene in soil DNA. CONCLUSION(S): The results of this study suggested that microbial community of soybean field were not significantly affected by cultivation of the transgenic soybeans.

        한반도 동부 농업생태계에 따른 지표배회성 무척추동물의 군집 특성 비교

        농업생태계에 따른 지표배회성 무척추동물의 군집 특성을 비교하기 위하여 한반도 동부의 3개 행정권역내에 분포하는 논, 밭, 과수원을 대상으로 선정하였다. 2013년부터 2015년까지 매년 2회에 걸쳐 pit-fall trap법을 사용하여 채집된 무척추동물을 통해 군집분석을 하고 기상자료를 포함하는 환경요인의 영향을 분석하였다. 전체 채집된 무척추동물은 13목 58과 172종 6,420개체이며, 분류군 중에서 벌목(38.26%), 메뚜기목(16.28%)이 우점하였다. 경상남도 출현 개체수가 2,983개체(46.46%)로 가장 많았으며, 경상북도가 풍부도(1.96), 다양도(1.51) 모두 높게 나타났다. 농업생태계 유형 중 논에서 69종 3,299개체(51.39%)가 조사되었으며, 종다양성 지수도 높게 확인되었다. 특히 우점종인 개미과의 포함유무가 종 다양성지수 분석에 미치는 영향은 미미한 것으로 보여진다. 곤충 분류군과 환경요인과의 정준상관분석 결과, 토양 pH, 토양온도와 토양함수율의 영향에 의해 매미목, 바퀴목, 흰개미목, 딱정벌레목과는 양의 상관관계로 구분되었고, 기온, 강수량, 전기전도도, 상대습도, 최저초상온도와는 음의 상관관계를 나타냈다. 무척추동물 군집은 계층적 군집화를 통해 생태계 유형별로 패턴화가 일어나는 것으로 확인되었다. 이러한 결과는 공간과 환경요인과의 상관관계가 무척추동물의 분포에 다소 영향을 주는 것으로 사료된다. To compare the features of ground-dwelling invertebrates according agroecosystems, we selected paddy fields, dry fields, orchards in the Eastern region of Korea. The surveys were performed by using pit-fall traps twice per year from 2013 to 2015. Total 6,420 individuals of 172 species belonging to 13 orders, 58 families were investigated in the Eastern region, the species of Hymenoptera (38.26%), Orthoptera (16.28%) accounted large portion of the communities. In the geographical observation, invertebrates were caught was 2,983 individuals in Gyeongsangnam-do, the diversity index of Gyeongsangbuk-do community was higher than of the others and abundance and species richness of paddy field were higher than from dry field or orchard. To understand the relation between taxonomic groups and environmental factors, we carried out the canonical correspondence analysis and hierarchical clustering. As a result, Homoptera, Blattaria, Isoptera, and Coleoptera were positively related to soil pH, soil temperature, and moisture contents, and negatively related to the others. Invertebrate community also were patterned dependently by type of ecosystems. This results were shown that distribution of invertebrates is a few influenced the relationship of the space habituated invertebrates and environmental factors.
