        구기자술의 저장에 따른 성분변화

        Fruit of Lycium chinense Miller was known to Korean as traditional medicine that has effective components for strengtherning function of human body, especially liver. To study characteristics of the wine and Yakju made by fruit of Lycium chinense Miller, general components such as organic acid, methanol, ester, fusel oil and SO₂ were analyzed after storing them for 10 and 20 days at 20℃ and 30℃, respectively. Organic acid contents of the wine were mainly lactic, malic, tartaric ann acetic acid. Ester and fusel oil contents of the wine were 4.0∼5.0 ㎎% and 12∼30 ㎎%, respectively, and the values were smaller than those of the Yakju. Methanol content of the wine was 25∼27 ㎎%. Storage temperature in terms of the quality of the wine and Yakju was better in 20℃ than 30℃. Treatment of the wine and Yakju either by adding 100 ppm of SO₂ or heating at 60℃ for 30 min was better in quality than control for storage.

        한방에서 강장과 보간 등에 효과가 있는 것으로 알려진 구기자를 원료로 술을 양조하여 그 일반성분, 유기산, 메탄올, ester, 휴젤유 및 $SO_2$ 등을 조사하였고, $20^{\circ}C$와$30^{\circ}C$에서 20일 저장하면서 위의 성분분석과 관능 검사를 하였다. 보당형 구기자술 중의 유기산량은 젖산, 능금산, 주석산 및 초산 등이 주를 이루었으며 ester 및 휴젤유는 각각 $4.0{\sim}5.0\;mg%$와 $12{\sim}30\;mg%$ 그리고 methanol은 $25{\sim}27\;mg%$이었다. 약주형 구기자술의 ester 및 휴젤유는 보당형보다 많았다. 한편, 보당형 및 약주형 구기자술의 주질에 미치는 저장온도는 $30^{\circ}C$보다 $20^{\circ}C$가 좋았으며 살균 처리법은 $SO_2$ 100 ppm 첨가 또는 $60^{\circ}C/30$분의 처리가 좋았다.         갯기름나물의 쿠마린에 의한 RAW 264.7 세포주의 Nitric Oxide 생성 저해활성

        During the screening for inhibitors of nitric oxide production in LPS-activated macrophage, RAW 264.7 cells. Five coumarins were isolated from chloroform extract of the root of Peucedanum japonicum. They were identified as praeruptorin A (1), xanthotoxin (2), psoralen (3), isopimpinellin (4), bergapten (5) on the basis of spectroscopic methods. The $IC_{50}$ values for nitrite production by activated macrophages were about $1.5\;{\mu}g/ml$ (1), $0.3\;{\mu}g/ml$ (2), $1.0\;{\mu}g/ml$ (3), $25\;{\mu}g/ml$ (4), $25\;{\mu}g/ml$ (5), respectively. However, the inducible nitric oxide synthase (iNOS) was not inhibited by treatment with these compounds. Their inhibitory effect on nitric oxide production was resulted from the supperssion of iNOS expression.

        대장균군 검사용 간이 시험지 개발

        대장균군 검사용 간이 시험지는 본 실험실에서 국내 최초로 고안, 개발하였으며 이 간이 시험지법은 현장 검사법의 하나로 대장균군이 내는 succinic acid dehydrogenase 때문에 tetrazolium salt가 환원되어 적색 반점을 형성하는 것을 이용한 방법으로서 이 간이 시험지의 제조는 대체로 종래의 표준 평판법과 거의 동일한 조성의 배지와 사약을 사용하여 여지에 흡착치킨 후, 건조시켜(60℃) 멸균한 것으로 표준 평판법과 어떤 상관관계가 있는가를 검토하였다. 이 간이 시험지의 제조에서는 bile salt No. 3를 deoxycholate로 대체하여 제조 원가를 절감하였고, 또한 일본에서 현재 시판되고 있는 제품과 품질 비교시험을 하여 더 좋은 결과를 얻었으며 종래의 표준 평판법과 비교하였을 때도 오히려 표준 평판법(24-48시간 배양)보다 빠른 시간(16-20시간 배양)내에 판정할 수 있는 이점이 있으며, 표준 평판법에서는 없어서는 안될 배지나 배양 접시, pipette등의 자료 및 기구가 일체 필요없고 언제 어디서나 현장에서 직접 시험할 수 있어 매우 간편하며 또한 저렴한 가격으로 제조할 수 있는 경제성이 높은 이점을 갖고 있다. The objective of this study was to develop a paper strip which could determine E. coli qualitatively and quantitatively in water, wastewater, drinks, or food. This paper strip method was a simple and rapid test method that determine E. coli by visual identification. In this study, nutrient culture media were formulated and characterized for optimum conditions. Paper strips were then prepared by impregnating into the media and dried at 60℃. The test procedure is quite simple to use. The paper strip was dipped into a sample, and excess sample was removed. The strip was then incubated at 37℃ for 16 to 20 hours and the number of colonies on the strip was counted. The color of the colony spots produced by microorganisms varied depending on the media formulation. Violet-red spots were produced by E. coli. The test method was simple, rapid and no special laboratory equipment was necessary for visual identification. Therefore, this test method is applicable to on-site tests such as field tests or home tests. The paper strip method was compared with the standard agar plate method and Japanese commercial product. The method of the economical preparation of test strips was studied for production on industrial scale.

      • 국내산 식물의 항균활성 검색

        한국에서 자생하는 125종의 식물에서 추출한 141가지 메탄올 추출물들에서 식품부패미생물인 Bacillus subtillis, Staphylococcus aureus, Escherichia coli, Enterobacter aerogenes 그리고 Pseudomonas aeruginosa에 대한 항균 활성도를 조사하였다. Bacillus subtillis에 대해 가장 강한 항균력을 보인 것은 오미자였으며 staphylococcus aureus 에 대해서 대황이, Pseudomonas aeruginosa에 대해서는 산사가 가장 강한 항균력을 나타냈다. 금은화는 Bacillus subtilis를, 산사는 Stophylococcus aureus를, 모과와 황금은 Pseudomonas aeruginosa를, 모감주나무꽃, 밤나무꽃, 장미꽃과 파꽃은 Enterobacter aerogenes를, 오미자는 Escherichia coli를 제외한 나머지 4가지 균주에 모두 항균력이 있었다 141 methanol extracts from 125 plant species which populate in Korea were screened for antimicrobial activity against various food-borne pathogens and food spoilage microorganisms. Those plants were selected from 3 different plant groups : traditional herbs, edible plants and flowers. The methanol extracts were tested by using the disk diffusion assay against five bacteria : Bacillus subtilis, Staphylococcus aureus, Pseudomonas aeruginosa, Enterobacter aerogenes, Escherichia coli. From the evaluation of the inhibition zone diameter of microbial growth, the most significant antimicrobial activity against bacillus subtilis, Staphylococcus aureus, Pseudomonas aeruginosa, Enterobacter aerogenes, Escherichia coli was observed from the extract of Schizandra chinensis (Turcz) Baill., Rheum officinale Baill., Schizandra chinesis (Turcz) Baill., Koelreuteria paniculata Lax and Crataegus pinnatifida Bunge, respectively. The extract from many plants - Koelreuteria paniculata Lax, Chaenomeles sinensis Koehne, Scutellaria bacicalensis Georgi, Castanea crenata Sieb. et Zucc., Rosa centifolia L;, Allium fistulosum L. var. giganteum Makino, Crataegus pinnatifida Bunge, Schizandra chinensis (Turcz) Baill., Lonicera japonica - showed antimicrobial activity all four tested bacteria.
