      • 가감목방기탕(加減木防己湯)이 흰쥐의 Adjuvant 유발 관절염에 미치는 영향

        Objectives : This study was to investigate the effect of Kagammokbanggi-tang on the Freund's Complete Adjuvant(FCA)-induced arthritis in rats. Methods : Arthritis was induced by intradermal injection of FCA into base of tail. Arthritic rats were divided into control(n=10) and sample(n=10) group. Control group was taken normal saline for twenty days and sample group was taken extracts of Kagammokbanggi-tang for same duration. Normal group(n=10), non-arthritic group, was injected with mineral oil and was taken normal saline for twenty days. Body weight, paw edema volume and ankle joint thickness were measured at 0, 10, 15, 20 days after treatment. $TNF-{\alpha}$, $IL-1{\beta}$, IL-6, $PGE_2$ in synovia were analysed by ELISA at 20 days after treatment. Histochemical investigation of NADPH-d in the PAG and histopathological study on the ankle joint were performed at 20 days after treatment. Results : Paw edema volume, ankle joint thickness, $TNF-{\alpha}$, $IL-1{\beta}$, IL-6, $PGE_2$, NADPH-d of sample group were significantly decreased compared with control group. Conclusions : These results indicated that Kagammokbanggi-tang has antiarthritic and analgesic effects, and inhibited expression of NOS on the progression of FCA- induced arthritis in rats.

        가미소염탕(加味消炎湯)이 Monosodium Iodoacetate로 유발된 흰쥐의 골관절염에 미치는 영향

        Objectives : This study was to investigate the effects of Gamisoyeoum-tang(Jiaweixiaoyan-tang) on the monosodium iodoacetate-induced osteoarthritis in rats. Methods : Monosodium iodoacetate induced arthritic rats were divided into control and treated group. Control group was taken distilled water for 20 days. Treated group was taken extracts of Gamisoyeoum-tang(Jiaweixiaoyan-tang) by orally for same duration. Normal group was injected with normal saline and was taken distilled water. Body weights were measured at 0, 5, 10, 15, 20 days after injection. At the end of experiment, gross and histopathological examination on the articular cartilages of the knee joints were performed. Contents of TNF-α, IL-1β, IL-6 in synovial fluids and proteoglycan contents of articular cartilages were analysed. COX-2 and iNOS immunohistochemical examination on the knee joints were performed. Results : 1. Body weights of the treated group were significantly increased compared with control group at 20 days after injection. 2. Grossly, the severity of osteoarthritis in the treated group were alleviated compared with control group. 3. PG contents in articular cartilages of the treated group were significantly increased compared with control group. 4. Histopathologically, degenerative and necrotic lesion of articular cartilages in the treated group were alleviated compared with those of the control group. 5. IL-1β contents in synovial fluids of the treated group were significantly decreased compared with control group. 6. Positive reactions of COX-2 in chondrocytes and synovial membranes of the treated group were decreased compared with the control group. Conclusions : On the basis of these results, we concluded that Gamisoyeoum-tang(Jiaweixiaoyan-tang) has anti-arthritic effects on the monosodium iodoacetate-induced osteoarthritis in rats. And it`s effects were related with reduced secretion of IL-1β and COX-2 from osteoarthritic chondrocytes and synovial membranes.

        활락단(活絡丹)의 Adjuvant 유발 관절염 진행 억제 효과

        Objectives: This study was to investigate the antiarthritic effect of Hwallak-dan(Huoluo-dan) on the complete freund`s adjuvant(CFA)-induced arthritis in Lewis rats. Methods: Arthritis was induced by intradermal injection of CFA into base of tail. Experimental groups divided into normal(n=10), control(n=10) and treated(n=10) group. Animals of control group received normal saline for twenty days, treated group received extracts Hwallak-dan(Huoluo-dan) for same duration, light mineral oil only injected non-arthritic rats were served as normal group. The incidence of arthritis and arthritic index were observed after treatment. Body weight, paw edema volume and thickness of ankle joint were measured at 0, 11, 14, 17, 20 days after treatment. White blood cell(WBC) counts in blood were analysed at 20 days after treatment and tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β), IL-6 contents in paw exudate were analysed by ELISA at 20 days after treatment. Histopathology on the ankle joint were performed at 20 days after treatment. Results: 1. Incidence of arthritis of treated group was 60% and control group was 100% at 20 days after treatment. 2. Arthritic index of treated group was significantly decreased compared with control group at 20 days after treatment. 3. Paw edema volume and thickness of ankle joint of treated group was significantly decreased compared with control group at 20 days after treatment. 4. Total WBC of treated group was significantly decreased compared with control group at 20 days after treatment. 5. Neutrophils of treated group was significantly decreased compared with control group at 20 days after treatment. 6. TNF-α content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 7. IL-1β content content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 8. IL-6 content content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 9. Histopathological arthritic index of treated group was significantly decreased compared with control group at 20 days after treatment. Conclusions: These results indicate that Hwallak-dan(Huoluo-dan) has inhibitory effect on the development and progression of CFA-induced arthritis in rats.

        진통산(痛散)이 흰쥐의 adjuvant 유발 관절염 및 척수내 C-fos 발현에 미치는 영향

        Objectives : This study was carried out to investigated the anti-pathogenetic and curative effects of Jintong-san(Chentong-san) on Complete Freund`s Adjuvant(CFA) induced arthritis in rats. Methods : In experiment, Thirty male rats were divided into non-treated normal group(n=12), control group(n=12) is distilled water treated for 15 days after onset of arthritis by CFA, treated group(n=6) is Jintong-san(Chentong-san) treated for 15 days after onset of arthritis by CFA. Body weight, paw edema volume, and thickness of ankle joint were measured at 0, 5, 10, 15 days after treatment. At 15 days after treatment, IL-1β, IL-6 and PGE2 level in Paw Exudate were analysed for anti-inflammatory effect, C-fos immunoreactive cells measurement for analgesic effect, and histopathological examination was performed on the ankle joint. Results : Paw edema volume, thickness of ankle joint, IL-1β, IL-6, PGE2, C-fos immunoreactive cells number of treated group was significantly decreased compared with control group and histopathologically, destructive lesions of articular cartilage and subchondral bony tissue, and degree of fibrous ankylosis in treated group were alleviated compared with those of control group. Conclusions : These results indicated that Jintong-san(Chentong-san) has inhibitory effects on the progression of CFA arthritis in rats, by it`s lowering effects on the secretion of IL-1β, IL-6 and PGE2, and analgesic effect by C-fos immunoreactive cells decrease.

        대강활탕(大羌活湯)이 Collagen으로 유발된 관절염의 발생 및 치료에 미치는 영향

        Objectives: This study was carried out to investigate the anti-pathogenetic and curative effects of Taeganghwal-tang(Daqianghuo-tang) on collagen-induced arthritis in Wistar rats. Methods: Experiment 1; Thirty male rats were divided into non-treated normal group, porcine type II collagen immunized control group, and collagen immunized and treated group medicated with extract of Taeganghwal-tang(Daqianghuo-tang) for 15days after collagen immunization. Body weight, paw edema volume, and thickness of ankle joint were measured at 0, 10, 15days after immunization. Arthritic score was evaluated at 11, 12, 13, 14, 15days after immunization. At 15days after immunization, the incidence of arthritis was evaluated, and serum TNF-α, IL-1β and anti-collagen IgG level were analysed, and histopathological examination was performed on the ankle joint. Experiment 2; Thirteen rats, that were shown the gross lesions of collagen-induced arthritis, were divided into control group and treated group medicated with extract of Taeganghwal-tang(Daqianghuo-tang) for 15days after onset of arthritis. Non immunized seven rats were served as normal group. Body weight, paw edema volume, and thickness of ankle joint were measured at 0, 5, 10, 15days after treatment. At 15days after treatment, serum TNF-α, IL-1β and anti-collagen IgG level were analysed, and histopathological examination was performed on the ankle joint. Results: Experiment 1; -. Incidence of arthritis was 60% in control group and 10% in treated group. -. Arthritic score of treated group(0.3±0.9 at 13days, 0.3±0.9 at 14days, 0.3±0.9 at 15days) was significantly decreased(P<0.01) compared with control group(3.0±2.7 at 13days, 3.61±3.1 at 14days, 3.9±3.4 at 15days) respectively. -. Paw edema volume(㎖) of treated group(1.90±0.11) was significantly decreased(P<0.01) compared with control group(2.55±0.53) at 15days after immunization. -.Thickness of ankle joint(㎜) of treated group(6.64±0.15) was significantly decreased(P<0.01) compared with control group(8.35±1.75) at 15days after immunization.-.Serum TNF-α level(pg/㎖) of treated group(46.00±25.90) was significantly decreased(P<0.05) compared with control group(97.52±49.97) at 15days after immunization.-.Serum IL-1β level(pg/㎖) of treated group(39.40±10.42) was significantly decreased(P<0.05) compared with control group(52.22±15.32) at 15days after immunization.-.Serum anti-collagen IgG level(㎎/㎖) of treated group(0.27±0.16) was significantly decreased(P<0.05) compared with control group(0.47±0.20) at 15days after immunization. Experiment 2; -.Paw edema volume(㎖) of treated group(3.12±0.28 at 10days, 3.12±0.27 at 15days) was significantly decreased(P<0.05) compared with control group( 3.72±0.49 at 10days, 3.62±0.48 at 15days) respectively.-.Serum TNF-α level(pg/㎖) of treated group(84.64±22.04) was significantly decreased(P<0.05) compared with control group(135.31±55.56) at 15days after treatment.-.Serum IL-1β level(pg/㎖) of treated group(58.16±7.28) was not significantly different compared with control group(66.75±10.68) at 15days after treatment.-.Serum anti-collagen IgG level(㎎/㎖) of treated group(0.78±0.09) was not significantly different compared with control group(0.87±0.08) at 15days after treatment.-.Histopathologically, destructive lesions of articular cartilage and subchondral bony tissue, and degree of fibrous ankylosis in treated group were alleviated compared with those of control group at 15 days after treatment. Conclusions: These results indicated that Taeganghwal-tang(Daqianghuo-tang) has the anti-pathogenetic effects on the onset of collagen-induced arthritis in rats, by it`s inhibitory effects on the production of anti-collagen IgG. And also, Taeganghwal-tang(Daqianghuo-tang) has inhibitory effects on the progression of collagen-induced arthritis in rats, by it`s lowering effects on the secretion of TNF-α.

        Canine herpesvirus 감염증의 병리발생에 관한 연구 I. 병리조직학적 및 전자현미경적 관찰

        This study was carried out to investigate the pathogenesis of canine herpesvirus(CHV) infection in dogs. The 17 puppies, one day old, delivered from CHV seronegative 3 dams were divided into two groups. The 13 puppies were inoculated intranasally with 1ml of CHV-KK inoculum($5{\times}10^{5.6}TCID_{50}/ml$) and 4 puppies were served as control. And then the puppies were sacrificed at 2, 4, 6 and 7 days after the treatment, and collected blood, nasal mucosa, trigeminal nerve, trigeminal ganglion, bone marrow, eye, brain and other major organs. These organs were examined histopathologically and electron microscopically. The platelets of puppies infected with CHV were dramatically decreased because of the damages of vascular endothelial cells. Histopathologically, necrotizing vasculitis and neuritis were proceeded the generalized focal necrosis of all organs. Necrotic changes in trigeminal ganglion, trigeminal nerve and ventroposteriomedial nucleus of thalamus were observed in 4 puppies infected with CHV. Herpesviral particles, various forms of maturation, were observed in endothelial cells of the alveolar capillary and hepatic sinusoid with electron microscopy. These results suggest that the generalized focal necrcsis of all organs including brain and eyes in canine herpesvirus infection were resulted from generalized vasculitis, and also the hemonecrotizing meningoencephalitis was related to the necrosis of trigeminal nerve pathway.

        한국에서의 canine herpesvirus감염실태에 대한 혈청역학적 조사

        This study was carried out to investigate the actual condition of canine herpesvirus(CHV) infection in Korea. A total of 338 serum samples were collected randomly from the breeding and companion dogs in the local areas in Korea. The serum samples were used to determine the actual condition of the canine herpesvirus infection in Korea using enzyme linked immunosorbent assay(ELISA). The mean prevalence of CHV infection in dogs was 37% and that of the breeding and companion dogs was 58% and 28%, respectively. The prevalence of CHV infection in Seoul, Kyung-gi, Chung Nam, Cheon Nam and Pusan was detected 23%, 28%, 18%, 28% and 70%, respectively. The prevalence of CHV infection in less and more than 6 months old dogs, and in male and female dogs was 26% and 40%, 42% and 33%, respectively. These results indicate that the incidence of CHV infection is high in Korea, especially breeding dogs and older dogs.

        Canine herpesvirus 감염증의 병리발생에 관한 연구 II. 면역조직화학적 관찰

        This study was carried out to investigate the pathogenesis of canine herpesvirus(CHV) infection in dogs. The 13 puppies were inoculated intranasally with 1ml of CHV-KK inoculum($5{\times}10^{5.6}TCID_{50}/ml$) and 4 puppies were served as control. And then the puppies were sacrificed at 2, 4, 6 and 7 days after the treatment, and sampled nasal mucosa, trigeminal nerve, trigeminal ganglion, bone marrow, eye, brain and other major organs for the immunohistochemical examination. Distribution of CHV antigens was limited in cytoplasms and nuclei of necrotic nasal epthelia at 2 days after infection. At 4 days after infection, CHV antigens were detected in vascular walls and peripheral nerves of nasal lamina propria, reticuloendothelial cells of spleen, interstitium of kidney, leptomeningeal vascular walls and alveolar walls, At 6 and 7 days after infection, CHV antigens were detected in all of the necrotic area. CHV antigens were also detected in vascular endothelial cells of various organs and in blood leukocytes from 4 days after infection. Among the six puppies in which necrotic lesions of central nervous system were observed, CHV antigens were detected in trigeminal ganglion, trigeminal nerve and ventroposteriomedial nucleus of four puppies and in spinal trigeminal nucleus of three puppies. These results indicate that the generalized focal necrosis of all organs including brain and eyes in canine herpesvirus infection were resulted from generalized vasculitis with leukocyte-associated viremia, and also the hemonecrotizing meningoencephalitis was resulted from spreading of CHV via blood and nerve trunk.

        간흡충 감염 햄스터의 담관암발생에서 small cell과 oval cell의 역할

        This study was carried out to examine the role of small cells and oval cells in cholangiocarcinogenesis in the hamsters infected with Clonorchis(C) sinensis. Forty two female Syrian golden hamsters were divided into two groups. Group I was for the induction of the cholangiocarcinoma, which was infected orally with C sinensis and given dimethylnitrosamine(15ppm) in drinking water for 4 weeks. Group II was served as control. More than 5 heads of hamsters in each group were sacrificed at 4, 7, 11 and 15 weeks after the beginning of the experiment. The livers were examined histopathologically, electron microscopically and immunohistochemically. The results obtained were as follows; 1. Cholangiocarcinomas were occurred in 1 of 6 animals at 11 weeks and in 4 of 6 animals at 15weeks after the beginning of the experiment. 2. Small cells and oval cells were proliferated around the portal triads from 4 weeks and peaked at 11 weeks, and slightly decreased after then. 3. The strong positive reaction to the $\alpha$-fetoprotein was shown in many of small cells and oval cells. But ductlike oval cells, which were arranged rosette form, showed week positive reaction to the $\alpha$-fetoprotein. 4. Most of small cells and oval cells showed negative reaction to the cytokeratin. But weak positive reaction in ductlike oval cells, and moderate positive reaction in cholangiocarcinoma cells were observed. These results suggested that cholangiocarcinoma induced by infection of C sinensis was believed to originate from the proliferated small cells around the portal triads which would be able to differentiate to the oval cells, ductlike oval cells, and cholangiocarcinoma cells gradually.

        감초부자탕(甘草附子湯)이 흰쥐의 Adjuvant 유발 관절염에 미치는 영향

        Objectives :This study was performed to investigate the effects of Gamchobuja-tang(gancaofu□zˇl-tang) on the Complete Freund`s Adjuvant(CFA)-induced arthritis in rats. Methods :Arthritis was induced by intradermal injection of CFA into the base of tail. Arthritic rats were divided into control(n=8) and Gamchobuja-tang(gancaofu□zˇl-tang) treated(n=8) group. Control group was taken distilled water for 15 days. Gamchobuja-tang(gancaofu□zˇl-tang) treated group was taken extracts of Gamchobuja-tang(gancaofu□zˇl-tang) by orally for same duration. Normal group(n=8) was injected with light mineral oil and was taken distilled water. Gross structure of hind paw were observed at every 5 days after treatment. Body weight, paw edema volume and ankle joint thickness were measured at 0, 5, 10, 15th day after treatment. Histopathological examination on the tarsal joint and distal tibia of the right hind paw were performed at the 15th day after treatment. White blood cell counts in blood were analysed at the 15th day after treatment. The content of interleukin-1β(IL-1β) and tumor necrosis factor-α(TNF-α) in paw exudate were analysed by enzyme-linked immunosorbent assay(ELISA) at the 15th day after treatment. Results :1. Grossly, the severity of arthritis in the Gamchobuja-tang(gancaofu□zˇl-tang) treated group were alleviated compared with control group. 2. Paw edema volume(ml) of Gamchobuja-tang(gancaofu□zˇl-tang) treat group was significantly decreased compared with control group. 3. Ankle joint thickness(mm) of Gamchobuja-tang(gancaofu□zˇl-tang) treat group was significantly decreased compared with control group. 4. Histopathologically, destructive lession of synovial membrane, articular cartilage and bony tissue in Gamchobuja-tang(gancaofu□zˇl-tang) treated group were alleviated compared with control group. 5. Total WBC(103/μl) of Gamchobuja-tang(gancaofu□zˇl-tang) treated group was significantly decreased compared with control group. 6. TNF-α content(pg/ml) in paw exudation of Gamchobuja-tang(gancaofu□zˇl-tang) treat group was significantly decreased compared with control group. Conclusions :On the basis of these results, Gamchobuja-tang(gancaofu□zˇl-tang) has anti-arthritic effects on the progression of CFA-induced arthritis in rat. And it`s effect is related with reduced secretion of TNF-α.
