        홍도 상록활엽수림의 식생과 토양특성

        홍도 상록활엽수림을 식물사회학적 방법으로 군락을 분류하고, 방형구법을 사용하여 구배분석을 실시하였다. 식물사회학적으로 군락을 분류한 결과 붉가시나무군락, 굴거리군락, 소사나무군락으로 분류되었다. 토양특성은 유기물, 전질소, 유효인산, 치환성 K, 치환성 Ca, 치환성 Mg, 양이온치환용량(CEC)의 함량이 6.49~13.71%, 0.19~0.46%, 26.19~10.63(㎎/㎏), 0.24∼0.56(cmol+/kg), 0.68~3.19(cmol+/kg), 0.83∼1.82(cmol+/kg), 9.74∼22.55(cmol+/kg)이며, pH는 4.04~4.14로 약산성토양인 것으로 분석되었다. 소사나무군락은 해발고가 가장 높고, 붉가시나무군락과 굴거리군락은 다소 낮은 해발고에 분포하는 것으로 나타났으며, 붉가시나무군락이 굴거리군락보다 전질소, CEC, 치환성 Ca 함량이 다소 많은 입지에 분포하는 것으로 나타났다. This study used a bio-social approach in classifying the vegetation communities of evergreen broad-leaved forest in Hongdo island of Korea, and the results are as follows: Quercus acuta community, Daphniphyllum macropodum community, and Carpinus coreana community. The composition of soil properties in the same areas are as follows: soil organic matter, total nitrogen, available phosphorous, exchangeable K, exchangeable Ca, exchangeable Mg contained, cation exchange capacity, and soil pH. The capacities of these chemical properties of the soil ranged from 6.49∼13.71%, 0.19∼0.46%, 26.19∼10.63(㎎/㎏), 0.24∼0.56(cmol+/kg), 0.68∼3.19(cmol+/kg), 0.83∼1.82(cmol+/kg), 9.74∼22.55(cmol+/kg), and 4.04∼4.14 respectively. The Carpinus coreana community was found in the high elevation areas; Quercus acuta community and Daphniphyllum macropodum community were also found in the low elevation. However, Quercus acuta community, compared with Daphniphyllum macropodum community, was found in the areas that contained more total nitrogen, cation exchange capacity, and exchangeable Ca.

        온도 및 지베렐린 처리가 말오줌나무 종자의 배발달 및 발아에 미치는 영향

        This study was conducted to determine types of seed dormancy in Sambucus racemosa subsp. pendula (Nakai) H.I. Lim & C.S. Chang, an endemic tree species of Korea, whose seeds have been considered difficult to germinate. Seeds of S. racemosa subsp. pendula were stratified at 25/15 or 5℃ for 0, 6, or 12 weeks (wks) and incubated at 15/6, 20/10, 25/15, or 30/20℃ (12/12 h) under 14 h photoperiod. To determine the effect of GA3 on seed germination of S. racemosa subsp. pendula, seeds were treated with 0, 500, or 1000 ppm GA3 and then germinated at 25/15 or 5℃. The change in embryo length was investigated at 25/15 or 5℃. Seeds given 12 wks of cold stratification germinated to 33.4% at 15/6℃ and to 25.4% for seeds given 6 wks of warm stratification + 12 wks of cold stratification at 20/10℃. At 25/15℃, seeds given 12 wks of warm stratification + 6 wks of cold stratification germinated to 26.0%, and to 28.2% for seeds given 12 wks of warm stratification + 12 wks of cold stratification at 30/20℃. Warm stratification alone did not germinate seeds throughout the experiment, regardless of the thermoperiod. Linear embryos began to grow after 60 days of incubation at 25/15 or 5℃. The embryo length at day 69 increased from 1.4 mm to 1.50 or 1.62 mm at 25/15 or 5℃, respectively. Embryos of S. racemosa subsp. pendula seeds grew better at 5℃ than at 25/15℃. Gibberellin was effective to break seed dormancy of S. racemosa subsp. pendula. Seeds treated with 500 ppm GA3 germinated up to 40.0% at 25/15℃ and to 62.7% for those treated with 100 ppm GA3 at 5℃. With these results, seeds of S. racemosa subsp. pendula have both nondeep complex and intermediate complex morphophysiological dormancy.

        Segregation and linkage of allozymes in megagametophyte tissues of pinus thunbergii parl

        As a first step in the study of genetics of Pinus thunbergii Parl., the inheritance and the linkage of the detected polymorphisms were elucidated. Megagametophyte tissues of individual seeds from a single clone were analysed by starch gel electrophoresis. Seven enzyme systems studied comprised a minimum of 17 isozyme loci. Among them, 11 were polymophic. Segregation of isozymes in megagametophytes of heterozygous maternal clones indicated no significant departure from the expected 1 : 1 ratio in each locus. Significant linkage, as attested by the 95% confidence interval, was recognized in six pairwise combinations. The pairs of the gene loci ME-B/LAP-A and ADH-B/MNR-C were closely linked with estimated mean recombination values of 0.153 and 0.175, respectively. The remaining tested pairs (ADH-B/ LAP-B, ME-B/MDH-C, MNR-B/MDH-C, and MNR-A/MNR-B) appeared to be weakly linked. The gene loci MNR-C, ADH-B, and LAP-B could be linearly arranged in one linkage group, while the loci LAP-A, ME-B, MDH-C, MNR-A, and MNR-B seemed to belong to the other linkage group. Considerable variation with respect to the estimated recombination frequencies was observed for the same pair of loci among some clones. A comparsion with other conifer species showed that the gene orders and distances were relatively well conserved among conifer species.

        울릉도 섬현삼( Scrophularia takesimensis Nakai) 개체군의 식생과 토양특성

        This study was carried out to analyze vegetation and soil characteristic, and ordination of Scrophularia takesimensis population, Ulleung Island. The Scrophularia takesimensis population was classified into Aster spathulifolius for. oharai dominant population, Vitis amurensis dominant population and Scrophularia takesimensis typical population. The Scrophularia takesimensis population was located in elevation of 1m to 6m, in Ulleung Island. In the study sites, soil organic matter, nitrogen, available phosphorous, exchangeable potassium, exchangeable calcium, exchangeable magnesium and exchangeable sodium concentration, and cation exchange capacity, and soil pH were 9.1~19.1%, 0.19~0.52%, 87.1~196.7mg/kg, 2.0~2.8cmol+/kg, 5.4~5.9cmol+/kg, 5.9~8.8cmol+/kg, 4.4~4.8cmol+/kg, 20.3~ 26.7cmol+/kg, and 6.8~8.0, respectively. The Vitis amurensis dominant population was mainly found in the high percentage of total nitrogen, organic matter, CEC in comparison with Aster spathulifolius for. oharai dominant population.

      • 진범( Aconitum pseudolaeve Nakai) 종자의 형태생리적 휴면타파에 전처리가 미치는 영향

        본 연구는 한국 특산식물 진범종자 휴면유형 및 배발달 조건을 밝히고자 수행하였다. 무처리 종자의 경우 15/6℃, 20/10℃, 25/15℃에서 14.0±2.83, 14.0±2.83, 2.7±4.81%의 낮은 발아율을 나타냈다. 5℃에서 4주간 습윤처리한 결과, 73.3±6.11, 61.3±6.11, 50.7±16.2로 발아율이 향상되었으며 8주 처리시 15/6℃에서 80.0±4.00의 높은 발아율을 나타내었다. 8주간 2주 간격으로 종자에 대한 배의 비율을 관찰한 결과, 5℃ 습윤처리시 0.26에서 0.65으로 증가한데 반해, 25/15℃에서는 0.45로 저조한 생장을 보여 저온습윤처리가 진범종자의 배생장에 효과적인 것을 확인하였다. 5℃, 25/15℃조건 500, 1,000ppm 지베렐린(GA3)을 처리한 결과 모든 처리구에서 14% 미만의 발아율을 나타내 지베렐린 처리는 진범종자의 휴면타파에 효과적이지 않은 것으로 판단되었다. 따라서 진범종자의 휴면유형은 형태 생리적 휴면이며, 휴면타파에는 저온습윤 처리가 필요한 것으로 판단된다. This study was conducted to serve information for the research of embryo growth requirements and a type of seeds dormancy of A. pseudolaeve Nakai, a Korean endemic species. The germination percentage of untreated seeds in control plot was 14.0±2.83, 14.0±2.83, 2.7±4.81% at 15/6℃, 20/10℃, and 25/15℃. After four weeks of cold stratification, final germination percentage of seeds was improved to 73.3±6.11, 61.3±6.11 and 50.7±16.2 at 15/6, 20/10, and 25/15℃. After eight weeks germination rate was the highest 80.0±4.00 at 15/6℃. Embryo growth was monitored for eight weeks in every two weeks intervals in the seeds incubated at 5℃, 25/15℃. Embryo ratio(embryo/seed) increased from 0.26 to 0.65 at 5℃ where as 25/15℃ incubated seeds was less embryo growth percentage. Therefore, Cold stratification was effective in embryo growth of A. pseudolaeve. On the other hand, seed incubating at 5℃, 25/15℃ after treating in Giberellic acid(GA3-500, 1,000ppm), germination percentage improved better 500ppm than control. However, only 13.3±1.33% of treated seeds were germinated. This indicates that Gibberellic acid(GA3) was less effective in breaking dormancy of seed in A. pseudolaeve. Therefore, seeds of A. pseudolaeve required cold stratification for dormancy break and possess deep complex morpho-physiological dormancy.

      • 건조 방법과 저장 온도에 따른 할미꽃 종자의 발아

        봄철 관상식물로 활용되는 할미꽃의 종자는 실온보관 수명이 6주 정도로 알려져 있으며 장기저장이 어려운 것으로 알려져 있다. 본 연구는 할미꽃 종자의 실온보관 수명을 밝히고 장기저장 기술 개발을 하고자 실시하였다. 상온보관 기간에 종자 무게 변화는 5.158g에서 8주 후 4.988g, 16주 후 4.793g으로 변화하였고, 발아율은 각각 82±4.9%, 72.5±13.42%, 70.7±8.08%였다. 기건 기간에 따라 수분함량과 발아율은 모두 감소하였으나 처리 모두 70% 이상의 높은 발아율을 보였다. 15℃, RH 15%, 4주간 건조처리 한 결과 무게는 4.0714g 감소하였으며 발아율은 73.3±1.15%였고, 건조 후 -18℃, 12주 저장 후발아율은 72.0±8.16%였다. 건조 단독처리, 건조 및 저온 복합처리 결과 수분함량과 발아율 모두 감소하였으나 70% 이상의 높은 발아율을 보였다. 할미꽃 종자는 실온 보관 수명이 6주 정도로 알려져 있었으나, 본 실험 결과 16주의 실온보관이 가능하다. 또한, 10℃ 이하의 저온건조저장이 어려운 것으로 알려져있었으나, 본 실험 결과 12주의 저온건조저장(-18℃)이 가능하다. 따라서, 할미꽃 종자에 대하여 저온건조저장법에 의한 장기저장기술 개발연구가 요구된다. For long-term storage possibility of Pulsatilla koreana(Yabe ex Nakai) Nakai ex Nakai seeds according to drying and keeping at cold temperature, water content and germination ratio were compared at room temperature drying and cold-temperature storage after drying. Depending on storage condition at room-temperature(25℃), average weight were changing from 5.158g to 4.988g, 4.798g and germination ratios were studied 82±4.9%, 72.5±13.42%, 70.7±8.08%. Although water content was decreasing, germination ratios were showing more than 70%. Drying treatment was oven-drying(15℃, RH 15%) for 4 weeks. After drying, the weight was 4.0714g that was decreased. Germination of drying seeds was 73.3±1.15%, Germination of .18℃ 12 weeks storage seeds with drying was 72.0±8.16%. According to drying and low-temperature treatment, water content were decreasing but still germination ratios were more than 70%. Pulsatilla koreana(Yabe ex Nakai) Nakai ex Nakai seeds were known that life expectancy was 6 weeks at room temperature, but 16 weeks life was possible was shown in this study. Also the seeds were known that was difficult to be storage at less 10℃ and long-term storage, but it was possible to preserve 12 weeks at drying low-temperature and to be long-term storage in this study.

        잎이 강건하고 화색이 엷은 좀비비추 ‘청나래’ 육성

        We report a technique for obtaining seeds of ‘Cheongnarae’ (a variety of Hosta minor cultivated at the Korea National Arboretum in 2019) by artificial crossing of H. minor, collected from Jeju Island, as the female parent and H. minor ‘Krossa Regal’, cultivated outside Korea, as the male parent. Among the individuals obtained after sowing, those that showed different morphological characteristics, such as leaf shape and color, were selected. Among these individuals, those with thick leaves and light-colored flowers were further selected. Vegetative propagation and cultivation steps were repeated for evaluation of the characteristics. ‘Cheongnarae’ has relatively thicker leaves and a darker color (RHS 137A) than the control variety ‘Black Hills’ and has wings at the point where the petiole meets the leaf blade. The shape of the leaf blade edge is clearly sinuous, and the flower color is close to white (White N155B). ‘Cheongnarae’ cultivated in this way can be used for pot plant or gardening.
