        단독주택용지의 이용실태와 효율적 이용방안 - 전주시 공영택지개발지구를 중심으로

          This is a study doing for the purpose of taking out an efficient utility program as analysing non-efficient utility conditions of land and grasping its problems which are happened from the public-land development district. I suggest better programs as the following after making a field-investigation and analysing development situations of 3 public-land development districts developed by Jeonju city.<BR>  The city should establish the rate of land use to meet the local character due to dividing the use in detail on industrial classification from a city-plan stage and improve investors" earnings due to making the theme-type commercial area.<BR>  The city should fix the rule on parking-zone establishment which is a formal contract necessary only to obtain a license and to be completed.<BR>  The city should make in a group among same categories of business and remodel in a house on the black-hearted business section in blank space or the earnings-fallen restaurants.<BR>  The damaged-landscape facilities should restore through the housetop landscape even in order to prevent from a heat-island phenomenon of the city.<BR>  It is necessary to put a graft in the local culture owing to introducing Han-brand concept into the building style in Jeonju city.<BR>  The city should train the experts to suggest a development model.

        국내 유류오염지역에서의 석유계총탄화수소에 의한 비발암 인체위해성평가 전략

        Human health risk assessment for petroleum, oil and lubricant (POL) contaminated sites is challenging as total petroleum hydrocarbon (TPH) is not a single compound but rather a mixture of numerous substances. To address this concern, several TPH fractionation approaches have been proposed and used as an effective management tool for the POL-contaminated sites in many countries. In Korea, there are also recognized needs to establish a reliable and cost-effective human health risk assessment strategy based on the TPH fractionation method. In order to satisfy the social and institutional demand, this study suggested that the comprehensive risk assessment strategy based on a newly modified TPH fractionation method with 10 fractions, the Korean Standard Test Method (KSTM)-based analytical protocol and a stepwise risk assessment framework should be introduced into the domestic contaminated land management system. Under the proposed strategy, POL-contaminated sites can be effectively managed in terms of human health protection, and remedial cost and time can be determined reasonably. In addition, more researches required to increase our understanding of environmental risks and improve the domestic management system were proposed.

        Sweet BV 병행 치료 후 척수 손상 환자의 감각 분절 호전 1례

        Objective : Patients with spinal cord injury are increasing in numbers. However, there is no reliable treatment guide in both conventional & complementary medicine. Also, there are not much clinical case of patients with spina cord injury in oriental medical field. We investigated effect of sweet BV on subacute stage patient with spinal cord injury. Method : 31-year old female patient with spinal cord injury was treated with herb medicine(TID), electro arcupunture (BID), sweet BV injection(QOD), Physical treatment(QD), and conventional-medicine. Result : We had a satisfactory result with using sweet BV injection. The patient's ASIA grade improved from 34 to 52. And Frankle classification of the patient shifted from A to B. Conclusion : We reach a conclusion Using Sweet BV improve the sensation of patient with spinal cord injury. And more study about this disease is needed.

        Streptomyces phospholipase D의 정제를 위한 면역친화 크로마토그래피의 개발

        Streptomyces somaliensis가 생산하는 phospholipase D (PLD)를 정제하기 위하여 펩티드 항체 결합 면역 친화 크로마토그래피용 칼럼을 개발하였다. 단백질 구조 예측 프로그램과 Streptomyces PLD X-선 결정구조를 참조하여, S. somaliensis PLD의 1차 구조로부터 항원특성이 높고. 표면에 위치하는 것으로 예상된 5종류의 펩티드들을 epitope로 선정한 뒤, 이에 대한 항체로 면역친화 크로마토그래피용 칼럼을 제작하였다. 배양 농축액을 칼럼에 통과시켜 정제한 활성 분획을 SDS-PAGE 및 Western blot 결과, 칼럼 종류에 따라 순수한 PLD또는 35 kDa의 단백질 불순물만을 포함하는 PLD 정제 분획을 보여 면역친화 칼럼의 높은 항원결합 특이성을 보여주었다. 그러나 수용액상에서 PLD 자체의 구조적 불안정성 때문에 정제 후 PLD의 특이적 활성 및 정제 수율은 낮았다. An immunoaffinity chromatography for the specific binding of Streptomyces somaliensis phospholipase D (PLD) that is considered as an industrially potential enzyme was developed. By using the protein structure prediction programs and the X-ray crystal structure of a Streptomyces PLD, 5 different epitopes with high antigenicity that are predicted to locate on the surface of the S. somaliensis PLD were selected and then synthesized for the preparation of antipeptide antibodies. Each purified rabbit IgG was coupled with NHS-activated Sepharose to prepare the immunoaffinity resins. After one-step purification of the culture concentrate on the antipeptide IgG-coupled Sepharose column, SDS-PAGE and the Western blot analysis of the purified samples showed that purification of PLD on the affinity columns was satisfactory, indicating that the peptide design using the structural information of Streptomyces PLDs was rational. However, the purified PLD in the solution aggregated rapidly, which resulted in poor specific activity and low purification yield.

        가정방문프로그램을 통한 범죄예방모델에 관한 연구

        본 연구는 복지방문프로그램에 가정폭력 예방이라는 관점을 접목하여 복지방문프로그램을 통한 가정폭력 예방이라는 융합 모델을 제시해 보고자 하였다. 이를 위하여 국내외 선행연구와 미국의 HFA와 NFP를 검토하였으며, 현재 가정방문프로그램을 운영하고 있는 기관의 담당자 8명을 상대로 심층면접을 진행하였다. 그 결과 미국 등의 사례와는 달리 우리나라에서는 복지방문프로그램과 가정폭력관리프로그램이 완전히 분리되어 있고 이를 융합한 프로그램이 없다는 점, 복지방문프로그램이 비교적 사전적이고 대상가정의 호응이 높은 반면, 가정폭력방문프로그램은 사후적이고 대상가정의 거부감이 높다는 점 등을 알 수 있었다. 따라서 선제적이고 대상가정의 거부감을 줄이기 위한 방안으로 복지방문프로그램에 가정폭력예방이라는 관점을 접목한 융합적 프로그램의 개발이 필요함을 제안하고, 이러한 융합모델의 내용을 대상가정 선정, 종사자 역할, 프로그램내용 영역으로 나누어 제시하였다. This study attempted to present a convergence model of domestic violence prevention through welfare visiting programs by grafting the perspective of domestic violence prevention into welfare visiting programs. For this purpose, precedent studies and HFA and NFP in the US were reviewed, and in-depth interviews were conducted with 8 people in charge of organizations operating home visiting programs. As a result, unlike cases in the United States, the welfare visiting program and domestic violence management program are completely separated, and there is no program that combines them. It was found that the domestic violence management program was ex-post and that the target families had a high sense of rejection. Therefore, to prevent in advance and to reduce the objection of target families, it is suggested that the development of a convergence program that combines the perspective of domestic violence prevention with the welfare visiting program is necessary. We present the contents of this convergence model in area of target family selection, worker roles, and program contents.

        소양인(少陽人) 비만방(肥滿方)이 생쥐의 비만 억제에 미치는 영향

        Objectives: In order to investigate the effects of So-yangin Biman-bang(here in after referred to BM) on the obese gene and obese inhibitory, C57BL/6 mice were distinctively induced by high fat diet. Methods :C57BL/6 mice were divided into four groups(normal, high fat diet with control, high fat diet with Reductil(here in after referred to RDT), high fat diet with BM extract) and have been fed for 8 weeks. Through observing the process, the weight change of the adipocytes in body and liver, total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, glucose, leptin change in the serum, the expression of β3AR and leptin gene in 3T3-L1 cell and primary adipocyte cell. Results :The following results have been found in this study. 1. Losing weight have significantly been shown. 2. The weight change of the body adipocyte and liver have been decreased but no significance has been found. 3. With close observation on the change of blood, HDL-cholesterol has positively been increased in numbers but the level of glucose, LDL-cholesterol, triglyceride have distinctively been decreased. 4. The manifestation of β3AR in 3T3-L1 cell and primary adipocyte cell have shown a definite increase in numbers but the manifestation of leptin has shown certain decrease in its amount. Conclusions :As results of this, So-yangin Biman-bang is thought to be effective in metabolic syndrom and lipid metabolism disorder that can possibly be occurred by obesity control and treatment. The clinical research will be needed in order to apply to So-yangin constitution in the future.

        포유동물의 신유두 및 신우상피에 대한 비교형태학적 연구

        Anatomical features of the renal papilla and pelvis and ultrastructures of the epithelium covering these areas in four species of mammals were studied by means of light, scanning and transmission electron microscopy. In terms of the morphology of mammalian kidney types distinguished by Sperber(1944), Pfeiffer(1968) and Schmidt-Nielsen(1977), the kidneys of animal species used in this experiment were; 1) the mouse kidney with the fornix between a long conical papilla and the funnel-shaped pelvis, 2) the guinea pig kidney with the peripelvic column and pelvic pouch between a short conical papilla and the funnel-shaped pelvis, 3) the dog kidney with the peripelvic column and pelvic pouch between the crest-shaped papilla and the funnel-shaped pelvis, and 4) the cattle kidney which is divided into multiple renculi with minor and major calyces and pelvis. The renal papilla was lined with the simple or pseudostratified columnar epithelium which covered the inner zone of the renal medulla. The epithelial cells with numerous short microvilli on the surface contained a few organelles. In the mouse, the fornix was lined with one to two cell-layered cuboidal epithelium which covered the outer zone of the renal medulla and a part of the cortex. The epithelial cells of the fornix with numerous short microvilli or microridges on the surface had well-developed organelles. In the guinea pig, the peripelvic column was lined with the simple cuboidal or low columnar epithelium which covered the outer zone of the renal medulla. The epithelial cells with numerous short microvilli on the surface contained well-developed organelles. The pelvic pouch was lined with the pseudostratified columnar epithelium which was composed of four kinds of cells; the secretory cell with small electron-dense granules (310 nm), the secretory cell with large granules (720 nm) showing various electron densities, the mitochondria-rich cell with a single cilium, and the basal cell. Pelves of the mouse and guinea pig, peripelvic column, pelvic pouch and pelvis of the dog, and minor and major calyces and pelvis of the cattle were lined with the transitional epithelium. The fusiform vesicles in the superficial cells of the epithelium were highly developed in the dog, relatively well developed in the mouse and guinea pig, and poorly developed in the cattle. From the above findings, it is suggested that the transport of solutes and water of the urine in the pelvic cavity can take place through the epithelia covering the renal papilla and fornix of the mouse, papilla and peripelvic column of the guinea pig, and papillae of the dog and the cattle. And specialized cell types in the epithelium of the guinea pig pelvic pouch, two kinds of secretory cells and mitochondria-rich cell with a single cilium, could have peculiar functions in the renal pelvis, respectively.

        만성위염 환자의 유문부 Gastrin 세포의 분포 및 Gastrin 농도

        N/A This study was conducted to investigate the influence of chronic gastritis on the distribution of gastrin (G)-cells and gastrin levels in antral mucosa and blood in Korean patients. Fifty-one subjects were divided into three groups according to histological findings of biopsied specimens by endoscopy: normal subjects, patients with chronic superficial gastritis and patients with chronic atrophic gastritis. The G-cells in the antral mucosa were stained by the immunohistochemical method with gastrin antiserum and the number of G-cell per unit area (1 mm breadth of a 5 ㎛-thick sectioned mucosa) was counted. Gastrin levels in the antral mucosal tissue and plasma were measured by means of radioimmunoassay. The results were as follows: 1) The number of G-cells and the antral tissue gastrin level in normal subjects were 106±36.6 and 10.6±6.2㎍/g, respectively. Plasma gastrin concentrations of the normal subjects in fasting and postprandial states, 20 min and 40 min after ingestion of a meat meal, were 63.1±24.5, 141.8±73.8 and 126.0±62.5 pg/ml, respectively. 2) In patients with chronic superficial gastritis, fasting and postprandial plasma gastrin concentrations increased significantly compared with those of the normal subjects, while the number of the G-cells and antral tissue gastrin were not significantly different from the normal values. 3) In patients with chronic atrophic gastritis, the number of G-cells and antral tissue gastrin levels tended to decrease, but fasting and postprandial plasma gastrin concentrations tended to increase compared with the normal values. 4) No G-cells were observed in the region with intestinal metaplasia. The above results indicate that patients with chronic gastritis showed a marked increase in plasma gastrin concentration while the number of antral G-cells and antral tissue gastrin level decreased.

        베타세포 손상에 대한 오배자 추출물의 세포보호작용

        오배자의 수용성 추출물은 alpha-glucosidase의 활성억제를 통해 장관에서의 탄수화물 분해를 방해하여 포도당흡수를 억제 함으로서 식후 혈당농도 (postprandial blood glucose)를 낮추는 효과가 있음이 보고된 바 있다(Shim et al., 2003). 이에 따라 본 연구에서는 오배자 추출물이 베타세포에 대한 보호작용을 함으로 항 당뇨효과를 나타낼 수 있는지를 관찰하고자 하였다. 이를 위하여 흰쥐 (Sprague-Dawley rat)에 오배자 추출물을 지속적으로 투여하는 동안 streptozotocin (STZ)으로 당 뇨병을 유도한 후 이자조직과 혈당의 변화를 관찰하였다. STZ에 의한 당뇨유발은 신생쥐에 STZ을 투여하는 모델 (신생쥐 /STZ)과 osmotic pump에 의해 일정량의 STZ이 지속적으로 투여되는 방법을 이용하였다. 신생쥐/STZ 모델에서 오배자를 투여하지 않은 대조군에서는 외분비 선포조직의 일부 소엽에서 완연한 염증반응과 조직퇴화현상이 관찰되었으며 심한 림 프세포의 침윤이 관찰되었다. 특히 이러한 염증소엽의 이자섬 (islet)들은 매우 작아져 있거나 몇 개의 세포무리로 구성된 경우도 있었다. 특히 염증이 없는 이자소엽에 존재하는 일부 이자섬들에서도 림프구침윤에 의한 인슐린염증현상을 비롯 하여 베타세포의 손상에 의한 조직변형이 나타났다. 이에 비하여 오배자를 투여한 동물의 이자조직은 전술한 바와 같은 염증성 이자소엽은 거의 관찰되지 않았으며, 이자섬에서도 염증반응이 발견되지 않았다. 이때, 대부분 이자섬들이 중심부 베타세포-주변부 알파세포의 세포배열과 정상 크기를 유지하고 있었다. Osmotic pump를 실험동물의 피하에 이식하여 STZ (15 mg/kg/day)을 48시간 동안 일정한 농도로 투여한 당뇨병모델에서는 오배자를 투여하지 않은 경우 대부분 동물들 이 고혈당 (¤200 mg/dl)을 나타냈다. 이들의 이자의 조직소견은 대체로 신생쥐/STZ모델과 유사 하였으나 일부 이자섬 조 직들에서 나타나는 인슐린염증의 정도가 더 심한 것이 많았으며, 알파세포의 이상증식에 의한 소위 ‘변형이자섬(remodeled islet)’ 또는 ‘거대신생이자 섬 (giant neogenic islet)’이 관찰되었다. 이 모델에서 오배자를 투여한 실험동물의 이자조직에서는 변형된 이자섬 및 거대신생이자 섬의 출현이 드물었으며, 고혈당증이 지연되어 나타났다. 이와 같은 결과들은 오배자 추 출물이 식후혈당을 조절함으로 베타세포에 대한 당독성 (glucotoxicity)을 낮출 뿐 아니라 항염증작용에 의한 베타세포보호 작용이 있음을 시사하는 것이다. We have previously reported that aqueous extract of gall from Rhus chinensis, known as “Obaeja”, inhibited rat intestinal alpha-glucosidase and suppressed postprandial hyperglycemia by delaying digestion and absorption of intestinal carbohydrate (Shim et al., 2003). This led us to speculate that obaeja could be involved in ameliorating beta-cell injury by lowering glucotoxicity. In the present study, we thus examined the protective effect of obaeja on pancreatic beta-cell damage along with its anti-diabetic effect in streptozotocin-induced animal models. Streptozotocin was administered to rat pups (neonate/STZ model), or to adult rats with a lower dose using osmotic pump (osmotic pump/STZ model) for inducing beta cell death and diabetes. Obaeja was given to those rat pups after weaning in neonate/STZ model, or 2 weeks before subcutaneous implantation of osmotic pump to rats of the other latter model. In the diabetic control rats of the neonate/STZ model, which were not fed with obaeja, some pancreatic islets demonstrated a destruction of beta cell mass with insulitis 2 weeks after weaning, while some larger and irregular islets were formed by proliferation of alpha cells. In particular, we found some pancreatic lobules showing a severe inflammation and degeneration of islet and acinar tissues in this model. Islets in these inflammatory lobules were smaller in size with only few cells. In contrast, any inflammatory responses and insulitis were not observed in pancreas of the rats fed obaeja in this model. The islets in those rats maintained their normal profiles and islet cell population. Such anti-cytotoxic effect was also monitored in the diabetic rats of osmotic pump/STZ model. Especially, occurrence of hyperglycemia in the obaeja fed rats was delayed by 25~30 days than that of diabetic control rats in this model. Taken together, these results imply that regulation of postprandial blood glucose level by obaeja feeding may ameliorate a secondary injury caused by glucotoxicity.
