        전기충격과 화학적 돌연변이원 N-Methyl-N'-Nitro-N-Nitrosoguanidine의 병행처리에 의한 방선균과 효모의 돌연변이 유발

        방선균(Streptomyces) 포자와 효모(Saccharomyces) 반수체 및 배수체 세포를 대상으로 전기충격(AC 38V/1.3 cm)이 화학적 돌연변이원인 N-methyl-N'-nitro-N-nitrosoguanidine(NTG)의 세포치사효과와 돌연변이 유발에 미치는 영향을 조사하였다. 방선균의 경우 전기충격 단독 처리로는 180분 처리하였을 때 생존율이 100%이었으나 960분 처리한 경우에는 모두 사멸하였다. 전기충격과 NTG처리를 병행한 바 방성균의 경우 180분 처리시, NTG 단독 처리시보다 생존율이 72%에서 48%로 감소되었고, 반수체 효모의 경우 40분 처리시 8%에서 3%로, 배수체 효모의 경우 25%에서 10%로 각각 감소되었다. 전기충격과 NTG에 의한 영양요구 돌연변이 형성율에 있어서는 전기충격이 NTG에 의한 돌연변이율을 120분 처리 후 1.8%에서, 13.6%로, 반수체효모의 경우 40분 처리후 2.4%에서 4.8%로 각각 증가시켰다. The effect of electric shock (AC 38 Vll.3 cm) on the lethal effect and induction rate of mutation of chemical mutagen N-methyl-N'-nitro-N-nitrosoguanidine (NTG) was examined by using the spores of Streptomyces and the cells of haploid and polyploid yeast strains of Saccharomyccrs. Spores of Sireptomyces were all alive after 180 min of electric shock, hut all dead after 960 min-treatment. When the spores of Streptomyces or the cells of haploid and polyploid yeast were treated with electric shock and NTG, the electric shock increased the lethal effect of NTG; the survival rate of Streptomyces dropped from 72 to 48% after 180 min-treatment and those of haploidand polyploid-yeast decreased from 8 to 3% and 25 to lo%, respectively, after 40 min-treatment. The electric shock also increased mutation rates of Streptomyces and haploid yeast. from 1.8 to 13.6%' after 120 min-treatment and from 2.4 to 4.8% after 40 min-treatment, respectively.

        산업용 효모 Hybrid의 선별을 위한 우성선별표지로서의 Aureobasidin A 내성유전자의 이용

        교배와 원형질체 융합을 통한 배수체인 야생형 산업균주의 개발을 위하여, hybrid의 선별표지로서 우성의 선별표지인 aureobasidin A 내성이 사용될 수 있는 지를 알아보고자 하였다. 선별배지에서 aureobasidin A의 최적농도는 야생형 균주인 경우 SD와 YPD 배지에서는 0.5 ㎍/mL 이상이었고, SG와 YPG에서는 0.2-0.3 ㎍/mL 이었다. 한편 호흡결여돌 연변이주는 야생형 균주보다 훨씬 높은 농도의 aureobasidin A에도 내성이 있음을 나타내었다. 우리는 K114/YIP균주의 전분분해 능력이 배수체 야생형 산업 균주에 전달 될 수 있는지를 이 방법을 통하여 관찰하였다. 반수체 영양요구성 균주 K114/YIP에 aureobasidin에 대한 내성을 부여하는 pAUR112가 도입된 균주와 야생형 균주 KL 혹은 C6와의 rare-mating 후 aureobasidin A 함유 배지에서 성장한 hybrid를 분리할 수 있었다. Hybrid는 전분분해 능력을 함유하고 있었을 뿐 아니라 두 양친의 특성을 동시에 지녔으며, 전자현미경 관찰 결과에서도 hybrid는 양친주의 특성을 모두 갖는 것으로 나타났다. For the strain improvement of the industrial polyploid yeast strain through hybridization and protoplast fusion, a dominant selection marker other than a recessive marker such as the auxotrophic marker was required for the selection of the resulting hybrids. In the present investigation, the aureobasidin A resistant gene was tested in relation to whether it can be used as the dominant selectable marker for the isolation of hybrids of the yeast Saccharomyces. The plasmid pAUR112, carrying the gene responsible for resistance to aureobasidin A, was introduced into the haploid yeast strain K114/YIp. From the rare-mating between polyploid C6 and haploid K114/YIp carrying pAUR112, many hybrids were obtained from the agar medium containing 0.5 μg/ml of aureobasidin A. The hybrids exhibited characteristics derived from both of the parental strains; and the cell sizes of the hybrids were larger than those of the parental strains. These results showed that the aureobasidin A resistant gene could be successfully used as the dominant selectable marker for the isolation of yeast hybrids resulting from rare-mating.

        We characterize the behavior of newly isolated antibiotics suppression mutant. Abs721 mutant sporulated well but could not produce any pigment on R_2YE as well as minimal medium. The genetic locus of abs721 was determined near to cysA allele. The locus is similar to absA. Compare to absA mutant, abs721 mutant sporulated less and growth on minimal plate is inhibited little bit.

        The author isolated mutant candidate which mapped around 8 o'clock region. It was located clockwise to uraAl, which is very simmilar to bldG location. This B155 mutant did mot develop any aerial mycelium at all on glucose supplied medium, however, it developed some aerial mycelium on the maltose supplied medium.

        Genetic mapping technique was applied to new two bald mutants. The location of the one of the mutant seems to be near bldA, and that of the other mutant might be upper right portion of the circle. The reason why many bld mutants map to bldA region was discussed, as well as possible application of genetic mapping technique to other streptomyces spp. than S.coelicolor.

Since bldA was mapped counterclockwise to cysA, the candidate which mapped counterclockwise to cysA was transduced with phage containing wild type bldA gene clone. The candidates might be the alleles of bldA, because they were complemented by bldA clone.

        DWe made a library of J1501 chromosomal DNA. Then transformed the abs8752(antibiotics non-producer) with the library. Several colonies were found which restores the actinorhodin & undecylprodigiosin production. The isolated clone Pl-α8 also restores the methylenomycin producing ability.

There is a good chance that p1-α6 includes a component of pleiotrophic regulatory pathway.
