        소풍산물추출물이 DNCB로 유발된 접촉성피부염에 미치는 영향

        Sopung-san(SPS; prescription No. DO27), a herbal medicine prescribed by oriental hospital of Daegu fanny University, has been used as cure for pruritus and psoriasis. To evaluate anti-inflammatory effect of Sopung-san, we treated Sopung-san extract in animal model system induced contact hypersensitivity. Contact hypersensitivity, a local inflammatory response of the skin, was induced by 1% DNCB on the right ear of BALB/C mouse. Sopung-san was prepared as water extract and administrated everyday for 2 weeks per oral. A right ear of mouse was potently swelled by 1% of DNCB treatment, but a mouse ear thickness was significantly reduced by Sopung-san after 2 weeks. treatment. Sopung-san reduced IgG in serum obtained from blood of 1 % DNCB-treated mouse. IgE in serum was not changed by DNCB and/or Sopung-san treatment. $IL-1{\beta}$ and IL-6 from ear tissue were significantly increased by DNCB, Dut were not reduced by Sopung-san. From these results, anti-inflammatory effect of Sopung-san, especially reduction of ear swelling, might be partly due to reduction of IgG in serum.

        Effects of Isatidis Radix and it's Active Component, Tryptanthrin on the Production of Inflammatory Mediators in Lipopolysaccharide-activated Raw264.7 Cells

        Objectives : 판람근(板藍根)은 십자화과에 속하는 대청(大靑) 또는 숭남의 근(根)을 건조한 것이다. 본 연구는 판람근(板藍根)이 청열해독(淸熱解毒)함에 근거하여, LPS로 활성화된 Raw264.7 cell에서 판람근(板藍根)과 그 성분중의 하나인 tryptanthrin이 염증매개물질에 미치는 효과를 살펴보고자 하였다. Methods : 세포생존율은 MTT, nitric oxide (NO)는 Griess reagent를 사용하여 측정하였으며, 각 단백질의 발현량은 Western blot 방법을 사용하였으며, cytokine 및 cyclooxygenase-2 (COX-2)는 ELISA방법을 사용하여 측정하였다. Results : LPS는 NO 및 prostaglandin E2 (PGE2)를 유의하게 상승시켰으며, 판람근(板藍根)추출물 (IRE) 및 tryptanthrin 은 이들을 유의하게 억제하였다. 그러나 판람근(板藍根)의 또 다른 성분인 indigo는 유의한 결과를 나타내지 못하였다. IRE와 tryptanthrin은 inhibitory kappa B alpha의 인산화를 억제하여, nuclear factor-${\kappa}$B (NF-${\kappa}$B)의 핵으로의 전위(轉位)를 억제하여, iNOS 및 cytokine을 억제하였다. IRE와 tryptanthrin의 PGE2 억제는, COX-2의 발현억제에서가 아니라, COX-2의 활성을 억제함에서 기인하였다. Conclusion : 이러한 결과는 판람근(板藍根)이 NF-${\kappa}$B pathway를 경유하여 iNOS의 발현 및 COX-2의 활성을 억제함을 나타내며, 이러한 판람근(板藍根)의 항염증효능은 일부 tryptanthrin의 작용에서 기인함을 시사한다.

        산국 잎과 줄기의 유효성분 분리 및 특성 연구

        Chrysanthemum boreale Makino (C. boreale) is widely distributed in Asian countries, and has traditionally been used to treat various inflammatory diseases including bronchitis. In this study, we aimed to isolate biologically active compounds from leaves and stems of C. boreale. Chemical components were purified by column chromatograpy and recyclic HPLC, and characterized from their spectral data (IR, MS, NMR). Biological activity experiments were conducted for Farnesyl-protein transferase (FPTase) activity, apoptosis and nitirc oxide (NO) release. As a results, three sesquiterpene lactones were isolated. Compound 1 (4-methoxy-8-O-acetyl-10-hydroxy-2,11(13)-guaiadiene-12,6-olide) showed strong cytotoxic activities having an average growth inhibition of 50% (GI<sub>50</sub>) value of 1.89 ㎍/㎖against human colon adenocarcinoma cells. Compound 1 also showed a low half maximal inhibitory concentration (IC<sub>50</sub>) value of 10 ㎍/㎖ for NO release. In the caspase 3 activity, compound 1 and compound 2 (8-O-(2-carbonyl-2-butyl)-3,10-dihydroxy-4,11(13) -guaiadiene-12,6-olide) exhibited 94% and 90% apoptosis inhibition activity, respectively. Compound 3 (4,8-O-diacetyl -10-hydroxy-2(3),11(13)-guaiadiene-12,6-olide) showed a strong inhibitory effect on FPTase activity with 90% inhibitory activity at a concentration of 100 ㎍/㎖. These results clearly show the presence of lactone compounds in the leaves and stems, which may partially contribute to the pharmacological activity of C. boreale.

        HaCaT 세포의 산화 스트레스로 인한 세포자멸사에서 정향의 보호효과

        Objective : Caryophylli Flos has been used in Korean medicine to relieve vomiting and pains caused by chills that make fluid circulation difficult. This study was designed to investigate the protective effect of ethanol extract of Caryophylli Flos (CF) in hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>)-induced apoptotic cell death in human keratinocyte HaCaT cells. Methods : CF was prepared by extracting 200 g of Caryophylli Flos in 2 L of ethanol for 48 h. Cell viability was measured by MTT assay, and the protein expression was monitored by Western blot analysis. Apoptosis was determined by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. Reactive oxygen species (ROS) was measured using fluorescent dye, and reduced glutathione (GSH) was determined with a colorimetric commercial kit. Results : CF protected HaCaT cells from cell death caused by oxidative stress after H<sub>2</sub>O<sub>2</sub> treatment. H<sub>2</sub>O<sub>2</sub> amplified generation of ROS and induced depletion of GSH, whereas these changes in ROS and GSH were inhibited by GF treatment. In addition, H<sub>2</sub>O<sub>2</sub> resulted in apoptosis as assessed by TUNEL assay and the expression of apoptosis regulator proteins. However, cells treated with CF showed a decrease in TUNEL-positive cells and restored the reduced expression of procaspase-9, -3 and PARP. Conclusion : This study showed cytoprotective effects of CF by anti-apoptotic activity while exerting antioxidative activity in H<sub>2</sub>O<sub>2</sub>-treated HaCaT cells. These results suggest that CF could be beneficial in skin damage caused by oxidative stress.

        NF-κB 저해를 통한 브로콜리 잎 추출물의 PGE<sub>2</sub> 저해효과

        Objective : Broccoli is edible green plant that has a wide variety of health benefits including cancer prevention and cholesterol reduction. However, leaves of broccoli are not eaten and are mostly left as waste. This study was conducted to evaluate the effects of the broccoli leaf extract (BLE) on prostaglandin E<sub>2</sub> (PGE<sub>2</sub>) production related to nuclear factor kappa B (NF-κB) signaling in lipopolysaccharide (LPS)-activated macrophages. Methods : BLE was prepared by extracting dried leaf with ethanol. Cell viability was determined by 3-(4,5- Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. PGE<sub>2</sub> and inflammatory cytokines were detected by enzyme-linked immunosorbent assay (ELISA). Expression level of each protein was monitored by Western blot analysis. Results : In LPS-activated Raw264.7 cells, PGE<sub>2</sub> release into culture medium was dramatically enhanced compared to control cells. However, increased PGE<sub>2</sub> was attenuated dose-dependently by treatment with BLE. Inhibition of PGE<sub>2</sub> production by BLE was due to the suppression of cyclooxygenase-2 (COX-2) expression determined by Western blot analysis. BLE also inhibited the production of inflammatory cytokines such as interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α). Inhibition at PGE<sub>2</sub> and cytokine was mediated from inhibition of nuclear translocation of NF-κB due to the repression of inhibitory kappa B alpha (IκBα) phosphorylation and degradation. Conclusion : This study showed that BLE exerted inhibitory activities against PGE<sub>2</sub>, which is critical for the initiation and resolution of inflammatory responses, and that inhibition of PGE<sub>2</sub> was mediated by suppression of NF-κB signaling. These results suggest that the waste broccoli leaves could be used for controlling inflammation.

      • tert-butyl hydroperoxide로 유도된 위 상피세포 독성에 대한 치자의 수치 효과

        Objectives : Gardeniae Fructus is the dried mature fruit of Gardenia jasminoides Ellis that has been used as a Korean medicine for treating digestive disorders, liver disease and various inflammatory disorders. In this study, we aimed to evaluate the processing effect on protective activity of Gardeniae Fructus in tBHP-induced AGS gastric epithelial cells. Methods : The processed Gardeniae Fructus was made by adding ginger juice and alcohol to the Gardeniae Fructus or by roasting Gardeniae Fructus at 150℃ and 250℃ for 5, 10, and 20 minutes. The antioxidant activity was examined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,20-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS) assays. Cytoprotective activity was examined using MTT assay after treating cells with GFE and tert-butyl hydroperoxide (tBHP). To determine acid neutralizing activity, the excess HCl was immediately titrated with 0.1 N NaOH (end point pH 3.5). Results : The antioxidative capacity of processed Gardeniae Fructus was higher than that of untreated Gardeniae Fructus. Cytoprotective and acid neutralizing activities were also higher in the processed Gardeniae Fructus than in the untreated Gardeniae Fructus. Compared to 57.8% of untreated Gardeniae Fructus, the processed Gardeniae Fructus containing 40% ginger juice and 10% rectified alcohol showed 64.3% and 69.3% of cell viability, respectively. Gardeniae Fructus, roasted at 150 ℃ for 5 minutes, had the highest cell protection effect with a cell viability of 77%. Conclusions : These results show that proper processing conditions of Gardeniae Fructus may be beneficial in improving the cell protection effect of Gardeniae Fructus in tBHP-induced gastric epithelial cells.

        현호색(玄胡索)과 초초현호색(醋炒玄胡索)의 여드름균에 대한 항균효과 비교

        Objectives : Corydalis tuber has been used for promoting blood circulation and for relieving pain in Oriental medicine. In the present study, we focused on the antimicrobial activity of Corydalis tuber and compared its antimicrobial activity with the processed Corydalis tuber. Methods : Processing of Corydalis tuber was accomplished by immersing in 5% of acetic acid for 12 h and then by roasting at $250^{\circ}C$ for indicated time periods(0-30 min). Minimum inhibitory concentration(MIC) and the zone of growth inhibition were determined against Propionibacterium acnes(P. acnes). Results : The methanolic extracts of Corydalis tuber showed potent antimicrobial effect(MIC 62.5 ${\mu}g/ml$). Its alkaloidal component, dehydrocorydaline, also exhibited antibacterial activity(MIC 25.0 ${\mu}g/ml$). After processing of Corydalis tuber, its inhibitory effect on the growth of P. acnes was significantly enhanced compared with that of unprocessed Corydalis tuber. Furthermore, elevated content of dehydrocorydaline was found in the processed than the unprocessed Corydalis tuber. However, the different roasting minutes effected on antimicrobial activity. The best roasting time of Corydalis tuber was 10 min, while roasting for the time above 15 min resulted in diminishing antimicrobial activity. Thus, it was concluded that the standardized processing condition of Corydalis tuber should be established to obtain enhanced antimicrobial(P. acnes) activity. Conclusion : For antimicrobial effect against P. acnes, the best processing condition of Corydalis tuber is immersing in 5% of acetic acid for 12 h and by roasting at $250^{\circ}C$ for 10 min.

        황련(黃連)이 구강암 세포에서의 세포자멸사에 미치는 영향

        Objectives : The aim of this study was conducted that CRE (Coptidis Rhizoma Extract) induces apoptosis in YD-10B cells, human oral squamous carcinoma cell line. Methods : In this study, YD-10B cells were exposed to CRE (0.03-0.30 mg/ml), for 6-24 hours. We measured the effects of CRE on the changes of cell viability and cell membrane, TUNEL assay of CRE-treated YD-10B cell. Results : In this study, CRE caused a decrease of viability in YD-10B cells, human oral squamous carcinoma cell line. When YD-10B cells were treated with CRE, cells showed dose-dependent manner apoptotic cell death. Conclusions : These results suggest that CRE may be potential therapeutic approach in the clinical management of oral squamous cell carcinoma.

        Nuclear Factor kappa B 억제를 통한 인진추출물의 inducible Nitric Oxide synthase 및 Cytokine 억제효과

        Herba Artemisiae Capillaris is the dried bud of Artemisia capillaris Thunb, which has been used for expelling heat to loosen the bowels and normalizing gallbladder function to cure jaundice in traditional oriental medicines. In the present study, we evaluated the anti-inflammatory effects of the aqueous extracts of Herba Artemisiae Capillaris (HAC) in LPS-activated Raw 264.7 cells. Cells were treated with $1\;{\mu}g/ml$ of LPS 1 h before adding HAC extract. Cell viability was determined by MTT assay, and the relative level of NO was measured with Griess reagent. TNF-$\alpha$, IL-$1{\beta}$, and IL-6 cytokines were detected by ELISA. During the entire experimental period, all three doses of HAC extract (0.03, 0.10 and 0.30 mg/ml) had no significant cytotoxicity. LPS-activated cells showed increased NO levels and iNOS expressions compared to control. However, these increases were dramatically attenuated by treatment with HAC extract. Moreover, the inhibitory effects of HAC extract occurred in a dose-dependent manner. In addition, HAC extract reduced the translocation of $NF{\kappa}B$ into nuclear. HAC reduced production of IL-$1{\beta}$ and IL-6 by LPS, although it had no effects on TNF-$\alpha$. These results demonstrate that liquiritigenin exerts anti-inflammatory effects, which results from the inhibition of $NF{\kappa}B$ activation in macrophages, thereby decreasing production of iNOS and proinflammatory cytokines. Taken together, these results indicate that the aqueous extracts of Herba Artemisiae Capillaris warrant further development as an anti-inflammatory agent for the treatment of gram-negative bacterial infections.
