        Stevia 절편체 종류의 배양에 따른 식물체 재분화

        This study was conducted to determine the optimal concentrations of plant hormones (2,4-D, picloram, dicamba, NAA, kinetin) and the suitable explants among seeds, hypocotyl, and cotyledons on calls formation and plant regeneration of stevia(Stevia rebaudiana Bertoni). The frequency of cellus formation was higher in the young leaf-explants then the older ones, and in the seeds then the hypocotyls and cotyledons on MS medium with 1mg/L 2,4-D. After transfer of seed-derived stevia callus producing embryogenic callus on plant-regeneration medium, the frequency of plant regeneration from callus was 23.8% in MS medium with 1mg/L NAA and 3mg/L kinetin.

        水稻 葉組織 燐脂質의 主要 脂肪酸 遺傳分析

        A seven-parent diallel cross of rice was analyzed for fatty acid compsition of phospholipid from leaves. The analysis of variance revealed that differences among parents and F₁ hybrids were highly significant for fatty acid. The variances of parent vs. F₁ hybrids were highly significant for all characters except for linoleic acid. Both general and specific combining ability were highly significant and non-additive genetic variance predominated for fatty acid. In the F₁ generation, over dominance was observed for all fatty acids. Direction of dominance was positive for unsaturated fatty acid, whereas it was negetive for saturated fatty acid. The number of effective gene groups was estimated as one for palmitic acid and stearic acid, and two for oleic acid and linolenic acid. Broad sense heritability was fairly high but narrow sense heritability was low.

        베타아밀로이드로 유도된 신경소교세포 사멸에 대한 루테올린의 보호효과 연구

        Objectives : The purpose of this study is to investigate neuroprotective effects and molecular mechanisms of luteolin against β-amyloid (Aβ25-35)-induced oxidative cell death in BV-2 cells. Methods : The protective effects of luteolin against Aβ25-35-induced cytotoxicity and apoptotic cell death were determined by MTT dye reduction assay and TUNEL staining, respectively. The apoptotic cell death was further analyzed by measuring mitochondrial transmembrane potential and expression of pro- and/or anti-apoptotic proteins. To elucidate the molecular mechanisms underlying the protective effects of luteolin, intracellular accumulation of reactive oxygen species, oxidative damages, and expression of antioxidant enzymes were examined. Results : Luteolin pretreatment effectively attenuated Aβ25-35-induced apoptotic cell death indices such as DNA fragmentation, dissipation of mitochondrial transmembrane potential, increased Bax/Bcl-2 ratio, and activation of c-Jun N-terminal kinase and caspase-3 in BV-2 cells. Furthermore, Aβ25-35-induced intracellular formation of reactive oxygen species and subsequent oxidative damages such as lipid peroxidation and depletion of endogenous antioxidant glutathione were suppressed by luteolin treatment. The neuroprotective effects of luteolin might be mediated by up-regulation of cellular antioxidant defense system via up-regulation of γ -glutamylcysteine ligase, a rate-limiting enzyme in the glutathione biosynthesis and superoxide dismutase, an enzyme involved in dismutation of superoxide anion into oxygen and hydrogen peroxide. Conclusions : These findings suggest that luteolin has a potential to protect against Aβ25-35-induced neuronal cell death and damages thereby exhibiting therapeutic utilization for the prevention and/or treatment of Alzheimer`s disease.

        문화생활을 즐기는 현대인의 증가와 함께 온라인 티켓 거래가 활발하게 이뤄지고 있다. 문화생활에 대한 수요가 증가하고, 온라인을 통한 티켓 거래가 활발해지는 만큼 불법 거래 사례와 피해 또한 커지고 있다. 이러한 불법 거래 방지를 위해 본 논문에서는 기존 모바일 티켓팅 시스템의 QR코드에 티켓 고유식별번호와 난수를 결합하고, 사용자의 지문 인식과 사용 시간 제한 등의 기능을 추가한 불법거래 방지 티켓팅 시스템을 제안한다.

        감초분말 첨가 파운드 케이크의 품질 및 항산화 활성

        감초분말 첨가량을 0-8%로 달리하여 파운드 케이크를 제조한 후 물리·화학적 품질, 항산화 특성 및 소비자 기호도를 비교하였다. 감초분말을 첨가한 파운드 케이크의 pH와 수분함량은 각각 6.76-6.84, 13.96-17.80% 범위에서 유의적으로 감소하였다(p<0.05). 케이크의 무게와 높이는 감소한 반면, 굽기 손실률은 증가하였다(p<0.05). 한편 명도(L*)는 감초분말의 첨가량이 증가할수록 유의적으로 감소하였고(p<0.05) 적색도(a*)는 유의적으로 증가하였다(p<0.05). 대조군 케이크의 경도는 0.46 kgf으로 유의적으로 가장 높은 값을 나타내었으며(p<0.05), 감초분말을 첨가량의 증가할수록 단계적으로 감소하여 8% 첨가군의 경도는 0.29kgf로 유의적으로 가장 낮았다(p<0.05). 항산화 특성을 나타내는 DPPH와 ABTS radical 소거능은 유의적 차이를 나타내며 증가하였고(p<0.05), 두 지표간 상관관계가 매우 높은 것으로 나타났다. 소비자 선호도 검사 결과, 4% 첨가군이 모든 평가항목에서 유의적으로 높게 평가되어 케이크의 관능품질과 건강 기능성 효과 등을 고려할 때 감초분말 4%를 첨가한 파운드 케이크가 가장 적절한 것으로 판단된다. We investigated the feasibility of incorporating licorice powder as a value-added food ingredient in pound cake, a model system of convenient food products. The pH and moisture content gradually decreased with increasing levels of added licorice powder (p<0.05). The weight and height of the cakes decreased, and the baking loss increased as higher amounts of licorice powder were included in the formulation (p<0.05). The lightness and hardness both decreased, whereas browning increased significantly (p<0.05) with the addition of licorice powder. In addition, we observed a significant increase (p<0.05) in 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities that correlated well with the licorice powder content. In consumer acceptance tests, the addition of 4% licorice powder had the most favorable effect on consumer preferences for all the attributes examined. Based on these observations, we recommend supplementing pound cakes with 4% (w/w) licorice powder to take advantage of its functional properties without sacrificing consumer acceptability.

        중산간지에서 냉수처리가 벼 품종의 생육과 수량에 미치는 영향

        본 연구는 내냉성 연구의 기초자료를 제공하고자 우리나라 벼 20개 품종을 대상으로 냉수를 수구, 중구, 배수구, 자연구로 구분 처리하여 벼의 생육 및 주요형질간의 상호관계를 검정한 바 그 결과를 요약하면 다음과 같다. 냉수관개가 수도 생육에 미치는 영향을 보면 냉수 처리시 자연구 대비 냉수 유입구인 수구, 중구, 배수구에서 출수지연, 간장단축, 이삭추출억제, 수장단축, 수당립수의 감소 및 임실율이 저하되었고 그 정도는 중생종이 조생종보다 더 크게 나타났으며 수수의 경우는 냉수 처리에 따른 차이가 없었다. 수량의 경우 수장과 수당립수, 임실율 등 sink의 크기가 감소하여 수량의 감수를 가져왔다. 간장과 이삭 추출도와의 관계를 보면 간장과 이삭 추출도와는 정의 상관관계를 나타내었다. 품종에 따른 수량과 타형질 간의 상관관계를 보면 간장과 고도의 정의 상관을 나타내었으며(r=0.6954∗∗), 임실비율과도 고도의 정의 상관을 나타내었다(r=0.5797∗∗). 또한 추출도와도 유의상관이 인정되었으며(r=0.5066∗) 출수기와는 부의 상관이 인정되었다(r=−0.4826∗). 적고정도는 조생종의 경우 유묘기에는 금오벼를 제외한 모든 품종이 1∼3의 범위에 있어 강한 편이었으나 분열기에는 3∼5정도로 다소 약해졌으며 중생종의 경우 유묘기에는 약한 경향이었고 분얼기에는 다소 강하게 나타났다. 수량구성요소를 살펴보면 주요품종이 모두 냉수처리 시 이삭당 영화수, 등숙비율, 천립중이수구>배수구>중구>자연구 순으로 감소하였고, 등숙비율의 경우 타형질에 비해 감소폭이 현저하였다. 수구로부터 거리에 따른 수량의 구성요소와 수량의 감소는 공시품종 모두 수구에서 수량이 가장 낮게 나타났으며 중구와 배수구로 갈수록 감소의 폭은 적은 경향이었다. 소백벼 와 오대벼 가 상미벼 와 조령벼 에 비해서 수량의 감소는 더 현저하게 나타나는 경향이었다. 냉수처리 시 중산간지 주요 품종의 현미품질은 수구에 가까울수록 청미 발생율이 증가하였으나, 사미 등의 경우 일정한 경향이 없었다. This study was conducted to obtain the basic information on rice cold damage and relationship to be connected with cold tolerance. The 20 rice varieties were screened to cold tolerance under low water temperature condition. The water gradient were controlled to 17∘C at the inlet and 21∘C at outlet of screening field. In the relationship between cold water irrigation and growth of rice varieties, effect on medium maturing cultivars were higher than early maturing cultivars. In relationship of shorting of culm length and panicle exsertion, culm length and panicle exsertion showed positive correlation. Yield of cultivars and culm length and percent of fertile grain showed positive correlation. However, heading date presented negative correlation. In the early maturing cultivars, except Kumobyeo , all cultivars range of discoloration value (1-3) was resistant. In the medium maturing cultivars, Donghaebyeo was resistant in both the seedling stage whereas Hwajinbyeo was resistant only in tiller stage. Cold water irrigation reduced spikelet number per panicle and percent of filled grain. Unhulled rice yield was increased according to water temperature gradient from inlet to outlet. There was varietal differences in head rice recovery by cold water treatment.

        베타아밀로이드로 유도된 신경세포 사멸과 기억력 손상에 대한 밀기울추출물의 보호효과

        Objectives : The aim of this study is to examine the neuroprotective effect of wheat bran extract (WBE) against β-amyloid (Aβ)-induced apoptotic cell death in SH-SY5Y human neuroblastoma cells and memory impairment in triple transgenic animal model`s of Alzheimer`s disease (3xTg AD mice). Methods : In SH-SY5Y cells, MTT assay and TUNEL staining were conducted to evaluate the protective effect of WBE against Aβ25-35-induced neurotoxicity and apoptosis. Alterations in mitochondrial transmembrane potential (MMP), expression of proapoptotic Bax and antiapoptotic Bcl-2 proteins, cleavage of PARP, and brain-derived neurotrophic factor (BDNF) levels were analyzed to elucidate the neuroprotective mechanism of WBE. To further investigate the memory enhancing effect of WBE, Morris water maze test was performed in 3xTg AD mice. Results : In SH-SY5Y cells, WBE protected against Aβ25-35.caused cytotoxicity and apoptosis as shown by the restoration of cell viability in MTT assay and inhibition of DNA fragmentation in TUNEL staining. Aβ25-35. induced apoptotic signals such as dissipation of MMP, decreased Bcl-2/Bax ratio, and cleavage of PARP were suppressed by WBE. Moreover, WBE up-regulated the protein levels of BDNF, which seemed to be mediated by activation of cAMP response element-binding protein (CREB). In 3xTg AD mice, oral administration of WBE attenuated learning and memory deficit as verified by reduced mean escape latency in water maze test. Conclusions : WBE protects neuronal cells from Aβ25-35.induced apoptotic cell death and restores learning and memory impairments in 3xTg AD mice. These findings suggest that WBE exhibit neuroprotective potential for the management of AD.

        RS4 형태의 옥수수 저항전분이 첨가된 쿠키의품질 특성

        인산화 가교방법에 의새서 RS4 옥수수전분을 제조하고 이를 밀가루의 5, 10, 20%로 대체하여 쿠키를 제조하고 품질 특성을 비교하였다. RS4 옥수수전분의 첨가량이 증가할수록 반죽 RS4는 낮아졋음 반죽의 밀도은 RS4 옥수수 전분 20% 첨가구에서 낮아지는 경향을 보였지만 대조구 대비 RS4 옥수수전분 20%의 경우 약간 작아지는 경향은 있었지만 유의적인 차이점은 없었다.(P<0.05). 쿠키의 수분함량은 RS4 옥수수전분의 첨가량이 늘어날수록 증가하는 경향을 나타내었으며 색도는 첨가량이 늘어날수록 L값이 높아졌으며 a, b값의 변화에는 유의차가 없었다.(P<0.05). 쿠키의 경도(hardness)및 부서짐성(fracturability)는 첨가량이 늘어날수록 유의적으로 낮아졌다(P<0.05). 관능평가 결과 쿠키의 색상은 20% 첨가구에서는 조금 높은 점수를 받았으나 유의차는 없었으며 풍미는 대조구나 RS4 옥수수전분을 5%, 10%, 20% 첨가한 구나 유의차가 없었다. 조직감이나 맛도 RS4 옥수수전분을 첨가 할수록 일부 높아지는 경향을 보였지만 유의차는 없었다.(P<0.05). 전체적인 기호도도 RS4 옥수수전분을 20% 첨가한 구가 3.7로서 대조구 3.2보다 약간 높아지는 경향을 보였으나 유의차는 없었다(P<0.05). 이상의 결과를 보면 쿠키의 품질이나 기호도에 영향을 주지 않고 RS4 옥수수전분을 밀가루 대비 20%까지 첨가하여 RS함량을 추가로 14.7% 상승시킨 쿠키를 제조할 수가 있었다. Effects of RS4 type resistant com starch on the quality characteristics of cookies were investigated by physicochemical, instrumental and sensory evaluation The resistant starch was made by cress-linking of com starch as following; corn starch sluny was annealed at pH 2.0 and 50℃ for 2 h followed by the cross-linking reaction. The cross-linking reaction was performed at sot for 12 h in the presence of 1.2%st.ds NaOH, 10%/st.ds sodium sulfate and 10%/st.ds of sodium trimetaphosphate (STMP)sodium tripolyphosphate (STPP) mixture, Dietary fiber content or the resistant starch was estimated to be 73.8% by the AOAC method. FOr quality characteristics, dough pH decreased with the increase of the resistant starch content and spreed factor decreased a little at 20%, of the resistant starch, The moisture content and L value of cookies increased with the increase of the resistant starch content. However; the hardness and fracturability decreased with the Increase of the resistant starch content In the sensory evaluation, no significant differences were observed between the two cookies with or without the resistant starch, The results of this study suggest that the RS4 type resistant com starch can be 1I good ingredient to increase dietary fiber content in cookies without changes of their qualities.

        고추 草長의 遺傳에 關한 硏究 Ⅰ. 雜種强勢와 形質相關

        This study was performed to obtain basic informations for breeding early maturing varieties in red pepper. Eight parent varieties and twenty eight diallel crosses between them were used as materials. Heterosis and heterobeltiosis for main stem length, branch length, plant height and main stem diameter, corelations between 4 plant height characters and corelations between plant height characters and days to first flower were investigated. From the analysis of variance, the 1% level significances were observed among parents and F₁s in main stem length, branch length, plant height and main stem diameter. The means of F₁s were significantly longer than those of parents in all plant height characters. Degree and direction of heterosis and heterobeltiosis varied greatly with different crosses and most of F₁ hybrids showed positive partial dominace or over dominance in all plant height characters. Average degree of dominance for main stem length and branch length were partial dominant and those for plant height and main stem diameter were almost complete dominant. In F₁ hybrids, positively significant corelations between main stem length, branch length, plant height, main stem diameter and days to first flower were observed except between main stem length and branch length and between branch length and days to first flower. Highly significant corelations between mid - parents and F₁ hybrids were observed in all plant height characters.
