        단삼 등 천연물의 항암작용

        The cellular growth inhibition of 20 natural products was screened using SRB (sulforhodamine B) assay against 4 human cancer cell lines(SNU-1, SNU-C₄, Hep3B, Kato III). Ethanol extracts of Salvia miltiorrhiza, Saussurea lappa and Chelidonium majus showed potent anticancer activity among them, and further, it was fractionated into methylene chloride, hexane and methanol. Methylene chloride and methanol fraction of Salvia radix showed significant inhibitory activity against 4 human cancer cell lines. The effect of Salvia miltiorrhiza on anticancer activity in vivo models was evaluated with methylene chloride fraction of Salvia miltiorrhiza. Life span of ICR mice implanted with sarcoma-180 was increased by 40-61% and BDF₁ mice implanted with L1210 was increased by 66-89% upon intraperitoneal administration with methylene chloride fraction of Salvia miltiorrhiza. Based on these result, we suggested that Salvia miltiorrhiza showed anticancer activity on the in vivo and in vitro models

        난소절제 흰쥐에서 Estrogen의 항비만 효과

        Obesity is a chronic disease that is increasing in prevalence and that poses a serious risk for the hypertension, osteoporosis, heart disease, diabetes mellitus and certains forms of cancer. This study was performed to develop of obesity animal model and to assess the pharmacological assay for the rats of 8 weeks or 4 days after ovariectomization treated with estradiol for 8 weeks on the body weight, fat weight and food intake. The body weight, fat weight and food intake increased in the ovariectomized rats. In the rat of 8 weeks after ovariectomization treated with estradiol (250mg/100g) 8 weeks, the body weight decreased significantly (p<0.05). In the rats of 4 days after ovariectomization treated with estradiol 8 weeks, the body weight decreased significantly (p<0.05). These results suggest that estrogen plays a role in regulation body weight response to food intake and fat weight.

        죽염의 약리작용 평가

        Bamboo salt has been used for the purpose of precaution and treatment of certain diseases including cancer. Therefore, present study was carried out to ascertain the effects of bamboo salt upon anti-cancer, anti-hypertensive, and anti-diabetic activities as well. To examine the anti-cancer activity of bamboo salt, ICR mice implanted with 1 X 10^6 cells of sarcoma 180 intraperitoneally had been treated daily with bamboo salt A, crude salt, and reagent-grade NaCl (0.2, 1.0, and 2.0 g/kg, p.o.) for 60 days using adriamycin (2 mg/kg) as a positive control. Neither survival rate nor body weight had been significantly influenced by all the treatments indicating that bamboo salt A did not exert the anti-cancer effect on ICR mice. Anti-hypertensive activity was examined in spontaneously hypertensive rats (SHR) which had been administered with bamboo salt A, crude salt, and reagent-grade NaCl (0.1, 0.5, and 1.0 % in drinking water) for 28 days using hydralazin (2 mg/kg) as a positive control. Blood pressure and heart rate were measured at 1, 3, and 4 weeks after the starting date. Significant anti-hypertensive activity was not observed in any treated group compared to the positive control group. In order to determine if bamboo salt had anti-diabetic activity, rats in which diabetes had been induced by streptozotocin (45 mg/kg, i.m.) were treated daily with bamboo salt A, crude salt, and reagent-grade NaCl (0.2, 1.0, and 2.0 g/kg, p.o.) for 28 days using insulin (50 U/kg, s.c.) as a positive control. Blood samples were taken and analyzed at 1, 2, and 4 weeks after the starting date. Bamboo salt did not cause any decreasing effect on the blood glucose levels. These results clearly demonstrated that bamboo salt A did not exert anti-cancer, anti-hypertensive, or anti-diabetic activities in the present experimental animals.

        천연물의 항염증작용에 관한 연구

        These studies were conducted to investigate the antiinflammatory effects and the mechanism of action of natural products. We used the methods of carrageenan induced foot edema , PAF (platelet activating factor) induced foot edema , inhibition test of vascular permeability, inhibition test of white blood cell migration, formation of granuloma and adjuvant induced arthritis to examine the antiinflammatory effects of Angelica gigas, Ledebouriella seseloides, Ginkgo biloba and Bamboo salt (Jukyom). The oral administration of the water extract of Angelica gigantis radix, the methanolic extract of Ginkgo folium and the aqueous solution of Bamboo salt showed antiinflammatory effect on carrageenan and PAF induced foot edema in SD rat at a dose of 1 g/kg. The same administration of methanolic extract of Ginkgo folium also inhibited the vascular permeability in mice. The aqueous solution of Bamboo salt inhibited the formation of granuloma in SD rats at a oral dose of 1 g/kg. Angelica gigantis radix seems to have antiinflammatory effect by inhibition of PAF.

        HepG2 2.2.15 세포주를 이용한 가자, 지유, 복분자, 대황의 B형 간염바이러스 증식 억제 효과

        This study was undertaken to test for antiviral activity of the aqueous extracts prepared from 4 medicinal plants of Korea (Terminalia chebula, Sanguisorba officinalia, Rubus coreanus, Rheum palmatum). Aqueous extracts were assayed for the inhibition of hepatitis B virus (HBV) replication by measurement of HBV DAN and surface antigen (HBsAg) levels in the extracellular medium of HepG2 2.2.15 cells. All extracts decreased the levels of extracellular HBV virion DNA at concentrations ranging from 64 to 128mcg/ml and inhibited the production of HBsAg dose-dependently. Among the 4 tested plants, Terminalia chebula exhibits the most prominent anti-HBV activities. Our findings suggest that these 4 medicinal plants may have potential to develop as specific anti-HBV drugs in the future.

        오수유가 N-nitro-L-arginine methyl ester로 유발한 고혈압흰쥐의 심혈관계에 미치는 영향

        The present study examined the effect of a methanol extract of Evodiae Fructus on the cardiovascular function in N-nitro-L-arginine methyl ester (NAME)-induced hypertensive Wistar rats after treatment over 6 weeks. In rats treated with NAME, blood pressure, weight of heart, aorta media thickness and media/lumen ratio significantly (p<0.05) increased, whereas coronary flow and heart rate of isolated heart significantly (p<0.05) decreased compared with control group at 6 weeks. In rats treated with NAME and Evodiae Fructus, blood pressure, aorta media thickness and media/lumen ratio signiflcantly(p<0.05) decreased compared with NAME treated group at 6 weeks. These results suggest that Evodiae Fructus is applicable to the treatment of hypertension and vascular hypertrophy.

        홍화씨의 경골골절치유에 미치는 영향

        We investigated the effect of safflower-seed on fracture healing of fracture model in rat. Fracture healing was evaluated by examining the degree of wound healing macroscopically, radiography, bone histomorphometry and biochemical examination. After 1,3,5,7 days, the wound healing was accelerate in safflower-seed diet group. Radiography does not reveal the difference in fracture healing between two groups. After 2 weeks, safflower-seed had a significant, stimulatory effect on external callus formation(p<0.05). But after 4,6,8 weeks, no difference was observed between normal and safflower-seed diet group in callus size. Urinary hydroxyproline, osteocalcin and total alkaline phosphatase decreased significantly (p<0.05) in safflower-seed treated group at 2 week after fracture.

      • Culture of Rat Embryos During Early Organogenesis

        포유동물의 최기형성을 연구하는데 가장 많이 사용되는 동물인 설치류중 랫트의 배자를 이용하여, egg-cylinder기를 지나 head-fold가 이루어지는 초기체절기인 9.5일경부터 뇌, 눈, 귀, 심혈관계가 형성되는 시기인 11.5일까지 2일(48시간)동안, 체외에서 배양하였다. 그 배양의 최적조건은 실험실마다 다르기 때문에 최적조건확립을 목적으로 이 실험을 행하였다. 온도는 37℃?.2℃로 유지시키고 가스는 배양초기인 9.5일~10.5일 동안을 50% O₂, 5% CO₂, 90% N₂가스를 공급하여 10.5일부터 6시간 동안은 20% O₂,5% CO₂, 55% N₂가스를 배양계에 공급 했을 때 가장 좋은 결과가 나왔다. 배양배지로서는 랫트 IC혈청을 분리하여 열비동화시켜서 사용하였다. 이종혈청이나 합성배지를 첨가해서 배양할 경우는 랫트 혈청에 비해 성장이 좋지 않은 것으로 관찰되었다. 동물실험법의 대체법으로서 배자양법의 기본조건을 확립하여 최기형성 평가 방법으로서의 가능성을 제시하였다. Rat embryos were cultured out of the mother from the head-fold stages, 9.5 days to 11.5 days during which they start to develop the brain, eyes, ears and cardiovascular system etc. We principally did the basic experiment in order to establish the best condition of the rat whole embryo culture in our laboratory. The temperature in the culture system was maintained 37℃ ±0.2℃ for 48 hrs. The culture was carried out in humidified atmosphere of the air, intially, the bottles were equilibrated with 5% O₂, 5% CO₂, and 90% N₂ gas mixture. After 22 or 24 and 29 or 30 hrs the cultures were reequilibrated with 20% O₂, 5% C0₂, 75% N₂ and 40% 0₂, 5% CO₂, 55% N₂ respectively. Various types of sera were tested and for the purpose of minimizing the quantity of rat serum, artificial medium was also tried and it was determined that rat serum supported normal growth over a period of 48 hrs, based on total growth analysis and structural comparisons with in vivo specimens.

      • 천연물의 항염증작용 검색에 관한 연구

        본 연구에서는 여러가지 in vivo 동물모델을 이용한 항염증제의 효력 검색법을 확립하여 항암증제의 효력시험 수탁고시에 따른 시험법을 확립하고, 비교적 부작용이 적은 천연물을 대상으로 항염증작용 검색 및 ㅡㄱ 기전을 연구하고자 하였다. 대표적인 급성 염증 모델인 rat에서의 carrageenan 및 PAF 유도 족척부종법, 염증반응 1기의 대표적인 현상인 백혈구 유주도를 측정하는 모질인 meuse에서의 zymosan운토 백혈구 유주 억제도 측정법, 염증만응 3기의 섬유화 현상을 측정하는 모델인 rat에서의 cotton pellet유도 육아종 형성 측정법, 만성 염즈으이 대표적 모델인 ratd에서의 adjubant 유도 관절염법 등 6종으 in vivo호력 시험법을 표준화하고 당귀와 원방풍 물 , 추출액, 은행엽메탕올 추출액, 죽염 수영액의 항염증작용을 검색하엿다. 당귀 물 추출액(1g/kg), 은행엽 메탄올 추출액 (1g/kg),죽염(1.5g/kg)은 carrageenan 및 PAF 유도 부종을 유의하게 억제하였고, 이중 은해엽 메틴올 추출액은 모세혈관 투과도를, 죽염은 육아종 형성을 유의하게 억제하였다. 검색한 천연물 중 zyinosan 유도 백혈굴 육주 및 adjuva띠:유도 관절염에 대해 항염증 작용을 갖는 물질은 얼었다. Carrageenan 유도 족척부종에 대한 항염증 작용 중은행13탄을 추출액과 죽염은 초기 및 후기 부종 전시간대(0.511r~6hr)에 걸쳐 항투종 작용이 있었으나 당귀 물 추출액은 carrageenan 유』A 부종 쵸기(1시간 이내)와 PAF 유도 부종에 대해 항루종 작용이 있어 carrageenan유도 분종의 초기 단계는 매개물질로 PAF가 관련한다는 점을 고려한다면 당귀의 항염증작용은 염증 매개 물질인 PAf의 작용을 억제하여 나'타날 가능성이 있을 것으로 사료된다. 이상의 실험을 통하여 염증반응 각 단계에 대한 항염증작용 겋색법을 화립하여 항염증제의 효력시험 수탁 고시에 따른 시혐법을 표준화하였고, 비교적 부작용이 적은 수종 천연물을 대상으로 항영킁제로의 이용 가능성롸 그 기전을 연구하였다. The antiinflammatory effects of fngeff-ca gigat fedeDoHriefBa sefefoifer, eiHfgo fifaD3 and Bamboo salt (Jukyom ) were investigated by in ri:same administration of methanolic extract of Ginkgo folium also inhibited the vascular permeability in - mice. The aqueous solution of Bamboo salt inhibited the formation of granuloma in SD rat at a oral dose -of Igfg. ABgelica gigantis rad;ix seems to have antiinflammatory effect by inhibition of PAF.
