        불발효차와 발효차의 간섬유화 억제 효능 비교

        Green tea and black tea are commonly drunk in the Asian and Western countries. It is known that the major tea catechins, (-)-epicatechin, (-)-epigallocatechin, (-)-epigallocatechin gallate, and (-)-epicatechin gallate are epimerized by fermentation processing. In this study, the effects of the extracts of the non-fermented and fermented teas on hepatic fibrosis were examined by means of the in vitro collagen secretion and collagen promoter assay. As a result, the treatment with green tea extract was found to reduce the secretion of collagen into the media, whereas black tea extract increased it, in the human hepatic stellate cell line.

        옻나무 플라보노이드가 수컷 백서의 성행동에 미치는 영향

        Rhus verniciflua Stokes (RVS) has been used as a food supplement and a traditional herbal medicine for a men's sexual enhancement. In this study, we prepared a flavonoid fraction (RWE) from a hot water extract of RVS and its influence on sexual behavior was studied in male rats which were orally administered varying doses of RWE for 2 weeks. All doeses of RWE stimulated sexual behavior in male rats such as reducing mounting latency and enhancing mount frequencies. However, intermission numbers were not changed and none of group can observe the ejaculation during behavioral testing time 5 min. Testosterone levels were increased about 0~53% and 92~164% by the treatment of RWE for 1 and 2 weeks, respectively. However, estrogen levels in male rats teded to decrease in a dose dependent manner of RWE. At the dose of RWE 200㎎/㎏, estrogen level was reduced to 77.5% and 70.3% of control after 1 and 2 weeks treatment. These findings suggest that the Rhus flavonoid fraction can stimulate the androgen-dependent male sexual behavior and it can applied to the material of functional food for enhancing the sexual function.

        옻나무(Rhus verniciflua STOKES) Flavonoid 분획 투여가 정자생성 및 생식관련 장기에 미치는 영향

        Rhus verniciflua Stokes (RVS) has been used as a food supplement and a traditional herbal medicine. In this study, we prepared various flavonoid fractions (RS, RW1, RW2 and RWE) from a hot water extract of RVS and their influence on male reproductive organs and spermatogenesis were studied in rats which were orally administered 200 ㎎/㎏ of them for 8 weeks. All experimental groups did not show any significant changes in body weight and blood clinical chemistry for liver function. Plasma testosterone level was elevated about 3.7, 5.2 and 6.3 folds in RW1, RW2 and RWE groups, respectively. The weights of testes and epididymides tended to increase slightly without the statistical significance in RW2 and RWE. The spermatozoon motility and epididymal sperm concentration were significantly increased (P < 0.05) in RWE and RW1, respectively, when compared to the control group. There was no significant difference in histology and apparent shape of testes and epididymides among the control and the experimental groups. Collectively, RWE showed effectively the elevation of plasma testosterone level, spermatozoon motility and the epididymal sperm concentration without the significant increase of testis and epididymides weights. When the component HPLC profile among the flavonoids fractions of RVS was compared, the ratio of components were only different. These findings suggest that the Rhus flavonoid fraction, particularly RWE, can stimulate the androgen-dependent male sexual function and it can be applied to the material of functional food for enhancing the sexual function.

        Normally, environmental toxicants are classified as endocrine disruptors if they interfere with regulation of cellular function by endogeneous steroids through inhibition of receptor binding and/or transcriptional activation. So, many studies have been performed about agonist/antagonist of hormone receptor to study mechanisms of endocrine disruptors. If toxicants affect steroid biosynthesis and/or degradation and alter hormone homeostasis, these also are classified as endocrine disruptors. But there are not many studies of the mechanisms of endocrine disruptors on the basis of alteration of steroid biosynthesis and/or degradation. Isolation and culture of Leydig cells from testis is one of methods for the steroidogenesis screening assays to evaluate a substance for altering steroidogenesis. Leydig cells were harvested using the method described by Klinefelter with modifications. Leydig cells were purified by perfusion of testis and incubation (34℃, 80cycles/minute, 20 minutes) with collagenase (0.25 ㎎/㎏), centrifugal elutriation, percoll gradient centrifugation and BSA multidensity gradient centrifugation. To confirm if this method is one of appropriate tools to evaluate a substance for altering steroidogenesis, ketoconazole, positive control was administered to purified Leydig cells. Ketoconazole (10??M and above) significantly reduced testosterone production in purified Leydig cells. From above results, we suggest that this method for steroidogenesis screening assay appears to be a appropriate tool to detect suspected compounds for altering steroidogenesis.

        간경변 백서 모델에서 아데노바이러스 벡터를 이용한 Matrix Metalloproteinase-3 유전자 치료

        목적: 간경변은 활성화된 간성상 세포에 의해 이상 증식된 세포외기질이 축적되는 것을 특징으로 하며 matrix metalloproteinase (MMP)에 의해 분해되는 것으로 보고되고 있다. 본 연구의 목적은 간경변 모델 흰쥐에서 MMP-3를 발현하는 아데노바이러스를 이용한 유전자 치료로 간섬유화가 경감되는지를 알아보고자 하였다. 방법: 몸무게 200~250 g의 6주령된 수컷 백서에 8주 동안 DMN을 복강에 주사하여 간경변 동물 모델을 만들었다, 일주일 후에 AdMMP3.GFP, Ad.GFP, PBS를 문맥에 주사한 후 매주마다 백서를 희생시켜 조직을 채취하였다. 간경변의 개선정도는 활성형의 간성상 세포지표인 a-SMA, Desmin, TGFβRⅡ의 면역화학적 염색, 간세포 성장 표지자인 PCNA 및 dry liver weight 측정을 통해 분석하였다. 결과: MMP-3를 주사한 군에서 Masson`s trichorm 염색 시의 교원질과 hydroxyproline의 양은 현저히 감소하였다. MMP-3올 주사한 군에서 α-SMA, Desmin, TGFβRⅡ 양성 세포수는 감소하며, PCNA 양성 세포수와 dry liver weight은 증가하였다. 결론: 본 연구의 결과를 종합해 볼 때 MMP-3를 이용한 유전자 치료가 비가역적으로 알려진 간경변을 최소한 경감시킬 수 있는 가능성을 보여주어 향후 항섬유화 분자치료적 관점에서 긍정적인 가능성올 제시하고 있다. Background: Liver cirrhosis is characterized by fibrous scarring and hepatocellular regeneration. Matrix metalloproteinases (MMPs) comprise a family of zinc-dependent enzymes that degrade the extracellular matrix (ECM) components. This study examined whether or not gene delivery of human MMP-3 can attenuate established liver cirrhosis in a rat. Methods: Rat liver cirrhosis was induced by an intraperitoneal injection of dimethylnitrosamine (DMN) three times a week for 8 weeks. The rats were infected once with either a recombinant adenovirus. AdMMP3.GFP, or a control adenovirus, Ad.GFP, into a portal vein and followed up for 3 weeks. In the rat liver tissues, the collagen content, histopathology and immunohistochemical staining were measured. Results: Liver fibrosis in the DMN induced cirrhotic rat was attenuated along with a diminished hydroxyproline content and increased dried liver weight after the gene delivery of AdMMP3.GFP. In addition, the number of activated hepatic stellate cells was lower whereas the proliferation of hepatocytes, which was confirmed by immunohistochemical staining using anti-proliferating cell nuclear antigen (PCNA) antibody, was observed in the AdMMP3.GFP infected rats, suggesting that human MMP-3 stimulated hepatocyte proliferation. Conclusions: These results suggest that the gene transfer of human MMP-3 in the liver attenuates established fibrosis and induces hepatocyte proliferation. Therefore, gene therapy using MMP-3 in liver cirrhosis might be a promising therapeutic option in the future. (Korean J Med 71:609-619, 2006)
