        The cytotoxic, mutagenic and antimutagenic activities against chemical direct mutagens such as mitomycin C, 4-nitroquinoline-N-oxide, 2,4,7-trinitro-9-fluorenone of the 70% ethanol extracts of 29 colored rice varieties and chuchung as a control were examined. The results obtained using authentic alkaline phosphatase activity as a growth representative of the indicator cell E. coli PQ 37 demonstrated that the extracts of 13 kinds of colored rice varieties including Jumlalocal and Jumlalocal-1 showed strong toxic effect on the cell growth. However the extracts of DK 1, SC-5, LK 1A-2-12-1-1 and wx 139-3-64-20-3-1 seemed to have stimulatory effects on the cell growth. The mutagenicity and antimutagenicity of the colored rice varieties were screened using SOS chromotest. The mutagenic activity was detected from Jumalocal-1, IR 17491-5-4-3-3 and Jumlalocal. On the contrary, 7 samples including LK 1-3-6-12-1-1, Parnkhari 203, Jumlalocal, wx 139-3-64-20-3-1, Muthumanikam, HP 883-1-1-1-B-1-1 and Jumlalocal-1 were shown to have antimutagenic acitivities against the chemical direct mutagens used in this study. 국내외에서 수집 재배한 29품종 유색미의 70% 에탄올 추출물로 세포독성, 변이원성, 화학적 직접변이원 mitomycin C, 4-nitroquinoline-N-oxide, 2,4,7-trinitro-9-fluorenone에 대한 항변이원성을 측정하였다. 세포 내재성 alkaline phosphatase활성을 지표로 유색미 추출물이 지시세포인 E. coli PQ 37의 성장에 미치는 영향을 조사한 결과 Jumlalocal, Jumlalocal-1을 포함하는 13종류의 유색미 추출물이 세포독성을 나타낸 반면 DK 1, SC-5, LK 1A-2-12-1-1및 wx 139-3-64-20-3-1등의 품종은 세포성장 촉진활성을 나타내었다. SOS chromotest 기법을 이용하여 유색미가 가지는 변이원성 및 항변이원성을 조사한 결과에서는 Jumlalocal-1, IR 17491-5-4-3-3 및 Jumlalocal 등의 품종이 변이원성을 유도하는 품종이었다. 화학적 직접변이원 mitomycin C, 2,4,7-trinitro-9-fluorenone 및 4-nitroquinoline-N-oxide 에 대해서 공통적으로 항변이원성 효과가 인정되는 유색미 품종은 LK1-3-6-12-1-1, Parnkhari 203, Jumlalocal, wx 139-3-64-20-3-1, Muthumanikam, HP 883-1-1-1-B-1-1, Jumla-local-1 등 7품종이었다.

      • 유색미 에탄올 추출물의 품종간 항산화 활성 변이

        갈색에서부터 흑자색까지의 색상을 가지는 국.내외에서 수집한 23종류 유색미의 항산화 활성, 색소체 함량의 변기 및 상관성을 검정하였다. 유색미 에탄올 추출물 시료가 나타내는 환원력은 일반미 품종인 추청에 비해서 거의 모든 품종에서 높게 나타났으며, 환원력이 특히 높은 품종은 LK 1-3-6-12-1-1, Elwee, DZ 78, Jumlalocal-1, SC-45 등이었다. 전자공여능이 높은 품종은 HP 883-1-1-1-B-1-1, HP 833-1-3-1-1-1, LK-2-7-12-1-1, DZ 78 등이었고 hydroxy radical 소거활성이 큰 품종은 DK-1, IR 1544-38-2-2-1-2-2, SC-5, SC-45 등이었으며, LK 2-7-12-1-1은 오히려 산화촉진효과를 나타내는 품종이었다. 자동산화에 의한 지질 과산화물 형성을 강하게 억제하는 품종들은 RGS No. 336, LK IB-2-1-1, LK 1B-4-12-1-1, LK 1A-2-12-1-1, LK 2-7-12-1-1, HP 833-1-1-1-B-1-1등이었고 Elwee, Jumlalocal-1, SC-45 등의 품종은 산화적인 활성을 나타내고 있었다. 23품종의 유색미 중에서 색소체의 함량이 가장 많은 품종은 Elwee이었으며, 색소체 함량은 환원력과 정의 상관성이 있었고, 추출물이 고농도로 처리되었을 때 높은 환원력을 보인 유색미 품종은 linoleic acid 자동산화반응에서 지질 과산화를 촉진하는 경향이 있었다. 유색미 23품종 중에서 SC-5는 모든 반응계에서 비교적 높은 항산화 활성을 나타내었다. Interrelation between the antioxidative activities of 70% ethanol extracts from bran fraction of 23 kinds of colored rice and chuchung as a control were examined. Antioxidative activities were evaluated by assaying reducing power, electron-donation ability to DPPH free radical, scavenging activity of hydroxy radical $({\cdot}OH)$ generated through Fenton reaction and inhibitory activity on lipid peroxidation using linoleic acid autoxidation system, respectively. Among 24 varieties of colored rice LK 1-3-6-12-1-1 had the strongest reducing power followed by Elwee, DZ 78, Jumlalocal-1 and SC-45 in decreasing order. The electron-donating ability to DPPH radical was higher in order of HP 883-1-1-1-B-1-1, HP 833-1-3-1-1-1, LK 2-7-12-1-1 and DZ 78. The hydroxy radical scavenging activity was higher in order of DK-1, IR 1544-38-2-2-1-2-2, SC-5 and SC-45 but LK 2-7-12-1-1 had oxidative effect. In the liaoleic acid autoxidation model system, RGS No 336, LK 1B-2-1-1, LK 1B-4-12-1-1, LK 1A-2-12-1-1, LK 2-7-12-1-1 and HP 883-1-1-1-B-1-1 exhibited strong antioxidative activities but Elwee, Jumlalocal-l and SC-45 showed to have oxidative effects. The rice variety of highest pigment content was Elwee and the next were RGS-No 336, IR 1544-38-2-2-1-2-2 and SC-5 with the order of higher content. The reducing power was correlated with the quantity of the pigment in the ethanolic extract of rice bran and SC-5 showed relatively high antioxidative activity in every results of antioxidative activity tests.

        유색미 겨 추출물의 품종간 발암과정 억제효과의 비교

        Anticarcinogenicity of 12 kinds of colored rice cultivars was investigated in vitro with Ilpumbyeo as a control. Pigment and hexane fractions were prepared from the rice brans through extraction with 80% ethanol containing 0.5% TFA or n-hexane to use subsequent evaluation of their anticarcinogenic effect by determining inhibitory activity against DNA strand scission and tumor promotion, as well as antimutagenic activity using SOS chromotest system. Antimutagenic activity of the pigment fractions was revealed to decrease in order of LK1B4-12-1-1 $gt; Sanghaehanghyulla $gt; HP833-1-3-1-1 $gt; Chokoto14 $gt; Jagwangdo $gt; Jajin $gt; Kilmheugmi $gt; Tiwarr-Tsi-C $gt; Heugjinmi $gt; Linsia-Shoea-Dau $gt; Suweon425 $gt; Suweon415. The activity of the hexane fractions decreased in order of LK1B4-12-1-1 $gt; Sanghaehyanghyulla $gt; Ilpunibyeo $gt; HP833-1-3-1-1 $gt; Chokoto14 $gt; Jagwangdo $gt; Jajin $gt; Linsia-Shoea-Dau $gt;Tiwan-Tsi-C $gt; Heugjinmi $gt; Suweon425, Kilimheugmi $gt; Suweoo415.         유색미 쌀겨추출물의 in vitro의 발암 억제효과

        As a preliminary experiment to investigate the antitumor activity of colored rice in vivo, inhibitory effect of solvent extracts from colored rice brans on DNA strand scission and tumor promotion was examined in vitro. Two colored rice cultivars, Sanghaehyanghyulla and Suwon 415 were compared with Chuchung as a control. The antimutagenic activity of each rice cultivars increased in order of Chuchung$lt;Sanghaehyanghyulla$lt;Suwon 415. However, the inhibitory activity of 70% ethanolic extracts against DNA strand scission increased in order of Sanghaehyanghyulla$lt;Chuchung$lt;Suwon 415. The results from the examination on the chloroform fractions demonstrated that Sanghaehyanghyulla did not show any significant inhibitory activity, and that Chuchung was unexpectedly somewhat more inhibitory than Sanghaehyanghyulla. Strong inhibition against DNA strand scission was observed in the chloroform fraction of Suwon 415, activity of which was almost same as that of the 70% ethanolic extract(about 50% inhibition). We also measured the inhibitory effect of 70% ethanolic extracts on tumor promotion by means of its actions that block the expression of Epstein-Barr virus early diffusible antigen induced by tumor promotor, indicating the increase of inhibitory activity in order of Chuchung$lt;Sanghaehyanghyulla$lt;Suwon 415. Although every chloroform fractions showed complete blockage of tumor promotion, Suwon 415 was found to be the only cultivar that showed the cytotoxicity-free antipromotion effect.

        조현병 외래 환자에서 야간식이증후군의 유병률과 관련요인

        연구목적 본 연구의 목적은 조현병 외래 환자를 대상으로 야간식이증후군(night eating syndrome : NES)의 유병률과 이와 관련된 임상적 요인들에 대해 살펴보는 것이다. 방 법 정신건강의학과 외래를 방문한 201명의 조현병 환자들을 14 문항의 자기보고 형식인 야간식이증후군 설문지(Night Eating Questionnaire : NEQ)로 평가하였다. 인구학적, 임상적 특징과 체 질량 지수(BMI)을 조사하였고, 주관적인 기분과 수면, 폭식과 체중 관련 삶의 질에 대해서는 각각 Beck's Depression Inventory(BDI), Pittsburgh Sleep Quality Index(PSQI), Binge Eating Scale(BES)와 Korean version of Obesity-Related Quality of Life Scale(KOQoL)를 이용하여 평가하였다. 결 과 조현병 외래 환자에서 야간 식이 증후군의 유병률은 10.4%(201명 중 21명)이었다. NES 군과 non-NES 군간에 나이를 제외하면 인구사회학적 및 임상적 특성, BMI에서는 유의한 차이를 보이지 않았다. NES 군이 Non-NES 군에 비해 보다 우울했고, 수면과 폭식 양상, 체중 관련 삶의 질에 있어서 의미 있는 장애를 나타냈다. '아침 식욕부진'과 '지연된 아침식사'(NEQ에서 NES 핵심 항목 5개 중 2개)는 두 군간에 차이가 없었으나, '야간 섭식', '야간 식욕항진' 그리고 '기분/수면'은 NES 군에서 더 손상되어 있는 것으로 나타났다. 결 론 본 연구는 조현병 외래 환자를 대상으로 NES의 유병률과 관련 요인에 대해 기술한 최초의시도이다. 비록 NES와 비만과의 연관성은 밝히지 못했지만, 본 연구의 결과는 NES가 정신 건강에 부정적 영향을 준다는 사실을 보여주었다. 향후 이러한 결과를 뒷받침할 수 있는 추가 연구가 필요할 것이다. Objectives : The aim of study was to examine the prevalence of night eating syndrome(NES) and its correlates in schizophrenic outpatients. Methods : The 14 items of self-reported night eating questionnaire(NEQ) was administered to 201 schizophrenic patients in psychiatric outpatient clinic. We examined demographic and clinical characteristics, body mass index(BMI), subjective measures of mood, sleep, binge eating, and weight-related quality of life using Beck's Depression Inventory(BDI), Pittsburgh Sleep Quality Index(PSQI), Binge Eating Scale(BES) and Korean version of Obesity-Related Quality of Life Scale(KOQoL), respectively. Results : The prevalence of night eaters in schizophrenic outpatients was 10.4%(21 of 201). Comparisons between NES group and non-NES group revealed no significant differences in sociodemographic characteristics, clinical status and BMI. Compared to non-NES, patients with NES reported significantly greater depressed mood and sleep disturbance, more binge eating pattern, and decreased weight-related quality of life. While 'morning anorexia' and 'delayed morning meal'(2 of 5 NES core components in NEQ) were not differed between groups, 'nocturnal ingestions', 'evening hyperphagia', and 'mood/sleep' were more impaired in NES group. Conclusions : These findings are the first to describe the prevalence and its correlates of night eaters in schizophrenic outpatients. These results suggest that NES has negative mental health implications, although it was not associated with obesity. Further study to generalize these results is required.

        두류의 항산화활성 검정

        Seventy percent ethanolic extracts from commercially available 13 legumes were made to investigated their antioxidative activities by determining the reducing power, inhibitory effect on lipid peroxidation, scavenging activity against both superoxide radical and hydroxyl radical, together with inhibitory activity toward mitomycin C-induced oxidative damage of DNA. High level of reducing powers were detected in Yepat, Sokpiri, Yuweol-bean and Jeokdu. Inhibitory effects on lipid peroxidafion were found ubiquitously in all extracts examined when employing the linoleic acid autoxidation system, whereas, only 3 legumes, Yepat, Namul-bean and Jeenuni-bean, were revealed marked inhibition in rabbit erythrocyte-ghost membrane lipid peroxidation system. Yepat, Namul-bean, Jeokdu and Jeenuni-bean showed great scavenging activities on superoxide radical, on the other hand, high level of hydroxyl radical scavenging activities were demonstrated in Sokpiri, Chungtae, Yepat and Jebi-bean. Ubiquitous inhibitory effects on mitomycin C-induced oxidative damage on DNA were found in all extracts tested, Among them, however, Yepat, Jeenuni-bean, Namul-bean, Nokdu and Jeokdu showed the higher level of inhibition. Taken together, we could assign Yepat, Jeokdu, Jeenum-bean and Sokpiri, for the legume species highly functional on overall antioxidative activity.

        뇌졸중 환자의 트레드밀 훈련 시 손잡이 유무 및 위치가 보행 및 균형에 미치는 영향

        Purpose: The purpose of this study was to examine the influence of a handrail (presence and position) on treadmill gait and balance in stroke patients during gait training. Methods: 39 patients with stroke (male 31, female 8) participated in this study. The training groups were classified into a no-handrail group (NHG), front handrail group (FHG), and bilateral handrail group (BHG). Each group comprised 13 subjects. The subjects were trained to walk in a straight path 30 minutes per day for 8 weeks. The Good Balance System was used to measure static balance and dynamic balance. To measure walking ability, timed up and go (TUG) was also assessed. Results: The NHG showed no significant differences in static balance, dynamic balance, and TUG. The FHG was significantly different in their medial-lateral speed of static balance, dynamic balance, and TUG. The BHG was significantly different in their static balance, dynamic balance, and TUG. Conclusion: These findings consider the effects of holding handrails concomitantly with changes in postural stability. We conclude that for training stroke patients, treadmill walking while holding handrails improves balance and gait more than treadmill walking without holding handrails. The resulting changes in muscle activity patterns may facilitate the transfer to a gait pattern. The results of this study suggest methods for training treadmill walking in stroke patients.
