        인삼정천탕(人蔘定喘湯)이 호흡기계(呼吸器系)에 미치는 영향(影響)

        Insamjungchuntang has been used in Korea for many centuries as a treatment for respiratory disease. The effect of Insamjungchuntang on tracheal smooth muscle is not known. The purpose of the present study is to determine the effect of Insamjungchuntang on histamine and acetylcholine induced tracheal smooth muscle contraction in rats and guinea pigs. Guinea pig (500 g, male) and Sprague Dawley rats (200 g, male) were killed by $CO_2$ exposure and a segment (8-10 mm) of the thoracic trachea from each rat and guinea pig was cut into equal segments and mounted 'in pairs' in a tissue bath. Contractile force was measured with force displacement transducers under 0.5 g loading tension. The dose of histamine (His) and acetylcholine (Ach) which evoked 50% of maximal response $(ED_{50})$ was obtained from cumulative dose response curves for histamine and acetylcholine$(10^{-7}{\sim}10^{-4}\;M)$. Contractions evoked by His ($ED_{50}$) and Ach $(ED_{50})$ were inhibited significantly by Insamjungchuntang. In guinea pig tracheal smooth muscle, the mean percent inhibition of acetylcholine induced contraction was $38.58\(p<0.05)\;after\;10{\mu}l/ml$ Insamjungchuntang, $90.75\(p<0.01)\;after\;30{\mu}l/ml$. Insamjungchuntang and $133.17\(p<0.01)\;after\;100{\mu}l/ml$ Insamjungchuntang. In rat tracheal smooth muscle, the mean percent inhibition of acetylcholine induced contraction was $10.0\(p<0.05)\;after\;10{\mu}l/ml$ Insamjungchuntang, $80.71\(p<0.01)\;after\;30{\mu}/ml$ Insamjungchuntang and $118.29\(p<0.01)\;after\;100{\mu}l/ml$ Insamjungchuntang. Also, in guinea pig tracheal smooth muscle, the mean percent inhibition of histamine induced contraction was $45.5\(p<0.01)\;after\;10{\mu}l/ml$ lnsamjungchuntang, and $93.17\(p<0.01)\;after\;30{\mu}l/ml$. lnsamjungchuntang $134.50\(p<0.01)\;after\;100{\mu}l/ml$ Insamjungchuntang. In rat tracheal smooth muscle, the mean percent inhibition of histamine induced contraction was $37.83\(p<0.01)\;after\;10{\mu}l/ml$ lnsamjungchuntang, $90.5\(p<0.01)\;after\;30{\mu}l/ml$ Insamjungchuntang and $135.17\(p<0.01)\;after\;100{\mu}l/ml$ Insamjungchuntang. Propranolol $(10^{-7}\;M)$ slightly but significantly attenuated the inhibitory effects of Insamjungchuntang. Following treatment with propranolol, the mean percent inhibition caused by $100{\mu}l/ml$. Insamjungchuntang fell to 46.42% in guinea pig induced by acetylcholine contraction and by $100{\mu}l/ml$ Insamjungchuntang fell to 5.43% (p<0.05) in rat induced by acetylcholine contraction and the mean percent inhibition caused by $100{\mu}l/ml$ Insamjungchuntang fell to 49.0% in guinea pig induced by histamine contraction and by $100{\mu}l/ml$ Insamjungchuntang fell to 48.6% (p<0.05) in rat induced by histamine contraction. Indomethacin and methylene blue $(10^{-7}\;M)$ did not significantly alter the inhibitory effect of lnsamjungchuntang. Also, I could find the effects of lnsamjungchuntang and Insamjungchuntanggamorphine on the tracheal smooth muscle in guinea pig and rat did not change significantly. These results indicate that Insamjungchuntang can relax histamine and acetylcholine-induced contraction of guinea pig and rat tracheal smooth muscle, and that this inhibition involves sympathetic effects.

        퓨린 港代謝物의 抗癌活性에 관한 MO 이론적 연구

        퓨린 港代謝物에는 沆癌活性인 것과 非活性인 것이 있는데, 이들을 EHT 方法으로 계산하여 면밀히 검토해 본 결과, 沆癌活性인 것은 DNA 염기의 당이 결합한 위치에 대응되는 위치인 N(9)에서 electron donating ability를 나타내지만, 非活性인 것은 이와 반대임을 알았고 N(9)와 당에 해당되는 위치 (10)간의 overlap population이 가장 작게 나타났다. 이것은 nuclotide level로 전환되어 沆癌活性을 나타낸다는 실험사실과 일치한다. EHMO calculations were performed on several antitumor active as well as inactive purine antimetabolites. Results showed that the N atom (position 9) which corresponds to the position of sugar (position 10) linkage in DNA bases has a net negative charge for antitumor active whereas it has a positive charge for inactive purines. It was also found that overlap population was the smallest for bond between atoms 9 and 10, which agrees with the experimental findings that antitumor activity is effected after conversion to nucleotide level.

        分子軌道論의 生物化學에의 應用 (第 2 報). 發癌物質과 DNA 鹽基와의 相互作用

        The interactions of chemical carcinogens, such as polycyclic aromatic hydrocarbons, dimethylaminoazobenzene (DAB) and its derivatives and heterocyclic compounds with tissue components, especially with deoxyribonucleic acid (DNA), were examined by means of simple Huckel method. Assuming that the formations of a loose molecular complex between the carcinogens and the tissue components are the first step of chemical carcinogenesis, the most proble orientation between the chemical carcinogens and adenine-thymine (A=T) pair or guanine-cytosine $(G\equivC)$ pair is determined. It has been found that, in the case of the formation of molecular complex between chemical carcinogens and A=T pair, the two atoms of K-region of the carcinogens and the atom of L-region in the proximity of their K-region are combined correspondingly with C-l' carbon atom in the sugar that is attached to thymine, N-1 nitrogen atom and C-5 carbon atom in the thymine part of A=T pair, while, in the case of that between the carcinogens and $G\equivC$ pair, the above three atoms of the carcinogens are combined correspondingly with C-8 carbon atom, N-9 nitrogen atom and N-3 nitrogen atom in the guanine part of $G\equivC$ pair. 發癌物質과 DNA 鹽基雙間의 分子錯物形成에서 可能性있는 配置(orientation)를 決定하였다. 아데닌-티민 염기쌍에서는 티민쪽에서, 구아닌-시토신 염기쌍에서는 구아닌쪽에서 分子錯物을 形成한다.

        말단 헤테로원자가 산소(O)와 황(S)인계의 1,5-수소원자 이동반응에 관한 MO 이론적 연구

        ${\beta}$-diketones, ${\beta}$-thioxoketones and ${\beta}$-dithioketones류의 tautomeric forms (6${\pi}$ 전자계)의 1,5-H shift에 관해서 MNDO 방법으로 전이상태 구조를 결정하고, 활성화 에너지 장벽에 미치는 구조적 안정성을 분석하였다. 그 결과 1전자항 ${\Delta}( 2\sum\limits^{occ}{\varepsilon}_i)^{\neq}$ 이 증가할수록 활성화 장벽이 큼을 알았다. 또한 말단 헤테로원자의 하전분리가 클수록 바닥상태 분자의 안정성이 크며, 활성화 에너지 장벽$({\Delta}E^{\neq})$이 크다. MO studies have been carried out on 1,5-hydrogen shifts between tautomeric forms (6${\pi}$ system) of ${\beta}$-diketones, ${\beta}$-thioxoketones and ${\beta}$-dithioketones by determining transition state and analyzing structural stability effects on the activation barriers using MNDO method. The barrier was found to increase with the stability of the ground state having greater charge separation and with the increase (less stabilization) in the one electron energy ${\Delta}(2\sum\limits^{occ}{\varepsilon}i)^{\neq}$ in the activation process.

        오호탕(五虎湯)이 백서(白鼠)의 호흡기계(呼吸器系)에 미치는 영향(影響)

        These studies were carried out to investigate the effects of Ohotang water extract on the inhibitory contractile action of acetylcholine in control rat. The results of these were as follows; 1. Contractile force of acetylcholine from trachea smooth muscle in control rat was significantly inhibited by Ohotang. 2. Dose-response of acetylcholine pretreated Ohotang in control rat was significantly inhibited. 3. Inhibitory contractile action of acetylcholine pretreated propranolol in control rat was significantly inhibited by Ohotang. 4. Contractile force of acetylcholine pretreated indomethacin from trachea smooth muscle in control rat was not significantly changed. 5. Inhibitory contractile action of acetylcholine pretreated methylene blue in control rat was not significantly changed.

        클라리스로마이신의 건강한 한국 성인에 대한 집단 약물동태

        The purpose of this study was to estimate the population pharmacokinetics of clarithromycin in healthy adult Korean and to investigate the factors if1uencing the pharmacokinetics of clarithromycin. The population pharmacokinetic parameters of clarithromycin were calculated with the data from healthy adult Koreans. A total of 798 plasma concentrations obtained from 78 subjects after administration of a single oral dose of 250 mg or 500 mg were used for the modeling. The concentration-time data were fitted to a one-compartment open model assuming a first-order absorption and elimination with no lag time. The correlations between various factors [such as sex, age, height, welght, serum creatinine (Scr) and dose ] and pharmacokinetic parameters were estimated with stepwise linear regression analyses. The selected covariates were incorporated in the population model of NONMEM, and the importance of each covariate was investigated by means of backwards elimination. The apparent clearance (CL/F) was significantly correlated to Scr and sex, and the apparent volume of distribution (Vd/F) was significantly correlated to Scr and height in a nonlinear relationship. The population values of Ka was 1.8 hr-1, CL/F was 37.71 L/hr Vd/F was 200 L and t1/2 was 3.68 hrs for a male Korean with 170 cm height and 1.0 mg/dL Scr.

        In this study By the time that TTX technology is adopted as railway rolling stock we analyzed quantitatively energy saving by reducing the power consumption with the reduction of the operation time through the speed improvement and suggested the necessity to introduce TTX technology in the domestic rail vehicles. The effect of energy saving by comparing and analyzing the power consumption during the operation by TTX Hanvit 200 and 8200 electric locomotives to pull trains on the same line was suggested and the efficiency of the main devices(i.e C/I) of Hanvit 200 was compared and analyzed by measuring the power consumption by a single unit. For improving KORAIL management environment reducing energy usage is an urgent challenge its measures for solving them are constantly considered in many areas. In addition at the time of improving the conventional track to speed up and changing the signals Tilting technology will be contributed to the management environment by enlarging the passengers' demand through the reduction of the operation time and saving energy using the existing infrastructure.

        Estimation of Activtiy Against Adenocarcinoma CA755 and Toxicity of Purines in Mice Using Physicochemical Parameter and Connectivity Index

        The nonempirical molecular connectivity indexes of a number of mono- and disubstituted purines were calculated. Very good correlations were obtained between anticancer activity (log 1/c) and toxic activity (log 1/$LD_{50}$) of tIhese compounds and their molecular connectivity indexes and physicochemical constants. Our structure-activity relationship is discussed briefly in relation to theories of general QSAR.
