        JSAP1 Interacts with Kinesin Light Chain 1 through Conserved Binding Segments

        KIF5는 2분자의 kinesin heavy chain (KHC)과 2분자의 kinesin light cham (KLC)으로 구성되며 미세소관과 직접 결합한다. KIF5는 여러가지 세포 내 소기관을 이동시키나 KIF5가 이동시키는 운반체가 어떻게 특이적으로 결합하는지는 아직 밝혀지지 못하였다. 본 연구에서 효모 two-hybrid system을 사용하여 KLC1의 tetratricopeptide repeats (TRP) 부위와 결합하는 세포 내의 단백질을 분리하였다. 결과 KLC1와 특이적으로 결합하는JNK/stress-activated protein kinase-associated protein 1 (JSAP1/JIP3)을 분리하였다. 이러한 결합은 KLC1의 TRP1, 2 영역과 JSAP1의 leucine zipper 영역이 결합에 관여하며, 또한 효모 two-hybrid assay에서 JSAP1은 KLC2와 결합하지만 신경세포에서 발현하는 KIF5A, KIF5C 그리고 모든 세포에서 발현하는 KIF5B와는 결합하지 않았다. 단백질간의 결합을 pull-down assay로 확인한 결과 KLC1은 glutathione S-transferase (GST)와는 결합하지 않으나 GST결한 JSAP1과는 결합하였다. 또한 생쥐의 뇌 파쇄 액으로부터 JSAP1 항체로 면역침강을 행한 결과 KLC1은 JSAP1과 같이 침강하였다. 이러한 결과들은 KLC1은 JSAP1과 결합하며, JSAP1은 KLC1의 수용체로세포 내 KIF5의 수송의 매개 단백질로 작용함을 시사한다. A conventional kinesin, KIF5/kinesin-I, is composed of two kinesin heavy chains (KHCs) and two kinesin light chains (KLCs) and binds directly to microtubules. KIF5 motor mediates the transport of various membranous organelles, but the mechanism how they recognize and bind to a specific cargo has not yet been completely elucidated. Here, we used the yeast two-hybrid system to identify the neuronal protein(s) that interacts with the tetratricopeptide repeats (TRP) of KLCI and found a specific interaction with JNK/stress-activated protein kinase-associated protein 1 (JSAP1/JIPP3). The yeast two-hybrid assay demonstrated that the TRP 1,2 domain-containing region of KLCI mediated binding to the leucine zipper domain of JSAP1. JSAP1 also bound to the TRP region of lac2 but not to neuronal KIF5A, KIF5C and ubiquitous KIF5B in the yeast two-hybrid assay. In addition, these proteins showed specific interactions in the GST pull-down assay and by co-immunoprecipitation. KLCI and KIF5B interacted with GST-ISAP1 fusion proteins, but not with GST alone. An antibody to JSAPI specifically co-immunoprecipitated KIF5s associated with JSAP1 from mouse brain extracts. These results suggest that JSAP1, as KLC1 receptor, is involved in the KIF5 mediated transport.

        한국재래산양의 전지골격에 관한 해부학적 연구

        The anatomical structure of the Skeleton of thoracic limb of thirty-one adult Korean native goats(body weight: 14~17kg) was observed after skeletal preparation, and the osteometry was performed in each bone. The results were as follows; 1. The thoracic limb of the Korean native goat was composed of scapula, humerus, radius, ulna, carpal bones, metacarpal bones, phalanges and sesamoid bones. 2. The scapula was flat and triangular in shape. There were no distinct tuber of spine and acromion in the spine. The subscapular fossa was deep and triangular in shape and the vertebral border was sigmoid form. The coracoid bone was formed as the coracoid process at the medial aspect of the supraglenoid tubercle but the clavicle wa.s not observed. The left and right scapular indexes were 57.92 and 58.31 and the glenoid cavity indexes were 89.23 and 86.82, respectively. 3. The greater tubercle of the humerus was devided into cranial and caudal parts. The third tubercle was observed and the face for the infraspinatus muscle was rectangular form. The left and right humerus indexes were 32.44 and 32.63, the head indexes were 94.13, 96.62 and the trochlear-epidondyle indexes were 67.32 and 65.81, respectively. 4. The radius and ulna were fused entirely except at the broad proximal and narrow distal interosseous spaces. The ulna was longer than the radius, and its reduced body and distal end were fused at the caudomedial surface of the radius. 5. The carpal bones were six in number. There were radial, intermediate, ulnar, accessory, second-third and fourth carpal hones in carpal bones. 6. The metacarpal bone was composed of a large metacarpal bone resulted from the fusion of the third and fourth metacarpal bones, and there was a metacarpal tubercle at the dorsolateral part of the proximal end. There were no vestiges of the second and fifth metacarpal bones. 7. The digits were composed of third and fourth digits and each digit was composed of the proximal, middle and distal phalanges. 8. The sesamoid bones were six in number. There were two at the fetlock joint and one at the coffine joint palmarly in each digit. 9. The ratios of the lengths among the scapula, humerus, antebrachium and metacarpal bone were 1.42 : 1.47 : 1.77 : 1.00 in the left and 1.42 : 1.45 : 1.77 : 1.00 in the right, respectively.

        한국재래산양의 후지골격에 관한 해부학적 연구

        The anatomical structure of pelvic limb, of thirty-one adult Korean native goats (Body weight: 14~17kg) was observed after skeletal preparation, and the osteometry was performed in each bone. The results were as follows: 1. The pelvic limb of the Korean native goat was composed of the hip bone, femur, patella, tibia, fibula, tarsal bones, metatarsal bone, phalanges and sesamoid bones. 2. The hip bone consisted of the ilium, ischium and pubis which fused each other, The gluteal surface of the ilium was directed dorsolaterally. The tuber sacrale and tuber coxae were formed at the dorsal apex and ventrolateral part of the iliac wing, respectively. The lesser ischiatic notch was deeper than the greater one. The ischiatic tubercles were triangular form and consisted of the dorsal, lateral and caudal ischiatic tubercles. The left and right hip bone indexes were 67.08, 66.20, the acetabular indexes were 93.78 and 92.10 and the obturator foramen. indexes were 53.84 and 54.77, respectively. 3. In femur, both of the greater and lesser trochanter were well developed but the third trochanter was not observed. The left and right femur indexes were 26.55 and 26.14, head indexes were 81.66 and 81.49 and the trochlear-epicondyle indexes were 42.47 and 41.63, respectively. 4. The patella was observed as an isosceles triangle with base lying proximal and the cranial surface was more convex. 5. The tibial shaft was sigmoid form and the popliteal notch was deep. There was a large nutrient foramen at the cranial aspect of the cranial intercondylar area. The tibial indexes were 22.09 in left and 21.10 in right. 6. The proximal extremity of the fibula was fused with the lateral condyle of the tibia but the distal one was observed independently as the malleolar bone. 7. The tarsal bones were five in number; the talus, calcaneus, centroquartal tarsal bone, first tarsal bone, and second-third tarsal bone. 8. The metatarsal bone was composed of a large metatarsal bone. resulted from the fusion of the third and fourth metatarsal bones, The structure of metatarsal bone was similar to the metacarpal bone but longer about 7mm. 9. The phalanges and sesamoid bones were similar to these of the thoratic limb. 10. The ratios of the lengths among the hip bone, femur, tibia and metafarsal bone were 1.71 : 1.54 : 1.73 : 1.00 in left and 1.68 : 1.53 : 1.72 : 1.00 in right, respectively.

        Nitrate가 가토 갑상선 기능에 미치는 영향

        The present study was carried out to investigate the effects of nitrate ($KNO_3$) on the thyroid glands in rabbits which were administrated $KNO_3$ of 1g/kg/day for 6 weeks. Growth rate, and serum levels for $T_4$, $T_3$ and TSH were observed every weeks. The histological changes and the weights of the thyroid glands were observed in 6 weeks. The results obtained were summarized as follows: 1. The mean growth rates of experiments were decreased significantly in the 1st week, but were increased a little from the 5th to 6th week compared with those of controls. 2. The serum levels for $T_4$ of the experimental group manifested significantly decreased values than those of control through the experimental term consistently and the serum levels for $T_3$ were greatly decreased in the 3rd and 4th weeks. 3. TSH contents of the serum were not changed through the experimental term. 4. The mean weight of the thyroid gland in the experiment was decreased significantly after 6 weeks compared with that of control. 5. In the experimental group, the color of colloid in the thyroid follicles revealed deeper eosinophilic and the heights of the follicular epithelial cells were taller than those of controls. The colloid in the thyroid follicles revealed depletion. As summarized above, the observations suggest that nitrate can be an antithyroid substance in rabbits and it leads the thyroid glands to hypofunctional state.

        저농도 일산화탄소가 흰쥐 미상핵에 미치는 영향에 관한 조직화학적 연구

        This study was undertaken to investigate the changes of enzyme activities resulted from low concentrated carbon monoxide poisoning on the caudate nucleus in rat. The activities of cytochrome oxidase, succinate dehydrogenase and lactate dehydragenase were observed histochemically, after the experimental animals were poisoned to 100ppm carbon monoxide for 8 hours every day from one day to 16 days. The materials were sliced from coronal section at the level of the optic chiasm and immediately frozen sections of $10{\mu}m$ thickness were cut on the cryostat at $-15^{\circ}C$ and incubated in the medium containing substrate for histochemical detection of cytochrome oxidase, succinate dehydrogenase and lactate dehydrogenase. The sections were mounted in glycerol gelatin and observed under light microscope. It was obtained that cytochrome oxidase activity decreased moderately and succinate dehydrogenase activity showed marked or moderate activity during entire poisoning period and lactate dehydrogenase activity showed marked or moderate activity from one to 8 days but recovered to normal condition at 16th day.

        베이지안 학습을 이용한 문서의 자동분류

        As the number of online documents increases enormously with the expansion of information technology, the importance of automatic document classification is greatly enlarged. In this paper, an automatic document classification method is investigated and applied to UseNet 20 newsgroup articles to test its efficacy. The classification system uses Naive Bayes classification algorithm and the experimental result shows that a randomly selected newsgroup arcicle can be classified into its own category over 77% accuracy. 정보통신기술의 비약적인 발전은 온라인으로 생성되는 전자문서의 양을 폭발적으로 증가시키고 있다. 따라서 수동으로 문서를 분류하던 종래의 방법 대신 문서의 자동분유 기술 개발이 특별히 요구되고 있다. 본 논문에서는 베이지안 학습 기법을 이용하여 문서를 자동으로 분류하는 방법을 연구하고, 20개의 유즈넷 뉴스그룹 문서들을 분류하도록 시험하였다. 사용한 알고리즘은 Naive Bayes Classifier이며, 구현한 시스템을 이용해 유즈넷 문서를 대상으로 자동분류를 실험한 결과 분류의 정확률이 약 77%로 나타났다.

        불완전한 지식에서 정리증명을 위한 확률추론

        본 논문은 논리문장으로 표현된 지식을 처리하는 정리증명 과정에서 증명이 완료되기 전에 잠정적 결론을 유도하는 확률추론 기법을 제시한다. 정리증명 과정 중에 베이지안 해석을 이용하여 지식을 갱신하는 방법을 제시하고, 의사결정 방법을 사용하여 시간에 민감한 사안에 대해 신속하게 대처할 것인지 아니면 고의로 미룰 것인지를 결정하는 방법을 밝힌다. We present a probabilistic reasoning method for inferring knowledge about mathematical truth before an automated theorem prover completes a proof. We use a Bayesian analysis to update beleif in truth, given theorem-proving progress, and show how decision-theoretic methods can be used to determine the value of continuing to deliberate versus taking immediate action in time-critical situations.
