본 연구에서는 sesquiterpene류의 생물학적 변환을 위한 생촉매로서 목재부후균을 선발하고 생물변환 가능성을 확인하였다. Sesquiterpene류 합성 전구체로 알려진 trans,trans-farnesol을 기질로 하여 총 19종의 목재부후균에 대해여 저항성 실험을 실시하였다. 기질의 농도를 달리하여 저항성 테스트를 실시하여 생물변환 가능성을 갖는 5종의 균주를 선발하였으며 선정된 균주를 이용하여 액체배지 조건에서 trans,trans-farnesol의 생물변환을 시도하였다. GC/MS를 이용하여 분석한 결과 Laetiporus sulphureus var. miniatus (jungh) Imaz, Coriolus versicolor (L. Fr). Prlar, Fomitopsis pinicola, Lampteromyces japonicas 등 4개의 균주에서 각각 nerolidol, eicosane, isoborneol, isocyclocitral 등의 대사산물을 확인할 수 있었다. 이와 같은 결과로 sesquiterpene류의 생물학적 변환을 위한 생촉매로서 목재부후균의 가능성을 확인하였다. In this study, we screened and evaluated possibility of wood rot fungi as biocatalyst for biotransformation of sesquiterpenes. Screening were performed to select the most promising microorganisms with ability to biotransformation the substrate trans,trans-fanesol. Trans,trans-farnesol which is synthesized precursor of sesquiterpenes was used for resistance test on 19 of wood rot fungi. From the 19 tested wood rot fungi, 5 were selected by resistance test on different concentration of trans,trans-fanesol. Biotransformation was performed with selected wood rot fungi on liquid culture. The metabolites detected by GC-MS analysis were nerolidol for Laetiporus sulphureus var. miniatus (jungh) Imaz and eicosane for Coriolus versicolor (L.Fr) Prlar and isoborneol for Fomitopsis pinicola and isocyclocitral for Lampteromyces japonicas. As the results, wood rot fungi could be potential biocatalyst for biotransformation of sesquiterpenes.
신이화(辛夷花, Magnoliae Flos)추출물의 화장품약리활성 및 항염증효과에 관한 연구

본 연구에서는 신이화에 함유된 물질을 열수와 에탄올을 이용하여 추출한 후 화장품약리활성 및 항염증효과를 확인하고, 화장품 및 바이오산업에 적용하기 위한 천연소재로써의 이용 가능성을 살펴보았다. DPPH 라디칼 소거능 실험결과 농도의존적으로 소거능 효과를 보이며, 열수추출물의 경우 5,000 ㎍/㎖ 농도에서 대조군인 BHA보다 우수한 효능을 보였다. SOD 유사활성에서는 농도가 증가할수록 유사활성이 증가하는 것을 확인할 수 있었고, 5,000 ㎍/㎖에서 열수추출물의 경우 85% 유사활성을 나타내었고, 에탄올 추출물의 경우 43%의 유사활성을 나타내었다. Xanthine oxidase 저해능 실험에서도 5,000 ㎍/㎖ 농도에서 대조군 BHA 보다 높은 저해능을 효여 우수한 항산화효과를 보였으며, 합성항산화제를 대체할 수 있는 천연항산화제로 사용이 가능할 것으로 사료된다. 항염증효과를 확인하기 위하여 NO 저해 활성 및 iNOS 발현량을 측정하였는데, 열수와 에탄올 추출물 모두 농도가 증가할 수로 NO 저해활성과 iNOS 발현량이 감소하여 우수한 항염증효과를 보였다. 이상의 결과에서 신이화열수추출물과 에탄올추출물은 우수한 화장품약리활성 및 항염증효과가 있음을 확인할 수 있었고, 안티에이징 및 트러블 피부 개선 화장품 소재로서의 가능성을 확인할 수 있었다. Existing pharmaceutical studies show that Magnolia biondii is effective in treating rhinitis and in reducing cholesterol, given its endogenous, volatile ingredients. The study herein seeks to assess the cosmeceutical activities and anti-inflammatory activities of Magnolia biondii extracts for possible application as cosmetic ingredients. The cosmeceutical and anti-inflammatory activities were investigated using hydroxyl radical scavenging, superoxide dismutase (SOD)-like activity, xanthine oxidase (XO) inhibition, cell viability, nitric oxide (NO) inhibition, and inducible nitric oxide synthase (iNOS) expression by Western blotting. Magnolia biondii extracts were identified to have antioxidant activities in hydroxyl free radical scavenging, SOD-like activity, and XO inhibition. In testing the anti-inflammatory activities of the extracts, NO production was inhibited in a dose-dependent manner. Additionally, in a dose-dependent manner, the Magnolia biondii extracts were able to suppress iNOS expression in LPS-stimulated RAW 264.7 macrophage cells. From these results, Magnolia biondii showed adequate potential for application in cosmetic production and related industries as well as a functional material.
완전분해와 전분해 방법에 의한 영산강,섬진강수계 퇴적물의 중금속농도 상관관계

본 연구에서는 완전분해와 전분해 전처리 방법에 따른 중금속 농도 사이에 대한 상관관계를 알아보고자 하였다. 영산강·섬진강 수계 하천 및 호소에서 총 43점의 퇴적물 시료를 채취하여, 두 방법으로 전처리하고 5성분의 금속(Cd, Cr, Ni, Pb, Zn)을 분석하였다. 전체적인 중금속의 평균농도는 Zn>Cr>Pb>Ni>Cd 순으로 나타났으며, 전분해 값은 완전분해 값의 35.0% (Cd), 53.8% (Cr), 66.2% (Ni), 64.4% (Pb), 76.4% (Zn)로 완전분해시 중금속 농도가 높게 나타났다. 호소의 중금속 농도가 하천의 농도보다 높게 나타났다. 완전분해와 전분해 농도 사이의 상관관계식을 구하였으며, 강한 상관관계를 가지는 것을 나타난 Zn 및 Ni의 경우 완전분해 농도가 증가할 경우 왕수에 의한 용출농도도 같이 증가하는 것으로 나타났다. 나머지 중금속들에서도 두 분해법에 의한 퇴적물의 중금속농도 사이에 보통이상의 선형상관관계가 있는 것으로 나타났다. 본 연구결과는 퇴적물의 불균질성으로 인해 제한적으로 응용될 수 있을 것으로 판단된다. In this study, the correlations of concentrations in sediment heavy metals between two pretreatment methods, total digestion and aqua regia digestion, have been investigated. Total 43 samples had been collected from streams and lakes in Yeongsan and Seomjin watersheds. They were decomposed in two pretreatment methods and five metal components (Cd, Cr, Ni, Pb, Zn) were analyzed. Overall average concentrations of heavy metals were Zn>Cr>Pb>Ni>Cd in the order. The concentrations of aqua regia digestion were 35.0% (Cd), 53.8% (Cr), 66.2% (Ni), 64.4% (Pb) and 76.4% (Zn) of the concentrations of total digestion and heavy metal concentrations of total digestion were higher. Heavy metal concentrations in sediments from lakes were higher than those from streams. The correlation equations between the concentrations of total digestion and aqua regia digestion were obtained. The concentrations of Zn and Ni, which showed strong linear correlations, increased in aqua regia digestion as the concentration in total digestion increased. The linear correlation coefficients between two digestion methods for most analyzed metals were above the average correlation. However, these results can be applied with limitations due to heterogeneity of sediments.
국내 담수퇴적물의 CODsed 분석방법 평가: CODMn법과 CODCr법의 신뢰성 및 상관성 비교

In Korea, the chemical oxygen demand(COD(sed)) in freshwater sediments has been measured by the potassium permanganate method used for marine sediment because of the absence of authorized analytical method. However, this method has not been fully verified for the freshwater sediment. Therefore, the use or modification of the potassium permanganate method or the development of the new COD(sed) analytical method may be necessary. In this study, two modified COD(sed) analytical methods such as the modified potassium permanganate method for COD(Mn) and the modified closed reflux method using potassium dichromate for COD(Cr) were compared. In the preliminary experiment to estimate the capability of the two oxidants for glucose oxidation, COD(Mn) and COD(Cr) were about 70% and 100% of theoretical oxygen demand(ThOD), respectively, indicating that COD(Cr) was very close to the ThOD. The effective titration ranges in COD(Mn) and COD(Cr) were 3.2 to 7.5 mL and 1.0 to 5.0 mL for glucose, 4.3 to 7.5 mL and 1.4 to 4.3 mL for lake sediment, and 2.5 to 5.8 mL and 3.6 to 4.5 mL for river sediment, respectively, within 10% errors. For estimating COD(sed) recovery(%) in glucose-spiked sediment after aging for 1 day, the mass balances of the COD(Mn) and COD(Cr) among glucose, sediments and glucose-spiked sediments were compared. The recoveries of COD(Mn) and COD(Cr) were 78% and 78% in glucose-spiked river sediments, 91% and 86% in glucose-spiked lake sediments, 97% and 104% in glucose-spiked sand, and 134% and 107% in glucose-spiked clay, respectively. In conclusion, both methods have high confidence levels in terms of analytical methodology but show significant different COD(sed) concentrations due to difference in the oxidation powers of the oxidants.
리튬이차전지 양극재인 유기라디칼 고분자의 산화법에 대한 연구

라디칼 고분자화 반응 후 산화법을 이용하여 나티트록실 라디칼 고분자를 제조하였다. 두 가지 방법으로 산화된 라디칼 고분자의 라디칼 농도를 electroparamagnetic resonance spectroscopy(EPR) 법과 UV-visible absorption (UV-vis)방법을 통하여 측정하고 이를 통해 과산화수소수-Na2WO4법으로 산화하였을 때 라디칼 농도가 97.6% 정도 높게 얻어짐을 확인하였다. 또한 UV-vis 법은 정량적인 분석이 어려우나 대략적인 라디칼 형성 정도를 평가하는데 유용함을 확인하였다. 제조된 유기라디칼 고분를 리튬이온전지 양극재로 적용한 결과 우수한 용량, 초기효율, 높은 속도 특성을 가짐을 알수 있었다. An organic radical polymer (ORP) was prepared by radical polymerization and following oxidation intonitroxyl radical. Two different oxidation methods were employed and their radical concentrations were measured usingelectroparamagnetic resonance spectroscopy (EPR) and UV-visible absorption (UV-vis) spectroscopy. From these measurements,H2O2-Na2WO4 oxidation method exhibited a complete oxidation, which resulted in 97.6% spin concentration. Also, it was revealed that convenient and cheap UV-vis measurement was useful for preliminary radical concentrationcomparison. After applied as a cathode material in lithium ion batteries, ORP electrode showed a high initial capacity(110 mAh g-1), a good initial efficiency (96%), a very high rate performance (70% charging during 1.2 min) and stablecycle performance.
한국 이유자돈에서 검출된 돼지 유행성 설사 바이러스의 유전자 서열 분석

Porcine epidemic diarrhea virus (PEDV), an enveloped single stranded RNA virus in the family Coronaviridae, causes acute viral enteric disease in piglets. Recently outbreaks of porcine epidemic diarrhea (PED) have been rare in Europe but frequent in Asia. In Korea, the increase of PED prevalence is showing specially in postweaning pigs. The purpose of this study was to investigatenucleotide sequence of nucleocapsid protein gene of PEDV field isolates from postweaning pigs in Korea and get more information about the viruses. A total of 15 postweaing pigs clinically suspected of PEDV infection by severe watery diarrhea and dehydration were used in this study. Viral RNA was extracted from small intestines and stools of the pigs. The N gene was amplified by nested RT-PCR, purificated, sequenced, analyzed and then compared with published sequences of other PEDV strains. Three PEDVs were isolated from the suspected postweaning pigs. The N gene of three PEDV field isolates consisted of 483 nucleotides. These PEDV field isolates showed nucleotide sequence homology range from 99.6% to 95% with Chinese strains, from 99.8% to 95.2% with Korean strains, from 97.3% to 95.7% with Japanese strains and from 96.5% to 95.7% with Belgium and British strains. The encoded pritein shared range from 98.8% to 95.6% with Chinese strains, from 99.4% to 95% with Korean strains, from 97.5% to 96.3% with Japanese strains, from 95.6% to 95% with Belgium and British strains. By phylogenetic tree analysis based on nucleotide sequence, three PEDV field isolates were clustered into two groups which were Chinese isolate groups and other Korean isolate groups. These results indicated that some of PEDV field isolates prevailing in Korean postweaning pigs may be associated with those of Chinese strains and other Korean strains.
