        조개류(類)에 기생(寄生)하는 흡충류(吸蟲類)에 관(關)한 연구(硏究) V. 동죽, 가무락조개 및 맛조개에서 검출(檢出)되는 극구흡충류(棘口吸蟲類)의 유충(幼蟲)에 대(對)하여

        해산이매패(海産二枚貝)의 흡충류감염종(吸蟲類感染種)과 패류(貝類)의 병해(病害)를 조사할 목적으로 서해안(西海岸) 금강하구(錦江河口) 내초도산(內草島産) 동죽, 가무락 조개 및 맛조개에서 극구흡충과(棘口吸蟲科)의 피낭유충(被囊幼蟲)을 검출(檢出)하였고, 맛조개에서 검출된 피양유충(被襄幼蟲)을 괭이갈매기에 감염(感染)시켜 16일(日)만에 성충(成蟲)을 얻었다. 1. 동죽 및 가무락조개에서는 Himasthla kusasigi의 피낭유충을 검출(檢出)하였으며, 기생율(寄生率)은 각각 70.4%, 85.7% 이었다. 2. 조직부위별(組織部位別) Himasthla kusasigi의 기생율(寄生率)은 족부(足部) 40.4%, 77.3%, 아가미 17.0%, 12.4%, 외투막 12.6%, 10.3% 순으로 기생(寄生)되었다. 3. 맛조개에서 Acanthoparyphium속(屬) 흡충(吸蟲)의 피낭유충(被囊幼蟲)을 검출(檢出)하였으며, 기생율(寄生率)은 63%, 조직부위별 기생율은 foot, mantle, gill 순(順)으로 기생(寄生) 되었다. 4. 맛조개에서 검출(檢出)된 피낭유충(幼蟲)을 괭이갈매기에 감염(感染)시켜 16일만에 성충(成蟲)을 분리하여 충체(蟲體)의 내부(內部)구조를 조사한바 Acanthoparyphium tyosenensis Yamaguti, 1938로 동정(同定) 되었다. 5. 동죽 및 가무락조개는 Himathla kusasigi의 새로운 중간숙주(中間宿主)로 추가되며, 본인(本人)(1968)등(等)이 개량조개에서 검출보고(檢出報告)한 Echinostoma속 흡충(吸蟲)은 Acanthoparyphium tyosenensis흡충(吸蟲)의 유충(幼蟲)임이 밝혀졌다. An experiment was carried out in order to reveal the infection species of trematode and the infection demage in the marine bivalves. Metacercaria of Echinostomatidae were found in Mactra veneriformis, Cyclina sinensis and Solen strictus in the vicinity sea of Naecho-do, the estuary of the Keum River in the western coast of Korea. The metacercaria detected from Solen strictus were fed by Larus crassiostris, at 16 days after infection, the adult worm could be seceded. The metacercaria of Himasthla kusasigi were detected from Mactra veneriformis and Cyclina sinensis, their infection rates were 70.4% and 85.7% respectively. The partial infection rates with respect to the body parts were 40.4% and 77.3% in the foot, 17.0% and 12.4% in the gill, 12.6% and 10.3% in the mantle in order. The metacercaria of Acanthoparyphium were found from Solen strictus. The total infection rate was 63% and the partial infection rate was 63% and the partial infection rate was shown foot, mantle and gill in order. The adult worm ceded from Larus crassiostris was indentified as Acanthoparyphium tyosenensis Yamaguti, 1938 from view point of its physical characteristics. Therefore, Mactra veneriformis, Cyclina sinensis would be added as new intermediate hosts, and Echoinostoma decteded from Mactra sulcataria which was studied by author(1969) was revealed as larvae of Acanthoparyphium tyosenensis.

        The marine environmental characteristics of the vicinity of Nampo sea area were investigated in order to assess the environmental variation and the ecological influences by the project of reclaimation from February to May, 1986. At flood tide, the water coming from the southwest of Kabampo turned to counter-clockwise and flowed to the in front of Kapbawi with the speed of 0.4-1.2 knot. Whileas, at ebb tide, the current turned to the clockwise in front of Kapbawi and then moved to the off sea with 0.4-1.3 knot. Although the levels of nutrient salts were relatively low, but the SS level was higher compared with that of other areas due to the reclaimation construction. As a whole, the procuctivity of this area is poor state compared with other coastal areas.

        뱀장어 아가미 울혈증에 관한 연구 III. NaCl처리가 울혈증 치료에 미치는 영향

        This experiment was carried out in order to elucidate the effect of NaCl(0.85%) acclimation on congestion symptoms in central venous sinuses(CVS) of gill filaments and on the alteration of blood constituents in cultured eels for 1, 2, 3, 4, 5, 6 and 7days after handling stress. After giving a handling stress on eels, the frequency of gill congestion was about 90%. It gradually decreased to 40% on the 3rd day, then increased to 60% on the 7th day in NaCl(0.85%) acclimation. The values of haematocrit(Ht), methemoglobin(Met-Hb), glucose (GLU), calcium($Ca^{++}$), lactic dehydrogenase(LDH), glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase(GOT) were lower than those of the control, whereas magnesium($Mg^{++}$) and haemoglobin(Hb) were higher than the control. However total protein(Tp) and albumin (ALB) levels similar to the control. When the fish after giving a handling stress were cultured in NaCl-free water as a control, the frequency of gill congestion and the value of blood components showed a similar pattern to that obtained from the NaCl acclimated experiments. However, in the early period(1-3day) after handling stress, NaCl acclimating slightly helped the natural tendancy of recovery. These results suggest that the frequency of gill congestion is not always reduced over the whole period of NaCl acclimation. However, the acclimation to NaCl solution may be a somewhat useful therapy for the gill congestion in stressed eels. 뱀장어에 인위적으로 스트레스를 가하여 아가미 울혈증을 발생시킨 후에 NaCl 처리에 따른 CVS의 울혈증의 치료효과 및 혈액성상의 변화에 미치는 영향을 조사하였다. 스트레스 직후에 고빈도의 발병율을 보였던 아가미 및 간의 울혈증은 염처리 후 3일째 까지는 감소되었으나, 4일 이후에는 별 영향이 없었다. 혈액성분중 스트레스 후 증가되었던 Ht, Met-Hb, $Ca^{++}$, GOT, GPT, LDH 및 glucose치 등과 감소되었던 $Mg^{++}$ 및 Hb치 등은 염처리 3일까지는 회복율이 촉진되었으나, 그 이후에는 큰 차이를 보이지 않았다. 그러나 알부민 및 총단백치는 염처리 기간동안 별 변화가 없었다. 이상의 결과는 스트레스에 의해 발병된 아가미 울혈증에 대한 염처리법은 초기 3일까지는 효과가 있으나, 그 이상의 기간에는 전혀 효과적이지 못함을 보여준다.

      • pUC8 플라스미드 운반체에 핵다면체 바이러스 DNA절편(일부 Pst 1 Genome)의 클로닝

        핵다면체 바이러스(Baculovirus, Nuclear Polyhedrosis Virus) 게놈 DNA는 조직배양 세포에 바이러스를 감염시켜 분리하였으며, 분자량은 약 127.6kb였다. BNPV DNA 게놈을 제한효소 Pst I으로 절단한 후 그들의 일부절편을 pUC8 플라스미드 운반체에 클로닝하여, E. coli JM83세포에 형질전환하였다. BNPV DNA 게놈을 Pst I 효소로 저단했을 때 16개의 절편으로 분리되었으며, 분자량범위는 1.01에서 25.06kb 사이였다. 그 중 6개의 Pst I 절편 DNA을 클로닝하고 다시 Pst I 효소로 절단하여 아가로스 겔 전기영동을 통하여 확인하였다. 이 결과로 viral DNA 게놈의 gene library는 약 37.5%였다. 또한 바이러스 DNA 절편이 클로닝된 형질전환체와 E. coli JM83 균주를 SDS-polyacrylamide gel 전기영동하였다. 이들 결과들은 단백질 밴드의 pattern에 있어서 어떤 차이점도 나타나지 않았다. The complete genome of Baculovirus (Nuclear Polyhedrosis Virus, BNPV), which was isolated from tissue culture cells infected with the virus, was digested with Pst I endonuclease and cloned in E. coli JM83 using the plasmid vector pUC8. The molecular weight of genome DNA of BNPV is 127.6kb. BNPV genome DNA digested by the Pst I enzyme was divided into 16 fragment, with the molecular sizes ranging from 1.01 to 25.06kilobases. Six Pst I DNA fragments were cloned and identified by digestion of the recombinant DNAs with Pst I enzyme by comparison of their electrophoretic mobilities in agarose gel. The gene library was showed about 37.5% of the viral genome. The total cell polypeptides of the all transformants and E. coli JM83 were analyzed on the SDS-polyacrylamide gel electrophoresis. These results didn't show any significient differences in protein band patterns.

      • 한국산 메기(Silurus asotus)의 질병에 관한 연구 Ⅱ. 비브리오병에 관하여

        발병한 양식 메기로부터 vibrio속 균주를 분리하였다. 분리된 균주는 생물학적 생화화적 특성에 기초하여 동정한결과 V.ordalii로 동정되었다. 분리균은 glucose, lactose, maltose와 salicine으로부터 산을 생산하였으며, arabinose, galactose, inocitol과 xylose는 이용하지 못하였다. 분리된 V.ordalii를 각각 KL-1, KL-2라 명명하였다. KL-1과 KL-2는 생리학적 특성이 유사하였다. 즉, pH 5-10, NaCl 0%-6.0%에서 발육하였으며, 또한 NaCl 7.0%, pH 10이상 그리고 pH5이하에서는 발육하지 않았다. KL-1균주를 건강한 메기에 인공 감염시킨 결과 양식장에서 발병된 증상과 동일한 출혈성 궤양이 유발되었다. 감염 24시간 후에 나타난 붉은 반점은 접종 부위 주변에서부터 확장되기 시작했으며, 감염 120시간 후부터는 배지느러미 부위까지 궤양이 확장되었다. 각각 온도에 따른 실험에서는 25℃에서 폐사율이 70% 로 나타났다. 약제 감수성 시험에서 KL-1과 KL-2 균주는 모두 GM, K, N, S와 SxT에 감수성을 나타내었으나, CF 및 L₂와 VA에는 저항성을 나타내었다. Two vibrio sp. strains were isolated from disease catfish(Silurus asotus). The present isolates were identified as Vibrio ordalii based on their biological and biochemical characteristics: they were positive for acid production from glucose, lactose, maltose, sucrose and salicine, while negative for arabinose, galactose, inocitol and xylose. They are named KL-1 and KL-2. KL-1 and KL-2 strains were similar to physiological characteristics : growth was observed at pH 5 to 10 and in 0% to 6.0% NaCl. Two strains did not growth at a concentration above 7.0% NaCl and pHIO. This bacterium was injected into health catfish hypodermically. Such injection was found 10 induce haemorrhagie ulcers very similar to those observed in naturally infected fish. At 24h post-infection, the red spot developed around the injection site and grew bigger to from a red Sport area. At 120h post-infection, the muscle ncerosis was extended near the ventral fin. The seventy percent lethal dosage was appeared to water temperature at 25℃. Two strains were tested for drug senistiveity by plate method. KL-l and KL-2 strains were sensitive to GM. K. N. S and SxT, but resistant to CF. L₂and VA.

        Acanthoparyphium tyosenense Yamaguti , 1939 의 생활사에 관한 연구

        1999년 3월부터 2000년 3월까지 서해안 만경강 하구역의 심포리지선 간석지에서 채집한 갯우렁이 (Lunatia fortuni)와 큰구슬우렁이 (Neverita didyma)에서 Cercaria yamagutii Ito,1957를 검출하고, 실험실에서 이매패류인 동죽 (Mactra veneriformis)에 감염시켜 피낭유충이 형성됨을 확인하였고, 동 실험에서 형성된 피낭 유충을 괭이갈매기 (Larus crassiostris)에 경구 감염시켜 10일 후 성충을 얻어 Aconthoparyphium tyosenense Yamaguti 1939임을 확인하여 지금까지 밝혀지지 않았던 이 충의 전 생활사를 밝혀냈다. 제1중간숙주는 해산 복족류인 갯우렁이, 큰구슬우렁이, Tympanotonus microptera와 Cerithides (Cerithidea) largillierti, Cerithidea (Cerideopsilloa) cingulata이고, 제 2중간숙주는 이매패인 동죽, 개량조개, 맛조개, 바지락 이며, 종숙주는 괭이갈매기와 물오리의 일종인 Melanitta fusca stejnegeri이다. 갯우렁이와 큰구슬우렁에서 검출한 Cercaria yamagutii를 실온 25℃ 전후에서 동죽이 들어있는 수조에 넣은 후 30분 이내에 입수공을 통하여 체내에 침입하였으며, 5시간 경과시 꼬리가 탈락되고 둥글게 피낭을 형성하기 시작했으며, 감염후 30시간 (14일)에는 피낭의 크기 300∼360㎛, 탈낭유충 790∼800×300∼310㎛ 크기로 활발히 신축 운동하는 성숙한 유충으로 발육되었다. 자연산 이매패에서 Acanthoparyphium tyosenense의 피낭유충 기생율은 동죽 99.5% , 맛조개 76.3% , 바지락 37.0%이었고, 피낭유충의 부위별 기생율은 동죽의 경우 검출된 피낭유충 총 18,064개중 족부 근육 62.9% (11,368개), 아가미 23.6% (4,258개), 내장낭 13.5% (2,435개)의 기생율을 나타냈다. 피낭유충은 숙주 종이나 크기와 관계없이 나타났다. Cercaria yamagutii Ito, 1957 (C. yamagutii) was found in Lunatia fortuni (L. fortuni) and Neverita didyma (N. didyma) collected from the tideland of Sim-po located at the estuary of the Mankyong River, Chonbuk. It was finally confirmed that the parasite is Acanthoparyphium tyosenense Yamaguti, 1939 (A. tyosenense) and its life history was clarified in this study. Mactra veneriformis (M. veneriformis) was artificially infected with C. yamagutii isolated from L fortuni and N. didyma. It began to intrude into M. veneriformis through the inhalent canal. Five hours after infection, the tails of the cercaria began to be separated from the main body and the cercaria started to form cysts. Mature cysts were formed 340 hours (14 days) after infection. The cysts were 300∼360㎛ in diameter and the excysted metacercarias were 790∼800 ×300∼310㎛ in size. The metacercarias were administered orally to Larus crassiostris (L crassiostris), and adult worms of 84.5∼112.5 ×55∼65㎛ were found full of eggs with 2.20∼3.70㎜ long and 0.40∼0.59 ㎜ wide after 10 days. In a field study, it was observed that the infection rate of A. tyosenense is 99.5% in M. veneriformis, 76.3% Solen strictus (S. skrictus), and 37% Ruditapes philippinarum (R. philippinarum). No difference was found among different host sizes. It was concluded that the first intermediate hosts of A. tyosenense Yamaguti were L fortuni, N. didyma, Tympanotonus microptera, Cerithidea (Cerithidea) largillierti, Cerithidea (Cerideopsilloa) cingulata, the second intermediate hosts M. veneriformis, S. strictus and R, philippinarum, and the final hosts L. crassiostris and Melanitta fusca stejnegeri.

        소비자의 와인 선택속성에 따른 기대-불일치가 구매행동에 미치는 영향

        This study is about the impacts of expectation disconfirmation in wine attribute on the consumer’s buying behavior especially as the wine industry grows rapidly it shows high level of competition among the firms the study will be suggest competitiveness with in effective marketing strategic. Therefore, the study choose five wine selection attribute which as product factor, preparation factor, buying information factor, environmental factor and value factor for analyzing that the impact of before wine purchase and after wine purchase achievement on expectation disconfirmation on the a buying behavior. First, the wine selection attribute factor impacts as positive aspect on consumer’s after purchase achievement. Second, the before wine purchase expectation factor was not impact on the consumer’s expectation disconfirmation. Third, the study indicates that when the after wine purchase achievement is high the expectation disconfirmation show as positive aspect. Forth, the demographic factors shows significance differences within product factor, purchase information factor, environmental factor and value factors. Fifth, according to difference verification result shows different levels of purchase information factors with the education level, age and income.

        한국산 메기의 질병에 관한 연구 2 . 비브리오병에 관하여

        Two vibrio sp. strains were isolated from disease catfish(silurus asotus). The present isolates were identified as Vibrio ordalii based on their biological and biochemical characteristics; they were positive for acid production from glucose, Iactose, maltose, sucrose and salicine. while negative for arabinose. galactose, inocitol and xylose. They are named KL-1 and KL-2, KL-1 and KL-2 strains were similar to physiological characteristics: growth was observed at pH 5 to 10 and in 0% to 6.0% NaCl. Two strains did not growth at a concentration above 7.0% NaCl and pH10. This bacterium was infected into health catfish hypodermically. Such injection was found to induce haemorrhagic ulcers very similar to those observed in naturally infected fish. At 24h post-infection, the red spot developed around the injection site and grew bigger to from a red sport area. At 120h post-infection, the muscle ncerosis was extended near the ventral fin. The seventy percent lethal dosage was appeared to water temperature at 25℃. Two strains were tested for drug senistiveity by plate method. KL-1 and KL-2 strains were sensitive to GM. K, N, S and SxT, but resistant to CF. L and VA

        연어 세포주의 전염성 췌장과사 바이러스의 감염 증상에 관한 연구

        CHSE(Chinook Salmon Embryo)-214 fish cell lines was cultured in Eagle`s minimal medium (MEM) supplemented with 10% fetal bovine serum and 2mM-glutamin. Optimum growth temperature of CHSE-214 cell line was 20℃. Infectious pancreatic necrosis virus(IPNV) was successfuly multiplied and showed the cytopathic effect in CHSE-214 cell line. Infection symptom of IPNV was observed with inverted phase contrast microscopy. At 6h-12hrs post-infection, the cells infected with IPNV were similer to normal cells. At 18-24hrs post-infection, the cells were somewhat round form and a little swollen form than normal cells. At 30hs post-infection, the cells were becoming more abnormal cells. At 48-68 post-infection, the infected cells were lysed and showed the severe cytopathic effect.
