        콩 Hypocotyl에서 Acetyl Soyasaponin $A_1$의 분리 및 구조 분석

        본 연구는 국내육성 콩 품종 및 전통식품의 우수성을 입증하고, 생리활성이 우수한 고품질의 신품종 육성을 위한 기초기반기술 확립의 일환으로 group A soyasaponin의 분리 및 정제에 관한 연구를 수행한 결과를 요약하면 다음과 같다. 1. 콩 종실로부터 hypocotyl을 분리하고 탈지한 후 automated solvent extractor(ASE)를 이용하여 MeOH 추출 후 flash column($150mm{\times}40mm$ i.d.)을 이용하여 총 14개의 fraction을 분리하였다. 2. Flash column으로 얻어진 8 및 9번 fraction(Fraction-I) 을 Luna $C_{18}$ semipreparative reverse phase column($250cm{\times}50mm$ i.d.)을 이용하여 Fast PCLC로 정제하여 미지화합물(unknown compound : UKC)(Fr-I-2)을 분리하였다. 3. Compound 1을 NMR($^1H-NMR$, 600 MHz; $^{13}C-NMR$, 100 MHz; DEPT), IR, UV 및 ESI-MS 분석을 통하여 구조를 동정한 결과 분자량(MW)이 1436.6이며, 분자식이 $C_{67}H_{104}O_{33}$인 group A 계열의 acetyl soyasaponin $A_1$으로 확인되었다. Soyasaponins are phytochemicals of major interest fur their health benefits. Chemical investigation of a soybean phytochemical concentrate resulted in the isolation and identification of triterpenoid saponins. The MeOH extraction of defatted hypocotyl separated from soybeans was peformed by the automated solvent extractor (ASE). Fractionation was performed on a flash column ($150mm{\times}40mm$ i.d.) packed with a preparative $C_{18}$ reverse phase bulk packing material $(125\AA,\;55-105{\mu}m)$ and monitored at 210 nm, and collected 14 fractions. Consequent Fsat preparative column liquid chromatography (Fast PCLC) was performed for the purification of Fraction-I (Fr-I) collected from the fraction 8 and 9 of flash chromatography. Fsat PCLC was performed on a Luna $C_{18}\;10{\mu}m,\;100{\AA}$, semipreparative reverse phase column ($250cm{\times}50mm$ i.d.) for the purification of isolated unknown compound (Fr-I-2). Chemical structure of acetyl soyasaponin $A_1\;(MW:1436.6,\;C_{67}H_{104}O_{33})$ was identified and determined by a combination of extensive NMR ($^1H-NMR$, 400 MHz; $^{13}C-NMR$, 100 MHz; DEPT), IR, UV, and ESI-MS analysis.

        옥수수 종실 및 속대의 Phytosterol 동정과 함량 변이

        옥수수 종실(kernel)과 속대(cob)의 불검화물(ZML) 중 phytosterol의 조성과 함량의 변이를 구명하여 고품질 옥수수 신품종 육성 및 생리활성물질의 유용 소재화를 위한 기초자료로 활용하고자 본 연구를 실시하여 얻어진결과를 요약하면 다음과 같다. 1. 옥수수 종실과 속대의 포화지방산은 phytosterol 함량과 부의 상관관계의 경향이었고, 불포화지방산 중 stearic 및 linoleic acid는 정상관 경향을 보였으나, linolenic acid는 종실에서 정상관($r=0.652^*$), 속대는 부상관($r=-0.505^*$) 관계를 보였다. 2. 옥수수 종실의 불검화물을 TLC로 분리한 결과 band I (campesterol, stigmasterol, ${\beta}$-sitosterol), band II (${\Delta}^5$-avenasterol), band III (${\Delta}^7$-stigmastenol), 및 band IV (${\Delta}^7$-avenasterol)로 뚜렷하게 분리되었고, 속대는 band I~IV 이외에도 3종 이상의 band가 추가적으로 분리되었다. 3. 옥수수 종실과 속대에 함유된 phytosterol의 GC 분리 패턴을 확인한 결과 campesterol, stigmasterol 및 ${\beta}$-sitosterol의 분리능이 좋았으나, ${\Delta}^7$-avenasterol (RT 22.846), ${\Delta}^7$-stigmastenol (RT 22.852) 및 ${\Delta}^5$-avenasterol (RT 22.862)은 혼합물질 상태로는 분리가 되질 않았다. 4. 옥수수 종실의 평균 phytosterol 함량은 635.9 mg/100g, 속대는 273.0 mg/100 g으로 종실이 속대에 비해 약 2.4배 정도 phytosterol 함량이 높았다. 옥수수 종실의 phytosterol 조성은 ${\beta}$-sitosterol 80.05% > campesterol 10.5% > stigmasterol 9.46% 순이었으나, 속대는 ${\beta}$-sitosterol 59.43% > stigmasterol 31.72% > campesterol 10.98%으로 종실과 속대의 phytosterol 조성비는 다소 상이하였다. 5. 본 연구 결과를 토대로 판단할 때 옥수수 종실에 함유된 ${\beta}$-sitosterol, campesterol 및 stigmasterol의 생합성 경로에서 전구물질이 되는 ${\Delta}^7$-avenasterol, ${\Delta}^7$-stigmastenol 및 ${\Delta}^5$-avenasterol이 옥수수 속대에서 검출되는 것으로 보아 옥수수 종실의 phytosterol은 속대에서 합성되어 종실로 전이되는 물질인 것으로 추정되었다.

        콩 종피의 이화학적 특성과 광택과의 관계

        콩 종피의 이화학적 특성과 종실광택과의 상호관계를 구명하고자 본 연구를 실시하여 얻어진 결과를 요약하면 다음과 같다. 1. 콩 종피 표면의 전자현미경으로 관찰 결과 L값이 높은 콩의 종피는 표면이 매끄럽고 pore의 크기가 작았으나 L값이 낮은 콩의 종피는 표면이 거칠고 pore가 비교적 큰 것으로 나타났다. 2. 종피두께가 $70{\mu}m$에 달할 때 까지는 종자의 광택이 증가되지만 그 이상에 달하면 오히려 종실광택이 감소되었고, 종피두께가 두꺼울수록 수분흡수율이 낮으며, 특히 수분흡수 초기에 경향이 뚜렷하였다. 3. 종실광택이 높을수록 수분흡수율은 증가되는데, 수분흡수 초기에는 종실광택의 영향이 작지만 시간이 경과됨에 따라 종실광택이 밝은 품종일수록 수분흡수율이 높았다. 4. 종피의 단백질은 종실광택과 부상관이 있었으나 단백질의 구성성분인 아미노산은 종실광택과 상관이 없었다. 5. 종피의 조지방은 종실광택과 부상관적 요인으로 작용하나 종피에 함유된 불포화지방산은 종피의 광택과 정상관이 있는 것으로 나타나 종피의 광택은 지방산중 불포화지방산의 조성비율이 높을수록 종실이 밝았다. 6.종피의 조섬유 함량은 종실광택과 이차선형관계가 인정되어 종실광택과 밀접한 관계를 맺고 있는 성분으로 판단되었다. 7. 종피의 중성단당류로는 fucose, rhamnose, glucose, mannose, galactose, arabinose, xylose가 함유되어 있었으며, 이들 중 arabinosefl xylose는 종실광택과 정상관이 있었으나 조성비가 가장 높은 galactose는 유의성이 없었다. Lipid and protein contents in whole soybean seeds have negative correlation (r=-0.693**), however, these components in seed coat showed positive correlation (r=0.746**). Fatty acids in whole soybean seeds were higher in the order of $C_{18:2}>C_{18:1}>C_{16:0}>C_{18:3}>C_{18:0}$, while those of seed coat were higher in the order of $C_{18:3}>C_{18:2}>C_{18:0}>C_{16:0}>C_{18:1}$. The average content of total amino acid in twenty Korean soybean varieties was 38,938.7 mg/100 g, while that of seed coat was 4,418.4 mg/100g. Glutamic acid showed the highest composition rate $(16.4\%)$ in whole soybean seeds, while glycine was the highest in seed coat and their composition rate was $23.8\%$. The surface of shiny-lustre seed coats was smooth and their pore size was observed smaller than dull-lustre ones. Significant quadratic regression was observed among seed coat lightness, seed coat thickness, protein, lipid, unsaturated fatty acid and crude fiber. Fucose, rhamnose, glucose, mannose, galactose, arabinose and xylose were detected as a neutral mono-saccharides in the seed coats. The arabinose and xylose showed significant correlation with seed coat lightness. The unsaturated fatty acid was significantly correlated with seed coat lightness (r=0.726**). Water absorption rate was low in the thick seed coat varieties, but the rate was high in the shiny seed coat varieties. From the obtained results, it was considered that the thinner and brighter seed coat varieties were much favorable to increase the water absorption rate than thicker and darker seed coat ones.

        We analyzed rutin content using mulberry genetic resources. They were grown under the same environment and conditions. Mulberry leaves were collected and then freeze-dried and powdered for rutin test. As a result, mean content of mulberry strains was 0.38 ± 0.17%, and the coefficient of variation (CV) was 45.7%. The variation was greatly severe. Among the tested strains, ‘Jangloe’ was showed the highest content of 0.97%, whereas ‘Sungsu 3’ was showed the lowest content of 0.01% respectively. Finally we selected rutin high-containing 6 strains. They are as follows. ‘Jangloe’, ‘Pumbo 6’, ‘Youjin’, ‘Kaeryangjeonjeon’, ‘Baekhak’, and ‘Pumbo 32’ which are more than twice of the overall average content.

        연구보문 : 풋옥수수 수확 후 예냉처리와 저온저장에 따른 품질 변화

        Quality attributes of fresh waxy corn (Zea mays L.) as affected by pre-cooling and storage temperature (4˚C and room temperature) were investigated in terms of weight loss, moisture, color, reducing sugar content, and sensory quality. Two varieties of waxy corn with white and black kernel color, Ilmichal and Heukjinjuchal respectively, were cultivated at the field of National Institute of Crop Science in 2007 and 2008 to determine the physiologically optimal storage condition for higher eating quality stored in refrigerator of 4˚C and room temperature. They are pre-cooled immediately after harvest with ice for 5 hours. Quality analyses were monitored during 10 days at 4˚C storage room comparing with those of non cooled and stored at room temperature. A higher weight loss over 15% observed in both of two waxy corn varieties that were stored at room temperature. But there was not significantly difference with variety or pre-cooling treatment. There was up to 10% increase in moisture content in kernels non pre-cooled and stored at room temperature. For all treatments, very small amount of free sugar were detected, however, there were no significant differences between treatments. Glucose concentration increased with time in kernels stored at 4˚C and the effect of pre-cooling was not significant. A general darkening in color of kernels and a trend to turn yellow was observed in the pre-cooled corns when they were steam-cooked. Total sensory scores of corns stored in 4˚C were better than those of samples stored in room temperature and there was not significantly difference with variety or pre-cooling treatment. As storage period was longer, shelf lifes of waxy corn in control and waxy corn treated with technique applied to store waxy corn, and the quality of waxy corn could be maintained and the storage expense could be saved during storage. It is required that further studies narrow down the effect of pre-cooling on fresh waxy corn during short time storage.

        벼종자 미랑 단백질의 프로테오믹스 연구를 위한 글루테린 저장 단백질의 제거방법

        본 연구는 벼종자 미량 단백질의 프로테오믹스 연구를 위하여 벼종자에 고 함량으로 존재하는 벼종자 글루테린 저장 단백질을 제거하는 방법에 관한 것이다. 따라서 본 연구는, (A) 벼종자에 액체 질소를 가하고 분쇄하여 벼종자 가루를 만드는 분쇄단계; (B) 상기 분쇄된 벼종자 가루를 물에 현탁하여 현탁액을 만드는 현탁단계; (C)상기 현탁액 중 미용해 물질을 제거하는 분리단계를 포함하는, 벼종자 미량 단백질의 프로테오믹스 연구를 위한 벼종자 글루테린 저장 단백질의 제거방법에 관하여 검토하였다. 본 연구의 결과, 단순하고 신속하며 저렴하고 효율적인 방법으로 미량 비글루테린 단백질들을 용이하게 동정할 수 있을 것으로 판단되었다. Abundant proteins often cause problems in proteome study. Glutelin family proteins (hereafter referred to glutelin) are present in rice proteome sample as over-whelming constituents with very high abundance. In order to increase the number of identified proteins in rice proteome study, we developed a newly improved method for sample preparation through the removal of glutelin. When the protein samples from rice seed were extracted by the conventional trichloroacetic acid (TCA) extraction method, glutelin accounts for about 60% of total rice seed proteins in SDS gels. Using our new water extraction method, glutelin consists of only about 10% of total proteins. After analyzing on a two-dimensional gel electrophoresis (2-DE), 937 protein spots were detected using the conventional TCA extraction method. On the other hand, 1240 proteins could be seen using the new water extraction method. The selectivity for non-glutelin and less abundant protein by the water extraction method was also confirmed by ESI-Q/TOF mass spectrometry analysis. Thus, the new water extraction method developed here can be efficiently used to study the proteome analysis of rice storage seed.

        서해안 간척지 담수호 및 유입지천에서의 수질 특성

        This study investigated water quality of freshwater lakes and inflow streams in the six west reclaimed lands (Shihwa, Hwaung, Sukmoon, Iweon, Nampo and Saemangeum) once every two months from April to October in 2015. The measured pH and electrical conductivity of the water samples ranged 7.6~9.2 and 0.004~0.426 ds m-1, respectively. The concentrations of dissolved oxygen (6.9~10.5 mg L-1), biochemical oxygen demand (0.2~18.1 mg L-1), chemical oxygen demand (3.4~100.6 mg L-1), total organic carbon (0.2~8.9 mg L-1), suspend solid (2.4~86.3 mg L-1), total nitrogen (0.2~4.4 mg L-1), and total phosphorus 0.03~0.21 mg L-1 also varied in a wide range. Correlation of water quality analysis with concentration of EC between COD and TOC was higher. Recently, watershed management system necessary to sustainable water quality monitoring of agricultural water for water environmental conservation in water pollution. It is need to be ensure the water quality of reliable agricultural water in the watersheds. Our results can provide some useful information for deciding water quality management plans at freshwater lake and watersheds.
