        미세먼지 PM10으로 손상을 유도한 각질형성세포에서 청각 (Codium fragile) 추출물의 항노화 효과

        Objectives : Fine dust caused by environmental pollution cause oxidative damage and skin aging. In this study, The possibility of using the Codium fragile extract (CFE) as an anti-aging product for skin improvement was evaluated by confirming the protective effect of skin cells from PM10 (particulate matter 10) through inhibition of ROS and MMPs. Methods : In this study, elastase and collagenase inhibitory activities were evaluated. Cell viability was evaluated by treating keratinocytes (HaCaT cell line) with CFE at various concentrations. The cytoprotective effect from PM10 in keratinocyteswas evaluated using the 3-[4,5-dimethylthiazol]-2-yl]-2,5-diphenyl-tetrazoliumbromide (MTT) assay. ROS (reactive oxygen species) was measured in keratinocytes damaged by PM10 using DCF-DA (2′,7′-dichlorofluorescin diacetate) fluorescence staining. As an anti-aging effect of CFE, MMP-1 (matrix metalloproteinase-1) and MMP-1 (matrix metalloproteinase-9) inhibitory activities were evaluated. Results : As a result, CFE decreased the activity of elastase and collagenase. As a result of evaluating the toxicity of CFE, it is non-toxic at a concentration of 10 to 80 ㎍/㎖. Although cell viability of HaCaT cells treated with PM10 decreased, cell viability increased by 38% when treated with CFE 80 ㎍/㎖. Also, ROS decreased by 8.4%, and MMP-1 and MMP-9 decreased at CFE 80 ㎍/㎖. Conclusions : CFE showed excellent cell protection effect, and it is considered that it can be used in anti-aging products for skin improvement by effectively inhibiting ROS and MMPs from keratinocyte damage caused by fine dust.

        자외선 B로 산화적 손상이 유도된 HS68 세포에 익모초 추출물의 효능 평가

        Objectives : The aim of this study is to investigate anti-inflammation of Leonurus sibiricus methanol extract against UVB-damage in fibroblast. The skin is continuously exposed to damage from environmental stresses. UV radiation causes a variety of biological effects especially on the skin, including inflammation and photoaging. Methods : In this study, we tried to search for Leonurus sibiricus which exhibit protective activities against UVB-induced cytotoxicity and oxidative cell death, NO and $PGE_2$ production. HS68 cells were exposed to UVB ($120mJ/cm^2$) and treated with various concentrations (0, 0.25, 0.5, 1, 2, 4, $8mg/m{\ell}$) of Leonurus sibiricus methanol extract for additional 24 h. Intracellular reactive oxygen species (ROS) levels generated by UV radiation were detected using a spectrofluorometer after DCF-DA staining. Also, HS68 cells were irradiated with UVB and then treated with Leonurus sibiricus methanol extract for 12 h. The lipid peroxidation was assayed by measuring the levels of 8-isoprostane secreted into the culture medium. Results : UVB-induced cytotoxicity and cell death were effectively suppressed by treatment of Leonurus sibiricus aqueous methanol extracts. Oxidative cell damage was mediated $PGE_2$ in UVB-induced HS68 fibroblast cell, which was significantly inhibited by treatment with Leonurus sibiricus extracts. Also, the protective effect of these extract seemed to be mediated by inhibited intracellular ROS generation and lipid peroxidation in dose-dependent manner. Conclusion : These results suggest that Leonurus sibiricus aqueous methanol extracts may have anti-aging effects new functional materials against oxidative UVB stress-mediated skin damages.

        잘피 추출물의 UVB로 손상을 유도한 각질형성세포에 대한 항염 효능

        Objectives : In order to confirm whether extracts of different parts of Zostera marina (ZM), a marine flowering plant, can be used as cosmetic ingredients, this study evaluated their cytotoxicity and cytoprotective effects against ultraviolet B (UVB). Inflammatory responses induced by UV stimuli are also associated with the aging of the skin. Methods : We investigated the effects of ZM extracts on cells through the water soluble tetrazolium salt-1(WST-1) assay for cell viability. In order to investigate the anti-inflammatory effects, we evaluated the suppression of Cyclooxygenase-2 (COX-2) expression by ZM extracts in HaCaT cells with UVB-induced damages, and also evaluated the production of Prostaglandin E2 (PGE2) in RAW 264.7 cells with LPS-induced damages. Results : High cell viabilities above 90% were observed in all types of ZM extracts, except for whole ZM extract at 0.5 mg/ml; in keratinocytes with UVB-induced damages, the cell viabilities were above 80% when treated with all types of ZM extracts. We confirmed their anti-inflammatory effects by investigating the suppression of inflammatory mediators. In keratinocytes with UVB-induced damages, COX-2 expression decreased in the experimental group treated with ZM extract. Similarly, in RAW 264.7 cells where inflammation was induced with LPS, the biosynthesis of PGE2 was inhibited. Conclusion : These results suggest that ethanol extracts from Zostera marina may have value as the potential anti-inflammatory medicinal plant. Also based on the abovementioned results, ZM extract protects skin cells from UV-induced damages, and thus can be used in topically applied products for skin protection.

        논문 : 국내 유니버설 디자인 연구동향에 대한 내용분석 연구

        현대 다원화사회 및 민주사회의 진전은 인위적 창조물에 대한 일대 변화를 요구하고 있고 이러한 변화는 인간의 존엄성과 다양성을 존중하는 유니버설디자인 패러다임을 부각시켰다. 이 패러다임은 전세계적으로 지역적 특성에 따라 달리 파급되어왔으며 최근 이러한 동향을 국제적으로 파악하는 움직임이 일어나고 있다. 본 연구의 목적은 국내 유니버설디자인 연구의 국내 확산 동향을 파악하고자 하였으며 이는 융복합을 지향하는 현대사회의 학제간 교육을 고무시키는데 의미가 있다. 본 연구방법은 내용분석 방법으로서 문헌유형, 연도, 전공영역, 연구대상, 연구내용별 분석하였다. 연구결과, 국내 UD문헌은 344건이었으며 그중 학술지 논문을 대상으로 전공영역 9개의 학계로 구분되었다. 연도별로는 실내디자인학계에서 도입되어 점차 비디자인계인 교육과 의료계까지 보급되었다. 학계별 UD연구에서 연구대상은 공공시설을 위주로 실시되었으며 연구내용별로는 UD평가연구가 가장 많았다. 민주화 다원화 사회는 더욱 성숙될 것이므로 이러한 동향에 앞으로의 유니버설 디자인은 양적, 질적 확산이 지속되리라 전망된다. The advancement of multicultural society and democracy in modern society demands radical and immense changes in artificial creatures. These changes have presented a universal paradigm that respects the human dignity and diversity. Currently studies on UD in differently regions & countries have emerged. Recently, the research has been doing a trend of development in the world. The purpose of this study is to identify the trend of universal designs researched in Korea. As a study method, contents analysis has been utilized using academic published journal article that were from 1997 until 2013. Sixty five academic journals were analyzed according to by type of literature, year, author’s major area, research target & content. It was introduced in interior design filed and it was mostly introduced in the spatial fields of interior designs and architectures. It has been gradually expanded to design circles and further to non-design circles. It expected that this diffusion will be continued because multicultural and democratic society will become more mature in future.

        DNCB로 유발된 알레르기성 접촉피부염에 황련해독탕이 미치는 영향

        Objectives: HRHDT has been known as a useful prescription with antibiotic, anti-inflammatory, antioxidative and immunosuppressive activity. To evaluate anti-inflammatory effect of HRHDT, we treated HRHDT-skin in Balb/c mice model induced contact hypersensitivity. Methods: Contact hypersensitivity, a local inflammatory respinse of skin, was induced by spreading the back skin of Balb/c mice with 1% DNCB. HRHDT-skin was prepared by dissolving 3% 1,3-butylene glycol extract of HRHDT in solution and treated 2 weeks on the back skin. Results: HRHDT-skin significantly reduced TEWL and erythema by 0.4-1% of DNCB treatment compared with control group. HRHDT-skin reduced IgE on serum obtained from blood of DNCB-treated Balb/c mice. Histopathological examination showed that thickening of the epidermis, hyperkeratosis and the infiltration of inflammatory cells were found in Balb/c mice under conventional circumstances. Conclusions: These results showed that HRHDT-skin could be used as a pharmaceutical material with antiinflammatory effects by reducing IgE in contact hypersensitivity dermatitis Balb/c mice by DNCB.

        화장품 융합 기술을 이용한 천연 부산물의 소재 개발 및 적용방안- 커피 부산물의 화장품 소재로서의 가능성을 중심으로 -

        자원 절약 인식의 부족으로 인한 자원의 낭비, 현대 인류공동체 전체가 가진 비이상적 습관들, 소모적인 문화들이 오늘날의 심각한 환경문제를 야기시키고 있다. 그리하여 현대사회는 고도의 산업화로 인해 각종 산업폐기물과 일상생활에서 쉽게 버려지는 쓰레기들로 인한 환경오염의 문제를 중요한 문제로 보고 있다. 환경에 대한 경각심이 커지면서 자원재활용이라는 인식을 가져왔고 다양한 분야에서 재활용에 대한 연구가 활발히 이루어지고 있다. 본 연구는 커피 부산물로부터 획득한 오일 추출물의 화장품소재로서 활용가능성을 확인하기 위해 추출물이 함유된 에멀션의 시간과 보관 온도에 따른 제형 안정성을 확인하였다. 커피 오일 추출물이 함유되어 있는 에멀션의 pH, 점도를 측정하고 Turbiscan LAB을 이용하여 입자의 안정성을 확인하였다. 에멀션은 28일 동안 4, 25, 40℃ 항온조에 보관하여 일주일 간격으로 측정하였다. 그 결과 커피 오일 추출물 0.5, 1.0% 농도로 함유한 에멀션에서 보관 기간에 따른 물성의 변화가 미미하여 시간과 보관 온도에 따라 안정한 것으로 확인되었다. 상기의 결과를 통해 커피 부산물 오일은 콜로이드 에멀션 제형에서 안정적인 화장품 원료로서 응용이 가능할 것으로 사료된다. 본 결과가 재활용 소재에 대한 적극적 활용의 본보기로서 화장품 산업에서도 폐자원에 관한 인식이 긍정적으로 확대되기를 기대한다. Recently in Korea awareness of the country’s exhibition industry has expanded and a range of support and promotional strategies have been put forward. Environmental pollution is one of the most pressing issues of our time. Industrial waste and waste that are neglected in our daily lives are inevitable byproducts of industrialization. As more and more people became alert to global environment, resource recycling also gained popularity. As a result, many researches on recycling are being conducted across different fields. In order to use the coffee residue extract as a cosmetic ingredient, this study was conducted to evaluate its antioxidant effect. We confirmed the formulation stability of the emulsion containing coffee residue oil extract. We studied the pH, viscosity and particle dispersion of the emulsion containing coffee residue oil extracts were measured using a Turbiscan LAB. The emulsions were measured at one-week intervals in a thermostat chamber(4, 25, 40℃) for 21 days. We measured the stability of the pH, viscosity and emulsion particles using the Turbiscan LAB in a thermostat chamber for 21days. The formulations to which the coffee residue extract were added were considered to be stable, due to their physical property changes over time during storage. The results suggest that the coffee residue oil extract could be used in cosmetics colloid emulsion. Finally, we will hope enabling the expansion of the competitiveness of recycling in the residue materials from natural productut focussing on the present state of the cosmetic industry.

        스피룰리나 추출물의 모유두세포 보호 효능 및 세포증식에 관한 연구

        모유두세포는 모발의 성장과 주기를 조절하며, 자외선, 온도 변화, 호르몬, 산화적 스트레스 등에 의해 세포증식이 감소될 수 있으며 탈모의 원인과 직접적으로 관련된다. 본 연구는 스피룰리나의 모유두세포의 보호효과 및 세포 증식 효능을 평가하기 위해 phycocyanin 정량 분석 및 추출물의 자체의 세포 독성, UVA로부터의 세포보호, 상처회복 평가를 위한 세포증식 실험을 수행하였다. UPLC 성분 분석을 실시한 결과 물을 용매로 하여 추출하였을 때 phycocyanin이 26.66μg/ml 함량으로 나타났다. 모유두세포에 MTT assay를 실험한 결과 0.05, 0.10, 0.50 mg/ml의 농도로 처리하였을 때 농도의존적으로 세포생존율이 감소하였다. UVA로 활성산소종을 유도한 모유두세포에서 스피룰리나를 처리한 후 ROS 형광 세기가 감소되었다. 또한 모유두 세포에 상처를 유도한 후 세포증식능을 평가한 결과 스피룰리나를 처리후 24시간 뒤 모유두세포의 세포증식된 면적이 증가한 것을 확인하였으며 특히 0.10mg/ml의 농도에서 높은 효능을 나타내었다. 결과를 종합해 볼 때 스피룰리나 추출물이 모유두세포에 항산화제로서의 세포보효과를 나타내고, 세포증식을 활성화 시키는 것으로 판단되며 두피보호와 탈모 개선을 위한 다양한 제품에 활용할 수 있을 것으로 기대된다. Dermal papilla cells control the growth and cycle of hair, and cell proliferation can be reduced by ultraviolet rays, temperature changes, hormones, and oxidative stress, which is directly related to the cause of hair loss. In this study, to evaluate the protective effect and cell proliferation efficacy of dermal papilla cells of spirulina extract, quantitative analysis of phycocyanin and cell proliferation experiments were performed to evaluate the cytotoxicity of the extract, cytotoxicity from UVA, and wound healing. As a result of UPLC component analysis, phycocyanin was found to be 26.66 μg/ml when extracted using water as a solvent. As a result of MTT assay on dermal papilla cells, cell viability decreased in a concentration-dependent manner when treated with concentrations of 0.05, 0.10, and 0.50 mg/ml. ROS was reduced after spirulina treatment in dermal papilla cells induced by UVA. In addition, as a result of evaluating the cell proliferation ability after inducing a wound to the dermal papilla cells, it was confirmed that the cell proliferation area of the dermal papilla cells increased 24 hours after spirulina extract treatment. These results indicate that spirulina extract exhibits a cell-boosting effect as an antioxidant on dermal papilla cells and activates cell proliferation, and is expected to be utilized in various products for scalp protection and hair loss reversal.

        구기자 종자 초임계유체 추출물의 화장품소재로서의 가능성 평가

        연구는 구기자 종자를 초임계유체 추출한 후 in vitro 미백활성과 항균 실험을 실시하였으며, 추출물을 에멀션에 적용하여 안정성 평가를 수행하기 위해 pH, 점도, 에멀션 입자를 관찰하였다. 그 결과 미백활성의 경우 750 μg/mL에서 81.86 %의 미백 활성을 나타내었으며, 항균효과의 경우 Staphylococcus epidermidis, Candida albicans, Escherichia coli 균에서 항균효과가 나타났다. 또한 추출물을 함유한 에멀션 물리적 안정성 평가에서 점도와 pH가 28일 동안 큰 변화가 없었으며 유화물 입자 관찰 시 경시변화에 따른 합일과 분리현상이 확인되지 않았다. 이러한 결과는 화장품소재로서의 구기자 씨앗 추출물이 유용되게 활용될 수 있는 가능성을 가진 것으로 평가된다. The purpose of this study was to investigate the possibility of applying cosmetic material about extracted from Lycii Fructus seed supercritical fluid. This study was conducted to evaluate the anti-melanogenesis effect and antimicrobial activity about the extracts from Lycii Fructus seed. These researches studied for emulsion physical stability of pH, viscosity, particle size from emulsion containing Lycii Fructus seed extract. As a result, the supercritical fluid extract from Lycii Fructus seed significantly inhibited melanin synthesis by 81.86 % at the concentration 750 μ g/mL. Antimicrobial effects of extract was determined against Staphylococcus aureus, Candida albicans, Escherichia coli. except by Staphylococcus epidermidis. The physical stability of viscosity and pH on the emulsion containing Lycii Fructus seed extract were stable for 28 days. Emulsion containing Lycii Fructus seed extract did not change particles at observation into optical microscope. These results suggest that extracts from Lycii Fructus seed may have value as the potential cosmetic formulation.
