        벼 밀양 23호 $\times$ 기호벼 재조합 자식계통의 지역에 따른 품질 특성 관련 QTL 분석

        M/G RIL 164계통과 그 유전자지도를 이용하여 지역에 따른 벼의 품질과 관련된 양적형질 유전자좌(QTL)를 분석한 결과를 보면 다음과 같다. M/G RIL 164계통의 지역에 따른 단백질함량, 아밀로오스 함량, 지방산함량 및 식미평가치에 있어 빈도분포는 정규분포에 가까운 연속변이를 보였으며, 양친의 범위를 벗어나는 초월분리 현상을 나타내었다. 또한 지역에 따라 품질형질의 분포범위의 폭이 다양하게 나타났으며, 단백질함량은 원주>익산>대구의 순으로, 아밀로오스함량, 지방산함량 및 식미평가치는 대구>익산>원주의 순으로 나타났다. 품질에 관련된 QTLs분석에 있어 단백질함량과 관련하여서는 8개의 QTLs를 확인하였으며, 1번 염색체에서 2개, 3번, 6번, 7번 염색체에서 각각 1개, 8번 염색체에서 3개의 QTLs를 확인할 수 있었으며, 이들 8개의 QTLs가 설명할 수 있는 표현형 변이는 $6.0\~15.2\%$로 나타났다. 아밀로오스함량과 관련하여 6번 염색체에서 1개, 7번 염색체에서 2개의 QTLs를 확인하였다. 3개의 QTLs가 설명할 수 있는 표현형 변이는 $7.3\~24.4\%$로 나타났다. 지방산함량과 관련하여서는 2번과 6번 염색체에서 각각 깨, 3번과 7번 염색체에서 각각 1개의 QTLs를 분석하였으며, 6개의 QTLs로 설명할 수 있는 표현형 변이는 $5.5\~14.0\%$를 보였다. 식미평가치와 관련된 QTLs는 2번과 6번 염색체에서 각각 1개, 7번과 8번 염색체에서 각각 2개의 QTLs가 분석되었으며, 그 6개의 표현형 변이는 $5.5\~10.3\%$로 나타났다. The purpose of this study was to locate the quantitative trait loci (QTL) associated with quality properties in the recombinant inbred lines derived from the 'Milyang 23' and 'Gihobyeo' cross. Four quality-related traits; protein content, amylose content, fat acid content and sensory value were measured. Eight QTLs for protein content were detected on chromosomes 1 (two loci), 3, 6, 7 and 8 (three loci), each accounting for $6.0\%\~15.2\%$ of the phenotypic variation. Three QTLs for amylose content were detected on chromosomes 6 and 7 (two loci), each explaining from $7.3\%\;to\;24.4\%$ of the phenotypic variation. Six QTLs for fat acid content were detected on chromosomes 2 (two loci), 3, 6 (two loci) and 7, each explaining form $5.5\%\;to\;14.0\%$ of the phenotypic variation. Six QTLs for sensory value were detected on chromosomes 2, 6, 7(two loci) and 8 (two loci), each accounting for $5.5\%\~10.3\%$ of the phenotypic variation.

        水稻品種의 二面交雜에 의한 量的形質의 遺傳分析 Ⅳ. 자포니카 水稻品種의 source 및 生長解析關聯形質에 대한 組合能力

        The combining ability of source related characters such as leaf area index, leaf weight ratio, specific leaf weight, specific leaf area, and growth analysis related characters such as crop growth ratio, relative growth ratio, and net assimilation ratio were studied on the F₁ generation of 15 crosses from the partial diallel cross of 6 Japonica rice varieties. The effects of general combining ability(GCA) and specific combining ability (SCA) were highly significant for leaf area index, leaf weight ratio, specific leaf weight, specific leaf area, crop growth ratio, relative growth ratio and net assimilation ratio in the F₁ generation and the GCA of leaf weight ratio and net assimilation ratio was greater than SCA. The effects of GCA and SCA were different by characters, but the GCA and SCA to specific leaf area and leaf weight ratio were quite great. Cheolweon 34 and Ishiokamochi 7 as parents were quite effective for increasing specific leaf area and RAC 3 was quite good for increasing crop growth ratio and net assimilation ratio.

        연구보문 : 벼 육성연대에 따른 주요 품종의 수량 및 미질관련 특성의 품종변이

        This experiment was carried out to get the basic breeding informations regarding varietal variation and correlation of the growth characters, grain yield components, and physico-chemical properties related to grain quality of the major rice varieties by the released year and ecological type. The days from seeding to heading and culm length of the varietal group released after 2000 eras were shorter than those of the varietal group released before 1970 eras. The varieties released after 2000 eras showed higher grain yield due to the increments of the numbers of panicles per hill and spikelets per panicle. The varieties released by recently showed shorter and thiner of the length and width for the brown rice. The protein content and grain quality evaluation value of the varietal group released recently were so much improved compared to the varietal groups released before 2000. The yield component related characters affects to the grain yield by the varietal groups classified by the released year were different according to the characters of the yield components. The correlation between grain quality evaluation value and protein content, amylose content showed by negatively significant.

        연구보문 : 벼 재조합자식계통의 초기급속등숙속도 년차간 변이에 따른 아밀로그램 특성변이

        밀양 23호와 기호벼의 재조합자식 164계통에 대하여 2002년 부터 2005년까지 4년간 초기급속등숙속도의 년차간 표준오차를 기준으로 품종군을 분류하여 품종군별 아밀로그램 특성과 상관관계를 분석한 결과 년차간 변이가 작을수록 급속등숙형이 며 년차간 변이가 클수록 완만등숙형 이었다. 년차간 변이가 작을수록 호화온도가 낮았으며 년차간 변이가 클수록 취반점도를 제외한 다른 아밀로그램특성 RVA가 낮았고 반면에 년차간 변이가 작을수록 높았다. 초기급속등숙속도 년차간 변이 기 준 모든 품종군에서 강하점도와 취반점도는 고도의 부 상관을 보였고 년차간 변이가 큰 제4품종군을 제외한 모든 품종군에서는 호화온도와 취반점도는 유의한 정상관을 보였다. Amylogram properties such as peak viscosity, hot viscosity, cool viscosity, breakdown, pasting consistency and setback were investigated by the Rapid Visco Analyzer and interpreted by the varietal groups classified by the year variation of standard error of the rapidity of grain filling(RGF) which was calculated by the percentage of grain weight at 15 days after heading to 40 days after heading during four years from 2002 to 2005. The 164 rice recombinant inbred lines from the cross of Milyang23 and Gihobyeo were used to get the informations regarding amylogram properties and correlations among amlogram properties by the varietal groups based on year variation of RGF. The varietal group which has small year variation of the RGF showed fast grain filling, low gelatinization temperature and high RVA(rapid visco analyzer unit) of amylogram properties, otherwise varietal group which has big year variation of the RGF showed slow grain filling, low RVA of amylogram properties. Negative significant correlations were found between breakdown and setback in all varietal groups based on the year variation of RGF, however positive significant correlations were found between gelatinization temperature and setback in all varietal groups except varietal group 4 whcih has big year variation of RGF.

        水稻品種의 二面交雜에 의한 量的形質의 遺傳分析 Ⅰ. 자포니카 水稻品種의 雜種强勢 및 主要形質間의 相關

        This work has been performed to get the useful informations regarding the degree of heterosis and correlations between quantitative characters related to sink and source in Japonica rice varieties. Heterosis for days to flowering in F1 generation was mostly significant to late maturing, however, cross combination in which Ishiokamochi 7 crossed with Shin # 2 showed hetersois for days to flowering to early maturing. Degree of heterosis for culm length in F1 was greater than that of in F2 generation and the direction of heterosis and heterobeltiosis was tall culm length in case of cross combination in which Ishiokamochi 7 crossed with Samnambyeo. Almost all F1 generation in which had Ishiokamochi 7 as parent showed high heterosis for the number of panicles, 1000 grain weight and number of spikelets per panicle. Degree of heterosis to the productivity score in F₁, generation mostly showed high and positive direction. Significant positive heterosis and heterobeltiosis for leaf area index was found in the cross combination involved Ishiokamochi 7, Shin # 2 and Samnambyeo as their parent. In general, genotypic correlation coefficient was higher than environmental correlation coefficient and characters such as 1000 grain weight, number of spikelets per panicle and productivity score showed highly significant genotypic correlation with yield. The genotypic, phenotypic and environmental correlation coefficients between dry matter and harvest index in F1 generation showed highly negative significant. Significant positive genotypic and phenotypic correlation were found for crop growth rate and relative growth rate.

        특수 유색미, 향류미 및 한국 재래종 벼 종자의 층위별 항산화 및 간보호 활성

        한국 재래종 벼 15품종의 쌀을 세 층, L₁층(중심부부터 92%이상), L₂층(중심부로부터 81-91%), L₃층(중심부로부터 80%이하)로 나누어 항산화활성과 간보호활성 및 ferulic acid 함량 평가를 위한 재료로 사용하였다. 갈락토사민을 투여한 흰쥐에서 이들 세 층의 시료를 급여함으로써 혈중 MDA를 측정함으로써 항산화활성을 측정하였다. 그 결과 anthocyanin을 함유하는 수원 425호 및 상해향혈나의 품종이 가장 높은 항산화활성을 나타내었다. 또한 간보호활성을 측정하기 위하여 ALT 및 AST의 활성을 측정했을 때 L₁층에서만 몇몇 품종에서 활성이 나타났다. 따라서 쌀의 생리활성 물질의 표면 부위에 집중적으로 분포함을 알았다. 간보호활성의 경우에도 수원 425호 품종이 가장 강한 효과를 나타내었다. Ferulic acid의 함량 평가에서도 표면으로부터 안으로 진입할수록 급격히 감소함을 알 수 있었다. 결과적으로 수원 425호의 품종은 건강미의 개발을 위한 중요한 대상이 될 수 있는 것으로 평가되었다. Fifteen varieties of Oryza sativa mainly including those of Korean native rice were exactly cutted into three layers such as L₁layer(the outer part of 92% from rice center), L₂layer(the part of 81~91% from the rice center) and L₃layer(the inner part of 80% from the rice center) We biologically evaluated the antioxidative effect on the every layer(L₁, L₂and L₃) of all the varieties, by observing malondialdehyde (MDA) produced by galactosamine in the mouse. L₁layer of some varieties showed significant antioxidative effect, while L₂- and L₃layer didn′t show the activity. It was also suggested that coloured rice(Suwon 425 and Sanggaehangbyeolna) showed stronger activity than other general rices, probably due to high contents of anthocyanins. Successively, we evaluated antihepatotoxic effect, based on the determination of serum ALT- and AST activity. Some varieties of only L₁layer, except for L₂and L₃layer, significantly decreased the serum ALT- and AST activity. This finding indicate that oral diet of some raw rice are able to protect hepatotoxicities. Among all the samples tested, L₁layer of Suwon 425 showed the strongest antihepatotoxic effect. From quantitative analysis on ferulic acid derivatives, it was found that the more it enters from the surface into the rice center, the more those secondary metabolites contents were highly reduced. These findings above suggested that Suwon 425 could be a promising candidate for the development of health rice food.
