서울 시내 PC방(Internet Cafe)의 세균분포와 항생제 내성 양상

본 연구는 PC방의 세균 오염 실태를 조사하기 위한 목적으로 2004년 7월${\~}$12월까지 서울시내에 있는 PC방(Internet Cafe)들 중에서 34곳을 표본으로 선정하여 연구를 수행하였다. 총 세균수를 측정한 결과, 키보드에서는 $9.0{\times}10^4$ CFU/ml, 마우스에서는 $2{\times}10$ CFU/ml, 그리고 화장실 손잡이에서는 $5{\times}10^3$ CFU/ml의 세균이 검출되었다. 이는 화장실 손잡이보다 키보드와 마우스에서 세균에 대한 오염정도가 더 심한 것을 의미한다. PC방에서 항생제에 대한 내성을 지닌 7개의 균주를 분리해 내었는데, 2개의 균주는 methicillin과 erythromycin에 대한 내성을 나타내었고, 5개의 균주는 gentamicin, ampicllin, cefotaxim, chloramphenicol에 대해서 내성을 나타내었다. 이들 균주들을 동정한 결과, 2 균주는 Staphylococcus aureus로 동정되었고, 4 균주는 Actinobacillus ureae로 동정되었으며. 나머지 1개의 균주는 Pasteurella multocida로 동정되었다. Pasteurella multocida와 Actinobacillus ureae는 잠재적인 병원균으로 이전에 Pasteurella ureae로 알려졌던 Actinobacillus ureae는 인간의 상부 호흡기에 드물게 공생하는 것으로 알려졌으며, Pasteurella multocida는 많은 가축의 비인두강 안에 존재하는 평범한 정상 균총을 구성하는 세균인 것으로 알려졌다. 항생제 내성에 대한 연구 결과 Staphylococcus aureus는 erythromycin과 methicillin의 혼합제에 대해서 $100\;{\mu}g/ml$ 이상의 높은 항생제 내성을 나타내었고, Pasteurella multocida와 Actinobacillus ureae는 gentamicin, ampicillin 에 대하여 $100\;{\mu}g/ml$ 이상의 높은 항생제 내성이 있음을 알 수 있었다. This study was performed to investigate the hygiene condition of PC room(internet cafe) in Seoul Korea. From July 2004 to December, 34 samples were collected, there's an average of $9.0{\times}10^4$ CFU/ml on keyboards, $2{\times}10$ CFU/ml on mouse and $5{\times}10^3$ CFU/ml on door konbs toilets, suggesting that keyboards and mouse are more contaminated than toilet door knobs. Seven antimicrobial resistant strains were isolated from PC Rooms. Two isolates were resistant to methicillin and erythromycin, while five isolates were resistant to gentamicin, ampicllin, cefotaxim, and chloramphenicol. By identification, these strains were identified as Staphylococcus aureus (2 strains). Actinobacillus ureae (4 strains) and Pasteurella multocida (1 strain), respectively. Pasteurella multocida and Actinobacillus ureae are potentially pathogenic bacteria. Actinobacillus ureae, formerly, known as Pasteurella ureae, is an uncommon of the upper respiratory tract in humans. Pasteurella multocida is a part of the normal flora in the nasopharynx of many domestic animals. We concluded that Staphylococcus aureus is highly resistant to erythromycin and methicillin over $100\;{\mu}g/ml$, while Pasteurella multocida and Actinobacillus ureae is highly resistant to gentamicin, ampicillinover over $100\;{\mu}g/ml$.

목적: 자궁경부 편평상피세포암에서 array CGH법을 이용하여 유전자 발현의 양상을 조사함으로써 자궁경부 정상조직과 암 조직에서 유전자 발현의 차이에 대하여 알아보고자 하였다. 연구 방법: 자궁경부 편평상피세포암 조직 15예를 사용하였다. 조직을 파라핀에 고정하고, 10㎛씩 절단하여 헤마톡실린과 에오신염색을 한 다음, 미세절제술을 이용하여 DNA를 추출하였다. 1,440개의 human BACs으로 구성된 BAC array에 Cy5-dCTP와 Cy3-dCTP로 각각 표지된 정상과 자궁경부 편평상피세포암의 DNA를 교잡시켰으며, 정상과 환자군의 변이가 0.25배 이상 변화된 BAC clone에 포함된 유전자를 선별하였다. array CGH법에서 발현의 변화를 보인 유전자는 중합연쇄효소반응을 이용하여 그 유전자의 발현정도를 확인하였다. 결과: 자궁경부 편평상피세포암 70% 이상에서 SKI, FLJ13941, ZC3H3, GSDMDC1, PP3856, EEF1D,SOLH, LOC146325, FLJ36208, PIGQ, RAB40C, CYHR1, KIFC2, FOXH1, PPP1R16A, GPT, LOC113655, RECQL4, LRRC14, LRRC24, MGC70857, KIAA1688, PRKCZ, C1orf86, LOC440554, HDAC9, WDR8, TP73, KIAA0495, ESRRA, HSPC152, PRDX5, FLJ37970, RPS6KA4, TSC2, PKD1, RAB26, TRAF7 등 유전자를 포함하는 BAC clone들의 증폭과 PTPRG, LOC389432, SPATA19, LOC283172, KIAA0087, LOC442614, LOC386610 등 유전자를 포함하는 BAC clone들의 감소를 발견하였으며, 중합연쇄효소반응을 통하여 GSDMDC1, ABCF3, PCOLN3, SOLH, PRKCZ 등 유전자의 증폭과 NR3C2, CSF2, MSH6, MTR, PDE1C, ELF5, TXNDC10, SOD1 등 유전자의 감소를 발견하였다. 결론: 자궁경부암 조직에서 Array CGH를 이용하여 유전자발현의 양상을 조사한 결과, BAC clone내에 존재하는 유전자의 발현변화에 관한 정보를 얻을 수 있었다. 이는 자궁경부암의 암화과정에 대해 이해하는데 도움이 되리라 생각한다. Objective: Cervical cancer has long been linked to the sexually transmitted human papillomavirus (HPV), and the oncoproteins E6 and E7 disrupt the functions of tumour suppressor genes, resulting in genetic alteration. It was shown that loss of heterozygosity at 6p is a common genetic alteration in cervical cancer. However, the molecular genetics of cancer have only recently been understood, and for the development of cervical cancer additional genetic alterations in host cell genes are required. The present study has identified the differential changes of the cervical cancer-associated genetic alterations by a genome-wide array based comparative genomic hybridization (array-CGH). Methods: We analyzed 15 cases of cervical cancer from St. Mary`s hospital of The paraffin-fixed tissue samples were microdissected under microscope and DNA was extracted by the procedures of proteinase K digestion and chloroform extraction. Array-based CGH and genomic PCR were carried out with statistical analyses such as hierarchical clustering and Gene Ontology. The BAC array used in this study consisted of 1,440 human BACs, the space among the clones were approximately 2.08 megabase (Macrogen, Seoul, Korea). Results: All of 15 cases of cervical cancer showed specific gains and losses. The analysis limit of average gains and losses was 53%. A significant positive correlation was found between 1p36.32, 3p14.2, 3q27.1, 7p21.1, 8q24.3 and 11q13.1 changes through the cervical carcinogenesis. The high-level of gain regions, BAC clones encoded GSDMDC1, RECQL4, TP73, ABCF3, ALG3, HDAC9, ESRRA and RPS6KA4 genes. Frequently gained BAC clones encoded genes were PRSS8, FUS, COL18A1, PCOLN3, MAFG and ASPSCR1. The genes encoded by frequently lost BAC clones were PTPRG, GRM7, ZDHHC3, EXOSC7, LRP1B and NR3C2. Also, hierarchical clustering of the expression data readily distinguished genomic alterations in cervical cancer. A subset of cellular processes from each gene was clustered by Gene Ontology database. Conclusion: Using Array-CGH, genomic alterations related to cervical cancer were identified to determine whether induction of chromosomal imbalances occurs prior to carcinogenesis. The high resolution of array-CGH combined with human genome database would give a chance to find out possible target genes present in the gained or lost clones.

Background and aims: The Apolipoprotein E is a ligand of both the protein component LDL receptor as well as the apo E LDL-Receptor related protein (LRP). It modulates the receptor binding of lipoproteins, with the apolipoprotein E found on cell surfaces as its component, thus serving an important role in the lipid metabolism by carrying out the intracellular transport of cholesterol in lipoproteins. The gene for apolipoprotein E is the product of three common genotypes as well as many more rare alleles. The common genotypes are ε2, ε3, and ε4, and are expressed in the three phenotype isoforms of E2, E3, and E4. In the event that E4 is the main component, a rise in the cholesterol level, as the result of down-regulation of the LDL receptor, is observed. Therefore, those samples with E4 genotypes are known to be in much higher risk of coronary artery disease than those with ε3/ε3, while those with ε2 are in low risk (with the exception of hypertiglyceremai Ⅲ). The aim of this study is to analyze in patients with ischemic heart disease the role of aplipoprotein E alleles in order to seek its correlation with coronary artery disease, as well as to seek whether the polymorphism of apo E produces any differences in the severity of coronary artery disease according to plasma lipid levels. Methods: The subjects for study were 273 patients admitted to the Internal Cardiology Division of the Soonchunhyang University Hospital form December 1998 to February 1999. The subjects were divided into the two groups of which one was ischemic heart disease (IHD) experiment group totaling 105 (avg.60.1 years of age, male/female ratio = 69/36) and the control group totaling 168 (avg. 59.7 years of age, male/female = 73:95). The coronary angiogram was given to 127 subjects, and of this total, 94 have developed significant stenosis in the coronary artery. The stages of the analyzing of the apo E phenotype was first, the separation of DNA from the blood samples, subjecting it to the PCR from with 228 base pairs of expanded products were obtained. The band was determined by means of the reverse hybridization principle on the nitrocellulose strip. Results: From the 105 patients the distributions of apo E phenotypes were as follows: ε3/2(5.7%), ε4/2(1.9%), ε3/3(70%), ε4/3(20%), ε4/4(1.9%). The relative frequencies of each allele are as follow: ε2 (0.038), ε3 (0.833), ε4 (0.128). The results show as follows: ⅰ) The IHD experiment group to have a higher occurrence of ε4/3 phenotypes as well as ε4 alleles than the control group. ⅱ) Both the control group and IHD group showed the largest distribution of ε3/3 for phenotypes, and ε4 for alleles. ⅲ) The IHD group showed less ε2/3 phenotypes as well as significantly less allele frequency of ε3 in comparison to the control group. ⅳ) the IHD group showed a much lower level of HDL in comparison to the control group, while the LDL was significantly higher; samples including the apo ε2 showed a significantly higher level of HDL than those without. Among the control group, samples including apo ε2 showed a significantly higher level of TG (triglyceride) than samples without. No significant difference was found between the experiment apo ε4 sample and the control plasma lipid sample. ⅴ) No significant correlation was found between an apo E polynorphism and the number of involved arteries of a coronary angiogram. Conclusion: Between the experiment IHD group and control group were found differences in the frequency of alleles. The polymorphism of apo E alleles may contribute as a risk factor to the development of heart disease by involving itself in the metabolism and modulation of plasma lipids.
