희석액별 감성돔 , Acanthopagrus schlegeli 정자의 단기보존 효과

Experiments were performed to find out the proper conditions of diluents for the short-term storage of sperm of black seabream, Acanthopagrus schlegeli. For milt collection, brood stock was reared in a recirculating seawater system. The following results indicated that short-term storage methods with fresh condition could be employed in black seabream sperm. When the black seabream`s serum was used as a diluent for the fresh storage, sperm activity index (SAI), fertilization rate and survival rate derived the best result in 7∼10 days of storage. pH 7 and 8 showed the highest SAI in the same storage condition. In order to keep high SAI and survival rate of the sperm, addition of 800 ppm neomycine into the diluent revealed the best storage results.
바지락, Ruditapes philippinarum의 먹이 섭취에 미치는 수온과 염분의 영향

여름철 고수온기와 집중강우로 인하여 염분의 급격히 감소하는 시기에 바지락의 일간먹이밀도 감소량, 여수율, 소화율을 조사하였다. 실험결과 바지락은 수온과 염분에 의하여 일간먹이밀도 감소량, 여수율, 소화율이 영향을 받는 것으로 나타났다. 수온의 경우 소형그룹의 27℃에서 각각 5.8×10⁴cells ml^(-1), 16.96 liter hr^(-1)g^(-1), 12.04×10^7 cells hr^(-1)g^(-1)로 가장 높은값을 보였으며 대형그룹에서도 27℃에서 각각 15.60×10⁴ ce11s ml^(-1), 11.85 1iter hr^(-1)g^(-1), 26.30×10^7 cells hr^(-1)g^(-1)로 가장 높은 값을 보였다. 염분에 따른 소형그룹의 일간 먹이밀도 감소량, 여수율과 소화율은 12‰에서 0.10×10⁴cells ml^(-1), 0.19 liter hr^(-1)g^(-1), 0.18×10^7 cells hr^(-1)g^(-1)로 가장 낮은값을 보였으며 대형그룹에서도 12‰에서 0.45×10⁴cells ml^(-1), 0.24 liter hr^(-1)g^(-1), 0.71×10^7 cells hr^(-1)g^(-1)로 가장 낮은값을 보였다. 본 실험결과 여름철 고수온과 저염분이 바지락의 대량 패사 원인으로 작용하며 안정적인 생산을 방해하는 요인임을 알수 있었다. The feeding, clearance rate, and ingestion rate of Manila clam (Ruditapes philippinarum) were investigated with small group (10.8?.58 mm in shell length) and large group (37.5?.8 mm in shell length) under combination in water temperature (24, 27, 33, 36?) With salinity (12, 15, 24, 27‰). The maximum feeding, clearance rate, and ingestion rate in small and large groups were 27? and 27‰, respectively. While, The minimum feeding, clearance rate, and ingestion rate in both groups were 36? and 12‰, respectively. There results were suggested that the feeding, clearance rate, and ingestion rate of Manila clam were significantly influenced by external factors such as water temperature and salinity. The reason for the mass mortality of Manila clam during the heavy rainfall in summer season can be explained by high temperature and low salinity.

피조개, Scapharca broughtonii (Schrenck) 정자의 동결보존 위해 동결높이에 따른 예비동결 (0, 2, 4, 6 cm), 희석제 (150 mM NaCl, 300 mM glucose, Ringer's solution)와 동해방지제(glycerol, dimethylsulfoxide (DMSO), dimethylacetamide (DMA), methaol) 에 따른 효과를 정자의 운동성과 생존율로 비교하였다. 피조개 정자 동결보존에 있어 예비동결은 액체질소표면으로부터 2 cm가 가장 효과적이었으며, 희석제로는 Ringer's solution이 동해방지제로는 DMSO 10% 농도가 가장 적합하였다. This study was conducted to investigate protocol standardization for spermatozoa cryopreservation of the Scapharca broughtonii (Schrenck). Among the freezing rates, freezing at a height of 2 cm above liquid nitrogen surface for 5 minute gave higher activity and survival rate. Among the various diluents, Ringer's solution was the best for S. broughtonii sperm cryopreservation. The suitability of cryoprotectants dimethylsulfoxide (DMSO), dimethylacetamide (DMA), glycerol and methanol were tested against three freezing rates. DMSO gave significantly higher activity and survival rates than others.
능성어, Epinephelus septemfasciatus와 자바리, E. bruneus의 정자동결보존에 관한 연구

The purpose of this research is to develop the simple freezing method for sperm cryopreservation in the Sevenband grouper and Kelp grouper. The motility and survival rate were checked to investigate the effects of diluent and cryoprotectant on sperm cryopreservation.In cryopreservation experiments, 300 mM glucose and 15% dimethyl sulfoxide (DMSO) were used as a diluent and a cryoprotectant, respectively. As a results of cryopreservation, the motility rate of Sevenband grouper was found to be more than 90%, and that of Kelp grouper was found to be more that 60% in sperm 5 months after cryopreservation.
삼세기(Shaggy sea raven, Hemitripterus villosus)의 carbonic anhydrase III에 관한 연구

본 논문에서는 삼세기(Shaggy sea raven, Hemitripterus villosus)를 실험 재료로 선택하여 Carbonic Anhydrase Isozymes (CAs) 중의 하나인 CA III에 대한 연구를 SDS-PAGE, Isoelectric Focusing (IEF), Western blot analysis의 방법을 통하여 진행하였다. SDS-PAGE와 Western blot 결과 삼세기 아가미, 혈액, 장, 간, 신당, 근육, 심장조직에서 CA III의 분자량인 30 kDa의 band가 확인되었다. 삼세기의 근육과 아가미에 대한 등전점 전기영동(IEF)과 Western blot analysis 결과 pI 7.0 부근에서 band가 형성되는 것을 확인할 수 있었다. 특히 SDS-PAGE와 IEF 실험 결과 삼세기의 아가미 조직에서 CA III의 발현량이 다른 조직들에 비하여 우세하게 나타났다. 이는 아가미가 다른 조직들과 달리 어류의 생체기관 중 유일하게 외부와 직접 접촉이 가능한 조직으로서 활성산소에 대한 손상을 최소화하기 위한 것으로 사료된다. Carbonic anhydrase isozymes (CAs) are widespread zinc-containing metalloenzyme family. The enzyme catalyzes the reversible interconversion of CO2 and HCO3. This reaction is the main role of CA enzymes in physiological conditions. CA III, one of the CA isozymes, has been identified in many tissues. It is distinguished from the other isozymes by several characteristics, particularly by a lower specific activity and by its resistance to acetazolamide. However, the physiological function of CA III in fish is unknown. In this study, we examined the detection of CAs in the Shaggy sea raven Hemitripterus villosus, using SDS-PAGE, isoelectric focusing (IEF), and western blot analysis. We detected a significant protein band with molecular weight about 30 kDa from the tissues of H. villosus by SDS-PAGE and western blotting. A specific band of CA III with pI 7.0 was detected by IEF and western blotting in gill and muscle. The immunoreaction of anti-CA III expressed in the gill of H. villosus was much stronger than other tissues. One explanation for this result is that the fish gill is the only organ that is exposed to the external environment and that plays an important role in acid-base relevant ion transfer, the transfer of H+ and/or HCO3 ?, for the maintenance of systemic pH. This is the first report on the identification of a carbonic anhydrse III-like protein from H. villosus.
