        김치에서 tetracycline 내성 유산균의 분리

        대구지역에서 수집한 50점의 김치 중에서 10점의 김치로부터 tetracycline내성 세균을 분리하였다. 이 균주들의 tetracycline에 대한 MIC는 25-100 mg/l 이상의 범위로 분포하였으며 다른 항생제에 대한 내성도 다양하였다. tet(M), tet(O)특이적 primer를 이용한 PCR에서 HJ9 한 균주에서만 tet(M)유전자가 검출되었으며, tet(M)은 플라스미드상에 존재하는 것으로 나타났다. HJ9 균주의 tet(M)부분 염기서열을 분석한 결과 기존에 보고된 Gram양성균의 tet(M)의 DNA 염기서열 및 아미노산 서열과 각각 90-99%, 94-100%의 높은 상동성을 보였다. 165 rRNA 염기서열을 분석한 결과 HJ9 균주는 Lactobacillus sakei로 동정되었다. 김치를 통하여서도 항생제 내성 유전자의 전파 확산이 가능할 것으로 생각된다. Tetracycline resistant bacterial strains were isolated from 10 batches of Kimchi among 50 batches collected in Taegu restrict. The MIC of tetracycline ranged between 25 and> 100 ㎖/l. Total genomic DNA preparation from all 10 tetracycline resistant lactic acid bacterial isolates were subjected to PCR amplification with class-specific primers for tet(M) and tet(O). In only one isolate, HJ9, tet(M) was detected. By Southern blotting and hybridization with a tet(M)-specific probe, the tet(M) gene of HJ9 isolate could be localized on a plasmid. The partial nucleotide sequence and deduced amino acid sequence of tet(M) of HJ9 showed 90-99% and 94-100% homology to those of Gram positive bacteria, respectively. With sequencing of 16S rRNA, HJ9 isolate from Kimchi was identified as Lactobacillus sakei. From these results, Kimchi can be considered potential vehicle for the spread of antibiotic-resistant lactic acid bacteria along the food chain to the consumer.

        Thidiazuron 처리에 의한 히어리나무의 기내번식

        This study was conducted to evaluate the effect of thidazuron(TDZ) on shoot proliferation and growth from axillary buds of 20-years-old Corylopsis coreana. Shoots proliferation was effectively achieved on WPM(Woody Plant Medium) supplemented with 0.03∼0.1mg/L TDZ. The highest shoot number(6.5$\pm$0.7) was obtained on 0.1mg/L TDZ treatment. On the TDZ medium shoots formed as clusters less than 1cm in height and therefore needed to subculture on GA$_{3}$ containing medium to induce elongation. In consecutive cultures, phenolic compounds were excreted at the proximal part of the explants and inhibited growth of the explants. Growth inhibition by the compounds was overcome using liquid and paper bridge culture system. About 60% of the elongated shoots rooted on half- strength MS medium containing IBA. Generally, IBA was mire effective on in vitro rooting than NAA with optimal range of 0.5mg/L to 1.0mg/L. Rooted plantlets were transferred in an artificial soil(vermculite) and acclimatized in high humidity greenhouse condition. Survival rate differed greatly depending on rooting types of the explants. Two types of rooting were observed. The first type was direct rooting from the explants. The second type was callus formation followed by rooting from the callus. The explants showing the 1st type rooting survived can be multiplicated in vitro by TDZ treatment followed by elongation with GA$_{3}$ and rooting with IBA.

        Peace포플러의 기내 배양시 발생하는 과수화 식물체의 조직적 특성

        목본류 생리학 연구에 있어서 중요한 수종인 Peace 포플러의 기초 연구자료를 얻고자 본 실험을 실시하였다. 액아 마디를 BA 0.2 mg/L처리된 MS 배지에 배양하여 shoot의 증식시 나타나는 과수화 된 shoot를 정상 shoot와 비교하였다. 과수화 된 shoot는 진한 녹색을 띠고, 잎은 두껍고 휘어져 있으며 투명하였다. 또 줄기는 정상적인 줄기보다 두껍고 길이가 짧았다. 조직학적 관찰 결과 과수화 된 잎은 책상조직의 발달이 부진하였으며 정상적인 잎에 비하여 책상조직과 해면조직 그리고 표피 세포 모두 불규칙하고 세포간극의 크기가 큰 것으로 나타났다. 또한 과수화 된 잎의 기공은 크기가 커서 단위면적 당 밀도가 낮았으며 공변세포와 그 주변 세포들이 변형되어 있었다. 과수화된 shoot의 직경은 정상적인 것보다 큰 것으로 나타났는데 이것은 피층과 수층 세포의 비대에 따른 것으로 관찰되었다. 한편 과수화 된 줄기는 정상 줄기에 비해 목질화 발달이 미약하여 형성층 구별이 어려웠다. We investigated the anatomical aspects of vitrification in peace poplar. Comparisons were made with regard to characteristics occurring between hyperhydric and normal shoots in shoot proliferation cultures on MS medium containing 0.2 mg/L BA. Compared with normal plants, hyperhydric plants had thick, curled, and dark green leaves. Hyperhydric stems were thicker and shorter than those of normal stems. When examined under the microscopes, the mesophyll palisade cells of hyperhydric leaves were vacuolated, whereas those of normal leaves contained normal and enriched vacuole with cytoplasm. Generally, the hyperhydric leaves showed poorly developed palisade parenchyma, and revealed irregular and bigger sized intercellular structures in both palisade and spongy parenchyma as well as epidermis cells compare to those of normal leaves. In addition, the hyperhydric leaves had lower stomatal density and bigger sized cell. Vascular tissues of hyperhydic stems were less differentiated because of poorly lignified xylem tissue. The greatly expanded cortical cells and pith appeared to be the main cause of thick stems as compared with normal stems.

        논문(論文) : 학질발작(학疾發作)의 주체와 기전에 대한 고찰(考察) -『소문(素問), 학론(학論)』을 중심으로-

        Objective : I would like to determine the main factors, in other words, the subjects that are responsible for febrile paroxysm and how they interact at the time of onset, based on the 「Discourse on Hak(학)」 chapter of『Huangdi Neijing(黃帝內經)』. Methods : First, the pathological mechanism of the paroxysm was examined as described in the text. Then the subjects in question were analyzed based on the contents of the text. Result : 1. Febrile paroxysm happens when the three factors coincide at the Fengfu(風府). The three elements are as follows: first defense qi(衛氣), second latent qi(伏邪) and third, external pathogen(新邪). 2. Fengfu(風府) is not a specific point, but a region which external pathogen(新邪) passes through during which the defense qi is deficient. Conclusion : Febrile paroxysm is not caused by either an internal pathogen or an external pathogen, but the interaction between the three elements of the internal and external pathogens and the condition of defense qi. Moreover, the site, Fengfu(風府), which the disease is manifested is not a specific point but a more general region where the pathogenic qi has invaded while defense qi has weakened.

        스마트 홈 시스템은 앞으로도 계속 기술의 발전과 수요가 증가하는 블루오션 시장이다. IT 시장의 주목을 받는 아이템을 다룬 만큼 이 작품이 높은 발전 가치와 시장성을 보유하고 있다고 볼 수 있다. 스마트 홈 시스템 구축을 통해 개인에게 최적화 된 라이프 스타일을 구축하고, 더 나아가 개인에게 맞는 환경을 설정하여 맞춤 라이프 연계 서비스를 제공한다. 더 나아가 주목받는 이슈인 인공지능 기술을 사용하여 스마트 기기들에 대한 지능형 제어 및 효율적인 관리가 가능하도록 한다. 게이트웨이 서버에 에어컨, 공기청정기 등 우리 실생활과 밀접한 기기들에 연결함으로써 기존의 기기들에 비해 중요한 기기들을 더 높은 빈도로 관리할 수 있다. 이 프로젝트는 스마트 홈의 기본이 되는 통합 제어시스템과 이를 위한 IoT 허브 시스템의 하드웨어를 모두 개발한 프로젝트로써 게이트웨이 서버로 대표 되는 하드웨어를 통해 스마트 기기의 상태를 모니터링 하다가, 특정 센서값을 받으면 액션을 취해줌으로써 스마트기기를 제어할 수 있다. 그리고 이들과 관련하여 IoT 기반의 다양한 기기들을 표준화 제어하기 위한 제어 시스템을 구축하고 이를 위한 소프트웨어도 함께 개발했다.

        서번트 리더십이 비윤리적 친조직 행동에 미치는 영향 : 일터영성의 매개효과

        TServant leadership emphasizes the importance of leadership in an increasingly competitive global environment. Servant leadership is a field of leadership that is drawing attention in that the organization contributes to organizational performance by serving and serving employees. This study was conducted with the aim of examining the relationship between workplace spirituality and unethical pro-organizational behavior of the leader, and verifying the intermediary effect of workplace spirituality in the relationship between Servant leadership and unethical pro-organizational behavior. The survey was conducted on 600 employees in Korea and the results of 475 of them were analyzed. As a measurement tool, Servant leadership, workplace spirituality, and unethical pro-organizational behavior were used to analyze and utilize the statistical package SPSS 23.0. and AMOS 21. First, it was found that servant leadership had a positive significant effect on workplace spirituality. In other words, it was verified that servant leadership has a direct positive relationship to all of the sub-elements of workplace spirituality, namely, inner life, meaningful work, and sense of community. Second, it was found that servant leadership had a positive significant effect on unethical pro-organizational behavior. This was confirmed to increase the unethical pro-organizational behavior of employees through the leader's servant leadership. Third, in the relationship between workplace spirituality and unethical pro-organizational behavior, workplace spirituality showed significant results. In other words, it was confirmed that the higher the sense of the follower's inner life and meaningful work, and the sense of community with colleagues and the local community, enhances the follower's unethical pro-organizational behavior. Lastly, it was confirmed that among the three sub-dimensions of workplace spirituality, the inner life and meaningful work except for the sense of community among the three sub-dimensions of the workplace spirituality were found to have a mediating effect. In this work, the following importance can be found: First, we explained the process by which the boss's leadership of Servant influences organizational effectiveness. This study identifies the effectiveness of the Servant Leadership presented in the previous prior study by showing that organizational members bring a higher level of organizational effectiveness when influenced by Servant Leadership. Next, the servant leader, who grows and develops followers' capabilities through understanding and adapting to environmental change, explained the theoretical process of contributing to the achievement of organizational goals by presenting a vision to employees and increasing trust with them. Secondly, we present theoretical implications for unethical pro-organizational behavior. The study focused on the process of why organizational members become immersed in unethical pro-organizational behavior, along with ethical results from previous studies, confirming that positive social exchange relationships and organizational identification motivate organizational members to act unethical to benefit the organization. We intends to present practical implications to corporate CEOs and human resource managers as follows: First of all, there is a positive relationship between servant leadership and workplace spirituality, so it is necessary to increase servant leadership to increase workplace spirituality that can increase the intrinsic motivation and work immersion of organizational members. Second, workplace spirituality can have positive effects such as loyalty or attachment to the organization through internal motivation, so the organization needs to train employees to understand the positive aspects of workplace spirituality. Thirdly, it is a management of unethical pro-organizational behavior. Unethical pro-organizational behavior is unethical in terms of 'pro-organizational', which in turn can greatly harm the organization's reputation and sustainability, which is dangerous. Therefore, it is important to understand what motivates employees to participate in unethical pro-organizational behavior, as well as to reduce and eliminate unethical pro-organizational behavior. To this end, ethical behavior should be encouraged, managers should not only maintain an appropriate balance between financial benefits and ethical standards through evaluation and promotion systems, but also reduce unethical pro-organizational behavior through performance evaluations or links to salaries. It is necessary to raise awareness of this by highlighting issues of unethical pro-organizational behavior toward organizations or employees through ethical training programs.
