전 골수성 백혈병 세초인 HL-60 세포를 model로 하여, 민간요법으로 사용되어져 부작용이 극히 적은 거승로 알려진 고려인삼의 구성 성분 중 주요성분이 ginseng (Panzx ginseng C.A. Meyer) saponin 및 ginsenoside Rh1, Rh2, Rh3, 비파 (Eriobotrya japonica L.) 잎의 성분들 중에서 항발암 및 항암성분으로 알려진 ursolic acid 및 oleanolic acid, 웅담중의 중요성분 성분인 lithocholoc acid 드잉 분화능력이 있는 지를 조사하고자 본 실험을 수행아였다. Retinoic acid를 처리한 결과 타 연구자들의 연구결과들처럼 높은 분화력을 관찰할 수 있었으며, dbcAMP 단독 처리군에서도 높은 분화효과를 나타냈었다. Dexamethasone 처리군에서는 분화효과를 거의 관찰할 수 없었으나,dexamethansone과 구조적으로 유사한 ursolic acid와 oleanolic acid는 보다 높은 분화력을 보였고 웅담성분의 중요성분인 lirhocholic acid는 높은 분화력을 나타냈었다. Ginseng saponin은 0.00375% (w/v)에서 20% 이상의 분화력을 보였으며, Ginsenoside Rh2와 Rh3는 높은 분화력을 나타냈다. The acute myelogeous leukemia cell line, HL-60 is good model to examine leukemia differentiation with nitro blue terazolium reduction assay. We investigated that effect of triterpene acids and ginseng saponin on differentiation of HL-60 cells. Differentiation of HL-60 cells was induced in proportion to ,olar concentration by dibutylyl cAMP, ginseng saponin, lithocholic acid, ginsenoside RH2, and ginsenoside RH3.

Cytogenetic and hematological analysis was performed in peripheral blood of cattle in the vicinity of Uljin nuclear power station and control area. The frequency of micronuclei(MN) in peripheral blood lymphocytes from cattle was used as a biomarker of radiobiological effects resulting from exposure to environmental radiation. An estimated dose of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the bovine lymphocytes exposed in vitro to radiation over the range from 0 Gy to 4 Gy. MN ratio in lymphocytes of cattle from Uljin nuclear power station and control area were 8.90/1,000 and 9.60/1,000, respectively. There were no significant differences in MN frequencies and hematological values in cattle between Uljin and control area.

소핵분석은 방사선의 생물학적 선량계로서 활용되고 있으나 이의 생성 기전은 아직까지 확실치 않다. 본 연구에서는 사람 림프구에 방사선을 조사한 후 DNA 손상회복 저해물질, Cytosine Arabinoside(Ara C)와 3-Aminobenzamide(3-AB) 그리고 Hydroxyurea(HU)를 특정 세포주기에 처리하고 소핵분석과 FISH기법을 이용하여 방사선에 의한 소핵 및 이수성의 정도를 구명하고자 하였다. 방사선 선량에 따라 소핵과 이수성의 빈도는 양반응 관계를 보이며 증가하였고 DNA 손상회복 저해물질 처리 후 소핵의 빈도는 모든 DNA 손상 회복 저해물질에 의해 증가하였으며 Ara C, 3AB, HU 순으로 나타났다. 이수성의 빈도는 HU와 Ara C에 의해서 크게 증가하였으나, 3AB는 아무런 영향을 주지 않았다. 또한 1번 염색체가 4번 염색체보다 방사선에 의한 소핵형성 및 이수성에 더 많이 관여되었다. 본 연구 결과, 방사선에 의한 소핵 및 이수성의 형성 과정은 여러 다른 기전이 관여하고 있음을 알 수 있었다. This study was carried out to examine the effect of the DNA repair inhibitors, Cytosine Arabinoside(Ara C), 3-Aminobenzamide(3AB) and Hydroxyurea(HU) on the frequencies of radiation-induced micronuclei(MNi) and aneuploidy. Irradiated lymphocytes(1-3Gy) were treated with DNA repair inhibitors, Ara C, 3AB and HU for 3 hours and CBMN assay - FISH technique with DNA probe for chromosome 1 and 4 was performed. The frequencies of x-ray induced MNi and aneuploidy of chromosome 1 and 4 were increased in a dose-dependent manner. Ara C, 3AB and HU enhanced the frequencies of radiation-induced MNi and the frequencies of radiation-induced aneuploidy of chromosome 1 and 4 were enhanced by HU and Ara C while no effect was observed by 3AB. The frequency of radiation-induced aneuploidy of chromosome 1 was higher than that of chromosome 4. These results suggest that there are different mechanisms involved in the formation of MNi and aneuploidy by radiation.

Cytogenetic and hematological analysis was performed in peripheral blood obtained from cattle bred in the high background radiation areas (HBRA, Goesan-gun, Cheongwon-gun,Boeun-gun) and a control area. The frequencies of gamma-ray induced micronuclei (MN) in the cytokinesis- blocked (CB) lymphocytes at several doses were measured in 3 cattle. An estimated dose of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the bovine lymphocytes exposed in vitro to radiation over the range from 0 mGy to 1,969 mGy. The measurements performed after irradiation showed dose-related increases in the MN frequency in each donors. The results were analyzed using a linear-quadratic model with a line of best fit of y = (0.0583 ± 0.0137)D + (0.0366 ± 0.0081)D² + (0.0093 ± 0.0015) (y = number of MN/CB cells and D = irradiation dose in Gy). MN rates per 1,000 CB lymphocytes of cattle from the Goesangun, Cheongwon-gun, Boeun-gun and the control area were 6.50 ± 2.72, 9.00 ± 4.50, 10.89 ± 4.23 and 9.60 ± 4.70, respectively. The MN frequencies of CB lymphocytes from cattle bred in 4 areas mean that the values are within the background variation in this experiment. The MN frequencies and hematological values were similar regardless of whether the cattle were bred in the HBRA or the control area.

The purpose of the present experiment was to investigate the micronuclei(MN) frequency in cytokinesis-blocked(CB) cells after various doses of gamma-rays in pig (Landrace, male, 3-month-old) and so to contribute to the clarification of the question whether these species are suitable as a target organism in the test system. The frequencies of binucleated cells, and gamma-ray-induced MN in CB cells at several doses were measured in three donors. The peaks of binucleated lymphocyte formation(22%) were found at a concentration of 2% phytohaemagglutinin(PHA) and $4{\mu}g/ml$ Cytochalasin B(Cyt-B) in pig at 72 hours after incubation. Measurements performed after irradiation showed a dose-related increases in MN frequency in each of the donors studied. When analysed by linear-quadratic model the line of best fit was $y=0.0183D+0.0124D^2+0.0133$(y = number of MN/CB cells and D=irradiation dose in Gy). In conclusion, the results demonstrate that it appears feasible to use pig as target organisms in the micronucleus test to estimate the cytogenetic damage caused by ionizing radiations or, potentially, chemical compounds.

Nowadays Sports Utility Vehicle is popular. Because of heavy opening effort, it has power tailgate system that is opened tailgate automatically. It is too convenient to opening tailgate but it is also easy to crash rear obstacles. So we decide to develop new technology that is un-touched rear obstacle detection power tailgate using sensors. From now on, I will explain how to research this project.

N/A Results : There was no significant relationship of VDR or ER genotypes to lumbar or femoral neck BMD, hormones, and bone turnover markers. However, after controlling for potential confounding factors, a statistically significant ER X genotype effect on femoral neck BMD (p=0.038) , but not on lumbar BMD was observed . Moreover, there was more significant effect on femoral neck BMD by an interaction of VDR B * ER X genotype (p=0.013) in multiple regression analysis . Conclusion : The ER X genotype was associated with femoral neck BMD in Korean postmenopausal women . An estimated dose of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the swine lymphocytes exposed in vitro to radiation over the range from 0 Gy to 4 Gy. When analysed by Iinear-quadratic model the line of best fit was y=0.0102D²+0.0250D+0.011[y=number of MN/cytokinesis-blocked (CB) cells and D=irradiation dose in Gy]. MN rates per 1,000 CB lymphocytes of pig from HBRA and control area were 9.20±3.88 and 11.00±2.98, respectively. There were no significant differences in MN frequencies and hematological values in pig between HBRA and control area.
