        맥아의 ${\alpha}-Amylase$ isozyme에 미치는 Red Light의 영향

        이조대맥(Hordeum distichum L. emend Lamark)에 적색광을 조사하여 발아일수에 따른 ${\alpha}-amylase$의 활성 및 isozymes에 미치는 적색광의 영향을 조사하였다. ${\alpha}-amylase$는 암소에서 비활성도가 발아 3일과 5일에 각각 355.0, 523.7 $units/mg{\cdot}protein$로 발아일수가 경과함에 따라 증가하였으며, 적색광조사에 의해서 비활성도가 발아 3일과 5일에 각각 암소에 비하여 48%, 15% 정도 증가하였다. CM-cellulose와 Sephacryl S-200에 의해 2종의 isozyme이 분리되었고 그 중 ${\alpha}-amylase$ I이 주 효소였다. ${\alpha}-amylase$ I, II의 활성비율은 암소에서 발아 3일과 5일에 다 같이 95 : 5 정도였으나 적색광조사구에서는 각각 60 : 40, 90 : 10으로 현저한 차이가 있었다. 특히 적색광조사에 의해서발아 3일에 ${\alpha}-amylase$ II의 활성비율이 현저히 높았다. This study carried out to change ${\alpha}-amylase$ activity and isozymes in barley during germination in the dark and red light. The specific activity of ${\alpha}-amylase$ increased during the germination in the dark, giving 355.0 and 523.7 units/mg protein at 3 and 5 days, and the activity was increased by the red light up to 48 and 15% at 3 and 5 days of germination, respectively. The ratio of ${\alpha}-amylase$ I and II was approximately 95 : 5 at both 3 and 5 days of germination in the dark while the different ratio was found by the red light i.e. 60 : 40 and 90 : 10 at 3 and 5 days of germination, respectively.

        건시제조 중 Invertase 의 정제 및 그 특성

        건시제조중 invertase의 활성 변화를 조사하고 정제된 invertase의 효소적 특성에 관하여 실험한 바 그 결과는 다음과 같다. 건조가 진행됨에 따라 invertase의 활성은 증가하여 건조 10일째 최고 활성을 나타내었으며 그 이후는 감소하였다. 건시에서 invertase를 250 mM potassium phosphate(pH 7.4)로 추출하여 (NH₄)₂SO₄염석, DEAE-cellulose 및 sephadex G-200 column chromatography의 과정으로 정제한 결과 정제도는 조효소액보다 약 27배 증가되었으며 회수율은 약 11%였다. 최적 pH는 sucrose, raffinose에 대하여 각각 5.0과 6.0으로 나타났으며 최적온도는 40℃였다. 열과 pH 안정성은 50℃까지 그리고 pH 5 부근에서 안정하였다. Sucrose에 대한 K_m값은 2.5 mM이었으며, 정제된 invertase는 polyacrylamide gel 전기영동상에서 하나의 band를 나타내었다. This study was conducted to determine invertase activity in persimmon during the drying process and characterize the purified enzyme. As drying proceeded, invertase activity increased until 10 days and decreased gradually afterwards. Invertase in persimmon fruit was extracted with 250 mM potassium phosphate sulfate buffer at pH 7.4. The enzyme was purified by means of ammonium sulfate fractionation, column chromatography on DEAE-cellulose and gel filtration on Sephadex G-200 column. The optimal temperature of enzyme was 40℃ and optimal pH was 5.0 and 6.0 for sucrose and raffinose, respectively. The enzyme was stable up to 50℃ and pH 3-6. The Km value of the enzyme, with sucrose as a substrate, was 2.5mM. Electrophoretic pattern of purified enzyme solution showed a single band.

        This experiment was carried out to study on the removal of astringency and production of tannin by investigation on the changes of polyphenolic contents during growth persimmon fruits (Diospyros Kaki Linn.) which were used in experiment were Fuyu of nonastringent varieties and Cheongdo Bansi. Daegu Bansi and Sangju Dungsi of astringent varieties. Polyphenolic contents in astringent varieties was high value in the early growth stage. And then rapidly decreased, and increased to reach at maximum in the middle of Aug. and Sep. before continuously decreasing. On the other hand polyphenolic contents in Fuyu variety was high in the early growth stage as astringent persimmon, but continuously decreased until harvest. Contents of polyphenolic substances per fruit continuously accumulated until the early day of harvest, and then decreased. Sangju Dungsi and Daegu Bansi were more difficult than Cheongdo Bansi in the removal of astringency, because in Cheongdo Bansi, soluble polyphenol contents was higher than insoluble polyphenol contents, and in Daegu Bansi and Sangju Dungsi, this was higher than that from the middle of Aug. to harvest time.

      • 감의 脫澁處理中 Pectin 含量과 Texture의 變化

        This experiment was made to investigate the changes of the contents of acetaldehyde, alcohol, tannin, and to study the texture changes of persimmon in relation to the contents of pection during the removal of astringency by CO_2. 1. During the removal of astringency of persimmon fruits, the contents of soluble tannin showed the rapid decrease in high concentration of CO_2, but the contents of ethanol and acetaldehyde showed the rapid increase in all treatments. 2. The contents of W.S.P. increased, but H.S.P. decreased in all treatments. 3. All texture paramenters of persimmon fruits except springness and adhesiveness were decreased druing the removal of astringency. 4. The contents of W.S.P. were good correlated with the changes of hardness in all treatments.

        표고버섯 균사체의 배양특성 및 Pilot Scale 생산

        Lentinus edodes 산련 1호의 최적배양 조건을 TGY 배지로 조사한 바 배양 최적온도 25℃, 초기 pH 4.0, 교반속도 300 rpm, 접종량 10.0%로 하고 산소 통기량을 1.0 v/v/m일 때 최적배양조건이었으며 산업용 배지로 대량생산하기 위한 액체배지에서 최적의 C/N ratio 13.1 : 1로 7일간 배양하였을 ?? 건조 균사체량은 18.8 g/ℓ을 얻었다. 또 수율 증가를 위하여 식물성장 호르몬인 2,4-D, BA를 첨가한 실험에서는 2,4-D 0.5 ppm을 첨가하여 배양하였을 때 대조구보다 1.1% 수율이 증가하였지만, BA를 첨가하였을 때 대조구에 비하여 수율이 감소하여 균사체 수율향상에 식물성장호르몬 BA는 효과가 없었다. The optimum conditions for the submerged mycelial culture of Lentinus edodes SR-1 were elucidated to be incubation temperature of 25℃, initial pH 4.0, agitation of 300 rpm, inoculation of 10.0%(v/v), and aeration of 1.0 v/v/m in TGY medium. The optimum C/N ratio and economic yield coefficient for the submerged mycelial culture were 13.1:1 and 0.45 respectively. As the plant growth hormones test, SCM medium containing 0.5ppm of 2,4-dichlorophenoxyacetic acid increased mycelial yield in 1.1%, but 6-benzylaminopurine was not effcetive.
