        천연 생약재 열수 및 알코올 추출물의 어병 세균에 대한 항균력

        감초 외 48종의 천연 생약제로부터 열수 및 알콜 추출액을 제작하여 19 균주 어병 세균을 대상으로 항균력을 조사한 결과, 알콜 추출액은 22종, 열수 추출액은 16종이 어병 세균에 대하여 항균력을 나타내며 이 중 13종의 생약재는 알콜 및 열수 추출액 모두 8㎜ 이상의 저지대를 나타내어 항균력이 있는 것으로 나타났다. 열수 추출물에서 항균력을 나타내는 약재 중 그람 음성균에 감수성이 양호한 약재는 저지대 8 ㎜ 이상을 나타내는 애엽, 개옻나무, 연교, 지유, 파엽, 대황 및 황금이었으며, 그람 양성균에 항균력이 양호한 약재는 삼지구엽초, 육계 및 보골지 등 이었다. 그리고 오매, 황련, 계혈등, 상백피, 오배자 및 오미자는 저지대가 8㎜ 이상으로 그람 음성,양성균 모두에 항균력이 있었다. 알콜 추출물중에서 그람 음성균에 항균력을 나타내는 것은 백작약, 오매, 선모 및 황금 등으로 저지대가 8㎜ 이상으로 측정되었으며, 그람 양성균에 항균력을 나타내는 것은 감초, 계혈등, 단삼, 상백피, 육계 및 보골지 등으로 저지대가 8㎜ 이상이었다. 또한 그람 음성, 양성균에 모두 8㎜ 이상의 저지대를 나타내는 것은 애엽, 개옻나무, 황련, 지유, 오배자 및 오미자 등이었다. 그러나 이 중에서 오배자 열수 추출물의 어병 세균에 대한 발육 저지대가 32㎜로 그람 음성, 양성균 모두에 가장 뛰어난 항균력을 나타내어 다른 생약재와 다른 광범위 생약재인 것으로 나타났다. 애엽, 황금, 지유, 오매, 황련 및 오배자 열수 ·알콜 추출물은 tetracycline에 내성을 나타내는 균주에 감수성을 나타내었다. Hundreds of medicinal herbs have been using for the purpose of diseases treatment and immune enhancement for human being and other animals including fishes. Among them, 49 species of medicinal herbs were selected and tested for antibacterial activities against 19 strains of fish pathogenic bacteria in different 4 species. The 49 medicinal herbs were extracted by water and ethyl alcohol. The extracts were freeze dried and some paper discs from the extracts were prepared for the evaluation of antibacterial activity. The tested pathogenic bacteria were 5 strains of Edwardsiellu tarda, 5 strains of Vibrio sp., 4 strains of Lactococcus gawieae, 1 strain of Lactococcus raffinose. 1 strain of Streptococcus parauberius, and 3 strains of Streptococcus iniue. The Galla rhois (Obaeja), Gaeonnamu and Hwangleyon showed antibacterial activities on both gram negative and gram positive fish pathogenic bacteria. The Youkgae, Sangbaekpi, Bogolji and Gamcho showed very effective antibacterial activities on gram positive pathogens while Jiyu, Aeyoeb and Yeonkyo showed very effective on gram negative pathogens.

        잉어류 바이러스성전신괴사증바이러스 (VSNCV) 백신 투여에 대한 잉어의 면역반응

        양식 잉어류에서 심각한 피해를 야기하고 있는 VSNCV에 대한 백신을 제작하며 잉어에 투여한 후 특이적 및 비특이적 면역반응을 조사하였다. 백신은 포르마린처리백신 (FKV)과 가온처리백신 (HKV)를 제작하여 건강한 잉어에 0.2㎖씩 복강주사 하였으며, 2주 후 동일한 방법으로 boost처리하였다. 백신 투여 후 비특이적 면역반응 중 혈청 라이소자인 활성과 대식세포의 chemiluminescent (CL) reponses은 1차 주사 후 및 boost 처리 후 2일째부터 7일째까지 유의적인 증가를 나타내었으며, 이후 대조구 수준으로 감소하였다. ELISA법으로 항체가를 조사한 결과 대조구를 제외한 FKV와 HKV 투여구에서 주사 후 2주 째부터 boost처리 후 6주 째까지 유의적으로 증가하였으며, 그 중에서도 boost 처리 후 2주 째에 VSNCV로 공격 실험한 결과 FKV 투여구에서는 20%의 누적폐사율을 나타내어 방어력이 인정 되었으나, HKV투여구에서는 70%의 누적폐사율이 나타나 방어효과가 거의 없는 것으로 나타났다. VSNC is a viral disease causing significant economic losses in cultured carp Ciprinus carpio L. in Korea. Carps were immunized with prepared vaccines against VSNCV and examined specific and nonspecific immune responses. Carps were injected by 0.2㎖ of formalin-killed vaccine(FKV), heat-killed vaccine(HKV) or E-MEM, respectively and dealt with boost with same way two weeks later. The lysozyme activity of serum and chemiluminescent reponses of head-kidney leucocytes showed increased responses during 2-7 days post-first injection(pfi) and post-boost(pb) in the vaccinated fish, and then decreased to the level of control. As measured by ELISA, vaccinated groups showed a significant increase in VSNCV-specific serum antibodies between 2 weeks pfi and 6 weeks pb with a peak at 2 weeks pb. Results of the virus challenge showed that the fish vaccinated with FKV have induced protective immunity, white HKV injection hardly provided protection.

        대농갱이 암,수의 성장 차이

        대농갱이는 동자개과의 담수어로 고부가가치를 창출 할 수 있는 새로운 담수 양식 대상종으로 주목받고 있으나 현재까지 성장도 등 양식에 대한 기초정보가 밝혀지지 않았으며, 본 연구에서는 인공으로 생산한 자 ? 치어를 사육하면서 암 ? 수의 성장 차이를 비교 ? 조사하였다. 암수의 정확한 성장 차이를 구명하기 위해 본 실험은 자어 초기 조직검사에 의한 조사와 치어기부터 표지법에 의한 조사로 구분하여 실시하였다. 조직학적 판별에 의한 암수간 성장비교 시험 결과 부화 후 50일까지는 수컷과 암컷의 성장차이는 없는 것으로 나타났으나, 부화 80일 후에는 수컷(체장 72.0±6.7㎜, 체중 3.71±6.7g이 암컷(체장 64.3±6.7㎜, 체중 3.71±0.99g)보다 성장이 빠른 것으로 나타나 대농갱이는 부화 후 50일에서 80일 사이에 암 ? 수의 성장차이가 시작되는 것으로 사료되었다. 부화 80일 이후 조사는 표지법이 개체 성장에 미치는 영향을 조사한 후 본 실험에 착수하였으며 표지구는 대조구와 사이에 유으적인 성장의 차이를 보이지 않았다. 개체간 성장차이는 조직학적 판별 결과와 동일한 경향으로 수컷이 암컷보다 성장이 유의적으로 빨랐으며(P<0.05), 시험 종료 후 암컷과 수컷의 크기는 수컷이 평균 체장 17.57㎝, 체중 52.19g으로 암컷의 평균 체장 11.02㎝, 체중 16.26g보다 체장은 약 1.5배, 체중은 약 3배이상 큰 것으로 나타났다(p=0.05). 체중으로 본 성장곡선은 지수성장을 보였으며, 수컷은 Y=0.0024X^(1.738)(R^(2)=0.985), 암컷은 Y=0.0405X^(1.0316)(R^(2)=0.983)의 지수식에 밀접하게 수렵되었고 성장의 차이는 사육기간이 길어질수록 크게 나타났다. The Ussurien bullhead, Leiocassis ussuriensis, of the family Bagridae has recently been targeted for freshwater culture in Korea. In this paper we investigated sexual dimorphism in body size in rearing condition using specimens produced artificially. Tissue sections of gonads were made between 20 and 80 days after hatching to identify sex of the specimens investigated. A difference in body size between sexes was not observed in fry until they were 50 days old. Male fry 80 days old had attained 81.62f7.21 ㎜ total length, and females 72.84±8.62 ㎜ total length. After 80 days the difference in body size was analysed by tagging. Applying the tag by cutting fin tips had no affect on the growth of the fishes (P< 0.05). The growth in body size of males was significantly faster (P< 0.05) than in females, and males attained about 1.5 times more in standard length and about 3 times more in body weight than females at the end of 320 days. The growth curve of body weight was exponential and the body weight was expressed by Y = 0.0024X^(1.738) (R² = 0.985) and Y = 0.0405X^(1.0316)(R² = 0.983) in male and female, respectively, observed at monthly intervals.

        Chemotaxonomic Significance of Taxifolin-3-O-Arabinopyranoside in Rhododendron Species Native to Korea

        Genus of Rhododendron has been used in traditional medicine since ancient times and is known to be effective in immune function, inflammation, and cold symptoms. And the reason for this activity is the flavanonol type among flavonoids in the genus of Rhododendron. Among the flavanonol types, Taxifolin-3-O-arabinopyranoside was isolated from the root of native R. mucronulatum in Korea, and the structure was finally identified through HPLC, LC-MS/MS, <sup>1</sup>H-NMR, and <sup>13</sup>C-NMR. Taxifolin-3-O-arabinopyranoside is a compound mainly found in R. mucronulatum, a representative species of the genus of Rhododendron, and exhibits antioxidant, anti-inflammatory, and anti-atopic activities. In this study, Taxifolin-3-O-arabinopyranoside was chemotaxonomic significant in 5 species of the genus Rhododendron native to Korea (R. mucronulatum, R. mucronulatum var. ciliatum, R. schlippenbachii, R. yedoense var. Poukhanense, R. japonicum for. Flavum). Compared with the existing literature, Taxifolin-3-O-arabinopyranoside was identified for the first time in 4 species of Rhododendron except for the R. mucronulatum.

        Implementation of a revolute-joint-based asymmetric Schönflies motion haptic device with redundant actuation

        <P><B>Abstract</B></P> <P>In this work, a revolute joint-based asymmetric Schönflies motion (SM) haptic device with 4-Degree-of-Freedom (DOF) force feedback capability is developed. The SM haptic device is composed of a redundantly actuated parallel sub-module which has translational 3-DOF output motion, a pantograph limb which takes the role of providing 1-DOF rotational output motion, and a revolute joint allowing the relative motion between them. All five DC motors without gearhead are placed on the ground by employing proper parallel transmission linkages for power transmission. The large singularity-free workspace and the improved kinematic characteristics are secured by redundantly actuating the 3-DOF sub-module. Thus, the SM haptic device has excellent features such as the unlimited 1-DOF rotational output motion, minimal friction, minimal inertia, and large dexterous workspace. Mobility analysis, kinematic modeling, singularity analysis, optimal design, and linear inertia modeling of the SM haptic device are conducted. Then a prototype with two operational modes such as gravity compensation and linear inertia compensation modes is implemented. Through friction measurements, motion tests for gravity and/or linear inertia compensation modes, and virtual wall experiments, it is confirmed that the prototype possesses minimal friction as well as good gravity and linear inertia compensation performances sufficient for the high-quality haptic device applications such as medical training, robot-assisted surgery, etc.</P>

        Parameters for successful pig islet isolation as determined using 68 specific-pathogen-free miniature pigs

        <P>Abstract: Background: </P><P>Islet cell transplantation is a novel therapeutic modality for the cure of diabetes. Pig islet cells are an attractive substitute for human islet cells; however, they are known to be particularly difficult to isolate because of a weak islet capsule and a tendency to be fragmented during enzymatic digestion. Therefore, parameters favoring successful pig islet isolation were investigated using specific-pathogen-free (SPF) miniature pigs.</P><P>Methods: </P><P>Sixty-eight SPF miniature pigs were used for islet isolation. Birth weight, body weight, age, sex, pregnancy history, and the fasting blood glucose levels of each pig were determined. Each pig’s general condition was assessed with regard to feeding status and physical activity. Pancreas procurement was performed by one surgical team. Anesthesia duration, operation duration, procedure quality, and perfusate type were recorded. After pancreatectomy, a biopsy was performed for islet density analysis. Decapsulation, cannulation duration, degree of distension, and cold ischemic time were assessed. During islet isolation, pancreas weight, digestion time, and digested tissue proportion were recorded. Isolation results were evaluated by total islet equivalents (IEQ), islet equivalents per gram of pancreas (IEQ/g), isolation index, islet recovery rate, purity, and visual grade. To identify the predictors of higher islet isolation yield, we performed binary logistic regression analysis with significant (P < 0.05) variables from the univariate analysis.</P><P>Results: </P><P>The pigs were categorized into high (n = 34) and low yield (n = 34) groups according to the median IEQ/g or total IEQ values. Body weight and age were significantly different between the two groups. Being male or a positive history of pregnancy in females was factors favoring successful islet isolation. General condition assessments failed to estimate islet isolation results. Long anesthesia duration, which might have caused ischemic injury to the pancreas, negatively affected islet isolation results. Decapsulation, cannulation duration, and subsequent pancreas distension were significantly important in successful islet isolation. Inter-lot variability of Liberase was not observed because of screening processes performed before purchase. Isolation index and islet recovery rate correlated well with islet yields.</P><P>Conclusions: </P><P>Multivariate analysis using total IEQ and IEQ/g as outcome variables indicated that age older than 2, being male and moderate distension by Liberase injection are major determinants influencing successful islet isolation.</P>

        Intraperitoneal Bronchogenic Cyst Misidentified as Gastric Submucosal Tumor

        Most bronchogenic cysts originate in the mediastinum, while 15% to 20% occur in the lung parenchyma. According to the literature, most intrapulmonary cysts occur in the lower lobes. But, they are rarely located in an extrathoracic site, such as subdiaphragmatic retroperitoneal area. We describe a paraesophageal intra-abdominal bronchogenic cyst, first considered as gastric submucosal tumor. Resection was successfully realized by laparoscopy. A 20-year-old female was admitted to our hospital with incidentally detected gastric submucosal tumor. At operation, there was a 2.5×2 ㎝ sized cystic tumor that was isolated from the gastric wall. We performed tumor resection laparoscopically. The postoperative course was uneventful and the patient was discharged on the second postoperative day. Histological examination reported a bronchogenic cyst. We, herein, report this case.

        Discrepant Bowel Perforation from a Primary Lesion after Chemotherapy of Diffuse Large B Cell Lymphoma

        Diffuse large B cell lymphoma is the most common type of non-Hodgkin’s lymphoma, representing approximately one-third of all cases and involving the gastrointestinal tract in about 18%. With the development of modern chemotherapeutic regimens and advances in medical care, the prognosis for malignant lymphoma can be excellent. However, because of the aggressive adjuvant therapy required, complications such as bowel perforation may be fatal. In cases of chemotherapy for malignant lymphoma, we should keep in mind the possibility of perforation of the bowel after chemotherapy. Early detection is important to save patients.
