      • 젤라틴 제조시 추출온도가 젤라틴의 물리적 특성에 미치는 효과

        본 실험에서는 돈피를 원료로 acetic acid에 산침한 후, 열수 온도를 달리하여 추출해 낸 젤라틴의 점도, 겔강도, 색도 등의 물리적인 성질을 비교하였다. 산침 후 무게는 원료무게의 2배 이상 증가하였으며 팽윤율은 평균 115.9%로 증가하였고, 산침 후 두께는 평균 2.02mm, 두께의 팽윤율은 152.6%로 증가하였다. 추출율은 온도에 따라서 60℃<100℃<80℃<121℃ 순으로 나타났으며 추출온도별로 비교하면 동일한 온도, 동일한 전단속도에서 점도는 121℃<100℃<60℃<80℃ 순으로 높게 나타나 고온에서 추출하였을 경우 점도가 낮아짐을 알 수 있었다. L-value는 100℃에서 74.78로 가장 높게 나타났으며 121℃에서 가장 낮은 51.96을 보여주었다. a-value는 80℃에서 -1.35로 가장 낮은 값을 나타내었으나 60℃, 100℃, 121℃에서는 유의차가 없었다. b-value는 100℃에서 6.46으로 가장 높게 나타났으며, 80℃에서 3.78으로 가장 낮은 값을 나타내었으나 60℃와 121℃간에는 유의성이 발견되지 않았다. Gel 강도측정 결과를 보면 견고성(hardness)은 121℃<100℃<60℃<80℃순으로 크게 나타났으며, 응집력(cohesiveness)은 추출 온도별로 유의성을 나타내지 않았고 평균 0.66을 나타내었다. 탄성력(elasticity)역시 평균 0.977g/cm²로 추출온도별 유의성은 나타나지 않았다. 부서짐성(brittleness)은 견고성과 마찬가지로 121℃<100℃<60℃<80℃순으로 80℃가 가장 높았다. 그리고 부착성(adhesivenss)의 경우 추출온도별 유의성은 100℃와 121℃에서는 나타나지 않았고, 60℃와 80℃에서만 나타났다. 따라서 추출온도가 낮을수록 견고성과 부착성이 높은 것으로 나타났다. The physical properties of gelatin extracted from pork skin were investigated by determmation of expanding rate of skin, viscosity, gel strength and color depending on extraction temperature. Pork skin during acid treatment was expanded to about 152.6%. The extract rate was increased by 60℃<100℃<80℃<121℃. Viscosity of extracted gelatin sol was increased by 121℃<100℃<60℃<80℃. L-value was highest at 100℃(74.78) and lowest at 121℃(51.96). a-value was lowest at 80℃(-1.35), but there were no significant difference between temperatures of 60, 100 and 121℃. b-value was highest at 100℃(6.46) and lowest at 80℃(3.78). Hardness of gel was increased by 121℃<100℃<60℃<80℃ and cohesiveness was about 0.66 whereas elasticity was about 0.977g/cm³, Brittleness was increased by 121℃<100℃<60℃<80℃, and it was highest at 80℃, which showed similar trend with hardness. Adhesives were no significance difference between 100 and 121℃. From these results, it was achieved that gelatin becomes hard and adhesive when the extraction temperature is low.

        Anti-arthritic Effects of <i>Ephedra sinica</i> S<small>TAPF</small> Herb-Acupuncture: Inhibition of Lipopolysaccharide-Induced Inflammation and Adjuvant-Induced Polyarthritis

        <P>Anti-inflammatory and anti-arthritic effects of water distillates of <I>Ephedra sinica</I> S<SMALL>TAPF</SMALL> (ES), in herb-acupuncture, on the inflammatory responses of arthritis was investigated using phorbol 12-myristate 13-acetate (PMA)/lipopolysaccharide (LPS)-induced human macrophage and adjuvant-induced arthritic rat. The luciferase reporter vectors driven by the tumor necrosis factor (TNF)-α and cyclooxygenase-2 promoters were transiently transfected into U937 cells, which were then differentiated and stimulated by PMA and LPS, respectively, to develop an in vitro anti-inflammation assay system. The luciferase activities, observed in the activated U937 cells, were significantly inhibited by ES herb-acupuncture, compared to those of PD98509 and berberine. To evaluate ES herb-acupuncture as a novel anti-arthritic therapy, a polyarthritic rat model was developed using heat-killed <I>Mycobacterium tuberculosis</I>, and 50 μl of ES distillate was subcutaneously injected into the ST36 acupoint on each knee joint. While the articular indexes of arthritic rats were evidently decreased by ES herb-acupuncture, their body weights did not regain their initial levels. This may be due to the accelerating effects of ES on weight-loss and fat consumption. The mRNA expressions of TNF-α and interleukin (IL)-6 genes, which were closely stimulated in the arthritic rat joints, were found to be restored to the normal levels through the ES treatment. In the case of IL-1β, the recovery was not significant but substantial. The anti-arthritic effect of ES herb-acupuncture was not found in the ES-treated/non-acupoint group. In conclusion, the ES herb-acupuncture into the ST36 acupoint was found to be effective in alleviating the inflammatory response and thus arthritic symptoms in adjuvant-induced arthritic rats.</P>

        흰쥐의 Adjuvant 관절염에 대한 자하거(紫河車) 약침의 효과

        Rheumatoid arthritis (RA) is a chronic inflammatory autoimmune disease. characterized by leukocyte infiltration, a chronic inflammation of the joint, a pannus formation and the extensive destruction of the 3Iticular caJ1ilage and bone. Several proinflammatory cytokines such as tumor necrosis factor-${\alpa}$(TNF-${\alpa}$), interleukin-1${\beta}$ (IL-1${\beta}$) and interleukin 6 (IL-6) have been implicated in the pathological mechanisms of synovial tissue proliferation, joint destruction and programmed cell death in rheumatoid joint. In the Korean traditional medicine, Hominis placenta (HP) as an herbal solution of herb-acupuncture has been widely used to treat the inflammatory diseases including RA. In order to study the medicinal effect of HP herb-acupuncture on rheumatoid joint, an adjuvant-induced arthritis (AlA) was generated by the injection of 1.5 mg uf Mycobactelium tuberculusis. emulsified in squalene, 10 the base of the tail of Sprague-Dawley(SD) rats. After onset stage of polyarthritis, HP was daily injected to the Zusanti (ST36) acupuncture points in both of rat lags and the expression pattems of cytokines such as TNF-{\alpa}$, IL-1${\beta}$, and 1L-6 at the knee joint were analyzed using immunostaining and RT-PCR. The HP herb-acupuncture was found to be effective to alleviate the arthritic symptums in adjuvant-induced arthritic rats as regards the joint appearance and the expression profiles of inflammatory cytokines. In conclusion, therapeutic effects of HP herb-acupuncture on the rat with AlA might be related to anti inflammatory activities of the hurb-acupuncture.

        금연치료 요법으로의 침 자극에 대한 신경학적 기전

        목적: 흡연에 따른 건강적 피해가 많이 알려져 있음에도 불구하고 특히 여성 및 청소년 계층의 흡연이 증가하고 있으며 폐암 사망자수는 급격히 늘어나고 있는 상황이다. 완전한 금연이 실패하는 주요 이유는 담배의 중독성에서 비롯되며 금단현상을 극복하지 못해 일어나는 것으로 알려져 있다. 흡연의 중독성은 담배 주요성분 중의 하나인 니코틴(nicotine)에 의해 유발되며 따라서 모든 금연 요법 및 금연 치료보조 제들은 이 니코틴 작용을 어떻게 효과적으로 억제 또는 대체하느냐에 초점을 맞추고 있다. 최근 금연치료 요법으로 한방 침이 주목 받고 있으며 본 논문을 통해 한방 침의 금연효과에 대한 신경학적 기전을 고찰하고자 한다. 방법: 금연을 돕는 전형적인 보조 치료제 및 치료법이 몇 종류 개발되어 사용 중에 있으며 대표적으로 니코틴 대체 요법(nicotine replacement therapy, NRT)이나 항우울제로 사용되는 bupropion 등을 들 수 있다. 이 치료 방법들은 뇌의 도파민계 신경전달 체계에 영향을 미쳐 금연 효과를 발휘하게 되는데 실질적인 금연 성공확률은 그리 높지 않은 것으로 알려져 있다. 따라서 침의 자극에 대한 도파민계 신경전달 조절효과를 중심으로 고찰함으로써 침의 금연효과에 대한 의과학적 기전을 설명하고자 하였다. 결과: 침자극은 우수한 금연효과를 가져올 수 있는 치료법으로 기존의 금연 치료요법 및 치료제 들을 보완할 수 있는 보다 확실한 치료요법 중의 하나이며 특히 금연 후에 오는 금단현상을 효과적으로 완화시키는 작용을 한다. 그리고 이 같은 효과는 부분적으로 도파민계를 비롯한 신경전달계를 조절함으로써 가능한 것으로 판단된다. 결론: 본 논문을 통해 니코틴에 의한 금단현상의 신경학적 기전과 금연과 관련된 신경전달체계에 대한 침자극의 효능에 대해 고찰하였으며 기존의 금연보조치료법을 대체할 수 있는 우수한 의학적 치료법으로써의 침치료법을 제시하였다.

      • Larvicidal Activity of Apiaceae Plant Essential Oils and Their Components Against mosquitoes, Aedes albopictus.

        The larvicidal activity of 12 Apiaceae essential oils and their constituents was evaluated against third instar larvae of mosquitoes, Aedes albopictus. Of the 12 Apiaceae essential oils, Dill (Anethum graveolens), Caraway seed (Carum carvi), Cumin (Cuminum cyminum), Carrot seed (Daucus carota), Parsley (Petroselinum sativum) and Ajowan (Trachyspermum ammi) oils at 0.1 mg/ml exhibited 90% larval mortality after 2 days. Analysis by gas chromatography-mass spectrometry led to identification of 8, 3, 13, 12 compounds, respectively. in the oils of Dill (Anethum graveolens), Caraway seed (Carum carvi), Cumin (Cuminum cyminum), Ajowan (Trachyspermum ammi). The compounds thus identified were tested individually for their larvicidal activities against Aedes albopictus. At this same concentration, the individual constituents tested, α-phellandrene, α-Terpinene, ρ -cymene, (+)-Limonene, γ-Terpinene, Thymol, Carvacrol resulted in 100% mortality after 2 days. At this same concentration, the individual constituents tested, (+)-carvone, cuminaldehyde, neral, trans-anethole resulted in 80% mortality after 2 days.

        Purpose: The purpose of this study is to clearly classify the roles of Physician assistants and to identify obstacle factors and necessary factors for the operation of the system when they are performing the roles of Physician assistants. Methods: This is a descriptive research study to analyze the roles of Physician assistants and to understand the expected roles of Physician assistants according to each medical personnel (doctor, nurse, Physician assistants). Results: The obstacles to work are the lack of legal protection, and unclear work guidelines, and the matter necessary for the operation of the system is the legal protection. There were statistically significant differences in health collection ability, physical examination, drug response monitoring, and reporting in terms of the differences in role expectations considered by medical professionals regarding the roles of Physician assistants. Conclusion: Physician assistants need to develop various educational programs and materials, cultivate the ability to perform Physician assistants, and develop an advisory role through effective problem-solving between departments and increase the quality and satisfaction of Nursing care.

      • The Inhibition Study of the Aliphatic Compounds Against Acetylcholinesterases of the Pinewood Nematode, Bursaphelenchus xylophilus

        The pinewood nematode, Bursaphelenchus xylophilus is a serious forest pathogen in many countries including Japan, China and Korea. To minimize the environmental problems caused by synthetic chemicals broadly utilized in the control of B. xylophilus, we estimated the nematicidal potency of 63 aliphatic compounds against B. xylophilus by measuring their inhibition activity against acetylcholinesterases (ACE, EC 3.1.1.7) of B. xylophilus (BxACEs). In the primary inhibition assay using B. xylophilus crude protein, C6, C9, C10 2E-alkenal C12 alkanoic acid were observed the > 45% BxACE inhibition rate and they were subsequently estimated the inhibition rate against three recombinant BxACEs. Whole compounds showed the high inhibition rate against BxACE-1 and BxACE-2. Interestingly, C6 2E-alkenal and C12 alkanoic acid exhibited the high inhibition rate against BxACE-3 which would be insensitive to ACE inhibitors.

      • Insecticidal Activity of Apiaceae Plant Essential Oils and Their Constituents against Rice Weevil, Sitophilus oryzae

        We evaluated insecticidal activity of 11 Apiaceae plant essential oils and their constituents against rice weevil, Sitophilus oryzae.. Of 11 Apiaceae plant essential oils tested, dill (Anethum graveolens), carvi (Carum carvi), caraway seed (Carum carvi) and cumin (Cuminum cyminum) essential oils demonstrated strong fumigant toxicity against adults of rice weevil. Components of 4 active compounds were analyzed by GC and GC-MS, and fumigant toxicity of individual compound was also tested. Among test compounds, carvone, dihydrocarvone, carveol, linalool oxide, neral, cuminaldehyde, and anethole demonstrated the strong insecticidal activity. The toxicity of blends of constituents identified in 4 active oils indicated that carvone, and cuminaldehyde were major contributors to the fumigant toxicity of the artificial blend.
