Lymphoma is one of the most curable types of cancer. However, drug resistance is the main challenge faced in lymphoma treatment. Peroxisomal acyl-CoA oxidase 1 (ACOX1) is the rate-limiting enzyme in fatty acid β-oxidation. Deregulation of ACOX1 has been linked to peroxisomal disorders and carcinogenesis in the liver. Currently, there is no information about the function of ACOX1 in lymphoma. In this study, we found that upregulation of ACOX1 promoted proliferation in lymphoma cells, while downregulation of ACOX1 inhibited proliferation and induced apoptosis. Additionally, overexpression of ACOX1 increased resistance to doxorubicin, while suppression of ACOX1 expression markedly potentiated doxorubicin-induced apoptosis. Interestingly, downregulation of ACOX1 promoted mitochondrial location of Bad, reduced mitochondrial membrane potential and provoked apoptosis by activating caspase-9 and caspase-3 related apoptotic pathway. Overexpression of ACOX1 alleviated doxorubicin-induced activation of caspase-9 and caspase-3 and decrease of mitochondrial membrane potential. Importantly, downregulation of ACOX1 increased p73, but not p53, expression. p73 expression was critical for apoptosis induction induced by ACOX1 downregulation. Also, overexpression of ACOX1 significantly reduced stability of p73 protein thereby reducing p73 expression. Thus, our study indicated that suppression of ACOX1 could be a novel and effective approach for treatment of lymphoma. [BMB Reports 2019; 52(9): 566-571]

The asymmetric three-cabin structure is one of the favourite choices in urban utility tunnel engineering, as it can provide a higher space utilization and clearer classification for pipelines. However, few work has been done on utility tunnel in such special cross-section during undercutting construction until now. Besides, as the loose soil layers such as sand and silt are often encountered during the utility tunnelling, its influence on surface settlement become one of the main concerns. This study focuses on the influence of the construction scheme on the surface settlement details for an asymmetric three-cabin utility tunnel. A 3D finite element method (FEM) model was established including both the stratum and a utility tunnel based on the Beijing Daxing International Airport (BDIA) Expressway Urban Utility Tunnel project. With this model, the behaviour of the surface settlement with the undercutting construction of the utility tunnel was investigated, and the causes and values of surface settlement in five construction stages were analyzed. By changing the construction spacing and sequence of mid and side cabins, the surface settlement and structural deformation were compared. The numerical results demonstrated a strong correlation between the cross-section shape and surface settlement characteristics. And the symmetry line of the surface settlement trough was offset by 1 m to the side with the larger cross-section. The second (excavation of upper bench in mid-cabin) and fourth (excavation of upper bench in side-cabin) stages caused the largest surface settlement, which were 41.9% and 18.07% of the total settlement, respectively. In addition, after optimizing the field tunnelling scheme, the surface settlement was reduced by 31.9% using the side-tunnel first construction sequence. The proposed numerical model is able to predict the settlement characteristics in each construction stage, which is important and provides a basis for further studies on the surface settlement mechanism and optimal design of the asymmetric three-cabin utility tunnelling scheme.

<P>In order to exploit the 430-cavity in the active sites of neuraminidases, 22 zanamivir analogs with C-1 and C-4 modification were synthesized, and their inhibitory activities against both group-1 (H5N1, H1N1) and group-2 neuraminidases (H3N2) were determined. Compound <B>9f</B> exerts the most potency, with IC<SUB>50</SUB> value of 0.013, 0.001, and 0.09 μM against H3N2, H5N1, and H1N1, which is similar to that of zanamivir (H3N2 IC<SUB>50</SUB> = 0.0014 μM, H5N1 IC<SUB>50</SUB> = 0.012 μM, H1N1 IC<SUB>50</SUB> = 0.001 μM). Pharmacokinetic studies of compound <B>9f</B> in rats showed a much longer plasma half-life (<I>t</I><SUB>1/2</SUB>) than that of zanamivir following administration (po dose). Molecular modeling provided information about the binding model between the new inhibitors and neuraminidase, with the elongated groups at the C-1-position being projected toward the 430-loop region. In order to find a relationship between firefly luciferases structure and bioluminescence spectra, we focus on excited substrate geometries which may be affected by rigid luciferases. Density functional theory (DFT) and time dependent DFT (TDDFT) were employed. Changes in only six bond lengths of the excited substrate are important in determining the emission spectra. Analysis of these bonds suggests the mechanism whereby luciferases restrict more or less the excited substrate geometries and to produce multicolor bioluminescence.
c-Cbl-Mediated Neddylation Antagonizes Ubiquitination and Degradation of the TGF-β Type II Receptor

Transforming growth factor β (TGF-β) is a potent antiproliferative factor in multiple types of cells. Deregulation of TGF-β signaling is associated with the development of many cancers, including leukemia, though the molecular mechanisms are largely unclear. Here, we show that Casitas B-lineage lymphoma (c-Cbl), a known proto-oncogene encoding an ubiquitin E3 ligase, promotes TGF-β signaling by neddylating and stabilizing the type II receptor (TβRII). Knockout of c-Cbl decreases the TβRII protein level and desensitizes hematopoietic stem or progenitor cells to TGF-β stimulation, while c-Cbl overexpression stabilizes TβRII and sensitizes leukemia cells to TGF-β. c-Cbl conjugates neural precursor cell-expressed, developmentally downregulated 8 (NEDD8), a ubiquitin-like protein, to TβRII at Lys556 and Lys567. Neddylation of TβRII promotes its endocytosis to EEA1-positive early endosomes while preventing its endocytosis to caveolin-positive compartments, therefore inhibiting TβRII ubiquitination and degradation. We have also identified a neddylation-activity-defective c-Cbl mutation from leukemia patients, implying a link between aberrant TβRII neddylation and leukemia development.

From the viewpoint of engineering applications, the prediction of the failure of bogies plays an important role in preventing the occurrence of fatigue. Fatigue is a complex phenomenon affected by many uncertainties (such as load, environment, geometrical and material properties, and so on). The key to predict fatigue damage accurately is how to quantify these uncertainties. A Bayesian model is used to account for the uncertainty of various sources when predicting fatigue damage of structural components. In spite of improvements in the design of fatigue-sensitive structures, periodic non-destructive inspections are required for components. With the help of modern nondestructive inspection techniques, the fatigue flaws can be detected for bogie structures, and fatigue reliability can be updated by using Bayesian theorem with inspection data. A practical fatigue analysis of welded bogies is utilized to testify the effectiveness of the proposed methods.

Previously candidate gene approach revealed estrogen receptor (ESR) locus was associated with increased litter size. In this study, PvuII polymorphisms of ESR gene was detected by PCR-RFLP, and ESR locus was evaluated for its association with reproductive tracts components in the Large $White{\times}Meishan$ ($LW{\times}M$) F2 offspring. Ninety seven gilts with reproductive tracts components records and 136 offspring with performance traits records were genotyped and the results were used to estimate allele substitution effects. The results showed that two alleles (A and B) were identified, and 121 bp fragments were observed for the AA genotype and 65 bp and 56 bp fragments for the BB genotype; the length of uterine body (LUB) of BB gilts were significantly shorter than AA gilts', the additive effect was -1.762 cm; the uterine weight (UW) of AB gilts were significantly lighter than AA gilts' with the additive effect -18.058 g; no significant associations of ESR alleles with ovulation rate (OR), length of uterine horn (LUH), length of uterine cervix (LUC), weight of two ovaries (OW), volume of uterine lumen (VUL), length of oviduct (LO) were observed. BB genotypes gilts need significantly less days to 100 kg ($D_{100kg}$) than AA genotypes (p<0.01), the additive effect was per copy of B allele. Allele B is also favorable for average daily gain (ADG), with additive effect 0.015 kg/d (p<0.05). There was no difference between genotypes for backfat thickness at the 13th rib (SF13), loin meat height (ELMH), and loin meat percentage was estimated (ELMP), individual birth weight (IBW) and teat number (TN).

The novel AgCl/Ag2SO3 hybrids as an efficient photocatalyst had been fabricated by an in situ synthetic method. The correlations between the structure and the photocatalytic properties of the as-fabricated hybrids were analyzed. Experimental results exhibited that with increasing the amount of Ag2SO3, the degradation rate of the as-obtained samples was firstly increased and then decreased under the visible light irradiation. When the mass ratio of AgCl to Ag2SO3 was 1:2, in 30?min, it displayed the highest degradation rate of 99.2% for rhodamine-B, which was obviously higher than 46.1, 60.5, and 14.6% of pure AgCl, Ag2SO3, and TiO2 (P25), respectively. Similar results could be found in degradation of methyl orange. It had the maximum of 97.4% in 90?min, which was higher than 55.2, 48.7, and 12.7% of pure AgCl, Ag2SO3, and P25, respectively. Moreover, the as-prepared hybrids possessed the enhanced separation and transfer of photo-generated electron?hole pairs compared to the pure samples. In addition, the holes and superoxide radicals played the dominant role and the hydroxyl radicals played the secondary role during the process of photocatalytic degradation.

The Ag2SO3/NaNbO3 hybrids have been fabricated via a facile method at room temperature. Several methods such as X-ray diffraction, UV-Vis diffuse reflectance spectroscopy, scanning electron microscopy, electrochemical impedance spectroscopy and X-ray photoelectron spectroscopy were used to characterize the samples. Moreover, the photocatalytic activity of the samples was assessed by degradation of rhodamine B and methyl orange under the visible light illumination. Experimental results indicated the photocatalytic degradation efficiency of the as-fabricated hybrids was first increased and then decreased with increasing the dosage of NaNbO3. When the molar ratio of Ag2SO3 to NaNbO3 was 1:0.7, the as-fabricated composites had the best photodegradation efficiency of 96.4% in 30 min for rhodamine B and 97.1% in 60 min for methyl orange, respectively. These were obviously higher than that of pure samples. Furthermore, Ag2SO3 was conjugated with NaNbO3 via chemical-bonds rather than physical contact. In addition, the possible photocatalytic degradation mechanism was also provided and the main roles during the process of photocatalytic degradation were played by holes.
