        Loss of EMP2 Inhibits Melanogenesis of MNT1 Melanoma Cells via Regulation of TRP-2

        Melanogenesis is the production of melanin from tyrosine by a series of enzyme-catalyzed reactions, in which tyrosinase and DOPA oxidase play key roles. The melanin content in the skin determines skin pigmentation. Abnormalities in skin pigmentation lead to various skin pigmentation disorders. Recent research has shown that the expression of EMP2 is much lower in melanoma than in normal melanocytes, but its role in melanogenesis has not yet been elucidated. Therefore, we investigated the role of EMP2 in the melanogenesis of MNT1 human melanoma cells. We examined TRP-1, TRP-2, and TYR expression levels during melanogenesis in MNT1 melanoma cells by gene silencing of EMP2. Western blot and RT-PCR results confirmed that the expression levels of TYR and TRP-2 were decreased when EMP2 expression was knocked down by EMP2 siRNA in MNT1 cells, and these changes were reversed when EMP2 was overexpressed. We verified the EMP2 gene was knocked out of the cell line (EMP2 CRISPR/Cas9) by using a CRISPR/Cas9 system and found that the expression levels of TRP-2 and TYR were significantly lower in the EMP2 CRISPR/Cas9 cell lines. Loss of EMP2 also reduced migration and invasion of MNT1 melanoma cells. In addition, the melanosome transfer from the melanocytes to keratinocytes in the EMP2 KO cells cocultured with keratinocytes was reduced compared to the cells in the control coculture group. In conclusion, these results suggest that EMP2 is involved in melanogenesis via the regulation of TRP-2 expression.

      • 전기투석을 이용한 합성폐수중의 영양염류제거 특성

        NaCl수용액을 이용한 농축액/탈염액 농도비에 대해 실험한 결과, 농도비 1은 전력효율이 높은 가장 효율적인 농도비였으며, 전기전도도와 TDS는 90%이상의 제거율을 나타났다. 한계전류는 탈염액의 농도와 농축액의 농도가 높을수록 증가하였다. 반면에 농축액의 농도보다 탈염액의 농도가 한계전류에 더 큰 영향을 주었다. 효율적인 농도비를 혼합용액에 적용한 결과, 제거율에 있어서 암모니아성 질소와 인산염 인, 두 이온만이 존재하는 경우가 가장 효율적인 것으로 나타났다. 또한, 암모니아성 질소는 인산염인보다 높은 선택투과성을 가졌다. As the results of experiments on various conditions concentrated solution/diluted solution concentration ratio using NaCl solution, concentration ratiol was found to be the most effective in ter of high electric power efficiency. Conductivity and TDS removed more than 90%. Limiting current was increased with the high diluted solution concentration and concentrated solution concentration. But diluted solution concentration had influence more than concentrated solution concentration in limiting current. The efficient concentration was employed for nutrient removal, reduction rate was found to efficient to exist only NH_(4)^(+)-N and PO_(4)^(3-)-P. Also, NH_(4)^(+)-N was had high section permeability than PO_(4)^(3-)-P

      • 전이 금속 촉매에 의한 히드록시 관능성기를 갖는 전도성 고분자의 합성

        단량체인 4-hydroxy-4-isopropyl-1,6-heptadiyne 은 3-브로모프로핀과 마그네슘으로부터 제조된 Grignard 시약과 이소부탄산 메틸과의 반응에 의해 합성되었으며 IR 및 H-NMR 등의 분광학적 방법으로 확인하였다. 히드록시 관능성기를 갖는 단량체는 ?? 또는 ?? 계 촉매와 여러가지 조촉매에 의해 환상 중합되었으며 ??를 단독 촉매로 하여 벤젠에서 중합시켰을 때 64% 의 중합 수율을 보였다. 중합체의 구조는 분광학적 방법에 의해 확인되었으며, 고분자는 톨루엔, 클로로벤젠, 에테르, 아세톤 등 대부분의 유기 용매에 잘 용해되었고 pellet 상태에서 요오드로 도핑하였을 때 ??의 전기 전도도를 보였으며 분자량은 폴리스티렌을 표준 시료로 하여 보정하였을 때 GPC 상에서 8500 ~ 11000 의 범위에 있었다.

      • 6차 교육 과정에 따른 고등학교 공통 과학 (화학 부분) 교과서의 탐구 활동 분석

        In order to obtain the fundamental data for developing an ideal chemistry curriculum in the common science, four kind of common science textbooks were analyzed. Particularly, inquiry activities part was analyzed by the three dimension framework which consists of inquiry content dimension, inquiry process dimension and inquiry context dimension. The average volume of chemistry part in the common science text books was 13% (56 page). In the analysis of the inquiry content dimension of inquiry activities, the total number of themes in four kinds of textbook had similar inquiry activities. For the analysis of inquiry process dimension, 'interpreting data and formulating generalizations' category(39.1%) was most emphasized and the categories of 'observation and measuring'(30.8%), and 'building, testing and revising the theoretical model'(16.5%), 'seeing a problem and seeking ways to solve it'(13.5%) follow in order. As for the analysis of the inquiry context dimension, the scientific context was 82%, the individual context 1.8%, the social context 7.8% and the technical context 7.8%. It showed that the proportion of STS related contents in inquiry activities was 18%. For the purpose of common science text book in the 6th curriculum, it is expected to increase the proportion of the high level inquiry process and the proportion of STS related contents.

      • 고등학교 공통 과학내 화학 영역을 중심으로 한 STS 내용 분석

        Science-technology-society(STS) has been concerned with a reform in science education as a new direction and goal for science education. STS was introduced in the Fifth curriculum an was more emphasized in the Sixth curriculum. In this study, The STS materials in the chemistry part of common science textbook were analyzed. In the chemistry part of common science textbooks, average 14% of the total page content was devoted to the STS materials. The STS materials were included 23.8% in the chapter of reactivity of materials, 14.17% in the control factors on reaction kinetics, 10.7% in exothermic reaction and endothermic reaction, 10% in elements with similar reactivity. The STS materials are analyzed by STS topics by Piel(1981). 34.6% of STS materials were the topics on effects of 'socialization of science', 31.8% on 'technological developments', 16% on 'environmental quality and utilization of natural resources', 13% on 'energy', 3.6% on 'human engineering', and there was no topics on 'space research and national defense', and 'population'. The topics of 'socialization of science' and technology developments were covered in all chapters. The topics of 'environmental quality and utilization of natural resources' and 'energy' were covered in 3 chapters. The activities of STS program in the common science textbook were mostly consists of example study, inquiry research, data analysis. However there was no role play and practical work.

      • 전이금속촉매에 의한 환상중합고분자의 합성에 관한 연구

        The cyclopolymerization of 4-ethyl-4-hydroxy-1,6-heptadiyne was carried out by transition metal catalysts such as MoCl_(5) or WCl_(6)-based catalysts. The catalytic activity of MoCl_(5) for this cyclopolymerization was greater than that of WCl_(6). The polymerization of 4-ethyl-4-hydroxy-1,6-heptadiyne by MoCl_(5) alone gives a quantitative yield of polymer. WCl_(6)-based catalyst system was not effective in this polymerization. The obtained polymer was generally insoluble in common organic solvents. The IR spectrum of poly (EHH) showed the stretching band of the carbon-carbon double bond at 1590-1650 cm^(-1) indicate a high conjugation. The thermal properties and oxidative stability of poly(EHH) were also studied. The electrical conductivity of the I2 doped poly(EHH) was 1.2 - 2.5 × 10^(-4) Scm^(-1). 본 연구에서는 4-위치에 에틸기와 히드록실기를 동시에 갖는 1,6-heptadiyne 유도체를 합성하여 이를 여러가지 전이 금속 촉매를 사용하여 합성하였다. MoCl_(5) - 단독 촉매를 사용할 경우 가장 높은 중합수율을 보였으며 WCl_(6)- 계 촉매의 경우 중합체가 얻어지지 않았다. 합성된 고분자는 여러 가지 분광 법으로 확인한 결과 주 사슬에 6각형 고리 구조를 가지면서 공액이중결합을 구조를 갖는 것으로 확인되었다. 중합된 고분자는 치환기의 효과로 여러 용매에 쉽게 용해되었으며 산화안정성도 높은 것으로 확인되었다. 합성된 고분자를 요오드로 도핑 시켜 전도도를 조사한 결과 1.2 - 2.5×10^(-4) Scm^(-1)를 갖는 것으로 확인되었다.

      • Poly[4-(t-butyldimethylsiloxymethyl)-1,6-heptadiyne]의 합성과 그 성질에 관한 연구

        4-(t-butyldimethylsiloxymethyl)-l,6-heptadiyne(BDMSH)은 triethylamine의 존재하에서 t-butyldimethylsilyl chloride와 관련된 alcohol을 반응시킴으로 제조하였다. BDMSH의 중합은 여러 가지 전이금속촉매에 의하여 수행되었으며 ?? 촉매가 가장 좋은 촉매효능을 갖는 것으로 판명되었다. Poly(BDMSH)의 구조는 ¹H- 및?? 적외선 및 자외선 분광학 등의 방법으로 확인 하였으며 conjugated된 이중결합과 환상구조를 갖고있는 주사슬로 되어있었다. Poly(BDMSH)는 300 ℃ 이상에서 열중량감소를 보이기 시작했으며 600 ℃이상에서의 잔류량은 5% 였다. Poly(BDMSH)는 클로로포름, 이염화메탄, 벤젠, THF, 톨루엔 등의 유기용매에 잘 용해되었으며 유리판 위에서 film으로 쉽게 casting되었다. 제조된 film은 상대적으로 취약하였으며 ?? 전기전도도를 나타내었다. Poly(BDMSH)의 산소 투과도와 산소/질소의 선택투과도는 각각 180 barrer와 2.4였다.

      • Synthesis and characterization of new blue-light-emitting polymers based on PPP and PPV having fluorene pendant at vinyl bridge

        <P>The new poly(arylene vinylene) derivatives, which are composed of biphenylene vinylene phenylene vinylene, biphenylene vinylene m-phenylene vinylene, terphenylene vinylene phenylene vinylene, and terphenylene vinylene m-phenylene vinylene as backbone and bulky fluorene pendants at each vinyl bridge, were designed, synthesized, and characterized. The obtained polymers showed weight-average molecular weights of 11,100–39,800 with polydispersity indexes ranging from 1.5 to 2.1. The resulting polymers were amorphous with high thermal stability and readily soluble in common organic solvents. The obtained polymers showed blue emission (λ<SUB>max</SUB> = 456–475 nm) in PL spectra, and polymer 4 containing terphenylene vinylene m-phenylene vinylene showed the most blue shifted blue emission (λ<SUB>max</SUB> = 456 nm). The double layer light-emitting diode devices fabricated by using obtained polymers as emitter emitted bright blue light. The device showed turn on voltage around 6.5 V and brightness of 70–250 cd/m<SUP>2</SUP>. © 2006 Wiley Periodicals, Inc.
