        An Analysis on the Research Trends on Online Learning Engagement Using CiteSpace

        Online learning engagement is a network application that places the homepage and related materials of a course on the Web to form a shared virtual learning space, so as to achieve a face-to-face(FTF) learning effect. Based on the studies published from 2000 to 2020 in influential international and Chinese journals in the field of online learning engagement including the core databases of Web of Science, this paper conducts co-citation and co-occurrence analysis of the literature, subjects, and keywords with the use of CiteSpace. In other words, the study tries to draw the knowledge map of the international research in the field in comparison with China by using CiteSpace which is a software designed to analyze existing literature by co-citation networks. The researchers aimed to reveal the general situation and development trends of the research on online learning engagement. It was found that the number of publications in the field increased and the numbers are given according to year. The most cited journals and articles are revealed. The paper is concluded with some implications for future related research. (Hoseo University)

        Reduction of Erythrocyte Fluid Adaptability Due to Cell Membrane Hardening Based on Single‑Cell Analysis

        Because of the rapid development of precision medicine, single-cell analysis has attracted increasing research attention, especially for erythrocyte, whose potential role in the formation of vascular plaque (atherosclerosis) has emphasized the importance of flow characteristics of single erythrocytes in bionic microfluidics. Based on the high incidence of vascular plaques among the elderly and those who have received blood transfusions, we hypothesized that cell membrane hardening changes the fluid adaptability of individual erythrocytes. This hypothesis was verified using an in vitro microfluidic technique based on an analysis of the flow morphology and cell trajectory of individual cells. A symmetrical microchannel was fabricated with a central stenosis to simulate a blood vessel containing plaque. During flowing through this microchannel, normal erythrocyte predominantly exhibited deforming, rotating, and lifting morphologies, resulting in discontinuous contact with the channel wall and a narrower distribution. Conversely, hardened erythrocytes exhibited rolling, swinging, and tumbling morphologies, resulting in stable and continuous contact with the channel wall and a wider distribution. These results indicate that cell membrane hardening decrease cell fluid adaptability on a microscopic scale. This research can offer some new insights into vascular plaques research from a bio-tribological and mechanical perspectives.

        Novel Mutations in the UNC13D Gene Carried by a Chinese Neonate with Hemophagocytic Lymphohistiocytosis

        Hemophagocytic lymphohistiocytosis (HLH) in different ethnicities has been described in the literature, but few cases in patients of Chinese descent have been reported. Here, we describe the case of a Chinese neonate presenting with HLH carrying novel, compound heterozygous mutations of the UNC13D gene, including [c.2295_2298delGCAG, p.Glu765Aspfs*27] in exon 23, c.-250C>T, c.1+30G>A, c.279C>T, c.888G>C, c.18+36A>G, c.20-48T>C, c.1977C>T, c.2296C>T, c.24-46C>T, c.26-9_26-8insC, c.2599A>G, c.28+48C>T and c.3198A>G, some of which have not been reported in the literature. Cytokine profile analyses were performed in this patient, and the results were consistent with our previous findings in HLH patients. Cytokine profile monitoring may be helpful in differentiating among various clinical phases of HLH.

        Study on the Mechanical Properties of Functional Graphene/Polyethylene Composites by Molecular Dynamics Simulation

        The mechanical properties of functionalized graphene/polyethylene (f- GRA/PE) composites and graphene/polyethylene (GRA/PE) composites were studied based on the crystallized models with the molecular dynamics simulation in this paper. The effects of GRA and f-GRA with different mass fractions on the mechanical properties of polyethylene were considered. The results showed that the adsorption capacity of polyethylene molecular chains of f-GRA is stronger, the compatibility of f-GRA/PE system is higher with decreasing the interface distance, and the dynamic property of molecular chain is weaker with reducing the radius of gyration of f-GRA/ PE. Furthermore, the reduction of mechanical energy consumption results in the increase of wear resistance of the system, which is more obvious when the mass fraction of f-GRA increases. Finally, the influence of different tensile rates on the composite system was studied. It was found that during the tensile process, the ability of the composite material to resist deformation in the stretching direction is enhanced, and the functionalized graphene inhibits the movement of the molecular chain due to the adsorption force, which improves the yield stress of the f-GRA/PE composite material, thereby increasing the tensile strength. Tensile rate has a certain effect on mechanical properties, and elastic modulus and yield stress increase with the increase of strain rate.

        A Rapid PCR-Based Approach for Molecular Identification of Filamentous Fungi

        In this study, a novel rapid and efficient DNA extraction method based on alkaline lysis, which can deal with a large number of filamentous fungal isolates in the same batch, was established. The filamentous fungal genomic DNA required only 20 min to prepare and can be directly used as a template for PCR amplification. The amplified internal transcribed spacer regions were easy to identify by analysis. The extracted DNA also can be used to amplify other protein-coding genes for fungal identification. This method can be used for rapid systematic identification of filamentous fungal isolates.

      • Research on Application of CGAR Algorithm in Wireless Network Routing Protocols

        In order to reduce the transmission delay of the data packet in the wireless network, so as to improve the performance of the network, this paper proposes CGAR algorithm of the coding gain aware routing protocol. The algorithm uses the network coding gain and expected transmission count of the wireless link, provides the time needed for the node to transmit a data packet from the new flow in the wireless link, and take it as the routing metrics. This metric takes into consideration the factors such as the transmission delay, the time of the node’s contention channel and so on. The experimental results show that this algorithm can reduce the transmission delay of the data packet and improve the performance of the network.

        Early IL-17A Prevention Rather Than Late IL-17A Neutralization Attenuates Toluene Diisocyanate-Induced Mixed Granulocytic Asthma

        Purpose: Interleukin (IL)-17A plays a critical role in the pathogenesis of allergic airway inflammation. Yet, the exact roles of IL-17A in asthma are still controversial. Thus, the aim of this study was to dissect the roles of IL-17A in toluene diisocyanate (TDI)-induced mixed granulocytic asthma and to assess the effects of neutralizing antibody in different effector phases on TDI-induced asthma. Methods: IL-17A functions in allergic airway inflammation were evaluated using mice deficient in IL-17A (Il17a−/−) or IL-17A monoclonal antibody (IL-17A mab, intraperitoneally, 50 μg per mouse, 100 μg per mouse). Moreover, the effects of exogenous recombinant IL (rIL)-17A in vivo (murine rIL-17A, intranasally, 1 μg per mouse) and in vitro (human rIL-17A, 100 ng/mL) were investigated. Results: TDI-induced mixed granulocytic airway inflammation was IL-17A-dependent because airway hyperreactivity, neutrophil and eosinophil infiltration, airway smooth muscle thickness, epithelium injury, dysfunctional T helper (Th) 2 and Th17 responses, granulocytic chemokine production and mucus overproduction were more markedly reduced in the Il17a−/− mice or by IL-17A neutralization during the sensitization phase of wild-type (WT) mice. By contrast, IL-17A neutralization during the antigen-challenge phase aggravated TDI-induced eosinophils recruitment, with markedly elevated Th2 response. In line with this, instillation of rIL-17 during antigen sensitization exacerbated airway inflammation by promoting neutrophils aggregation, while rIL-17A during the antigen-challenge phase protected the mice from TDI-induced airway eosinophilia. Moreover, rIL-17A exerted distinct effects on eosinophil- or neutrophil-related signatures in vitro. Conclusions: Our data demonstrated that IL-17A was required for the initiation of TDI-induced asthma, but functioned as a negative regulator of established allergic inflammation, suggesting that early abrogation of IL-17A signaling, but not late IL-17A neutralization, may prevent the progression of TDI-induced asthma and could be used as a therapeutic strategy for severe asthmatics in clinical settings.

        A Rapid and Effective Colony PCR Procedure for Screening Transformants in Several Common Mushrooms

        In the post-genomic era, gene function analysis has attracted much attention. Transformation is often needed to investigate gene function. In this study, an easy, rapid, reliable, and cost-effective colony polymerase chain reaction (PCR) method for screening mushroom transformants was developed: picking up a suitable amount of transformant’s tissue (1-10 lg) to 20 ll 0.25% Lywallzyme solution, and vortexing for 10 s followed by incubation at 34 ℃ for 15 min. Finally, 2 ll of the suspension was used as templates to perform PCR and single target bands were successfully amplified from respective transformants of Tremella fuciformis, Pleurotus ostreatus, and Pleurotus tuber-regium. This procedure could be widely employed for screening transformants in mushroom transformation experiments.
