Background/Aims Cytoplasmic alkalinization induced by activation of the Na+/H+anti porter which is stimulated upon the addition of growth-promoting agents, such as insulin, epidermal growth factor, phorbol ester, plays an essential role in the initiation on cell proliferation. In the present study the effects of amiloride, a specific and reversible inhibitor of Na+/H+antiporter, on the growth of human pancreatic carcinoma cell line, Capan-1 cells was examined and the effects of 5-fluorouracil (5-FU) were also studied. Cell cycle analysis was done to examine the mechanisms for the inhibitory effects of amiloride. Materials/Methods The growth of Capan-1 cells were examined by counting cell number on two and four days treatment with 1 μM, 5 μM, 10 μM, 20 μM, 40 μM, 80 μM, 160 μM amiloride, and 0.1 ㎍/㎖, 0.3 ㎍/㎖ 5-FU, after plating Capan-1 cells into 35-mm2 plastic dishes at d density of 10x104 cells/dish. The reversibility of the effects of amiloride was examined on two day to eight days treatment with 20 μM amiloride after seeding 2×104 cells/dish. Cell cycle analysis was done on the sells after four days treatment with 20 μM amiloride. Results Amiloride significantly inhibited the growth of Capan-1 cells in a dose-dependent fashion (p<0.05). The inhibitary effect of amiloride on the growth of Capan -1 cells was firstly shown at the concentration of 5 μM, which is not so higher than the concentration of 0.1-0.2 μM attainable by administration of usual dose of amiloride (5-10㎎). Forty-eight percent inhibition of growth was found at an amiloride concentration of 20μM after 4 days treatment, and ninety-three percent inhibition of growth was found at an amiloride concentration of 160μM after 4 days treatment. The inhibitory effect of amiloride on growth of Capan-1 cells was reversible since removal of amiloride by a media change after 48 hours treatment lead to significantly more growth than amiloride treated group (p<0.05). The reversibility of growth inhibition suggests that amiloride in not a non-specific cytotoxin for Capan-1 cells. Amiloride combined with 5-FU significantly inhibited the growth of Capan-1 cells in a dose-dependent fashion compared to an amiloride or a 5-FU alone (p<0.05). After four days treatment with 20 μM amiloride, the faction of cells in G0-G1 phase, S phase and G2-M phase was 47.3%, 35.8%, 16.9% respectively in the amiloride group (20 μM), and 44.3%, 37.1%, 18.6% in the control group. showing no significant differences between the two groups. Conclusions Amiloride significantly inhibited the growth of Capan-1 cells in a dose-dependent fashion, which was reversible. The reversibility of growth inhibition suggests that amiloride is not a non-specific cytotoxin for Capan-1 cells. The concentration of 5 μM, which is not so higher than the concentration of 0.1-0.2 μM attainable by administration of usual dose of amiloride (5-10㎎), which suggests amiloride or its analogues may be used alone or in conjunction with 5-FU for the treatment of pancreatic carcinoma. Further study is needed to clarify the effects of more potent analogues of amiloride on the growth of human pancreatic carcinoma cells.

<B>ABSTRACT</B><P>A triple-site variant (W17Q N90A L129F) of mannose-6-phosphate isomerase fromGeobacillus thermodenitrificanswas obtained by combining variants with residue substitutions at different positions after random and site-directed mutagenesis. The specific activity and catalytic efficiency (<I>k</I>cat/<I>K</I><I>m</I>) forl-ribulose isomerization of this variant were 3.1- and 7.1-fold higher, respectively, than those of the wild-type enzyme at pH 7.0 and 70°C in the presence of 1 mM Co<SUP>2+</SUP>. The triple-site variant produced 213 g/literl-ribose from 300 g/literl-ribulose for 60 min, with a volumetric productivity of 213 g liter<SUP>−1</SUP>h<SUP>−1</SUP>, which was 4.5-fold higher than that of the wild-type enzyme. The<I>k</I>cat/<I>K</I><I>m</I>and productivity of the triple-site variant were approximately 2-fold higher than those of theThermus thermophilusR142N variant of mannose-6-phosphate isomerase, which exhibited the highest values previously reported.</P>
A New Zero-Voltage Switching Half-Bridge Converter with Reduced Rectifier Voltage Ringing

Symmetric half-bridge (SHB) converter has been widely used in PC power supply application since it has no DC offset of magnetizing current and it has symmetric device stress. However, it is difficult for SHB converter to get high efficiency due to significant switching loss. To solve this problem, a variety of Zero-Voltage Switching (ZVS) half-bridge converters have been proposed including an asymmetric half-bridge (AHB) converter. They can reduce switching loss with ZVS. However, ZVS half-bridge converters have serious voltage oscillation problem across the secondary rectifier, which would require the snubber circuit. This oscillation is increasingly severe when large resonant inductance is used for wide ZVS range. The proposed converter solves this problem by separating resonant inductance for ZVS from resonance with the rectifier junction capacitance. It reduces energy dissipation on snubber and decreases conduction loss on rectifier. In addition, it has wide ZVS range with series-connection of main switches. Operational principle and analysis of the proposed converter are presented and confirmed by experimental results of a 240W prototype.
Impact of pigmented contact dermatitis on quality of life in Korean patients

Background: Acquired facial hyperpigmentory disorders may lead to psychological and social distress to the patients. However, little is known about the psychosocial burden of pigmented contact dermatitis (PCD) in Korean patients. Objectives: This study was aimed to examine the level of psychosocial impacts of pigmented contact dermatitis on quality of life (QoL) in Korean patients. Methods: A prospective study recruited the 25 patients with pigmented contact dermatitis and 32 patients with melasma as well as 30 healthy controls. Participants were asked to complete questionnaires regarding demographic characteristics, Dermatology Life Quality Index (DLQI) and Melasma Quality of Life Scale (MELASQOL). Results: The mean score of DLQI was increased in patients with pigmented contact dermatitis (13.76±3.81) than that of patients with melasma (6.63±0.83; P<0.001) and healthy controls (1.83±1.16; P<0.001). In addition, the mean score of the MELASQOL was also increased in patients with pigmented contact dermatitis (54.36±9.26) than that of patients with melasma (36.40±5.65; P=0.01) and healthy controls (17.33±8.91; P<0.001). Conclusion: Among various acquired facial hyperpigmentory disorders, pigmented contact dermatitis poses the serious negative impacts on QOL of patients. Therefore, careful attention associated with psychosocial factors should be considered when managing patients with pigmented contact dermatitis.

cDNA Library Construction of Aphis gossypii Using Gateway Cloning System

Aphis gossypii was widely distributed throughout the tropical, subtropical and temperate zone. The chemical control of A. gossypii is becoming problem because it was rapidly appeared resistance expression to chemicals. We will attempt to resolve the this problem using RNAi technique. Besides, RNAi technology can be helpful to study the target genes of A. gossypii. In this study we produce cDNA library construction using gateway cloning system for selecting target gene in order to control of A. gossypii using RNAi. As a result, the 100-400bp of insert size, which is appropriate for RNAi was confirmed. Most of insert gene is associated with A. gossypii, after that insert sequence was compared with DNA databases and EST databases using NCBI blast search. Consequentially, A. gossypii of cDNA library with the titer of 3.15x105 clones were completed. And we will perform the LR recombination to transfer cDNA library into TRV2 (tobacco rattle virus) vector with att site. Then, after performing transformation using Agrobacterium tumefaciens (GV 2260), we inoculated to cucumber with A. tumefaciens. An insecticidal effect or a repellent activity against A. gossypii by changing behavior in transgenic cucumber plants were conformed. Also, the selecting target gene in order to control A. gossypii using RNAi may be provided.
