Purpose: The purpose of this study is to report the genetic diagnosis of nine cases of Fabry in one family, either as carriers or patients. Methods: We conducted analysis of the α-galactosidase A gene and ophthalmologic examination of family members of a patient diagnosed with Fabry disease. Results: Our patient, his brother, and two male cousins had Fabry disease; his mother, three aunts, and his female cousin were Fabry carriers. Genetic study revealed deletion mutation (1235-1236delCT) at the α-galactosidase A gene in all subjects. Ophthalmologic examination detected whirl-like corneal opacity in all subjects, which is a typical characteristic of Fabry disease.

Purpose: To compare the clinical results of single-piece hydrophilic acrylic intraocular lenses (IOL) (Akreos AdaptTM, B&L) with those of three-piece hydrophobic acrylic IOL (Sensar?, AMO) after cataract surgery. Methods: This study was a prospective case-control study in which Akreos lenses were implanted for the experimental group and Sensar lenses were implanted for the control group. All surgical procedures were performed by the same surgeon. Best corrected visual acuity (BCVA), change of anterior chamber depth, degree of posterior capsule opacification (PCO), higher-order aberration, and contrast sensitivity (CS) were examined. Measurements were performed by masked examiners prior to the surgery and at 1 week, 1 month, 3 months, and 6 months, postoperatively. Results: There were no statistically significant differences in BCVA, CS or PCO between the two groups. The Refractive error was -0.17±0.67D in the experimental group and -0.02±0.41D in the control group at 6 months, postoperatively. During the first postoperative month, forward movement of both IOLs was detected. Afterward, both IOLs moved backwards until 6 months. Fourth-order spherical aberrations were significantly higher in the Akreos group (p<0.05) at 1 month, 3 months, and 6 months, postoperatively. Conclusions: The clinical results for Akreos IOL and Sensar IOL were similar in most aspects.

Purpose: To demonstrate the capability of anterior segment optical coherence tomography (OCT), to evaluate central corneal thickness (CCT) and to compare the results with those by Orbscan II and standard ultrasound (US) pachymetry. Methods: One examiner measured the CCT of 44 normal eyes of 22 subjects using anterior OCT, Orbscan II, and US pachymetry. Non-contact measurements by anterior segment OCT and Orbscan II were performed first, followed by contact measurements using US pachymetry. Three consecutive measurements were taken with each method and the mean values and correlations were analyzed. Results: The mean value of the CCT was 531.45±32.90 ?m with anterior OCT, 537.11±32.21 ?m with Orbscan II, and 548.18±34.17 ?m with US pachymetry. There was no statistically significant difference among the values obtained by the 3 instruments (P>0.05). CCT measurements by anterior segment OCT were highly correlated with Orbscan II and US pachymetry (P<0.001) measurements. Conclusion: CCT measurements by anterior segment OCT are highly correlated with Orbscan II or US pachymetry measurements. Using non-contact anterior segment OCT, a closer examination of the anterior segment including the cornea is possible. The measurement of the CCT using anterior segment OCT is applicable because result values are similar to measurements by Orbscan II or US pachymetry.

Conventionally, the nMOS output driver transistor has been used as the ESD protection device. The uniform conduction of all multi-finger transistor is the most challenging issue to achieve the full protection level. This paper reports the influence of metal line routing in I/O circuits of 64Mb DRAM chips. We performed circuit-level ESD simulation using iETSIM (illinois Electro Thermal SIMulator) with careful circuit modelling. The discharge path through power protection circuits, and coupling effects between VCC and VSS power lines are also presented.

<P><B>Abstract</B></P> <P>Glutathione reductase (GSR) is an enzyme that catalyzes the biochemical conversion of oxidized glutathione (GSSG) into the reduced form (GSH). Since the ratio between the two forms of glutathione (GSH/GSSG) is important for the optimal function of GSH to act as an antioxidant against H<SUB>2</SUB>O<SUB>2</SUB>, the contribution of GSR as an enzymatic regulatory agent to maintain the proper ratio is essential. Abalones are marine mollusks that frequently encounter environmental factors that can trigger the overproduction of reactive oxygen species (ROS) such as H<SUB>2</SUB>O<SUB>2</SUB>. Therefore, we conducted the current study to reveal the molecular and functional properties of a GSR homolog in the disk abalone, <I>Haliotis discus discus</I>. The identified cDNA sequence (2325 bp) has a 1356 bp long open reading frame (ORF), coding for a 909 bp long amino acid sequence, which harbors a pyridine nucleotide-disulfide oxidoreductase domain (171–246 aa), a pyridine nucleotide-disulfide oxidoreductase dimerization domain, and a NAD(P)(+)-binding Rossmann fold superfamily signature domain. Four functional residues: the FAD binding site, glutathione binding site, NADPH binding motif, and assembly domain were identified to be conserved among the other species. The recombinant abalone GSR (rAbGSR) exhibited detectable activity in a standard glutathione reductase activity assay. The optimum pH and optimal temperature for the reaction were found to be 7.0 and 50 °C, respectively, while the ionic strength of the medium had no effect. The enzymatic reaction was vastly inhibited by Cu<SUP>+2</SUP> and Cd<SUP>+2</SUP> ions. A considerable effect of cellular protection was detected with a disk diffusion assay conducted with rAbGSR. Moreover, an MTT assay and flow cytometry confirmed the significance of the protective role of rAbGSR in cell function. Furthermore, <I>AbGSR</I> was found to be ubiquitously distributed in different types of abalone tissues. <I>AbGSR</I> mRNA expression was significantly upregulated in response to three immune challenges: <I>Vibrio parahaemolyticus</I>, <I>Listeria monocytogenes</I>, and lipopolysaccharide (LPS), thus indicating its possible involvement in host defense mechanisms during pathogenic infections. Taken together, the results of the current study suggest that AbGSR plays an important role in antioxidant-mediated host defense mechanisms and also provide insights into the immunological contribution of AbGSR.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We identified a glutathione reductase homolog (AbGSR) from disk abalone. </LI> <LI> AbGSR resembled functionally important domain architecture of GSR family. </LI> <LI> Recombinant AbGSR confirmed its biochemical properties via enzymatic assays. </LI> <LI> First functional antioxidant properties assessment of a molluscan GSR. </LI> <LI> <I>AbGSR</I> expression was modulated upon induced pathogen stress in gill and hemocytes. </LI> </UL> </P>

<P>4-1BB (CD137) is expressed on dendritic cells (DCs) and its biological function has remained largely unresolved. By comparing 4-1BB-intact (4-1BB(+/+)) and 4-1BB-deficient (4-1BB(-/-)) DCs, we found that 4-1BB was strongly induced on DCs during the maturation and that DC maturation was normal in the absence of 4-1BB. However, DC survival rate was low in the absence of 4-1BB, which was due to the decreased Bcl-2 and Bcl-x(L) in 4-1BB(-/-) DCs compared with 4-1BB(+/+) DCs after DC maturation. Consistent with these results, 4-1BB(-/-) DCs showed an increased turnover rate in steady state and more severely decreased in spleen by injecting LPS compared with 4-1BB(+/+) DCs. When OVA-pulsed DCs were adoptively transferred to recipient mice along with OVA-specific CD4(+) T cells, 4-1BB(-/-) DCs did not properly migrate to the T cell zone in lymph nodes and poorly induced proliferation of CD4(+) T cells, although both DCs comparably expressed functional CCR7. Eventually, 4-1BB(-/-) DCs generated a reduced number of OVA-specific memory CD4(+) T cells compared with 4-1BB(+/+) DCs. To further assess the role of 4-1BB on DC longevity in vivo, 4-1BB(+/+) and 4-1BB(-/-) C57BL/6 were administrated with Propionibacterium acnes that develop liver granuloma by recruiting DCs. Number and size of granuloma were reduced in the absence of 4-1BB, but the inflammatory cytokine level was comparable between the mice, which implied that the granuloma might be reduced due to the decreased longevity of DCs. These results demonstrate that 4-1BB on DCs controls the duration, DC-T interaction, and, therefore, immunogenicity.</P>
