      • 돼지 卵子의 透明帶에 대한 單一클론抗體生産과 그 特性에 關한 硏究

        本 試驗은 單一클론抗體의 강한 特異性과 抗體性質의 不變性을 이용하여 發生學的 側面에서 哺乳動物 卵子의 透明帶의 機能과 構造를 이해하고, 또한 種特異的인 精子 受容體의 存在 및 生化學的 構造를 규명하기 위한 기초연구로서, 돼지 卵子의 透明帶를 免疫原으로 하여 BABL/c 생쥐로부터 單一클론抗體를 생산하고 그 특성을 구명하였던 바 다음과 같은 결과를 얻었다. 1) 3마리의 BABL/c 생쥐(YⅠ, YⅡ, ZI)에 돼지卵子의 透明帶를 免疫化하고, 複合抗體 生成을 확인한 후 생쥐의 脾臟細胞와 Myeloma(SP2/O-Ag14)를 polyethylene glycol를 融合을 실시한 결과 각각 25.8%, 54.5% 그리고 59.7%의 融合效率을 나타내었으며, ELISQ에 의해 陽性反應을 조사한 결과 각각 17.3%, 32.6% 그리고 6.2% 陽性反應 效率을 나타내었다. 2) YI에서 강한 陽性反應을 보인 6개의 well에 대한 cloning을 실시하고 抗體檢證을 행한 결과 20.8% ∼ 48.4%의 Cloning效率과 54.6% ∼ 82%의 陽性反應 效率을 나타내었다. 3) 강한 陽性反應을 나타낸 항체에 대해 sandwich ELISA法에 의해 isotype을 決定하였던바 2E93C(YⅠ), 3E83E7(YⅠ), 4E3(YⅡ)각각 IgG₂b, IgG₂a, IgM으로 확인되었다. 4) Isotype이 決定된 2E93C9(YⅠ), 3E84E7(YⅠ), 4E3(YⅡ)의 세포를 생쥐의 腹腔에 주사하여 얻은 腹水를 indirect ELISA法에 의해 titer를 決定한 결과 모두 1:400,000 이상의 높은 titer를 나타내었다. 5) 處理區로서 單一클론抗體의 腹水와 對照區로서 normal mouse serum이 각각 2%씩 함유된 배양액속에서 난자를 배양한 후 顯徵鏡下에서 관찰했을 때 對照區에서 배양된 난자의 표면은 정상적인 형태를 나타냈으나 處理區에서 배양된 卵子는 표면에 뚜렷한 沈澱層을 형성하였다. 6) 處理區와 對照區 卵子를 Rabbit anti-mouse IgG-FITC가 1% 함유된 배양액속에서 배양하고 洗滌한 후 螢光顯徵鏡下에서 관찰한 바 處理區의 卵子는 透明帶 주위에서 螢光이 나타났으나, 對照區에서는 나타나지 않았다. This study was carried out ot produce and characterize monoclonal antibodies against porcine zona pellucida, and undertaken as a basic study to develop immunocontraceptive vaccine and to investigate the function of zana pellucida in early fertilization process. The results obtained were summarized as follows: 1. Spleen cells of three BALB/C mice(YⅠ, YⅡ and ZI) which were immunized with porcine zona pelucida were fused with myeloma cells(SP2/O-Ag14) by polyethylene glycol. At the result of fusion, fusion efficiency was 25.8 , 54.5% and 59.7% and positive efficiency 17.3%, 32.6% and 6.2%, respectively. 2. Cloning efficiency was shown to be from 20.8% to 48.4% and positive efficiency of them were 54.6% to 82%. 3. Sub-isotypes of three strong positive antibodies, 2E93C(YⅠ), 3E83E7(YⅠ) and 4E3(YⅡ) were determined by sandwich ELISA method and shown to be as IgG2b, IgH2a and IgM, respectively. 4. The titers of three ascitic fluids containing antibodies, 2E93C9(YⅠ), 3E84E7(YⅠ) and 4E3(YⅡ) were determined by indirect ELISA and all of them showed above 1:400,000. 5. The layer of precipitate was observed on the zona incubated with medium containing 2% ascitic fluid of monoclonal antibody while the eggs treated with 2% normal mouse serum did not. 6. Porcine eggs incubated with medium containing 2% ascitic fluid of monoclonal antibody and followed by subsequent incubation with Rabbit anti-mouse IgG-FITC conjugate showed strong fluorescent light on the zona surface while the zona of normal mouse serum-treated eggs did not show fluorescence.

      • The effects of Sm and Y addition on the properties of YBCO thin film prepared by a DCA-MOD method

        Off-stoichiometric (Y<SUB>1</SUB>RE<SUB>x</SUB>)Ba<SUB>2</SUB>Cu<SUB>3</SUB>O<SUB>7-δ</SUB>; (x=0-0.4, RE=Y or Sm) films have been prepared on LaAlO<SUB>3</SUB> (100) single-crystal substrates by a metal-organic deposition using dichloroacetic acid as chelating solution. Coating solutions with excess Sm or Y were prepared by dissolving Y, Sm, Ba and Cu acetates in dichloroacetic acid and distilled water followed by refluxing, drying to obtain blue gel. The final precursor solution for dip coating was prepared by diluting blue gel in 2-methoxyethanol. Coated films were calcined at low temperature up to 500<SUP>o</SUP>C in flowing humid oxygen atmosphere. Conversion heat treatment was performed at 800<SUP>o</SUP>C for 2h in flowing Ar gas containing 1000ppm oxygen with a humidity of 9.45%. The viscosities of the precursor solution were increased as the content of added Sm or excess Y increased. The transport critical current density (J<SUB>c</SUB>) of (Y<SUB>1</SUB>RE<SUB>x</SUB>)Ba<SUB>2</SUB>Cu<SUB>3</SUB>O<SUB>7-δ</SUB>; film was enhanced with the addition of excess Sm and Y up to 20% and then decreased with further addition. Scanning electron microscope (SEM) observation showed that surface roughness was increased with the addition of excess Y.

      • F-free Y & Cu 전구용액 적용에 의한 YBCO coated conductors의 MOD 공정 개선

        Total Fluorine content in the precursor solution for MOD processing of YBCO coated conductors can be significantly reduced by synthesizing precursor solution with F-free Y & Cu precursor and Barium trifluoroacetate(TFA). It was shown that crack-free and uniform precursor films were formed after calcinations in humidified oxygen atmosphere. Less than 2 hours are required to finish the calcination process, and XRD measurement shows that $BaF_2,\;CuO,\;and\;Y_2O_3$ are major constituent of calcined precursor films. Film thickness after calcination was measured to be ${\sim}2.8$ um by applying slot-die coating method. In particular, addition of Samarium shows critical current of Ic=226 A/cm-w($Jc=3.4\;MA/cm^2$). Also discussed are recent developments in the reel-to-reel processing using F-free Y & Cu precursor solutions. It is shown that uniform and fast processing route to YBCO coated conductor with high Ic can be provided by employing F-free Y & Cu precursor solutions in MOD process.

        액체금속로 Y-구조물의 비탄성 열응력 해석 및 손상평가에 관한 유한요소해석

        LMFBR(Liquid Metal Fast Breeder Reactor) vessel is operated under the high temperatures of 500-550.deg. C. Thus, transient thermal loads were severe enough to cause inelastic deformation due to creep-fatigue and plasticity. For reduction of such inelastic deformations, Y-piece structure in the form of a thermal sleeve is used in LMFBR vessel under repeated start-up, service and shut-down conditions. Therefore, a systematic method for inelastic analysis is needed for design of the Y-piece structure subjected to such loading conditions. In the present investigation, finite element analysis of heat transfer and inelastic thermal stress were carried out for the Y-piece structure in LMFBR vessel under service conditions. For such analysis, ABAQUS program was employed based on the elasto-plastic and Chaboche viscoplastic constitutive equations. Based on numerical data obtained from the analysis, creep-fatigue damage estimation according to ASME Code Case N-47 was made and compared to each other. Finally, it was found out that the numerical predictio of damage level due to creep based on Chaboche unified viscoplastic constitutive equation was relatively better compared to elasto-plastic constitutive formulation.

      • 착유우의 사료섭취량과 음수량이 산유량과 분뇨 배설량에 미치는 영향

        본 실험은 우리나라에서 사육되고 있는 홀스타인 젖소 착유우의 사료 섭취량과 음수량이 산유량과 분뇨 배설량에 미치는 영향을 구명하기 위하여 수행하였다. 실험은 착유우 10두를 공시하여 수원의 축산연구소 낙농우사에서 봄, 여름, 가을, 겨울 등 4계절에 걸처 실시하였으며 그 결과는 다음과 같다. 1. 시험에 공시된 착유우의 건물섭취량은 평균 19.5kg으로 봄과 가을이 각각 23.9, 22.1kg으로 높았고, 여름과 겨울이 각각 17.0, 15.3 kg으로 낮은 섭취량을 보였다. 2. 착유우의 음수량은 평균 $77.2\ell$로, 여름이 $85.5\ell$로 가장 많았고 겨울이 $62.2\ell$로 가장 낮았으나 계절별 유의차는 보이지 않았다. 3. 착유우의 산유량은 평균 26.0kg으로 봄, 여름, 가을 및 겨울이 각각 30.8, 24.0, 25.4, 23.7kg으로 봄의 산유량이 가장 높았다(p<0.05). 4. 착유우의 분뇨 배설량을 계절별로 조사한 결과, 봄 64.4kg, 여름 63.5kg, 가을 60.4kg, 겨울 51.0kg으로 계절 간에 차이를 보였으며(p<0.05), 전체 평균 분뇨 배설량은 59.8kg이었다. 5. 착유우의 사료섭취량, 음수량, 산유량 및 분뇨 배설량과의 상관관계를 분석한 결과, 산유량과 음수량$(R^2=0.7742)$, 산유량과 사료 섭취량$(R^2=0.7459)$, 음수량과 뇨 배설량 $(R^2=0.7422)$, 사료 섭취량과 분 배설량$(R^2=0.6044)$, 산유량과 분 배설량$(R^2=0.692)$의 상관관계가 비교적 높았으나, 음수량과 분 배설량$(R^2=0.2950)$, 사료 섭취량과 뇨 배설량$(R^2=0.1985)$, 산유량과 뇨 배설량$(R^2=0.2335)$의 상관관계는 낮게 나타났다. 6. 따라서 산유량과 음수량, 산유량과 사료 섭취량의 상관관계식은 $Y=0.1919X_1+11.181(R^2=0.7742),\;Y=0.8568X_2+9.3067(R^2=0.7459)$(Y=milk yield $X_1=water$ consumption, $X_2=feed$ intake)로 추정할 수 있다. This experiment was conducted to determine the effect of feed intake and water consumption on milk yield and manure production in milking cows. The average feed intake(dry matter) of milking cows were 19.5kg/hd/d. Spring(23.9kg) and fall(22.1kg) feed intake rates when higher than in the summer(17.0kg) and winter(15.3kg/hd/d). The average water consumption of milking cows were $77.2\ell/hd/d$. Summer showed the highest value$(85.5\ell/hd/d)$ and winter showed the lowest value$(62.2\ell/hd/d)$. The average milk yield during spring, summer, fall, and winter was 30.8, 24.0, 25.4, and 23.7kg/hd/d, respectively. Milk yield during spring was found to be statiscally greater than for the other seasons. Manure production of milking cows during spring, summer, fall, and winter was 64.4, 63.5, 60.4, and 51.0kg/hd/d, respectively. Consequently, a relatively high correlation between milk yield and water consumption$(R^2=0.7742)$, milk yield and feed intake$(R^2=0.7459)$, water, consumption and urine production$(R^2=0.7422)$, feed intake and feces production$(R^2=0.6044)$, and milk yield and feces production$(R^2=0.6920)$ were observed in milking cows. The other hand, correlation between water consumption and feces production$(R^2=0.2950)$, feed intake and urine production $(R^2=0.1985)$, and milk yield and urine production$(R^2=0.2335)$ were found to be relatively low. Therefore, correlation equation between milk yield and feed intake, milk yield and water consumption can be estimated from : $Y=0.1919X_1+11.181(R^2=0.7742),\;Y=0.8568X_2+9.3067(R^2=0.7459)$(Y=milk yield $X_1=water$ consumption, $X_2=feed$ intake).

      • 중년층의 비처방약품 오남용 실태 연구

        Purpose: The purpose of this study was to grasp the status of abusing Over The Counter Drug (OTC Drug) among the middle-aged. Methods: The participants of this study consisted of 175 middle-aged people in Seoul and Gyeoggi-do. Data were collected using a questionnaire, and the study was conducted from September 10 to October 10, 2013. The data were analyzed by t-test, chi-square test, one-way ANOVA and Turkey. Results : As a result, 97 participants(55.4%) out of 175 participants of this study had taken over the counter drug within the past one year. Their average score of abusive behavior of OTC drug was 6.96±3.01 out of 16. There was significant abusive behavior of OTC drug relative to the difference in family monthly income.(F=3.455, p=0.20) and there was significant difference depending on the information source of the OTC Drug.(F=2.56, p=.044) Conclusions : In conclusion, nurses should educate middle-aged people so that they can use over the counter drug more safely and accurately based on these results. In addition, these results will be useful in further studies regarding middle-aged people’s drug abuse according to the preceding data.

      • 셀프리더십에 따른 간호사의 직무만족도

        In this study, we investigated how much job-satisfaction and self leadership nurses have. And making use of self leadership, we hope to enhance job- satisfaction and develop nurse organization. To find out the relationship between nurse's self leadership and Job-satisfaction, we carried out a sample survey : The subjects of our study are 150 nurses being in offices(hospitals) at the two university hospitals and three general hospitals in Seoul or Keong-gi province. Nurses agreed with and understood the purpose of our study. For analyzing the result, we used ANOVA of SPSS 14.0. The main results of our study are as follows : According to characteristic of subjects, level of self leadership has a meaningful difference statistically according to marriage, religion, department career, position. As for the marriage, the level of self leadership of the married are higher((F=19.95, p= .000) than the unmarried. . Level of self leadership perceived by subjects is an average 3.50 point. An average points of each factors are as follows : Self-compensation is 3.75 point, rehearsal is 3.57 point, self-expectation is 3.55 point, goal-setting is 3.53 point, self-criticism is 3.31 point, constructive thinking is 3.27 point. The total average is 62.9 point to 90 full marks. Level of self job-satisfaction perceived by subjects is an average 2.98 point to 5 full marks. Level of self job-satisfaction perceived by subjects is an average 2.98 point to 5 full marks. An average points of each factors are as follows : Interrelation is 3.42 point, interrelation between doctors and nurses is 3.35 point, job demand is 3.10 point, administration is 3.02 point, a professional position is 2.94 point, self control is 2.84 point, payment is 2.44 point. A coefficient of co-relation of self leadership and self job-satisfaction is .406 point, a meaning probability is .000 point and they seem to have a close interrelation. Rehearsal among factors of self leadership has the most interrelation with payment of self job-satisfaction. A professional position of self job-satisfaction has the most interrelation with self expectation(r= .314, p= .000). A job demand has the most interrelation with constructive thinking(r= .272, p= .001). A interaction has the most interrelation with self expectation(r= .272. p= .000). A administration has the most interrelation with constructive thinking(r= .411, p= .000). An interrelation between doctors and nurses has the most interrelation with self expectation(r= .385, p= .000). Our study shows there is a close relation between self leadership and job-satisfaction. Especially, self expectation and constructive thinking, the factor of self leadership, appear to affect the nurse's job-satisfaction. It appears that nurses feel to have a intensive job-satisfaction, having positive self leadership, a sense of purpose, and constructive thought. Thus, we need to steadily study self leadership, self control, and internal motivation as a new paradigm focused on self-control and internal motivation for their self-developments and advancement of nurses' organizations.

      • 20대여성의 유방자가검진에 대한 지식과 자기효능감의 관계연구

        The purpose of this study was to know relationship between important variables which influence lifestyle in college women`s health promotion, prove adjustable factors and provide basic data which develops nursing intervention health promotion program. The method of this study was descriptive correlational study. The convenience sample was 206 college woman who live alone in lodgings, dormitory, a reading room or off-campus housing. Research instruments were the Multidimensional Health Locus of Control(MHLC) developed by Wallston, Wallston & Devellis(1978), Interpersonal Support Evaluation List developed by Cohern, Hoberman(1983) and The Health Promotion Lifestyle Profile(HPLP) developed by Walker, Pender & Sechrist(1978) and the data was used after Factor Analysis. The result of this study was follows. 1. It can be found that the level of the college woman`s health promotion lifestyle was 2.46±.33 with higher fulfilling level: According to general characteristic, the level of health promotion lifestyle was 5% significant statistical differences by age and period of menstruation. 2. The level of health locus of control was 3.25±.31 with higher fulfilling level. 3. The interpersonal support level was 3.05±.44 with higher fulfilling level. 4. The level of interpersonal support turned out most convincing factor in health promotion lifestyle (9%). In addition to age factor, two factors was 10% power of explanation in whole health promotion.

      • Oxidation suppression characteristics of the YSZ coating on Mg<sub>2</sub>Si thermoelectric legs

        Oxide-based coating layers were investigated to suppress the oxidation of an intermediate temperature thermoelectric module that operates in a temperature range of 300-700<SUP>o</SUP>C. Four different oxides [alumina (Al<SUB>2</SUB>O<SUB>3</SUB>), yttria (Y<SUB>2</SUB>O<SUB>3</SUB>), 8mol% yttria-stabilized zirconia ((Y<SUB>2</SUB>O<SUB>3</SUB>)<SUB>0.08</SUB>(ZrO<SUB>2</SUB>)<SUB>0.92</SUB>, YSZ), and 20mol% samaria-doped ceria (Sm<SUB>0.2</SUB>Ce<SUB>0.8</SUB>O<SUB>1.9</SUB>, SDC)] were coated onto Mg<SUB>2</SUB>Si thermoelectric leg surfaces and their anti-oxidation characteristics were compared. While Al<SUB>2</SUB>O<SUB>3</SUB>, Y<SUB>2</SUB>O<SUB>3</SUB>, and SDC layers failed to prevent the progression of oxidation of the thermoelectric Mg<SUB>2</SUB>Si surface at 700<SUP>o</SUP>C, YSZ showed potential as an anti-oxidation coating layer for intermediate temperature thermoelectric modules. The apparent oxidation suppression characteristics of the YSZ layer on the Mg<SUB>2</SUB>Si thermoelectric material were studied by a microstructural investigation of the interface between the coated layer and the thermoelectric material. The reliability of the YSZ coating structure was investigated via thermal cycling tests from room temperature to 600<SUP>o</SUP>C. The practical adaptability of the YSZ anti-oxidation coating in terms of use for thermoelectric power-generation modules is discussed in detail with the results of variation in contact resistance.

      • Novel dentin phosphoprotein frameshift mutations in dentinogenesis imperfecta type II

        <P>Lee K‐E, Kang H‐Y, Lee S‐K, Yoo S‐H, Lee J‐C, Hwang Y‐H, Nam KH, Kim J‐S, Park J‐C, Kim J‐W. Novel dentin phosphoprotein frameshift mutations in dentinogenesis imperfecta type II.</P><P>The dentin sialophosphoprotein (<I>DSPP</I>) gene encodes the most abundant non‐collagenous protein in tooth dentin and DSPP protein is cleaved into several segments including the highly phosphorylated dentin phosphoprotein (DPP). Mutations in the <I>DSPP</I> gene have been solely related to non‐syndromic form of hereditary dentin defects. We recruited three Korean families with dentinogenesis imperfecta (DGI) type II and sequenced the exons and exon–intron boundaries of the <I>DSPP</I> gene based on the candidate gene approach. Direct sequencing of PCR products and allele‐specific cloning of the highly repetitive exon 5 revealed novel single base pair (bp) deletional mutations (c.2688delT and c.3560delG) introducing hydrophobic amino acids in the hydrophilic repeat domain of the DPP coding region. All affected members of the three families showed exceptionally rapid pulp chambers obliteration, even before tooth eruption. Individuals with the c.3560delG mutation showed only mild, yellowish tooth discoloration, in contrast to the affected individuals from two families with c.2688delT mutation. We believe that these results will help us to understand the molecular pathogenesis of DGI type II as well as the normal process of dentin biomineralization.</P>
