Production of α- and β-Galactosidases from Bifidobacterium longum subsp. longum RD47

Approximately 50% of people in the world experience abdominal flatulence after the intake of foods containing galactosides such as lactose or soybean oligosaccharides. The galactoside hydrolyzing enzymes of α- and β-galactosidases have been shown to reduce the levels of galactosides in both the food matrix and the human gastrointestinal tract. This study aimed to optimize the production of α- and β-galactosidases of Bifidobacterium longum subsp. longum RD47 with a basal medium containing whey and corn steep liquor. The activities of both enzymes were determined after culturing at 37oC at pH 6.0 for 30 h. The optimal production of α- and β-galactosidases was obtained with soybean oligosaccharides as a carbon source and proteose peptone no. 3 as a nitrogen source. The optimum pH for both α- and β-galactosidases was 6.0. The optimum temperatures were 35oC for α-galactosidase and 37oC for β-galactosidase. They showed temperature stability up to 37oC. At a 1 mM concentration of metal ions, CuSO4 inhibited the activities of α- and β-galactosidases by 35% and 50%, respectively. On the basis of the results obtained in this study, B. longum RD47 may be used for the production of α- and β-galactosidases, which may reduce the levels of flatulence factors.
Oncogenic NRF2 mutations in squamous cell carcinomas of oesophagus and skin

<P>Nuclear factor erythroid-related factor 2 (NRF2) encodes a transcription factor that induces expression of cytoprotective proteins upon oxidative stress and oncogenic NRF2 mutations have been found in lung and head/neck cancers that inactivate KEAP1-mediated degradation of NRF2. The aim of this study was to catalogue NRF2 mutations in other human cancers. For this, we analysed 1145 cancer tissues from carcinomas from oesophagus, skin, uterine cervix, lung, larynx, breast, colon, stomach, liver, prostate, urinary bladder, ovary, uterine cervix, and kidney, and meningiomas, multiple myelomas, and acute leukaemias by single-strand conformation polymorphism (SSCP) assay. We detected NRF2 mutations in oesophagus (8/70; 11.4%), skin (1/17; 6.3%), lung (10/125; 8.0%), and larynx (3/23; 13.0%) cancers. Of note, all of the 22 mutations except one were found in squamous cell carcinomas (SCCs) (95.5%). The mutations were observed within or near DLG and ETGE motifs that are important in NRF2 and KEAP1 interaction. All of the oesophageal SCCs and skin SCCs with the NRF2 mutations showed increased NRF2 expression in the nuclei. However, none of the SCCs from oesophagus and skin harboured KEAP1 mutation. Our study demonstrated here that NRF2 mutation occurs not only in lung and head/neck cancers, but also in oesophageal and skin cancers. Objectives: The purpose of this study was to review previous clinical studies on underlying mechanisms of sleep-related type-specific pathophysiological symptoms among the Sasang types.Methods and Procedure: We reviewed seven research databases from December 2003 to August 2015 with the keywords Sasang typology, constitution and sleep. The Sasang type-specific sleep-related symptoms were analyzed based on seven categories, including subjective sleep quality, sleep latency, sleep duration, habitual sleep efficiency, sleep disturbances, use of sleep medications and daytime dysfunction.Results: Total of twelve studies were included in the analysis. The Tae-Yang type showed low subjective sleep quality, long sleep latency and frequent awakening, and the So-Yang type showed long sleep latency. The Tae-Eum type presented high subjective sleep quality, short sleep duration, frequent snore, toss and turn during the sleep, and insomnia and low frequency of dream. The So-Eum type showed long sleep latency in male and high frequency of dream. The Eum type combining Tae-Eum and So-Eum types had higher subjective sleep quality, longer sleep duration and higher frequency of dream than the Yang type combining Tae-Yang and So-Yang types.Conclusions: This study reviewed type-specific sleep-related characteristics and discussed possible pathophysiological mechanisms involved. Differences in sleep characteristics among the Sasang types might stem from type-specific temperaments and require further study.

The egg development and early life history of Korean spotted sleeper, Odontobutis interrupta which is Korean endemic species from Sora-choen was investigated. The Korean spotted sleeper were caught at Sora-myeon, Yeosu-si, Jeollanamdo, from Korea at May in 2014. The fertilized eggs were 4.23 ± 0.05 mm in long diameter and had oil globules. Hatching time of the embryo began about 442 hr 14 min after fertilization under water temperature of 19.5oC. The newly hatched larvae were 4.27 ± 0.35 mm in total length and their anus were not yet opened. 3 days after hatching postlarvae was measured 6.20 ± 0.11 mm in total length. 10 days after hatching postlarvae was measured 6.69 ± 0.14 mm in total length.
Effect of Opuntia humifusa Extract on Indomethacin-induced Gastric Ulcer in Sprague Dawley Rat

Phytogenic agents have traditionally been used by herbalists and indigenous healers for the prevention and treatment of gastric ulcers. The prickly pear (Opuntia humifusa) is a member of the Cactaceae family, and is widely distributed in all of the semiarid countries throughout the world especially in the Mediterranean area and Central America. Most of Cactaceae family is known to be therapeutic properties for arthritis, cancer, and so on. In this study, we examined the antiulcer effect of Opuntia humifusa (OH) on the indomethacin (30 ㎎/㎏, p.o)-induced gastric ulcer in Sprague-Dawley (SD) rats. Oral administration of OH (50, 100, and 200 ㎎/㎏) once a day for 7 days dose-dependently prevented the indomethacin-induced gastric mucosal injury and accelerated the gastric ulcer healing. The pH of stomach inner-surface wall in OH pretreatment group were higher than the pH value of the control group. Gastric wall mucus was significantly preserved by the OH pretreatment at doses of 100 and 200 ㎎/㎏. These results indicate that OH has potent gastric cytoprotective and ulcer healing effects.
