        Temporal bone high resolution computed tomogram(TBHRCT)is a essential diagnostic tool in considering for cochlear implantation in patients with profound sensorineural hearing loss. This study was designed for an effort to provide and efficacy of TBHRCT in determination of inner ear malformations for the patients who have profound sensorineural hearing loss and to examine the relationships between TBHRCT findings and other preoperative evalutions for cochlear implantation in that patients. Authors examined 68 deaf patients to provide fundamental data for cochlear implantation. Inner ear abnormalities were founded in 13(19%) patients, among which 10(14.7%) patients had Mondini'dysplasia in which 5 patients were hypoplastic type and 5 patients were hydrops type. Other inner ear abnormalities were as follows : lateral semicircular canal hypoplasia ; internal auditory canal abnormality ; cochlear aqueduct widening ; vestibular aqueduct widening. No relationships were found between TBHRCT findings and other test results. : promontory test, K-WISC cognitive test etc.(Korean J Otolaryngol 39 : 8, 1996)

      • 부재료를 달리한 깍두기와 배추김치의 pH변화 및 관능검사

        To investigate the effect of individual seasoning on the guality during fermentation of Kakdugi and Chinese cabbage Kimchi(Kimchi below), the most popular seasonings such as sugar, galic, red pepper, ginger and salted anchovies were used. First of all Kakdugi and Kimchi which were soaked in 5% brine for 1 hour at 13℃ were found to have the best quality. At this condition, sugar, galic and salted anchovies hastened the aging of fermentation of Kakdugi and Kimchi, while red pepper prolonged it. But, ginger didn't have any effection that. The data showed that each seasoning doesn't have it's own role in sensory evaluation and all the experimental samples have their own fermentation period which has the highest score at pH 4.3.

        This study is concerned with the corporate image strategy of aviation company. Greater focus on this study has brought attention to corporate advertising. The corporate image strategy of aviation company may be divided into two types : software and hardware. the focus of this study is hardware image strategy and theoretical background is the theory of cognitive responses. On this basis, three image strategies of contribution to the corporate advertising are classified. 1) image strategy according to preemptive strategy, 2) image strategy by convenience, 3) image strategy by economic appeal. And then, field cases are presented. Each of these cases have a related advertising strategy. Finally, based on these case study, some guidelines are recommended for corporate manager involved in corporate advertising and further research directions are discussed

      • Metal Ion Dependence of Pyrophosphate-Dependent Phosphofructokinase

        Cr(H₂O)₄ PPi was synthesized to test two metal dependent phosphorylation theory of fructose 6-phosphate by pyrophosphate-dependent phosphofructokinase. Cr(H₂O)₄ PPi acted as a competitive inhibitor in the absence of second metal ion with two dissociation constants indicating the possibility of the presence of activation site by pyrophosphate. To test whether or not Cr(H₂O)₄ PPi acts as a substrate, [³²P]Cr(H₂O)₄PPi was synthesized and incubated in the presence of second metal ion. The results indicate that Cr(H₂O)₄PPi acts as a substrate in the presence of second metal ion with a thousandth of regular activity. After binding of Cr(H₂O)₄PPi , saturation of MgPPi did not recover the activity completely indicating tight binding of Cr(H₂O)₄PPi . In other words, only binding of pyrophosphate before Cr(H₂O)₄PPi protects the inhibition completely.

      • 과민성 대장 증후군 환자에서 Colpermin 투여의 임상 효과

        목적 : 과민성 대장 증후군은 장기능 이상의 증상군으로, 인지할 수 있는 기질적 위장관 병태 생리가 없는 경우로 기능적인 장관의 이상 경축과 가장 밀접한 관련이 있을 것으로 생각된다. 그러나 치료면에서는 아직 뚜렷한 방침이 정립되어 있지 않고 다양한 방법들이 제시되고 있는 실정이다. 저자들은 장용성 캅셀로 조제된 박하유 성분인 Colpermin® 의 임상 효과를 평가하고자 본 연구를 시행하였다. 대상 및 방법 : 1996년 2월부터 1996년 11월까지 경북대학교병원 소화기 내과를 방문하여 과민성대장 증후군으로 진단된 34명의 환자를 대상으로 Colpermin® 을 1회 2캅셀씩, 1일 3회, 경구로 2주간 투여하였다. 결과 : 투여전의 증상의 빈도는 복통 88.2%(30/34) 및 설사 73.5%(25/34)를 호소하는 경우가 가장 많았으며, 그 다음으로 변비 50.0%(17/34), 복부 팽만 47.1%(16/34), 고창 38.2%(13/34)순이었다. 약물 투여후 증상별 개선율은 설사를 보였던 환자들에서 호전 11예와 매우 호전 11예로 88.0%(22/25)의 가장 높은 개선율을 보였고, 그 다음으로 복통 73.3%(22/30), 고창 69.2%(9/13), 변비 64.7%(11/17), 복부 팽만 56.3%(9/16)의 순이었다. 종합적인 증상의 개선도는 현저한 개선이 2예(5.9%), 중등도 개선이 9예(26.5%), 약간 개선이 18예(52.9%), 불변이 4예(11.8%), 악화가 1예(2.9%)였으며, 종합적인 유효율은 85.3%(29/34)였다. 약물의 부작용으로 3예의 환자에서 각각 복부 팽만의 발생, 변비의 악화, 복통의 발생이 있었으나, 그 외의 환자들에서는 아무런 부작용도 관찰되지 않았다. 결론 : 과민성 대장 증후군의 약물치료에 있어서 Colpermin®이 유용한 약제가 될 수 있을 것으로 기대된다. Irritable bowel syndrome (IBS) is a disease characterized by a collection of symptoms of intestinal dysfunctions, without any recognized organic gastrointestinal pathology. This study was performed to evaluate clinical effectiveness of peppermint oil (Colpermin®) in the treatment of patients with IBS. 34 patients (18 males and 16 females) with IBS were treated with peppermint oil 0.4ml (as menthol 176㎎) three times daily for 2 weeks and the efficacy of the treatment was evaluated according to the degree of the improvement. The improvement of symptoms was 88.0% (22/25) in diarrhea, 73.3% (22/30) in abdominal pain, 69.2% (9/13) in flatulence, 64.7% (11/17) in constipation, and 56.3% (9/16) in abdominal distension, respectively. Overall effectiveness of subjective symptoms was 85.3% (29/34). Side effects of the drug were noted in only 3 cases, those were aggravation of constipation, development of abdominal distension and abdominal pain. As a result, Colpermin® might be considered as an effective drug in relieving symptoms of IBS.

      • Comparing Free Energy Diagrams of Enzyme-Catalyzed Reactions to Un-Catalyzed Reactions

        Generalized kinetics for comparing enzyme -catalyzed and un-catalyzed reaction kinetics within the traditional thermodynamic box examined with the aid of a new convention. If the concentration of substrate is set equal to its dissociation constants and the concentration of enzyme is adjusted so that rates of reaction are equal. then the catalytic power of the enzyme must be algebraically equal to the ratio of these two concentrations, or [S]/[E]. This singular, model-independent assessment avoids ambiguities associated with comparisons at the extremes of high and low concentration of substrate, and normalizes molecularities within and between reactions. Normalized free energy diagrams for differing mechanisms of enhancing rates of catalysis, such as stabilizing the transition-state or destabilizing the reactant ground-state, are identical which resolves recent and long term controversies and suggests that isolated changes in free energy diagrams without normalization may be misleading. The use of a diffusion-controlled Kcat/Km as a criterion for a perfectly evolved enzymes is challenged : (a) because diffusion-controlled kinetics may obtain also when the catalytic power is compromised, (b) because Kcat,/Km does not encompass a complete turmover and is insensitive to late partially rate-limiting steps, and (c) because rates of diffusion for different substrates and enzymes differ, which renders the endpoint of a comparison uncertain. As an altermative that is free of these limitations, a new criterion is offered: a Km which exceeds physiological concentrations of substrate by two-fold or more requires an enzyme to be operating physiologically within 50% of the diffusion-controlled limit.

        Ethyl-(4-hydroxyphenyl) oxamate Sodium Salt As a Strong Melanin Biosynthesis Inhibitor

        Preliminary data show that hydroxyaniline derivatives have strong inhibitory activities against mushroom tyrosinase. Twelve out of thirty-four synthesized hydroxyaniline derivatives demonstrated inhibitory activities, among which ethyl-(4-hydroxyphenyl)oxamate sodium salt (EHPONa) was found to be the most effective inhibitor. Inhibitory activities of hydroxyaniline derivatives were much higher than that of commercially available whitening agent arbutin, and comparable to that of kojic acid. Data showed that EHPONa x had a higher inhibitory activity of melanin biosynthesis than those of both kojic acid and arbutin. Based on the study of inhibition kinetics and cupric chloride effects on enzyme activity, we propose that 4-hydroxyl oxygen and oxamate oxygen moiety of EHPONa chelate the two cupric ions at the active site, and that phenol moiety interacts with the hydrophobic enzyme pocket. Accordingly, EHPONa may be a good candidate as a skin-whitening agent.

        Reaction Mechanism of Chloramphenicol Acetyltransferase

        Initial velocity, product inhibition and dead-end inhibition patterns suggest that chloramphenicol acetyltransferase catalyzes its reaction by a rapid equilibrium random mechanism, wherein the interconversion of central complex is a rate-limiting step in both forward and reverse reactions. The results from chemical modification and reactant protection experiments suggest that histdine and aspartic acid may be involved in catalysis and cysteine in the binding of acetyl CoA. Above proposal was confirmed further by pH dependence of the kinetic parameters. The data from pH study suggest that N₁-0H of histidine hydrogenbonded to aspartic acid accepts proton from hydroxyl group of C₃ of chloramphenicol concomitant with nucleophilic attack of the acetyl group carbon on acetyl-CoA. The proton in N₃ of histidine is then transfered to ^-S-CoA. Taken together, the above results strongly suggest that chloramphenicol acetyltransferase catalyzes its reaction via general acid-base catalysis using a single base as a porton shuttle.
