The aim of this study was to investigate protein profiles related to the induction of adipogenesis within the bovine longissimus dorsi muscle (BLDM) by proteomic analysis. We analyzed BLDM proteins at different growth stages to clarify the physiological mechanisms of marbled muscle development in 20 head of Korean native cattle (11 month: 10 head, 17 month: 10 head). BLDM proteins were analyzed by two-dimensional electrophoresis and image analysis. Villin 2 was specifically identified by mass spectrometry and a protein search engine. Villin 2 protein expression in BLDM decreased during the fat development stage in test steers. In a Western blot cell culture study of spontaneously immortal bovine muscle fibroblasts, the abundance of Villin 2 was shown to be down-regulated during differentiation into muscle. In 3T3-L1 mouse embryonic fibroblasts, Villin 2 was decreased during differentiation into adipocytes. The results suggest that Villin 2 may be related to the induction of transdifferentiation and adipogenesis in bovine longissimus dorsi muscle. [BMB reports 2012; 45(2): 102-107].

Background: ENCODE project revealed that nearby or distantly located non-coding DNA regulates the expression of coding genes. RegulomeDB is a new database that can be used to predict whether variants affect transcription factor binding and gene expression. We investigated the association between lung cancer risk and potentially functional polymorphisms of XRCC1 that were selected using RegulomeDB in a Korean population. Methods: 185 polymorphisms of XRCC1 were evaluated using RegulomeDB. There was strong evidence that 10 polymorphisms affected XRCC1 expression with scores of 1a-1f that were based on the RegulomeDB. rs2854510 polymorphism was rare in Asians. Eight polymorphisms were in strong linkage disequilibrium (LD). rs2854509, which was one of the 8 polymorphisms in LD, and rs7248167, which was not in the LD block, were genotyped using the Taq-Man® assay in 610 lung cancer patients and 607 age- and sex-matched controls. Additionally, 4 polymorphisms of XRCC1 (Arg399Gln, Arg280His, Arg194Trp, and -77T>C), which were investigated with regard to the association with lung cancer risk in previous studies, were also genotyped. Results: Two polymorphisms (rs2854509 and rs7248167) that were predicted to affect XRCC1 expression based on their RegulomeDB scores were not associated with lung cancer risk (P = 0.31 and 0.93, respectively). When stratified according to age, gender, smoking status, and tumor histology, the 2 polymorphisms of XRCC1 were not associated with lung cancer risk. Among the 4 polymorphisms that were previously studied, only XRCC1 Arg280His was significantly associated with lung cancer risk (dominant model, adjusted odds ratio = 0.61, 95 % confidence interval = 0.46-0.83, P = 0.002). Conclusions: Although RegulomeDB is an attractive tool for predicting regulatory potential of variants, the 2 polymorphisms that were selected using RegulomeDB were not associated with lung cancer risk.
Thermodynamics and Kinetics of Lysozyme Adsorption onto Two Kinds of Weak Cation Exchangers

The present work investigated the adsorption behaviors of lysozyme onto weak cation exchangers at different temperatures. The adsorption isotherm, adsorption thermodynamics and adsorption kinetics were studied. The results indicate that the adsorption of lysozyme onto acrylic acid copolymer based beads (Hydrolite D115) is spontaneous and exothermic, while that onto agarose based beads (CM Sepharose 6 Fast Flow) is also spontaneous, but endothermic. The pseudo second-order kinetic model fits well to the dynamic adsorption experimental data, and the kinetic results are also in concert with the adsorption thermodynamics.

The microemulsion formulation (hereafter formulation) of curcuma oil and its acaricidal efficacy against Tetranychus cinnabarinus Boisduval (Acari: Tetranychidae) were optimized in the laboratory to evaluate their spray effectiveness of oviposition inhibition and repellence. Ethovision XT6 was used to analyse the effects of the sublethal concentrations (LC 20 ) of curcuma oil and the formulation on the behaviors of T. cinnabarinus. The results showed that Tween-80 was the best surfactant, Isopropanol was the best co-surfactant and K m = 2:1 was the best condition for the formulation. The prepared microemulsions are stable under conditions of centrifugation and incubation for extended periods. The results showed that the effect of the spray bioassays of the formulation against T. cinnabarinus continuously increased during the experiment, but for curcuma oil almost no longer increase observed when the exposure time went beyond 24 h. Moreover, compared with curcuma oil (LC 50 = 0.716%), the spray bioassay of the formulation (LC 50 = 0.035%) was stronger against T. cinnabarinus. The repellency of the formulation to T. cinnabarinus was stronger with increasing exposure time, but that of curcuma oil declined after 12 h of exposure. The mobile distance of T. cinnabarinus treated with the formulation continuously declined during the experiment but that due to the curcuma oil almost no longer declined when the treatment time reached 12 h. The maximum mobile frequency of T. cinnabarinus treated by curcuma oil and the formulation was recorded at 6 h and 12 h, respectively. Thus, the formulation is a promising candidate as a botanical acaricide of green vegetables.
Backward Orientation Tracking Control of Mobile Robot with N Trailers

A feedback control law for orientation tracking problem of a mobile robot with N on-axle trailers isproposed in this paper. Based on a recursive designing process, the orientation tracking problem is converted intostabilizing the including angles on the designed reference value. The designing process of the control law is simpleand can be easily extended to any given N trailers system. With input-to-state stability on cascade form system, theclosed-loop system is proven to be asymptotically stable. Simulation experiment on a mobile robot with 4 trailersis implemented and the results demonstrate that the proposed control law works successfully.

We investigated the protective effects of chlorogenic acid (CGA), a polyphenol compound, on oxidative damage induced by UVBexposure on human HaCaT cells. In a cell-free system, CGA scavenged 1,1-diphenyl-2-picrylhydrazyl radicals, superoxide anions,hydroxyl radicals, and intracellular reactive oxygen species (ROS) generated by hydrogen peroxide and ultraviolet B (UVB). Furthermore, CGA absorbed electromagnetic radiation in the UVB range (280-320 nm). UVB exposure resulted in damage to cellularDNA, as demonstrated in a comet assay; pre-treatment of cells with CGA prior to UVB irradiation prevented DNA damage andincreased cell viability. Furthermore, CGA pre-treatment prevented or ameliorated apoptosis-related changes in UVB-exposedcells, including the formation of apoptotic bodies, disruption of mitochondrial membrane potential, and alterations in the levelsof the apoptosis-related proteins Bcl-2, Bax, and caspase-3. Our findings suggest that CGA protects cells from oxidative stressinduced by UVB radiation.

In this study, 29 simple sequence repeat (SSR) markers were used to analyze the genetic diversity and population structure of 125 rice accessions from 40 different origins in Africa, Asia, Europe, South America, and Oceania. A total of 333 alleles were detected, with an average of 11.5 per locus. The mean values of major allele frequency, expected heterozygosity, and polymorphism information content (PIC) for each SSR locus were 0.39, 0.73, and 0.70, respectively. The highest mean PIC was 0.71 for Asia, followed by 0.66 for Africa, 0.59 for South America, 0.53 for Europe, and 0.47 for Oceania. Model-based structure analysis revealed the presence of five subpopulations, which was basically consistent with clustering based on genetic distance. Some accessions were clearly assigned to a single population in which >70% of their inferred ancestry was derived from one of the model-based populations. In addition, 12 accessions (9.6%) were categorized as having admixed ancestry. The results could be used to understanding the genetic structure of rice cultivars from these regions and to support effective breeding programs to broaden the genetic basis of rice varieties.

Due to ongoing climate change, short-term extreme heat waves in the summer are expected to be more frequent. Insect eggs are sensitive to thermal stress. This raises the question of whether herbivore insects' thermal adaptability would be changed after a single extreme heat wave at the egg stage. In this study, we examined the developmental performance of Ostrinia furnacalis Guenée at 25 °C, 27 °C, 29 °C or 31 °C after a single extreme heat wave (42 °C) for 0 h (control), 1 h, 2 h, or 3 h at the egg stage. The results showed that O. furnacalis at the egg or larval stage was more sensitive to a single heat wave than it was at the pupae or adult stage. After a single heat wave, O. furnacalis showed a reduced egg-hatching rate or reduced larval survival rate, but the optimum temperature for egg hatching and larval survival was higher than that in the control. The upper temperature threshold and optimum temperature for larval development in the control were higher than that after a single extreme heat wave. Both male and female pupal weight decreased with increasing temperature, and pupal weight decreased faster in females than in males. The Cox proportional hazard model showed that when O. furnacalis developed at 25 °C, the instantaneous death risk of adults with a 3 h heat wave at the egg stage was higher than that of the control, but when O. furnacalis developed at 29 °C and 31 °C, the instantaneous death risk of adults after a heat wave was significantly lower than that of the control. Our study highlights the effect of a single heat wave on O. furnacalis eggs and the subsequent development of survival individuals.
