        Selective addition of CXCR3+CCR4-CD4+ Th1 cells enhances generation of cytotoxic T cells by dendritic cells in vitro

        Increasing importance is being given to the stimulation of Th1 response in cancer immunotherapy because its presence can shift the direction of adaptive immune responses toward protective immunity. Based on chemokine receptor expression, CXCR3+CCR4-CD4+ T cells as Th1-type cells were investigated its capacity in monocyte-derived dendritic cell (DC) maturation and polarization, and induction of antigen specific cytotoxic T lymphocytes (CTL) in vitro. The levels of IL-4, IL-5 and IL-10 were decreased to the basal level compared with high production of IFN-γ, TNF-α, and IL-2 in CXCR3+CCR4-CD4+ T cells stimulated with anti-CD3 and anti-CD28 antibodies. Co-incubation of activated CD4+ or CXCR3+CCR4-CD4+ T cells with DC (CD4+/DC or CXCR3+CD4+/DC, respectively) particularly up-regulated IL-12 and CD80 expression compared with DC matured with TNF-α and LPS (mDC). Although there was no significant difference between the effects of the CXCR3+CCR4-CD4+ and CD4+ T cells on DC phenotype expression, CXCR3+CD4+/DC in CTL culture were able to expand number of CD8+ T cells and increased frequencies of IFN-γ secreting cells and overall cytolytic activity against tumor antigen WT-1. These results demonstrated that the selective addition of CXCR3+ CCR4- CD4+ T cells to CTL cultures could enhance the induction of CTLs by DC in vitro, and implicated on a novel strategy for adoptive T cell therapy.       • 관절염을 가진 대상자의 가족지지, 자가 간호 및 삶의 질과의 관계

        Purpose : The purpose of this study was to investigate relationships among family support, self-care, and quality of life of patients with arthritis through a survey. That will be basic data to plan nursing intervention for promoting quality of life for patients with arthritis. Method : The convenient sample of this study consisted of 120 patients with arthritis and data were collected using questionnaires during August 2006 from two OPDs of rheumatoid centers in C University hospitals. The study instruments were used to measure Self Care (So, 1992), Family Support (Kang, 1984), and Quality of Life (Ro, 1988). The data were analyzed using ANOVA, Duncan test, and Pearson correlation. Result : The mean scores were 3.56 for family support, 4.16 for self-care, and 3.19 for quality of life. The level of self-care was significantly higher in those men, having an occupation, higher income, and living with family members. The level of family support was significantly higher in those men, having an occupation, living with family members, and having a helper. The level of quality of life was significantly higher for those men, having a higher income, and shorter length of illness. Three relationships were found to be positively correlated: 1) between the levels of family support and self-care (r=.53), 2) between the levels of self-care and quality of life (r=.55), and 3) between the levels of family support and quality of life (r=.54). Conclusion: Therefore, a nursing intervention program needs to be developed to promote family support, and self-care for quality of life for patients with arthritis.

      • 생쥐의 卵子와 初期胚子에 對한 Clomiphene Citrate의 細胞毒性에 關한 硏究

        In order to elucidate the cytotoxicity of clomiphene citrate, the rate of in vitro fertilization(IVF) of ova and the developmental rate of early embroys in A-strain mouse were examined. The rate of ova cleavaging to 2-cell stage by IVF was remarkably decreased(12.6%) at the concentration of 10μg/㎖ of clomiphene than that of the control(65.2%) when the ova fertilized in vitro were cultured for 24 hours in the untreated medium after treatment ova with clomiphene for 5 hrs. But there was no ovum cleavaging to 2-cell stage at 50μg/㎖ clomiphene except only a few 2-polar bodied ova. The developmental rate of mouse early embryos was decreased dose-dependently. Especially at 25μg/㎖ of clomiphene, the number of embryos cleavaging to blastocyst from 2-cell stage(41.4%) was decreased remarkably compared with that of the control(87.2%). And only a few embryos were developed to 8-cell stage at clomiphene concentration of 50μg/㎖ after 2-cell embryos were incubated for 72 hours in clomiphene-treated medium. Morphological changes such as fragmentation and fusion of blastomeres, cytolysis and developmental retardation were increased with dose-dependently. These results suggest that clomiphene has cytotoxic effect by the decrease of the rate of IVF, the developmental rate and the degenerative changes of murine ova and early embryos.

        The Prevalence and Characteristics of Brown Adipose Tissue in an $^{18}F$-FDG PET Study of Koreans

        Purpose The object of this study was to evaluate the prevalence and characteristics of brown adipose tissue (BAT) in Korean subjects using $^{18}F$-fluorodeoxyglucose positron emission tomography ($^{18}F$-FDG PET). Methods Six thousand and five consecutive $^{18}F$-FDG PET/CT scans of 5,115 patients (3,007 females and 2,108 males, mean age 53.5 years) were retrospectively reviewed. We characterized the nature of BAT, such as its location, and we assessed the influence of sex, age, body mass index (BMI), and temperature on BAT. Results The prevalence of BAT in Koreans in a single $^{18}F$-FDG PET/CT scan in average conditions was 1.07%. The BAT detection rate was higher in females than males (1.32% vs 0.73%), and also with younger age (7.94% vs 0.73%), lower BMI (BMI with BAT, 21.1 vs BMI without BAT, 23.15) and cold outdoor temperature (1.65% vs 0.49%). The most frequent location of BAT was the supraclavicular area (left, 0.91%; right, 0.88%) and ventral neck area (left, 0.62%; right, 0.63%). Conclusions The characteristics of BAT in Koreans are not different from those described for Caucasians. However, the low prevalence of BAT in our study might be related to some scan condition like ambient temperature, but further study is needed.

        Itraconazole 에 의한 급성간염 1 예

        A 51-year-old women, suffering from an onycomycosis of the toe nails, was treated with itraconazole, 100 mg daily. Fourteen weeks later, she complained of jaundice, nausea, and epigastric discomfort. Itraconazole treatment was stopped immediately. She did not use other drugs. There was no history of alcohol abuse, hepatitis or transfusion. The laboratory investigations revealed marked elevation of serum bilirubin, aspartate aminotransferase and alanine aminotransferase, Histologic findings of liver disclosed acute hepatic necrosis. There was no clinical or serologic evidence of viral hepatitis. One month later, the laboratory data become almost normal. It is suggested that itraconazole therapy was a causal factor in this case of drug induced hepatitis.

        In this study, Activated Sludge was operated for treatment of cosmetic wastewater and combined with sewage The results from this study are follows. During the experiments, average COD and BOD concentrations of raw wastewaters were ranged 690∼1,010 ㎎/l, 550 ∼ 880㎎/l respectively, and fluctuated sharply with time. COD removal efficiency was 73.6% at HRT 12hr. It is higher removal efficiency than HRT 4hr 62.6%, HRT 8hr 69.4% when cosmetic wastewater combine with sewage, COD and BOD removal efficiencies were higher than wastewater only, therefore treatment of combined sewage was effective.

        한국산 주요패류에 대한 독의 분포 , 특성 및 제독에 관한 연구 : 2 . 진주담치에서 추출한 PSP 의 분리 , 정제 및 특성에 관하여 2 . Purification and Characterization of PSP Extracted from Cultured Sea Mussel , Mytilus edulis

        韓國 南海岸에서 生産되는 主要 貝類中에서 PSP含量이 比較的 높은 진주담치를 試料로 擇하여 抽出한 毒素에 대한 pH變化와 加熱處理하였을 때의 毒素의 安定性을 檢討하고, 抽出된 毒素를 活性炭素處理, Bio-Gel P-2 및 Bio-Rex 70의 각 column chromatography로 分離精製하여 electrophoresis, TLC 및 HPLC로 分析한 結果는 다음과 같다. 1. 抽出된 PSP의 pH는 3.0부근이었으며 이 PSP를 121℃ 15分과 100℃로 2시간 處理하였을 때 約 20%가 파괴되었다. 2. PSP는 121℃에서 15分 處理하였을 때 pH 2.0∼4.0 범위에서는 安定하였으나 pH 4.5이상에서는 不安定하였으며 pH 6.0에서는 80%가 不活性化되었다. 3. 忠武一圓에서 生産되는 養殖 진주담치에서 分離된 PSP는 主로 GTX 1, 2, 3, 4이었으며, STX群 毒素는 GTX群의 約 1/40에 不過하였다. The Stability of PSP extracted from the intoxicated sea mussel, Mytilus edulis was evaluated by the change of heating conditions and pH of the PSP solution. Also the composition of the PSP extracted from the cultured sea mussel collected at Chungmu, Korea on March 12, 1986 was analyzed. The extracted PSP was stable over the range of pH 2.0 to 4.0, but it was unstable above pH 4.5. For example, the toxicity of extracted PSP of pH 3.0 was only decreased less than 20% by the treatment at 121℃ for 15min or at 100℃ for 2 hours, but it was decreased more than 80% by the same treatment when the pH of the PSP solution was adjusted to 6.0. The toxin was purified from the ethanolic extract of the digestive glands of the sampled sea mussel by Bio-gel P-2 and Bio-Rex 70 column chromatography. The toxic fractions obtained were analyzed by cellulose acetate membrane electrophoresis, TLC and HPLC. The compositional analytical results of the PSP, most of the toxins were certified as GTX_(1-4), while the toxicity of STX was only about 1/40 of that of GTX_s.

      • 2단 폭기방식에 의한 도시생활하수 처리에 관한 연구

        In this study, sewage were treated with Two-step Aeration System, it was analyzed treatment efficiency by hydraulic retention time(HRT) in a 2nd. aeration basin. As to the increase of HRT 4∼10hr in a 2nd. aeration basin, treatment efficiency was increased but in consideration of economical efficiency, HRT 8hr was most stable. At Two-step Aeration System of HRT 8hr, BOD treatment efficiency were 90.1%, 84.6%, respectively. At Two-step Aeration System of HRT 8hr, range of F/M ratio was 0.11∼0.17kg·BOD/kg·MLSS.d. At Two-step Aeration System of HRT 8hr treatment efficiencies and effluent concentrations of NH₄^+-N and PO₄-P were 57.3%, 10.5mh/l and 33.9%, 1.74mg/l, respectively.
