        Culture Tube Method for the Determination of Total Cholesterol in Egg Yolk Lipid

        A simple, reproducible , and accurate enzymatic method using a cholesterol assay kit was developed to quantify total cholesterol content in egg yolk. Total egg yolk lipid was extracted with hexane : isopropanol(3 : 2, v/v) mixture. Samples containing various amount of the total lipid(0-3mg) in optically identifical culture tubes were reacted for 10 min in a water bath (37$^{\circ}C$) with the enzyme solution (5ml) from the cholesterol assay kit. Cholesterol content of the reaction mixturesin culture tubes was spectrophotometrically determined by two different ways : (1) using the culture tube as a curvette(designate culture tube method ; CTM) and (2) the quartz cvette containing the reaction mixture transferred from the culture tube (designate standard cvette method, SCM). CTM revealed lower cholesterol content in 0.1-1.0mg lipid sample range that SCM did, but not significant. For more than 2.0mg lipid sample, CTM gave significantly (p<0.01) lower cholesterol content relative to that by SCM, suggesting that SCM give a false positive result from the sample containing more than 2 mg lipid due to the interference of absorbance by lipid dispersed in the reaction solution . Cholesterol content of less than 1.0mg lipid sample by CTM was proportional to the amount of lipid used, but its linear relationship was not seen in more than 2mg lipid sample. Thus, to determine the appropriate lipid amounts (mg) analyzed . A constant level (41$\mu\textrm{g}$/mg) of cholesterol concentration was observed from the sample containing 0.1-1mg lipid. after which the cholesterol level was dropped to less than 41$\mu\textrm{g}$ /mg. Cholesterol concentration in egg yolk samples quantified by CTM was in accordance with that by GC method. These results suggest that CTM is an useful method for the quantification of cholesterol in egg yolk lipid and other lipids as well.

        Anti-proliferative efficacy comparison of conjugated linoleic acid on human cancer cell lines

        The anti-proliferative efficacy of t,t-conjugated linoleic acid (t,t-CLA), c9,t11-CLA, and t10,c12-CLA was compared in several human cancer cell lines. Gastric NCI-N87, liver Hep3B, pancreas Capan-2, and lung NCI-H522 cancer cells were incubated with 50 μM CLA isomers over a period of 6 days. The t,t-CLA inhibited the growth of all cancer cell lines to different extents, but c9,t11-CLA and t10,c12-CLA inhibited or stimulated the growth of the cancer cell lines. NCI-N87 cells were the most sensitive to growth inhibition and apoptosis from all CLA isomers tested. In NCI-N87 cells, CLA isomers reduced the release of arachidonic acid (AA) via the inhibition of cytosolic phospholipase A2 (cPLA2 ) activity, consequently reducing the production of PGE2 through the inhibition of cyclooxygenase-2 (COX-2). The efficacies of CLA isomers were in the following order (from most to least effective): t,t-CLA, t10,c12-CLA and c9,t11-CLA. Overall, these results imply that the anti-proliferative efficacy of t,t-CLA on cancer cells, especially NCI-N87 cells, was greater than other CLA isomers due to its induction of apoptosis through the inhibition of cPLA2 and COX-2 activities.

        RWPE-1 전립선세포에서 eritadenine을 함유한 신령버섯균사체 액체배양물의 항염증효과 및 항산화효과에 의한 전립선비대증 관련 biochemical marker 개선 효과

        신령버섯균사체 액체배양 추출물(Agaricus blazei mycelial liquid culture extract, ABMLCE)은 TM-3 mouse Leydig 고환세포에서 testosterone (TS)의 생성을 촉진하였다. 이번 연구에서는 eritadenine (EA, 15.3 ㎎/100 g)을 함유한 ABMLCE가 RWPE-1 전립선세포에서 전립선비대증(Benign prostatic hyperplasia, BPH) 관련 핵심 효소인 5α-reductase 2 (5α-R2) 활성과 dihydrotestosterone (DHT) 함량을 감소시켰음을 보고한다. RWPE-1 전립선 세포는 24-well plate에서 ABMLCE (0~50 ppm), EA (0~10 ppm,), finasteride (FS 10 μM: positive Control)를 함유한 Keratinocyte serum-free medium (K-SFM)에서 24시간 배양하였다. 시료처리 세포배양액에 대해 5α-R2, superoxide dismutase (SOD), catalase (CAT), cyclooxygenase-2 (COX-2) 효소활성, TS, DHT, tumor necrosis factor-α(TNF-α)와 interleukin-1β (IL-1β)를 assay kit를 사용하여 분석하였다. 5α-R2 활성과 DHT 함량은 ABMLCE의 농도 의존적으로 유의성(p<0.05) 있게 감소되었고, SOD와 CAT 효소활성은 유의성(p<0.05) 있게 증가되었다. 반면에 COX-2 활성과 TNF-α 및 IL-1β의 함량은 감소되었다(p<0.05). 마찬가지로 EA의 효과도 ABMLCE 효과와 유사하였다. ABMLCE 50 ppm과 EA 10 ppm의 5α-R2 및 DHT 감소효과는 FS 10 μM 효과와 유사하였다. 이들 결과는 ABMLCE와 EA는 항염증효과와 항산화효과를 통하여 BPH 유발 핵심 인자인 5α-R2 효소활성과 DHT 함량을 감소시켰다. 또한 이 결과는 EA를 함유하는 ABMLCE는 인체 BPH 개선 소재로 활용할 수 있음을 의미한다. Now, we report that ABMLCE containing eritadenine (EA) as a minor constituent (15.3 mg/100 g) reduced 5α-reductase 2 (5α-R2) enzyme activity and dihydrotestosterone (DHT) content which are key constituents for the benign prostatic hyperplasia (BPH) inductions. RWPE-1 prostate cells were grown in a Keratinocyte serum-free medium (K-SFM) containing ABMLCE (0~50 ppm), EA (0~10 ppm,), and finasteride (FS 10 μM: a positive control) in a 24-well plate for 24 hr. Supernatants collected from cell-cultured media were used for the assay of 5α-R2, superoxide dismutase (SOD), catalase (CAT) and cyclooxygenase-2 (COX-2) enzyme activities, and for TS, DHT, tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) contents by their assay kits. The 5α-R2 activity and DHT content were proportionally reduced (p<0.05) to concentrations of ABMLCE. The SOD and CAT enzyme activities were significantly (p<0.05) elevated concomitant with ABMLCE concentrations, while COX-2, TNF-α and IL-1β showed reverse results (p<0.05). Similarly, the effects of EA were similar to those of ABMLCE. Efficacies of ABMLCE 50 ppm and EA 10 ppm in 5α-R2 and DHT reduction were similar to those of 10 μM FS. These results suggest that ABMLCE and EA reduced 5α-R2 and DHT through their anti-inflammatory and anti-oxidative actions. This implies that ABMLCE containing EA could be a beneficial material in the cure of BPH in humans.

        Culture Tube Method for the Determination of Total Cholesterol in Egg Yolk Lipid

        달걀 중의 cholesterol 정량에는 GC, HPLC 및 효소 방법이 이용되고 있지만 GC나 HPLC 방법은 시간과 장비의 문제점이 있어 간편한 효소법이 가장 많이 사용된다. 그러나 효소법을 난황지방에 직접 사용할 경우 효소에 의해 분해되지 못한 지방이 비색 측정을 방해하여 실제 보다 과량 검출되거나 반복간의 차이를 유발하여 실제의 함량을 측정하는데 큰 오류를 범할 수 있다. 따라서 본 연구에서는 효소법의 이와 같은 단점을 보안하고 실제로 실험실에서 쉽게 사용할 수 있도록 culture tube method(CTM)를 개발하였다. 효소의 제재로서는 시중에서 구입할 수 있는 cholesterol assay kit를 사용하였다. 난황에서 분리한 지방(0.1, 0.2, 0.5, 1.0, 2.0, 3.0㎎)을 culture test tube (540㎚에서 흡광도의 차이가 0.001 이하)에 각각 넣고 여기에 효소액 5ml를 가하여 37℃ 항온조에서 10분간 반응시킨 후 2가지 방법으로 540㎚에서 흡광도를 측정하였다. 첫째는 culture tube를 cuvette으로 사용하여 반응물이 흔들리지 않게 spectrophotometer에서 흡광도를 측정하였고(Culture tube method, CTM) 다음은 이것을 다시 정상 cuvette에 옮겨 흡광도를 측정하였다 (standard cuvette method, SCM : 이 경우 반응물이 혼합됨). 이 결과 난황지방 1.0㎎까지는 CTM에 의해 측정한 cholesterol 함량은 사용된 지방 함량에 비례하였지만 그 이상의 지방 함량에서는 비례하지 않았다. 그리고 CTM으로 측정된 cholestrol 함량은 SCM에 의한 결과 보다 낮은 수치를 보여 1.0㎎에서는 11%, 3.0㎎에서는 42% 차이(p<0.01)를 보였다. CTM에 이용 가능한 난황지방 함량을 구하기 위해 CTM에 의해 정량된 cholesterol 함량을 사용된 지방 함량으로 나누어 본 결과, 사용된 지방의 함량 1.0㎎ 까지는 ㎎당 cholesterol 함량이 약 41㎍ / ㎎으로 일정하였으나 2㎎ 이상인 경우는 그 이하로 떨어졌다. 이 결과로 미루어 상기 조건하에서는 사용 가능한 지방량의 한계는 l㎎으로 나타났다. CTM으로 측정한 실제 달걀 중의 cholesterol 함량은 GC법으로 측정한 cholesterol 함량과 일치하였다. 이상의 결과로서 난황에서 추출한 지방에 함유된 cholesterol의 함량을 cholesterol assay kit를 사용하여 CTM법으로 측정하기 위하여서는 지방 함량 0.5~1.0㎎에 효소액 5ml를 사용하는 것이 가장 바람직하다. CTM를 이용하면 숙련된 사람일 경우 적어도 하루에 50~100개의 시료를 분석할 수 있다.         In this paper, the lattice Boltzmann method is discussed for simulating droplet formation in a microchannel. Droplets are formed by the hydrodynamic instability on the interface between two immiscible fluids when two immiscible fluids are imported simultaneously in a microchannel. Shan & Chen model (1993), which is a lattice-Boltzmann model of two-phase flows, is used to treat the interaction between immiscible fluids.

        In this paper, the immersed boundary-lattice Boltzmann method is discussed for the two-dimensional viscous incompressible flow computations. This approach uses a new computational method, the lattice Boltzmann method for the computation of the flow field and the immersed boundary method to treat the body geometry. The method of feedback momentum forcing (Goldstein et al., 1993) is applied to satisfy the no-slip boundary condition on the immersed body. The driven cavity flows with the stationary and oscillating immersed bodies are employed as the test cases of this approach.
