        백서 Paraquat 두여에 의한 간과 폐 조직의 산화성 손상에서 Vitamin C와 Deferoxamine의 항산화 효과에 관한 연구

        Background: The toxicity of paraquat has been known to be caused by oxygen free radicals which leads to the lipid peroxidation and multiple organ failure. Although vitamin C has been known to be a potent antioxidant, recently there are numerous data which have shown that a low dose of vitamin C may act as a prooxidant due to the stimulation of the Fenton reaction with metal ions, which produces hydroxyl radicals. It has been reported that a deferoxamine in paraquat intoxication could reduce the production of the hydroxyl radicals by the inhibition of the Fenton reaction through the reduction of iron ion in tissue. The aim of this study was to evaluate the effect of the high and low dose of vitamin C and deferoxamine on lipid peroxidation and plasma TNF-α in paraquat intoxication. Methods: Female Sprague -Dawley rats were divided into seven groups: control group which was not given paraquat(20 mg/kg), P group which was given paraquat only, PVH group given paraquat and high dose of vitamin C(100 mg/kg), PVL group given paraquat and low dose of vitamin C(10 mg/kg), PVHD given paraquat, high dose of vitamine C and deferoxamine(100 mg/kg), PVLD given paraquat, low dose of vitamin C and deferoxamine, and PD given paraquat and deferoxamine. Animals were killed at 6 and 24 hours after treatment. Malondialdehyde(MDA), superoxide dismutase(SOD) and glutathione(GSH) contents, catalase activity, plasma TNF-α, and histologic changes in the lung and liver tissue were measured. Results: The lung histology in the PVH and PD or PVHD groups showed the significant decreases in the alveolar edema and interstitial thickness compared to the P group. The liver histololgy in the PVH and PVHD groups demonstrated marked differences in the central venous and sinusoidal dilatation compared to that of the P group. While the MDA levels of the lung and liver in the PVH and PD groups showed the significant reduction compared to that of the P group at 6 hours after treatment, all groups showed the significant changes compared to the P group at 24 hours. There was no significant change of the SOD levels of the lung and liver at 6 hours among all groups. At 24 hours, the SOD levels of the lung in PVH, PVL, and PVHD groups showed the significant increases compared to the P group. The increase of the SOD level in groups combined with deferoxamine, however, revealed a little reduction. The SOD level of the liver in PVH group only significantly increased compared to the P group at 24 hours. There was no significant change of the GSH level of the lung and liver among all groups at 6 hours. At 24 hours, the GSH level of the lung and liver were significantly increased in both PVH and PD group and PVH group, respectively, compared to the P group. Although the catalase activity of the lung was not significantly increased, that of liver was signiflcantly increased in both PVHD and PD groups compared to the P group at 6 hours. The catalase activities of the lung and liver were significantly increased in PVH, PD, and PVHD at 24 hours. The concentrations of the plasma TNF-α were slightly decreased at 6 hours and slightly increased at 24 hours compared to that of the P group, but they were not significant. Conclusion: This study showed that although the low dose of vitamin C had no effect, the high dose of vitamin C revealed a decrease of the MDA level and an increase of SOD, GSH, and catalase activity in the lung and liver tissues, and the effect of the high dose of vitamin C increased with time. The administration of the deferoxamine with or without high dose of vitamin C, however, significantly showed the inhibition of the lipid peroxidation and antioxidant effect and low dose vitamin C decreased the effect of deferoxamine. The effects of the vitamin C and deferoxamine on plasma TNP-α were not clearly shown.

      • 韓國人에서 血友病 B 遺傳子(血液凝固因子 Ⅸ)의 構造 및 制限酵素 Dde Ⅰ, MnI Ⅰ 切片길이 多形性에 關한 分子 遺傳學的 考察

        Hemophilia B, a human chromosome X-linked recessive disease, is a bleeding disorder resulting from defect or abnormality in blood coagulation factor IX. DNA-based prenatal diagnosis or carrier detection for hemophilia B in Korean has been developed by analyzing restriction fragment length polymorphisms(RFLPs). Two polymorphisms( Dde I and Mnl I) were investigated as follow. Genomic DNAs were extracted from blood of 50 females at the age of twenty. The primers were chemically synthesized by the method of phosphoamidite. Mnl I primers were derived from exon 6, while Dde I primers, from the flanking sequences of intron 1 of the factor IX gene. Genomic DNAs were amplified with Mnl I primers to generate 405 nts long fragments in all cases. They were digested with Mnl I to analyze the polymorphic site on agarose gel. No MnlI polymorphic site was found in all cases. Also, the genomic DNAs were amplified with Dde I primers to generate ca. 320 nts long fragments in all cases. These results are not correlated with those obtained from the Caucasian. It suggests that Korean could exhibit different patterns of Dde I and Mnl I polymorphisms in the gene for blood coagulation factor IX. Direct sequencing of the polymorpic sites will confirm the above sugestion.

        불완전 전뇌 허혈후 소생의 백서 모델을 이용한 재관류 뇌혈류량과 허혈성 뇌조직 손상도와의 관계

        Background: Experimental data indicate that low-flow reperfusion following prolonged cardiocirculatory arrest may aggravate early cerebral microcirculatory reperfusion disorders. We investigated the influence of cerebral reperfusion flow change to the ischemic histopathologic damage of brain tissue after incomplete forebrain ischemia in rats. Materials and method: Anesthetized Sprague-Dawley rats were undergone ligation of both internal carotid artery by microvascular clamp for 10 minutes. After release of the clamp, reperfusion was started with several different flow levels (0, 10, 20, 30, 50, and 100%) of internal carotid artery comparing to pre-clamping phase using flowmeter. After 15minutes of reperfusion, rat brains were prepared by perfusion-fixation with 3% formaldehyde. Under light microscopic examination of Hematoxylin-Eosin stained tissue slide, histopathologic damage was examined at cortex, putamen, and hippocampus regions. Categorical hisotopathologic damage scores were derived in each regions by manual counts of ischemic neurons. Result: The histopathologic damage scores were 0, 10.2±0.5, 7.6±1.5, 5.9±1.4, 5.0±2.8, 3.5±0.7, and 1.0±0.0 in control, 0, 10,20, 30, 50, and 100% reperfusion groups, respectively(p<0.05). Conclusion: Our results showed significant increment of brain histopathologic damage scores along with decreasing amount of cerebral reperfusion flow after incomplete forebrain ischemia. We believe restoration of reperfusion flow to pre-ischemic level would be a critical component in attenuation of brain ischemic damage.

        급성 충수돌기염 의증 환자의 진단에서 컴퓨터 단층촬영과 초음파검사의 비교 분석

        Background: This study was conducted to compare the accuracy of computered tomography(CT) and sonography(US) in diagnosing acute appendicitis and to analyze the radiologic factors which influence the result. Methods: We retrospectively studied the cases of 53 patients who were suspected of having appendicitis and who were examined by CT and US covered a three-year period. The final diagnosis was determined from the surgico-pathologic diagnosis and clinical follow-up for two months, as well as from the discharge diagnosis. Results: Thirty-eight(38) patients had acute appendicitis; 15 patients did not. In the analysis, CT and US, respectively, revealed sensitivities of 89.5% and 73.7%, specificities of 93.3% and 60.0%, positive predictive values of 97.1% and 82.4%, negative predictive values of 77.8% and 47.4%. Among patients diagnosed with acute appendicitis, 10 patients were discordant in the US and the CT examinations; CT findings were correct in 8 and US findings in 2. Among non-appendicitis patients, 5 patients were discordant in the US and the CT examinations; CT was correct in 5 and US in 0. Nighteen(19) patients had complications such as an abscess or a perforation. Among these patients, the degree and the extent of the complication were accurately evaluated by CT in 15 patients and by US in 5 patients. Conclusion: CT is more accurate than US in diagnosing and excluding acute appendicitis. The use of CT will substantially decrease unnecessary laparotomies and post-operation complications.

      • 배추(Brassica campestris L. var. pekinensis Makino)의 cDNA library 구축 및 상동성 비교

        Expressed sequence tag(EST) has a good value to discover a new gene or to study its structure. Some 20 ESTs were generated to obtain new genetic resources of chinese cabbage(Brassian campestris L. var. pekinensis Makino). Poly A+ RNAs were isolated from 10-day-old seedlings grown at 25℃ under the day light. cDNA gene bank was constructed using λ ZAP /cDNA synthesis /Giga Pack Gold Packing kit. About a million clones were able to obtain from the library. All the clones examined so far had insert DNAs. Nucleotide compositions of randomly selected clones were determined by the Sanger mthod. The DNA sequences were compared with those deposited in the GenePept and GenBank database to figure out nucleotide homologies. Two ESTs showed significant similarities to the enlisted sequences. They are chloroplast GADPH subunit of Arabidopsis thaliana and carbonic anhydrase of A. thaliana. The full DNA sequences of the two clones are being determined. The cDNA gene bank constructed. in this experiment will being used to isolate more genes induced by the light in the plant.

        예비유아교사의 음악적 자아개념 및 음악적 개념에 대한 실제이해와 음악교수 효능감과의 관계

        본 연구는 예비유아교사의 음악적 자아개념 및 음악적 개념에 대한 실제이해와 음악교수 효능감의 수준을 알아보고, 이들 간의 관련성과 영향력을 살펴보는 데에 목적이 있다. 연구 대상은 경상남도에 위치한 3, 4년제 대학의 유아교육과 학생 201명이었으며, 연구 도구는 Svengalis(1978)가 개발하고 Philips(2002)가 수정한 ‘음악적 자아개념 척도’, Kim, Lee, Kwon과 Park(2011)이 개발한 '유아교사의 음악적 개념 실제이해 측정 도구’, Riggs와 Enochs(1990)가 개발하고 방은영, 박찬옥(2005)이 수정한 ‘음악교수 효능감 검사도구’가 사용되었다. 연구결과를 살펴보면 첫째, 예비유아교사의 음악적 자아개 념과 음악적 개념에 대한 실제이해는 보통 수준인 것으로 나타났으며, 음악교수 효능감은 비교적 높은 것으로 나타났다. 둘째, 예비유아교사의 음악적 자아개념, 음악적 개념에 대한 실제이해, 음악교수 효능 감은 대체로 유의한 정적 상관을 보였다. 셋째, 예비유아교사의 음악적 자아개념과 음악적 개념에 대한 실제이해는 예비유아교사의 음악교수 효능감 전체에 영향력을 미치고 있었으며, 하위요인 중 음악교수 능력에 대한 신념은 예비유아교사의 음악적 자아개념이 더 큰 영향력을 미쳤고, 결과에 대한 기대감은 음악적 개념에 대한 실제이해만이 영향력 있는 요인으로 나타났다. This study aimed to (1) examine early childhood pre-service teachers’ self-concept in music, the actual level of understanding of musical content knowledge and music teaching efficacy, and (2) investigate the relationships and influence among them. The participants consisted of 201 undergraduate students, majoring in early childhood education at colleges in Gyeongnam, Korea. The measurements used in this study were Self-Concept in Music Scale (SCIM), the scale of Understanding of Musical Content Knowledge, and Music Teaching Efficacy Belief Instrument (MTEBI). The results of this study are as follows. First, the mean score of early childhood pre-service teachers’ self-concept in Music and actual understanding of musical content knowledge were about the average, besides their music teaching efficacy was higher than the average. Second, there was a positive relationship among early childhood pre-service teachers’ self-concept in music and music teaching efficacy, and the actual level of understanding of musical content knowledge. Third, early childhood pre-service teachers’ music teaching efficacy exerted a statistically significant influence on the self-concept in music and the actual level of understanding of musical content knowledge. Among the variables of early childhood pre-service teachers’ music teaching efficacy, 'the belief of music teaching ability' and 'the expectation of the results', pre-service teachers’ self-concept in music had more influence on 'the beliefs of music teaching ability' and actual understanding of musical content knowledge was the only influential factor on 'the expectation of the results'.

        가시오갈피의 생리·생태적 특성 (Ⅰ) : 산지별 순광합성, 기공증산, 기공전도도 Net Photosynthetic Rates, Stomatal Transpirations, Stomatal Conductances of Leaves

        이 연구는 가시오갈피 엽의 생리적 특성을 밝히고자 광합성, 기공증산, 기공전도도의 생리반응을 측정한 것으로 결과를 요약하면 다음과 같다. 1. 광보상점은 양엽이 약 30 μ mol m^(-2) s^(-1), 음엽이 약 15 μ mol m^(-2) s^(-1) 이었다. 2. 광포화점은 양엽이 약 1,000 μ mol m^(-2) s^(-1), 음엽이 약 300 μ mol m^(-2) s^(-1) 이었다. 3. 우리나라 및 중국산 가시오갈피 상엽의 순광합성속도는 약 8.0-8.8 μ mol m^(-2) s^(-1) 로 산지간에 유의차가 인정되지 않았다. 제주산 섬가시오갈피 상엽은 약 6.9 μ mol m^(-2) s^(-1) 로 다소 낮았다. 4. 모든 산지에서 중위엽의 순광합성속도는 상엽의 40-65%, 하위엽은 상엽의 30% 정도이었다. 그러나 섬가시오갈피 하엽은 상엽의 약 71%로 다소 높았다. 5. 상엽의 기공증산속도는 1.1-1.4 mmol H₂O m^(-2) s^(-1), 중위엽은 0.7-1.0 mmol H₂O m^(-2) s^(-1), 하위엽은 0.5-0.6 mmol H₂O m^(-2) s^(-1)로 산지별로 유의차가 없었다. 6. 기공전도도는 상엽이 약 70-90 mmol H₂O m^(-2) s^(-1)로 산지별 유의차가 인정되지 않았다. 그러나 제주 섬가시오갈피 상엽은 약 380 mmol H₂O m^(-2) s^(-1)로 타산지보다 현저히 높았다. This study was made on the physiological reactions of photosynthesis, stomatal transpiration, stomatal conductance of Acanthopanax senticosus of leaves. The resuits obtained are as follows : 1. The light compensation points were approximately 30μ㏖ m^(-2)s^(-1) in sun leaves and 15μ㏖ m^(-2)s^(-1) in shade leaves. 2. The light, saturation points were approximately 1,000μ㏖ m^(-2)s^(-1) in sun leaves and 300μ㏖ m^(-2)s^(-1) in shade leaves. 3. There was no significant between various mountains distributed, net photosynthetic rates were approximately 8.0 to 8.8μ㏖ m^(-2)s^(-1) in upper leaves. However, net photosynthetic rate in upper leaves of Acanthopanax koreanum in Jeju island was slightly low about 6.9μ㏖ m^(-2)s^(-1). 4. Net photosynthetic rate in middle leaves of all mountains was ranged from 40% to 65% of upper leaves, and that in lower leaves was approximately 30% of upper leaves. But, the net photosynthesis in lower leaves of Acanthopanax koreanum in Jeju island was slightly high about 71% of upper leaves. 5. There was no significant between various mountains distributed, stomatal transpirations were ranged from 1.1 to 1.4 mmol H₂Om^(-2)s^(-1) in upper leaves, 0.7 to 1.0 mmol H₂O m^(-2)s^(-1) in middle leaves, and 0.5 to 0.6 mmol H₂0m^(-2)s^(-1) in lower leaves, respectively. 6. There was no significant between various mountains distributed, the stomatal conductance in upper leaves was ranged from 70 to 90 mmol H₂O m^(-2)s^(-1) . However, stomatal conductance in upper leaves of Acanthopanax koreanum in Jeju island was approximately 380 mmol H₂O m^(-2)s^(-1), its remarkedly higher than any other mountain.

        응급실로 내원한 소아 장중첩증의 임상양상

        Back ground : Intussuseception is one of the most common diseases requiring abdominal emergency care in early childhood. The delay in diagnosis increases morbidity and, in particular, reduces the success rate of the non-surgical method of reduction. In this report, the authors determined the clinical characteristics of pediatric patients with intussusception, and the factors involved in delayed diagnosis and the efficacy of doppler ultrasonography in diagnosis. Methods: The authors performed a retrospective study of 44 cases in 42 patients with intussusception who had visited the emergency room of SamSung medical center from January 1st to December 31, 1996. Results: The sex ratio was equal and 64.3% of the patients were under 1 year of age. The prevalent complaints were vomiting(36.4%) and abdominal pain(33.8%). Ultrasonography was used in 11 cases, 3 of them were performed because of obscure symptoms and the rest were due to diagnostic delay. The average time past from visit to diagnosis was 4hours and 20 minutes, and there were 4 diagnostic delays. Air enema reductions were unsuccessful in 11 cases(25%) and that was related to hospital-visit delays and long time past from symptom onset to diagnosis. There were no motalities. Conclusion: To reduce delays in diagnosis and unnecessary operations, the use of doppler ultrasonography is recommanded In suspicious cases. And to reduce the hospital-visit delays, related to the failure on non-surgical reduction, the education about this disease is necessary.
