<P> Recombinant Escherichia coli whole cells harboring Bacillus licheniformis L-arabinose isomerase (BLAI) were harvested to prepare alginate-immobilized biocatalysts. The operational conditions for immobilization were optimized according to relative activity and the cell leakage of the immobilized cell. The optimal conditions are as follows: alginate concentration, Ca2+ concentration, cell mass loading, and curing time were 2% (w/v), 0.1 M, 50 g l-1, and 4 hours, respectively. After immobilization, cross-linking with 0.1% glutaraldehyde significantly reduced cell leakage. The immobilized whole cells harboring BLAI were very stable with 89% residual activity remaining after 33 days of incubation at 50°C and were much more stable than the free enzyme and cells. The results showed that immobilizing whole cells harboring BLAI is suitable for use as a biocatalyst in the production of L-ribulose, largely due to its high stability and low cost.</P>

A series of poly(lactic acid) (PLA), poly(butylene adipate-co-terephthalate) (PBAT) and glycidyl methacrylate- grafted poly(ethylene octene) (GPOE) blends and films with different GPOE content were prepared by melt blending and blowing film technique. The effect of GPOE on the rheological behavior, melt strength, crystallization behavior, crystallization morphology, miscibility, mechanical property, phase morphology, thermal stability and water vapor permeability were studied. The addition of GPOE improved melt rheological properties. Results of DSC showed that addition of GPOE encouraged the mobility of PLA molecular chains and enhanced crystalline ability. POM observations revealed that the addition of GPOE made the density of spherulite nucleation increase and the size of crystalline particles decrease. From DMA and SEM analysis, it was demonstrated that PLA/PBAT blend was an immiscible system and GPOE in the blend could improve compatibility between PLA and PBAT. Results of mechanical test showed that the PLA/PBAT/GPOE blends and films obtained had excellent mechanical properties. The elongation at break of 50/30/ 20 w/w/w PLA/PBAT/GPOE blend (477%) was higher by about 2.2 times than that of 70/30/0 w/w/w PLA/PBAT/ GPOE blend (220%). The tensile strength of all the PLA/PBAT/GPOE blends exceeded 31 MPa. The tensile strength reached 32.9MPa (MD) and 22.5MPa (TD), the elongation at break exceeded 210% and tear strength exceeded 140 kN/m for 50/30/20 w/w/w PLA/PBAT/GPOE film. With increasing GPOE content, thermal stability and water vapor barrier property also improved.

<P><I>Bacillus licheniformis</I> <SMALL>L</SMALL>-arabinose isomerase (BLAI) with a broad pH range, high substrate specificity, and high catalytic efficiency for <SMALL>L</SMALL>-arabinose was immobilized on various supports. Eupergit C, activated-carboxymethylcellulose, CNBr-activated agarose, chitosan, and alginate were tested as supports, and Eupergit C was selected as the most effective. After determination of the optimum enzyme concentration, the effects of pH and temperature were investigated using a response surface methodology. The immobilized BLAI enzyme retained 86.4% of the activity of the free enzyme. The optimal pH for the immobilized BLAI was 8.0, and immobilization improved the optimal temperature from 50 °C (free enzyme) to a range between 55 and 65 °C. The half life improved from 2 at 50 °C to 212 h at 55 °C following immobilization. The immobilized BLAI was used for semi-continuous production of <SMALL>L</SMALL>-ribulose. After 8 batch cycles, 95.1% of the BLAI activity was retained. This simple immobilization procedure and the high stability of the final immobilized BLAI on Eupergit C provide a promising solution for large-scale production of <SMALL>L</SMALL>-ribulose from an inexpensive <SMALL>L</SMALL>-arabinose precursor.</P>
Association of Six Susceptibility Loci with Prostate Cancer in Northern Chinese Men

Background/Aim: Six prostate cancer (PCa) susceptibility loci were identified in a genome-wide association study (GWAS) in populations of European decent. However, the associations of these 6 single-nucleotide polymorphisms (SNPs) with PCa has remained tobe clarified in men in Northern China. This study aimed to explore the loci associated with PCa risk in a Northern Chinese population. Methods: Blood samples and clinical information of 289 PCa patients and 288 controls from Beijing and Tianjin were collected. All risk SNPs were genotyped using polymerase chain reaction (PCR)-high resolution melting curve technology and gene sequencing. Associations between PCa and clinical covariates (age at diagnosis, prostate-specific antigen [PSA], Gleason score, tumor stage, and level of aggressiveness) and frequencies of alleles and genotypes of these SNPs were analyzed using genetic statistics. Results: Among the candidate SNPs, 11p15 (rs7127900, A) was associated with PCa risk (P = 0.02, odds ratio [OR] = 1.64, 95% confidence interval [CI] = 1.09-2.46). Genotypes showed differences between cases and controls on 11p15 (rs7127900, A), 11q13 (rs7931342, T), and HNF1B (rs4430796, A) (P = 0.03, P = 0.01, and P = 0.04, respectively). The genotype TG on 11q13 (rs7931342, T) was positively associated with an increased Gleason score (P = 0.04, OR = 2.15, 95% CI = 1.02-4.55). Patients carrying TG on 17q24 (rs1859962, G) were negatively associated with an increased body mass index (BMI) (P = 0.03, OR = 0.44, 95% CI = 0.21-0.92) while those with AG on HNF1B (rs4430796, A) were more likely to have PSA increase (P = 0.002). Conclusion: Our study suggests that 11p15 (rs7127900, A) could be a susceptibility locus associated with PCa in Northern Chinese. Genotype TG on 11q13 (rs7931342, T) could be related to an increased Gleason score, AG on HNF1B (rs4430796, A) could be associated with PSA increase, and TG on 17q24 (rs1859962, G) could be negatively associated with an increased BMI in Chinese men with PCa.

SiPMs are generally coupled into whole columns in gamma energy spectrum measurement, but the relationship between the distribution of whole SiPM columns and the energy resolution of the measured energy spectra is rarely reported. In this work, ∅ 3 3 inch NaI scintillator is placed on an 8 8 SiPM array, and the energy resolution of the 137Cs peak at 662 keV corresponding to the g-ray is selected as a reference. Each SiPM is switched to explore the influence of the number of SiPM arrays, distribution position, and reflective layer on the energy resolution of SiPMs. Results show that without coupling, the energy resolution is greatly improved when the number of SiPMs ranges from 4 to 32. However, after 32 slices (the area covered by SiPMs relative to the scintillator reaches 25.9%), the improvement in energy resolution and total pulse count is not obvious. In addition, the position of SiPMs relative to the scintillator does not exert much impact on the energy resolution. Results also indicate that by adding a reflective film (ESR), the energy resolution of the tested group increases by 10.38% on average. This work can provide a reference for the design and application of miniaturized SiPM gamma spectrometers

Background and Purpose Intravenous tenecteplase (TNK) efficacy has not been well demonstrated in acute ischemic stroke (AIS) beyond 4.5 hours after onset. This study aimed to determine the effect of intravenous TNK for AIS within 4.5 to 24 hours of onset. Methods In this pilot trial, eligible AIS patients with diffusion-weighted imaging (DWI)-fluid attenuated inversion recovery (FLAIR) mismatch were randomly allocated to intravenous TNK (0.25 mg/kg) or standard care within 4.5–24 hours of onset. The primary endpoint was excellent functional outcome at 90 days (modified Rankin Scale [mRS] score of 0–1). The primary safety endpoint was symptomatic intracranial hemorrhage (sICH). Results Of the randomly assigned 80 patients, the primary endpoint occurred in 52.5% (21/40) of TNK group and 50.0% (20/40) of control group, with no significant difference (unadjusted odds ratio, 1.11; 95% confidence interval 0.46–2.66; <i>P</i>=0.82). More early neurological improvement occurred in TNK group than in control group (11 vs. 3, <i>P</i>=0.03), but no significant differences were found in other secondary endpoints, such as mRS 0–2 at 90 days, shift analysis of mRS at 90 days, and change in National Institutes of Health Stroke Scale score at 24 hours and 7 days. There were no cases of sICH in this trial; however, asymptomatic intracranial hemorrhage occurred in 3 of the 40 patients (7.5%) in the TNK group. Conclusion This phase 2, randomized, multicenter study suggests that intravenous TNK within 4.5–24 hours of onset may be safe and feasible in AIS patients with a DWI-FLAIR mismatch.

<P>For the goal of superconducting magnet applications in the advanced testing device for high temperature superconducting (HTS) wire and sample coils, a wide bore conduction-cooled superconducting magnet with available warm bore of phi 186 mm and center field of 5 T for the background magnetic field applications was designed and fabricated and tested. A sample cryostat with two GM cryocoolers is inserted in the background magnet. The system allows measurements to be performed in a repeatable and reliable fashion. The detailed design, fabrication and thermal analysis are presented in the paper.</P>
