        According to the China tumor registry 2013 annual report, breast cancer, lung cancer, and ovarian cancer are three common cancers in China nowadays, with high mortality due to the absence of early diagnosis technology. However, proteomics has been widespreadly implanted into every field of life science and medicine as an important part of post-genomics era research. The development of theory and technology in proteomics has provided new ideas and research fields for cancer research. Proteomics can be used not only for elucidating the mechanisms of carcinogenesis focussing on whole proteins of the tissue or cell, but also seeking the biomarkers for diagnosis and therapy of cancer. In this review, we introduce proteomics principles, covering current technology used in exploring early diagnosis biomarkers of breast cancer, lung cancer and ovarian cancer.

      • Low Prevalence of HPV in Male Sexual Partners of HR-HPV Infected Females and Low Concordance of Viral Types in Couples in Eastern Guangdong

        Objective: To investigate the prevalence of genital high-risk human papillomavirus (HR-HPV) in male sexual partners of HR-HPV infected women and the concordance of viral types in couples in China, and comprehend the role of men play in HPV transmission to women. Methods/Materials: 94 asymptomatic women and their husbands from rural Chaozhou participated in epidemiologic screening for HPV infection. Cervical cells from females were collected for high risk HPV screening by real time-PCR, and they were positive for at least 1 of 13 HR-HPV subtypes, then these samples were genotyped. Approximately one mouth later, penile epithelial cells from 94 asymptomatic husbands were collected for HPV genotyping. At the same time, a cross-sectional study was conducted in 366 male patients from sexually transmitted disease (STD) outpatient clinic in China for the prevalence of genital HR-HPV infection in men having frequent sex behavior. Penial epithelial cells were collected for HPV 6/11 and HPV 16/18 detection by fluorescent real-time quantified PCR. Results: Among 94 couples, the prevalence of genital HR-HPV infection in men whose wife was positive for cervical HR-HPV was 5.32% (5/94). Only 2.63% (2/76) had the same high risk viral type presented by their wife. HPV 16 proved to be the most prevalent viral type in men and in couples. Of 366 male patients from STD outpatient clinic, the prevalence of HPV 16/18 infection in men with or without HPV 6/11 was 6.85% and 8.16%, respectively. The incidence of HPV 16/18 was higher in men aged more than 35 years than the young men (18-35 years). Conclusion: The prevalence of genital HR-HPV infection in male sexual partners of HPV-positive women in China was lower than that expected, and the concordance of high risk viral type between couples was extremely low. These data suggested that infected men consitute an important viral reservoir, contributing to transmission of HR-HPV to women and maintenance of infection, but HR-HPV infection may be less likely to persist in men than in women.

      • Compression and Aggregation-Resistant Particles of Crumpled Soft Sheets

        <P>Unlike flat sheets, crumpled paper balls have both high free volume and high compressive strength, and can tightly pack without significantly reducing the area of accessible surface. Such properties would be highly desirable for sheet-like materials such as graphene, since they tend to aggregate in solution and restack in the solid state, making their properties highly dependent on the material processing history. Here we report the synthesis of crumpled graphene balls by capillary compression in rapidly evaporating aerosol droplets. The crumpled particles are stabilized by locally folded, π–π stacked ridges as a result of plastic deformation, and do not unfold or collapse during common processing steps. In addition, they are remarkably aggregation-resistant in either solution or solid state, and remain largely intact and redispersible after chemical treatments, wet processing, annealing, and even pelletizing at high pressure. For example, upon compression at 55 MPa, the regular flat graphene sheets turn into nondispersible chunks with drastically reduced surface area by 84%, while the crumpled graphene particles can still maintain 45% of their original surface area and remain readily dispersible in common solvents. Therefore, crumpled particles could help to standardize graphene-based materials by delivering more stable properties such as high surface area and solution processability regardless of material processing history. This should greatly benefit applications using bulk quantities of graphene, such as in energy storage or conversion devices.         Metabolism Characteristics of Mycoplasma pneumoniae Infection in Asthmatic Children

        Purpose: Studies have shown that Mycoplasma pneumoniae (Mp) infection can aggravate symptoms in asthmatics. However, the mechanism by which Mp infection exacerbates asthma remains unclear. Metabolomics can help identify the mechanism of Mp aggravating asthma in children, thereby providing more a potential target for improving clinical treatment programs. In this article, we analyzed the metabolic level of patients to explain how Mp aggravates asthma in children. Methods: We divided the subjects into the asthma, Mp infection, asthma combined with Mp infection and healthy groups. Patients’ peripheral blood was collected for metabolic and interaction analysis. Cytokine levels were measured via serum and exhaled breath condensate (EBC). Results: A total of 150 participating subjects were divided into four groups after exclusion. We found out that there were different metabolic pathways between the healthy and disease groups. The major pathways of both asthma and asthma combined with Mp infection were valine, leucine and isoleucine biosynthesis; malate-aspartate shuttle was the main differential pathway for Mp infection. Moreover, even though three disease groups involved 81 metabolites at the same time, compared with asthma combined with Mp infection, 2 single disease groups still involved different amino acid pathways (phenylalanine, tyrosine and tryptophan biosynthesis; valine, leucine and isoleucine biosynthesis). Interaction analysis showed that Mp infection in asthmatic patients not only activated cytokines, but also activated Toll-like receptors (TLRs) 2 and 6. Finally, the levels of interleukin (IL)-4, IL-8, IL-13 and tumor necrosis factor-α in EBC with asthma combined with Mp infection were significantly higher than the 2 single disease groups. Conclusions: Mp infection in asthmatic children can cause changes in the levels of various amino acids in the body, which were enriched in the pathways such as valine, leucine and isoleucine biosynthesis. Palmitic acid can activate TLR2, and iloprost reduces IL-10 levels, ultimately leading to the increased airway inflammation.

        Microstructure evolution and effect on deuterium retention in oxide dispersion strengthened tungsten during He<sup>+</sup> irradiation

        Oxide dispersion-strengthened materials W-1wt%Pr<sub>2</sub>O<sub>3</sub> and W-1wt%La<sub>2</sub>O<sub>3</sub> were synthesized by wet chemical method and spark plasma sintering. The field emission scanning electron microscopy (FE-SEM) analysis, XRD and Vickers microhardness measurements were conducted to characterize the samples. The irradiations were carried out with a 5 keV helium ion beam to fluences up to 5.0 × 10<sup>21</sup> ions/m<sup>2</sup> under 600 ℃ using the low-energy ion irradiation system. Transmission electron microscopy (TEM) study was performed to investigate the microstructural evolution in W-1wt%Pr<sub>2</sub>O<sub>3</sub> and W-1wt%La<sub>2</sub>O<sub>3</sub>. At 1.0 × 10<sup>20</sup> He<sup>+</sup>/m<sup>2</sup>, the average loops size of the W-1wt%Pr<sub>2</sub>O<sub>3</sub> was 4.3 nm, much lower than W-1wt% La<sub>2</sub>O<sub>3</sub> of 8.5 nm. However, helium bubbles were not observed throughout in both doped W materials. The effects of pre-irradiation with 1.0 × 10<sup>21</sup> He<sup>+</sup>/m<sup>2</sup> on trapping of injected deuterium in doped W was studied by thermal desorption spectrometry (TDS) technique using quadrupole mass spectrometer. Compared with the samples without He<sup>+</sup> pre-irradiation, deuterium (D) retention of doped W materials increased after He<sup>+</sup> irradiation, whose retention was unsaturated at the damage level of 1.0 × 10<sup>22</sup>D<sub>2</sub><sup>+</sup>/m<sup>2</sup>. The present results implied that irradiation effect of He<sup>+</sup> ions must be taken into account to evaluate the deuterium retention in fusion material applications.

        Corrosion behavior and mechanism of CLAM and 316L steels in fl owing Pb – 17Li alloy under magnetic field

        The liquid lead-lithium (Pbe17Li) blanket has many applications in fusion reactors due to its good tritiumbreeding performance, high heat transfer efficiency and safety. The compatibility of liquid Pbe17Li alloywith the structural material of blanket under magnetic field is one of the concerns. In this study,corrosion experiments China low activation martensitic (CLAM) steel and 316L steel were carried out in aforced convection Pbe17Li loop under 1.0 T magnetic field at 480 C for 1000 h. The corrosion results on316L steel showed the characteristic with a superficial porous layer resulted from selective leaching ofhigh-soluble alloy elements and subsequent phase transformation from austenitic matrix to ferriticphase. Then the porous layers were eroded by high-velocity jet fluid. The main corrosion mechanism ofCLAM steel was selective dissolution-base corrosion attack on the microstructure boundary regions andexclusively on high residual stress areas. CLAM steel performed a better corrosion resistance than that of316L steel. The high Ni dissolution rate and the erosion of corroded layers are the main causes for thesevere corrosion of 316L steel.

      • Glulathione-S-transferases Gene Polymorphism in Prediction of Gastric Cancer Risk by Smoking and Helicobacter Pylori Infection Status

        Aim: To evaluate the association of glutathione S-transferases gene polymorphisms with the risk of gastric cancer, with reference to smoking and Helicobacter pylori infection. Methods: We conducted a 1:1 matched case-control study with 410 gastric cancer cases and 410 cancer-free controls. Polymorphisms of GSTM1, GSTT1 and GSTP1 were determined using PCR-CTPP. Results: The GSTM1 and GSTT1 null genotypes were significantly associated with the risk of gastric cancer after adjusting for potential confounding factors (OR=1.68, 95% CI=1.32-2.23 for null GSTM1, OR=1.73; 95% CI=1.24-2.13 for null GSTT1). The combination of null GSTM1 and null GSTT1 conferred an elevated risk (OR=2.54, 95% CI=1.55-3.39). However, no association was found for GSTP1 polymorphism The smoking modified the association of GSTM1 and GSTT1 null genotypes with the risk of gastric cancer. Conclusion: GSTM1 and GSTT1 null genotypes are associated with increased risk of gastric cancer, and smoking modifies the association.

        Plasma polymerized bio-interface directs fibronectin adsorption and functionalization to enhance “epithelial barrier structure” formation via FN-ITG β1-FAK-mTOR signaling cascade

        Background: Transepithelial medical devices are increasing utilized in clinical practices. However, the damage of continuous natural epithelial barrier has become a major risk factor for the failure of epithelium-penetrating implants. How to increase the “epithelial barrier structures” (focal adhesions, hemidesmosomes, etc.) becomes one key research aim in overcoming this difficulty. Directly targeting the in situ “epithelial barrier structures” related proteins (such as fibronectin) absorption and functionalization can be a promising way to enhance interface-epithelial integration. Methods: Herein, we fabricated three plasma polymerized bio-interfaces possessing controllable surface chemistry. Their capacity to adsorb and functionalize fibronectin (FN) from serum protein was compared by Liquid Chromatography- Tandem Mass Spectrometry. The underlying mechanisms were revealed by molecular dynamics simulation. The response of gingival epithelial cells regarding the formation of epithelial barrier structures was tested. Results: Plasma polymerized surfaces successfully directed distinguished protein adsorption profiles from serum protein pool, in which plasma polymerized allylamine (ppAA) surface favored adsorbing adhesion related proteins and could promote FN absorption and functionalization via electrostatic interactions and hydrogen bonds, thus subsequently activating the ITG β1-FAK-mTOR signaling and promoting gingival epithelial cells adhesion. Conclusion: This study offers an effective perspective to overcome the current dilemma of the inferior interfaceepithelial integration by in situ protein absorption and functionalization, which may advance the development of functional transepithelial biointerfaces. Graphical Abstract: Tuning the surface chemistry by plasma polymerization can control the adsorption of fibronectin and functionalize it by exposing functional protein domains. The functionalized fibronectin can bind to human gingival epithelial cell membrane integrins to activate epithelial barrier structure related signaling pathway, which eventually enhances the formation of epithelial barrier structure.
