        Comparison of volatile organic compounds from uninfested and Monochamus alternatus Hope infested Pinus massoniana Lamb.

        Monochamus alternatus is a destructive stem‐boring herbivore of Pinus massoniana, and the principal vector of pine wood nematode. To investigate the impacts of boring by M. alternatus larvae on the emission of volatile organic compounds (VOCs) from their host trees, the VOCs from uninfested and M. alternatus larvae infested P. massoniana trees were observed using a gas chromatograph–mass spectrometer. We detected 12, 9, 18 and 14 volatile organic compounds from infested xylem, infested phloem, uninfested xylem and uninfested phloem, respectively. In P. massoniana xylem, the boring of M. alternatus larvae induced cyclosativene, and inhibited 4‐carene, humulene, styrene, α‐phellandrene, β‐myrcene, β‐phellandrene and γ‐terpinene. The relative amounts of camphene, copacamphene, longicyclene, longifolene, tricyclene and α‐longipinene were significantly increased, and the relative amounts of α‐pinene and β‐pinene were significantly decreased by the boring behaviors of M. alternatus larvae. In P. massoniana phloem, the boring of M. alternatus larvae induced 2‐bornanone, copacamphene, longicyclene and α‐longipinene, and inhibited 2‐carene, 4‐carene, styrene, α‐phellandrene, β‐myrcene, β‐phellandrene, β‐pinene, γ‐terpinene and ο‐cymene. The relative amounts of camphene, caryophyllene and longifolene were significantly increased by the boring behaviors of M. alternatus larvae. The results indicate that the boring behaviors of M. alternatus larvae changed both the sorts and contents of the VOCs from P. massoniana trees.

        Identification of differentially expressed immunity‐related genes in Monochamus alternatus Hope (Coleoptera: Cerambycidae) larvae parasitized by Dastarcus helophoroides (Fairmaire) (Coleoptera: Bothrideridae)

        The pine sawyer Monochamus alternatus Hope (Coleoptera: Cerambycidae) is a serious pest of several Pinus species, and the ectoparasitoid larvae of Dastarcus helophoroides (Fairmaire) (Coleoptera: Bothrideridae) is an important natural enemy of this pest. The transcriptome of M. alternatus larvae was sequenced using the Illumina platform and immunity‐related genes were specifically analyzed. De novo assembly resulted in the identification of 24 241 unigenes, with a mean length of 1122 bp, in unparasitized M. alternatus larvae and 23 807 unigenes, with a mean length of 1140 bp, for parasitized larvae. Removal of redundant unigenes resulted in 26 095 all‐unigenes, of which 16 959 (64.99%) showed clear homology with some of the known genes in the National Center for Biotechnology Information nr database. Parasitization had notable effects on the transcriptome profile of M. alternatus larvae. In all, 2702 genes were differentially expressed in M. alternatus larvae after parasitization, with 1491 (55.18%) upregulated and 1211 (44.82%) downregulated. Moreover, expression levels of immunity‐related genes in M. alternatus larvae were markedly altered in response to parasitization by D. helophoroides. In conclusion, the transcriptome profiling data, especially the discovered of immunity‐related genes, help illustrate the molecular mechanisms of parasitism between D. helophoroides and M. alternatus and provide new insights into developing immunity regulation‐mediated control methods of M. alternatus.

        Knockdown of the olfactory co-receptor Orco impairsmate recognition in Tenebrio molitor (Coleoptera: Tenebrionidae)

        The yellow mealworm beetle, Tenebrio molitor (Coleoptera: Tenebrionidae), is a common insect pest that damages stored grain. The insect olfactory co-receptor (Orco) is a transmembrane protein localized on the surface of the dendrites of olfactory sensory neurons and plays an essential role in the olfactory perception. In this study, an Orco ortholog (named as Tmol\Orco) inT.molitorwas characterized. The deduced amino acid sequence of Tmol\Orco shared significant high identities with Orcos in other insect species from distinct Orders. Reverse transcription quantitative PCR (RT-qPCR) analysis showed that Tmol\Orco was predominantly expressed in the antennae of both male and female adults, and the difference between male antennae and female antennae was not significant. Double-stranded RNA (dsRNA) directly against the Tmol\Orco mRNA was synthesized and injected into the T. molitor male adults, RT-qPCR results demonstrated that the transcription level of Tmol\Orco was significantly reduced at 6 d after the injection. The mate-seeking behavior of Tmol\Orco-silenced beetles was significantly inhibited. Compare to the control individuals, the percentage of Tmol\Orco-silenced beetles successfully find mateswas significantly decreased and the average time they spent for searching were significantly increased. These results provided first direct evidence that suppression of Tmol\Orco transcription contributes to the decline in mate recognition of T. molitor

      • 8q24 rs4242382 Polymorphism is a Risk Factor for Prostate Cancer among Multi-Ethnic Populations: Evidence from Clinical Detection in China and a Meta-analysis

        Background: Evidence supporting an association between the 8q24 rs4242382-A polymorphism and prostate cancer (PCa) risk has been reported in North American and Europe populations, though data from Asian populations remain limited. We therefore investigated this association by clinical detection in China, and meta-analysis in Asian, Caucasian and African-American populations. Materials and Methods: Blood samples and clinical information were collected from ethnically Chinese men from Northern China with histologically-confirmed PCa (n=335) and from age-matched normal controls (n=347). The 8q24 (rs4242382) gene polymorphism was genotyped by polymerase chain reaction-high-resolution melting analysis. We initially analyzed the associations between the risk allele and PCa and clinical covariates. A meta-analysis was then performed using genotyping data from a total of 1,793 PCa cases and 1,864 controls from our study and previously published studies in American and European populations, to determine the association between PCa and risk genotype. Results: The incidence of the risk allele was higher in PCa cases than controls (0.222 vs 0.140, $P=7.3{\times}10^{-5}$), suggesting that the 8q24 rs4242382-A polymorphism was associated with PCa risk in Chinese men. The genotypes in subjects were in accordance with a dominant genetic model (ORadj=2.03, 95%CI: 1.42-2.91, $Padj=1.1{\times}10^{-4}$). Presence of the risk allele rs4242382-A at 8q24 was also associated with clinical covariates including age at diagnosis ${\geq}65$ years, prostate specific antigen >10 ng/ml, Gleason score <8, tumor stage and aggressive PCa, compared with the non-risk genotype ($P=4.6{\times}10^{-5}-3.0{\times}10^{-2}$). Meta-analysis confirmed the association between 8q24 rs4242382-A polymorphism and PCa risk (OR=1.62, 95%CI: 1.39-1.88, $P=1.0{\times}10^{-5}$) across Asian, Caucasian and African American populations. Conclusions: The replicated data suggest that the 8q24 rs4242382-A variation might be associated with increased PCa susceptibility in Asian, Caucasian and African American populations. These results imply that this polymorphism may be a useful risk biomarker for PCa in multi-ethnic populations.

        An Overview of Remote Sensing of Chlorophyll Fluorescence

        Besides empirical algorithms with the blue-green ratio, the algorithms based on fluorescence are also important and valid methods for retrieving chlorophyll-a concentration in the ocean waters, especially for Case II waters and the sea with algal blooming. This study reviews the history of initial cognitions, investigations and detailed approaches towards chlorophyll fluorescence, and then introduces the biological mechanism of fluorescence remote sensing and main spectral characteristics such as the positive correlation between fluorescence and chlorophyll concentration, the red shift phenomena. Meanwhile, there exist many influence factors that increase complexity of fluorescence remote sensing, such as fluorescence quantum yield, physiological status of various algae, substances with related optical property in the ocean, atmospheric absorption etc. Based on these cognitions, scientists have found two ways to calculate the amount of fluorescence detected by ocean color sensors: fluorescence line height and reflectance ratio. These two ways are currently the foundation for retrieval of chlorophyll-a concentration in the ocean. As the in-situ measurements and synchronous satellite data are continuously being accumulated, the fluorescence remote sensing of chlorophyll-a concentration in Case II waters should be recognized more thoroughly and new algorithms could be expected.

      • Expression of hPOT1 in HeLa Cells and the Probability of Gene Variation of hpot1 Exon14 in Endometrial Cancer are Much Higher than in Other Cancers

        To investigate the expression of hPOT1 in the HeLa cell line and screen point mutations of hpot1 in different tumor tissues a two step osmotic method was used to extract nuclear proteins. EMSA was performed to determine the expression of hPOT1 in the HeLa cell line. PCR was also employed to amplify the exon14 sequence of the hpot1 gene in various of cancer tissues. A SV gel and PCR clean-up system was performed to enrich PCR products. DNAStar was used to analyse the exon14 sequence of the hpot1 gene. hPOT1 was expressed in the HeLa cell line and the signal was gradually enhanced as the amount of extracted nuclear proteins increased. The DNA fragment of exon14 of hpot1 was successfully amplified in the HeLa cell line and all cancer tissues, point mutations being observed in 2 out of 3 cases of endometrial cancer (66.7%) despite the hpot1 sequence being highly conserved. However, the sequence of hpot1 exon14 do not demonstrate point mutations in most cancer tissues. Since hPOT1 was expressed in HeLa cell and the probability of gene point variants was obviously higher in endometrial cancer than other cancers, it may be involved in the pathogenesis of gynecological cancers, especially in cervix and endometrium.

        Life extension analysis of high speed ball bearing based on multi-parameter coupling

        The internal contact nonlinear characteristics caused by the quasi-static properties of high-speed ball bearing is taken as a breakthrough point. Based on the L-P fatigue life theory, the bearing parameters and the preload load are used as the constraint variables to establish an overall bearing life extension model. Then, combined with the influence of geometric parameters on the osculation, the influence mechanism between spin-to-roll ratio and the life of the bearing are studied under the individual variation of the geometric parameters. Finally, the variation law of the bearing life, frictional heat generation and spin-to-roll ratio under the coupling changes of various parameters, and the priority of each parameter on the bearing life sensitivity, are investigated. The results show that the bearing life could be prolonged when the initial contact angle and the number of rolling elements increase but the inner and outer raceway curvature radius coefficients of the bearing and the preload force reduce. And the priority order of each parameter to bearing life sensitivity is as follows: the inner and outer raceway curvature radius coefficients shows the strongest influence on the bearing life, followed by the number of balls, and then the initial contact angle. This research provides a reliable theoretical basis for the design of high-speed ball bearings and life extension technology.

        In this paper, we deal with the problem of uniqueness of meromorphic functions that share two small functions with their derivatives, and obtain the following result which improves a result of Yao and Li: Let f(z) be a nonconstant meromorphic function, k > 5 be an integer. If f(z) and g(z) = $a_1(z)f(z)+a_2(z)f^{(k)}(z)$ share the value 0 CM, and share b(z) IM, $\overline{N}_E(r,f=0=F^{(k)})=S(r)$, f${\equiv}$g, where $a_1(z)$, $a_2(z)$ and b(z) are small functions of f(z).

      • Cyr61/CCN1 Overexpression Induces Epithelial-Mesenchymal Transition Leading to Laryngeal Tumor Invasion and Metastasis and Poor Prognosis

        Background: To examine the expression of cysteine-rich 61 (Cyr61/CCN1) protein in laryngeal squamouscell carcinoma (LSCC) tissues, and its relationship with the tumor epithelial-mesenchymal transition (EMT), invasion, metastasis, and prognosis. Materials and Methods: Immunohistochemistry was used to detect the expressions of Cyr61, Vimentin (Vim), and E-cadherin (E-cad) in 88 cases of LSCC tissues and 30 cases of tumor-adjacent normal tissues. Vim and E-cad were used as mesenchymal and epithelial markers, respectively, to determine the relationship between Cyr61 expression and the EMT of LSCC cells. In addition, clinical and histopathological data were combined to analyze the relationship between the positive-expression rates of Cyr61, Vim and E-cad and LSCC invasion, metastasis and prognosis. Results: In LSCC tissues, Vim expression rate was significantly higher than that of the tumor-adjacent tissues, whereas E-cad expression rate was significantly lower than that of the tumor-adjacent tissues. The Vim expression rate was significantly higher in stages T3 and T4 than in stages T1 and T2 LSCC tissues, whereas E-cad expression rate was significantly lower in stages T3 and T4 than in stages T1 and T2 LSCC tissues. Compared to the group without lymph node metastasis, the Vim expression rate was significantly higher and the E-cad expression rate was significantly lower in the group with lymph node metastasis. The expression rate of Cyr61 was significantly higher in LSCC tissues than in the tumor-adjacent normal tissues. In addition, the Cyr61 expression rate was higher in stages T3 and T4 than in stages T1 and T2 LSCC, and higher in the group with lymph node metastasis than in the group without lymph node metastasis. The Vim expression rate was significantly higher in the Cyr61 positive group than in the Cyr61 negative group, whereas the E-cad expression rate was significantly higher in the Cyr61 negative group than in the Cyr61 positive group. Survival analysis indicated that survival rates of Cyr61 positive, Vim positive and E-cad negative groups were significantly lower than that of Cyr61 negative, Vim negative and E-cad positive groups, respectively. Conclusions: Cyr61 expression is closely associated with LSCC invasion and lymph node metastasis. Overexpression of Cyr61 may induce EMT and therefore leads to LSCC invasion and metastasis and poor prognosis. Cyr61 may become a new maker for clinical prediction of LSCC invasion and metastasis and a new target for LSCC treatment.

      • Prognostic Impact of Cyclin D1, Cyclin E and P53 on Gastroenteropancreatic Neuroendocrine Tumours

        Conventional classifications of gastroenteropancreatic neuroendocrine tumours (GEP-NETs) are rather unsatisfactory because of the variation in survival within each subgroup. Molecular markers are being found able to predict patient outcome in more and more tumours. The aim of this study was to characterize the expression of the proteins cyclin D1, cyclin E and P53 in GEP-NETs and assess any prognostic impact. Tumor specimens from 68 patients with a complete follow-up were studied immunohistochemically for cyclin D1, cyclin E and P53 expression. High cyclin D1 and cyclin E immunostaining (${\geq}$ 5% positive nuclei) was found in 48 (71%) and 24 (35%) cases, and high P53 staining (${\geq}$ 10% positive nuclei) in 33 (49%). High expression of P53 was more common in gastric neuroendocrine tumors and related to malignant behavior, being associate with a worse prognosis on univariate analysis (RR=1.9, 95%CI=1.1-3.2). High expression of cyclin E was significantly associated with shorter survival in the univariate analysis (RR=2.0, 95%CI=1.2-3.6) and multivariate analysis (RR=2.1, 95%CI=1.1-4.0). We found no significant correlation between the expression of cyclin D1 and any clinicopathological variables. Our study indicated a prognostic relevance for cyclin E and P53 immunoreactivity. Cyclin E may be an independent prognostic factor from the 2010 WHO Classification which should be evaluated in further studies.
