        Growth - Inhibiting Effects of Various Traditional Drinks of Plant Origin on Human Intestinal Bacteria

        A total of 28 traditional drinks derived from 23 plant species in 19 families were tested for their in vitro growth-inhibiting effects against Bifidobacterium adolescents, Bifidobacterium bifidum, Lactobacillus acidophilus, Lactobacillus casei, Clostridium perfringens, and Escherichia coil using a paper disc agar diffusion method under anaerobic condition. The responses varied with bacterial strain, plant species and tissue sampled. In a test with C. perfringens at 5 and 10 ㎎/disc, potent growth inhibition was produced from the extracts of Eucommia ulmoides stems, Pinus densiflora leaves and shoots, Thea sinensis leaves (green and oolong teas) and Zingiber officinale roots. All materials tested did not adversely affect the growth of bifidobacteria, lactobacilli, and E. coli. These results may be an indication of at least one of the pharmacological activities of these plant-derived drinks.

        EGFR 돌연변이 검출에 있어 PNA-Mediated Real-Time PCR Clamping과 직접 염기서열 분석법의 비교 분석

        Background: Although the gold standard method for research trials on epidermal growth factor receptor (EGFR) mutations has been direct sequencing, this approach has the limitations of low sensitivity and of being time-consuming. Peptide nucleic acid (PNA)-mediated polymerase chain reaction (PCR) clamping is known to be a more sensitive detection tool. The aim of this study was to compare the detection rate of EGFR mutation and EGFR-tyrosine kinase inhibitor (TKI) responsiveness according to EGFR mutation status using both methodologies. Methods: Clinical specimens from 112 NSCLC patients were analyzed for EGFR mutations in exons 18, 19, 20, and 21. All clinical data and tumor specimens were obtained from 3 university hospitals in Korea. After genomic DNA was extracted from paraffin-embedded tissue specimens, both PNA-mediated PCR clamping and direct-sequencing were performed. The results and clinical response to EGFR-TKIs were compared. Results: Sequencing revealed a total of 35 (22.9%) mutations: 8 missense mutations in exon 21 and 26 deletion mutations in exon 19. PNA-mediated PCR clamping showed the presence of genomic alterations in 45 (28.3%) samples, including the 32 identified by sequencing plus 13 additional samples (6 in exon 19 and 7 in exon 21). Conclusion: PNA-mediated PCR clamping is simple and rapid, as well as a more sensitive method for screening of genomic alterations in EGFR gene compared to direct sequencing. This data suggests that PNA-mediated PCR clamping should be implemented as a useful screening tool for detection of EGFR mutations in clinical setting.

        Comparison of Underwater and Overground Treadmill Walking Exercise

        The purpose of this study was to compare the effects of treadmill walking in underwater and overground which affects gait and physical function of people who have had a stroke. Twenty people after a stroke who have become hemiplegic over 6 months were participated. Participants were divided into two groups: underwater treadmill group(UTG) and overground treadmill group(OTG). The intervention was done 4 times per weeks for 6 weeks and 1 session lasted for 30 minutes. Gait and physical function elements were measured at baseline, at the middle(3 weeks) and at the end of the intervention(6 weeks). For the elements of gait, walking velocity, affected stance phase, affected weight bearing were assessed. For the elements of physical function, Short Form 8(SF-8) health survey was used. The result of this study showed that both groups improved similarly in walking velocity. However participants in UTG improved more than those in OTG in affected stance phase(p<.05), affected weight bearing( p<.05) and emotional aspect(p<.001). Based on the results of this study, it can be suggested that treadmill walking both in underwater and on the ground can be effective in improving hemiplegic gait and physical function of people who have had a stroke. The result also suggest that the underwater treadmill exercise can be more effective than overground treadmill in restoration of gait in people after stroke.

        광 버스트 스위칭 망에서 최소 경로비용 기반의 동적 경로배정 기법

        Optical burst switching networks usually employ one-way reservation by sending a burst control packet with a specific offset time, before transmitting each data burst Same. Due to such a Property, burst-contentions occur when multiple bursts contend for the same wavelength in the same output link simultaneously in a node, leading to burst losses, eventually degrading the quality of service. Therefore, in this paper, we propose a dynamic routing algorithm using minimum local state information in order to decrease burst-contentions. In this proposed scheme, if burst loss rate exceeds a threshold value at a certain node, a new alternative routing path is chosen according to burst priority and location of burst generation, which enables the contending bursts to detour around the congested link. Moreover, for reducing the effect of sending bursts on the primary path due to the alternative path, we also apply a minimum path-cost based routing on link-cost concept. Our simulation results show that proposed scheme improves the network performance in terms of burst loss probability and throughput by comparing with conventional one. 광 버스트 스위칭 방식은 제어 채널과 데이터 채널을 분리하여 버스트 제어 패킷을 제어 채널로 먼저 전송하고 Offset 시간 후에 도착할 데이터 버스트에 대한 데이터 채널을 예약하는 단방향 예약 특성을 가지고 있다. 그러나 이러한 특성 때문에 버스트 간에 데이터 채널을 예약하기 위한 경합이 발생하게 되며 경합에 실패한 버스트는 손실되어 심각한 QoS (Quality of Service) 저하를 초래한다. 따라서 본 논문에서는 버스트 손실을 최소화하기 위해 노드에서 발생되는 버스트 손실이 임계값을 초과할 경우 버스트 우선순위와 버스트가 생성된 근원지 정보에 따라 유동적으로 경로를 재설정하는 동적 경로 설정 기법을 제안하였다. 이와 같이 제안된 기법은 기존 경로로 전송되고 있는 버스트 흐름에 영향을 최소화하기 위하여 링크 비용을 고려한 최소 경로 비용을 기반으로 하고 있다. 또한, 제안된 기법의 성능 평가를 위하여 버스트 손실률, 버스트 처리율 및 평균 흡수 관점에서 기존의 기법과 비교 평가하였다.

        Safety Profiles and Antitumor Efficacy of Oncolytic Adenovirus Coated with Bioreducible Polymer in the Treatment of a CAR Negative Tumor Model

        <P/><P>Adenovirus (Ad) vectors show promise as cancer gene therapy delivery vehicles, but immunogenic safety concerns and coxsackie and adenovirus receptor (CAR)-dependency have limited their use. Alternately, biocompatible and bioreducible nonviral vectors, including arginine-grafted cationic polymers, have been shown to deliver nucleic acids through a cell penetration peptide (CPP) and protein transduction domain (PTD) effect. We utilized the advantages of both viral and nonviral vectors to develop a hybrid gene delivery vehicle by coating Ad with mPEG-PEI-<I>g</I>-Arg-S-S-Arg-<B>g</B>-PEI-mPEG (Ad/PPSA). Characterization of Ad/PPSA particle size and zeta potential showed an overall size and cationic charge increase in a polymer concentration-dependent manner. Ad/PPSA also showed a marked transduction efficiency increase in both CAR-negative and -positive cells compared to naked Ad. Competition assays demonstrated that Ad/PPSA produced higher transgene expression levels than naked Ad and achieved CAR-independent transduction. Oncolytic Ad (DWP418)/PPSA was able to overcome the nonspecificity of polymer-only therapies by demonstrating cancer-specific killing effects. Furthermore, the DWP418/PPSA nanocomplex elicited a 2.24-fold greater antitumor efficacy than naked Ad in vivo. This was supported by immunohistochemical confirmation of Ad E1As accumulation in MCF7 xenografted tumors. Lastly, intravenous injection of DWP418/PPSA elicited less innate immune response compared to naked Ad, evaluated by interleukin-6 cytokine release into the serum. The increased antitumor effect and improved vector targeting to both CAR-negative and -positive cells make DWP418/PPSA a promising tool for cancer gene therapy.</P>

      • Effect of Chinese Milkvetch Biomass as Green Manure Crop on Growth and Quality of Rice in Southern Korea

        Chinese milkvetch(CM) has been cultivated as green manure source for environment friendly farming in southern area. This experiment was conducted to produce good quality rice through studying the changes of yield, quality and leaf colors as affected by the amount of CM biomass retuned to paddy field. We growed rice, transplanted in June 10 with cultivar dongjinilho, in four different plots retuned green manure with 0, 1.5, 3.0, and 4.5 ㎏ m-2 CM biomass in May 30. Nitrogen amounts were 5.5g by 1.5㎏CM biomass, 11.1g by 3.0㎏, and 16.6g by 4.5㎏ in late May. There were no different SPAD values among treatments in 10 days after transplanting. However, the more the CM biomass from 20 days after transplanting, the higher the SPAD values were. the plot of CM biomass to 3.0 ㎏ m-2 had same heading date but the plot of 4.5 ㎏ m-2 was one day late heading date as compared with the others. There were no different plant height, number of panicle, panicle length, number of leaf and ripening ratio among the CM biomass amounts. The higher the CM biomass amount, the more likely the plant was lodge. the plot retuned CM fresh biomass 1.5 ㎏ m-2 showed the higher rice yield than the plot retuned 3.0 and 4.5 ㎏ m-2 in 2006, but all plots had similar rice yields in 2007. There was no different whiteness of rice among all treatments . The more the CM biomass amount retuned to plot, the higher the protein content of rice.

      • Characterization of a new commercial cultivar ‘Hae Sal’ by mono-mono crossing in Hypsizigus marmoreus

        A new commercial cultivar ‘Hae Sal’ of Hypsizigus marmoreus was developed by the method of mono-mono crossing between monokaryotic strains derived from KNR8047 and KNR8080. The parental strains are characterized by the property of high quality and a short period of time in cultivation respectively. The optimum temperature of mycelial growth was 25°C and that of fruiting body development was 15~16°C. The period of harvesting including primordia formation was 7.0 days shorter than that of control strain ‘Mangadak No.2’. The color of pileus and stipe surface was neutral-brown and pure white, respectively. The yield was 117.6±23.2g/850cc plastic bottle. Analysis of the genetic characteristics of the new commercial cultivar ‘Hae Sal’ showed a different profile as that of the control strain, ‘Mangadak No.2’, when RAPD (Random Amplified Polymorphic DNA) primer was used. This new cultivar ‘Hae Sal’ of Hypsizigus marmoreus is characterized by the improved quality and a short period of time in cultivation after scratching. It would be of much help to expand the cultivation area of Hypsizigus marmoreus.

        광 버스트 스위칭 망에서 장애에 둔감한 SCM 기반의 노드 구조 설계

        광 버스트 스위칭 망은 Ingress 에지 라우터에서 입력된 If 패킷을 모아서 데이터 버스트를 생성하고 과도한 프로세싱 오버헤드 없이 효율적인 버스트 교환을 수행하기 위하여 채널에 대한 예약을 목적으로 데이터 버스트보다 offset 시간 이전에 제어 패킷을 전송하여 파장 채널을 예약한다. 따라서 OBS 방식은 광 계층에서 버퍼에 의존하지 않고 단방향 예약 방식으로 빠르게 투명성을 지닌 광전송 경로를 설정할 수 있다. 그러나 링크 장애와 같은 망 장애가 발생할 경우 광 버스트 스위칭 망의 단 방향 예약 특성 때문에 데이터 버스트의 많은 손실이 발생하게 되어 트래픽 QoS에 심각한 영향을 초래하게 된다. 따라서 본 논문에서는 광 버스트 스위칭 망에서 장애 발생시에도 손실되는 버스트를 최소화하여 QoS를 보장할 수 있도록 Ingress 에지 라우터에서는 부 반송파 다중화 방식 기반의 버스트 생성 및 전송모듈 구조를 설계하고 코어 망에서는 신속한 장애 복구가 가능한 혼합형 장애 복구 기법을 제안하였다. 또한 제안된 기법의 성능 평가를 위하여 망 장애 발생에 따른 버스트 손실률, 버스트 처리율 및 망 자원 사용량 관점에서 기존의 기법과 비교 평가하였다. In optical burst switching(OBS) networks, the ingress edge router assembles packets in the same class queue into the appropriate size of burst. A burst control packet(BCP) is generated for channel reservation of corresponding data burst and sent earlier than the corresponding data burst with an offset time. Offset time is determined considering the number of hops from source to destination and the required quality of service(QoS). After offset time, the burst data is passed through tile pre-configured optical switches without any O/E/O conversion. But a failure in OBS networks may lead to the loss of bursts until the ingress nodes receive the failure indication signal. This results in a significant degradation in QoS. Therefore, in this paper, we propose a fault-tolerant node architecture based on sub-carrier multiplexing to reduce the effects of failure in OBS networks. The Performance of the proposed fault-tolerant node architecture exhibits considerable improvement as compared with the previous ones.
