        Application of Bone Marrow Mesenchymal Stem Cells Effectively Eliminates Endotoxemia to Protect Rat from Acute Liver Failure Induced by Thioacetamide

        Background: Endotoxemia is related to worse clinical outcomes in acute liver failure (ALF), but its management remains unsatisfactory. In this study, we aimed to assess whether the application of bone marrow mesenchymal stem cells (BMSCs) could eliminate endotoxemia and protect rats against ALF induced by thioacetamide (TAA). Methods: BMSCs were isolated from rats and identified by the specific morphology, differentiation potential, and surface markers. The optimal dose of TAA for this study was explored and TAA-induced ALF rats were randomized to three groups: the normal control group (Saline), ALF group (TAA + Saline), and BMSCs-treated group (TAA + BMSCs). The intestinal migration and differentiation of BMSCs was tracked in vivo, and intestinal permeability, endotoxin and inflammatory cytokines, histology, and mortality were analyzed. Moreover, we added the inhibitor of the PI3K/AKT/mTOR signaling pathway into the co-culture system of BMSCs with enterocytes and then performed CK and Villin expression experiments to assess the role of PI3K/AKT/mTOR signal pathway in the intestinal differentiation of BMSCs. Results: BMSCs migrated to the intestinal injury sites and differentiated into enterocytes, intestinal permeability was decreased compared with the ALF group. The higher expression of endotoxin and inflammatory cytokines were reversed after BMSCs transplantation in rats with ALF. Mortality and intestinal lesion were significantly decreased. Blocking the PI3K/AKT/mTOR signal pathway inhibited BMSCs’ intestinal differentiation in vitro. Conclusion: BMSCs can eliminate endotoxemia and reduce mortality in rats with ALF, and the PI3K/AKT/mTOR signal pathway is involved in intestinal differentiation. BMSCs transplantation could be a potential candidate for the treatment of endotoxemia in ALF. .       • Assessing the Diagnostic Value of Serum Dickkopf-related Protein 1 Levels in Cancer Detection: a Case-control Study and Meta-analysis

        Background: This study aimed to summarize the potential diagnostic value of serum DKK1 levels in cancer detection. Materials and Methods: Serum DKK1 was measured using enzyme-linked immunosorbent assay in a case-control study. Then we performed a meta-analysis and the pooled sensitivity, specificity, diagnostic odds ratio, and summary receiver operating characteristic (sROC) curves were used to evaluate the overall test performance. Results: Serum DKK1 levels were found to be significantly upregulated in gastric cancer as compared to controls. ROC curve analysis revealed an AUC of 0.636, indicating the test has the potential to diagnose cancer with poor accuracy. The summary estimates of the pooled sensitivity, specificity and diagnostic odds ratio in meta-analysis were 0.55 with a 95% confidence interval (CI) (0.53-0.57), 0.86 (95%CI, 0.84-0.88) and 12.25 (95%CI, 5.31-28.28), respectively. The area under the sROC was 0.85. Subgroup analysis revealed that the diagnostic accuracy of serum DKK1 in lung cancer (sensitivity: 0.69 with 95%CI, 0.66-0.74; specificity: 0.95 with 95%CI, 0.92-0.97; diagnostic odds ratio: 44.93 with 95%CI, 26.19-77.08) was significantly higher than for any other cancer. Conclusions: Serum DKK1 might be useful as a noninvasive method for confirmation of cancer diagnosis, particularly in the case of lung cancer.

        Experimental study on the mechanical property of coal and its application

        Brazilian splitting tests, uniaxial compression tests and triaxial compression tests are carried out on the coal samples cored from Shanxi group $II_1$ coal seam of Jiaozuo coal mine, Henan province, China, to obtain their property parameters. Considering the bedding has notable effect on the property parameter of coal, the samples with different bedding angles are prepared. The effects of bedding on the anisotropic characteristics of the coal seam are investigated. A geological geomechanical model is built based on the geology characteristics of the Jiaozuo coal mine target reservoir to study the effects of bedding on the fracture propagations during hydraulic fracturing. The effects of injection pressure, well completion method, in-situ stress difference coefficient, and fracturing fluid displacement on the fracture propagations are investigated. Results show bedding has notable effects on the property parameters of coal, which is the key factor affecting the anisotropy of coal. The hydraulic cracks trends to bifurcate and swerve at the bedding due to its low strength. Induced fractures are produced easily at the locations around the bedding. The bedding is beneficial to form a complicated fracture network. Experimental and numerical simulations can help to understand the effects of bedding on hydraulic fracturing in coalbed methane reservoirs.

        A new model approach to predict the unloading rock slope displacement behavior based on monitoring data

        To improve the prediction accuracy of the strong-unloading rock slope performance and obtain the range of variation in the slope displacement, a new displacement time-series prediction model is proposed, called the fuzzy information granulation (FIG)-genetic algorithm (GA)-back propagation neural network (BPNN) model. Initially, a displacement time series is selected as the training samples of the prediction model on the basis of an analysis of the causes of the change in the slope behavior. Then, FIG is executed to partition the series and obtain the characteristic parameters of every partition. Furthermore, the later characteristic parameters are predicted by inputting the earlier characteristic parameters into the GA-BPNN model, where a GA is used to optimize the initial weights and thresholds of the BPNN; in the process, the numbers of input layer nodes, hidden layer nodes, and output layer nodes are determined by a trial method. Finally, the prediction model is evaluated by comparing the measured and predicted values. The model is applied to predict the displacement time series of a strong-unloading rock slope in a hydropower station. The engineering case shows that the FIG-GA-BPNN model can obtain more accurate predicted results and has high engineering application value.

      • SHOPPING MALLATTRIBUTES ON CUSTOMER EQUITY

        One of the top tourist destinations in Korea during 2014 was Dongdaemun market, which is a representative traditional market that provides reasonably priced, fashionable products. Already constituting 43.1% of Korea’s foreign tourists, the number of Chinese tourists is expected to reach 14.88 million by 2020 at an average annual growth rate of 19.8%. The most important way to improve customer equity is by understanding a store’s attributes, so this study aims to investigate the effects of Dongdaemun fashion market’s store attributes on customer equity. A survey was conducted among Chinese tourists and structural equation modeling were used to test the hypotheses. The main findings of this study are: Store attributes appear to have an impact on all of the three drivers of customer equity. Value equity has a positive impact on the customer lifetime value (CLV). Also, brand equity and relationship equity were identified as having positive impacts on revisiting intention, regardless of the type of shopping mall. Based on these findings, we discuss implications for developing the customer equity of Dongdaemun’s traditional markets.

        Fabrication of polyacrylic acid-based composite binders with strong binding forces on copper foils for silicon anodes in lithium-ion batteries

        The commercial application of silicon (Si) anodes with high theoretical capacity is hampered by the poorcyclic stability because the huge volume change of Si during discharge/charge processes results in thepulverization of electrode materials and the electric contact loss of electrode materials with copper(Cu) foils. Binders play an important role for adhering active materials and conductive additives togetheronto Cu foils. Herein, we design and develop a three-dimensional networked composite binder (PAA-co-SN) via an amidation reaction between polyacrylic acid (PAA) and thiourea (SN). The granular anchors(CuxS) are constructed at the interface between PAA-co-SN binders and Cu foils, resulting from the reactionof S and Cu during the drying processes of Si electrodes. When used as binders for Si nanoparticles,they exhibit stable cyclic performance (1580 mAh g1 after 500 cycles). The anchoring mechanismbetween composite binders and Cu foils provides a strategy for the improvement of electrochemicalproperties of Si-based anodes for lithium-ion batteries.

        Correlation of NUF2 Overexpression with Poorer Patient Survival in Multiple Cancers

        Purpose NUF2 has been implicated in multiple cancers recently, suggesting NUF2 may play a role in the common tumorigenesis process. In this study, we aim to perform comprehensive meta-analysis of NUF2 expression in the cancer types included in the Cancer Genome Atlas (TCGA). Materials and Methods RNA-sequencing data in 31 cancer types in the TCGA data and 11 independent datasets were used to examine NUF2 expression. Silencing NUF2 using targeting shRNAs in hepatocellular carcinoma (HCC) cell lines was used to evaluate NUF2’s role in HCC in vitro and in vivo. Results NUF2 up-regulation is significantly observed in 23 out of the 31 cancer types in the TCGA datasets and validated in 13 major cancer types using 11 independent datasets. NUF2 overexpression was clinically important as high NUF2 was significantly associated with tumor stages in eight different cancers. High NUF2 was also associated with significantly poorer patient overall survival and disease-free survival in eight and six cancers, respectively. We proceeded to validate NUF2 overexpression and its negative association with overall survival at the protein level in an independent cohort of 40 HCC patients. Compared to the non-targeting controls, NUF2 knockdown cells showed significantly reduced ability to grow, migrate into a scratch wound and invade the 8 μm porous membrane in vitro. Moreover, NUF2 knockdown cells also formed significantly smaller tumors than control cells in mouse xenograft assays in vivo. Conclusion NUF2 up-regulation is a common feature of many cancers. The prognostic potential and functional impact of NUF2 up-regulation warrant further studies.

        Effects of Exogenous Hydrogen Sulfide on Apoptosis Proteins and Oxidative Stress in the Hippocampus of Rats Undergoing Heroin Withdrawal

        In this study, the mechanism of H_2S protection in the hippocampus of heroin-treated rats was investigated. Male Sprague-Dawley rats were randomly divided into three groups: a saline group, a heroin and saline group, and a heroin and sodium hydrosulfide group. According to the principle of increasing heroin dosage daily, heroin withdrawal was precipitated on day 9with an injection of naloxone (5 mg/kg, i.p.), and withdrawal symptoms were scored. The levels of cystathionine-β-synthase, H_2S, reduced glutathione and malondialdehyde, as well as the levels of cleaved caspase-3, Bax, and Bcl-2 proteins and the activities of superoxide dismutase, catalase, and glutathione peroxidase were assayed in the hippocampus. The results showed that exogenous H_2S alleviated heroin withdrawal symptoms. Moreover, exogenous H_2S not only increased cellular H_2S and the cystathionine-β-synthase protein level activity but also significantly improved heroin-induced oxidative stress. Protein expression of cleaved caspase-3 and Bax decreased, whereas Bcl-2 protein levels in hippocampus increased with exogenous H_2S. Exogenous H_2S alleviated heroin-induced rat hippocampal damage through antioxidant and antiapoptosis effects.
